This study investigates the relationship between geomagnetic storms and ionospheric total electron content(TEC)disturbances during interplanetary coronal mass ejections(ICME)events from 2000 to 2017.The research emplo...This study investigates the relationship between geomagnetic storms and ionospheric total electron content(TEC)disturbances during interplanetary coronal mass ejections(ICME)events from 2000 to 2017.The research employed statistical analysis methods to examine data from 56 ICME events and their corresponding ionospheric TEC disturbances.The results reveal that TEC disturbances preceded geomagnetic storms in 68%of SB-type events,particularly in mid-latitude regions compared to low-latitude regions.Specifi cally,the duration of positive ionospheric TEC disturbances is signifi cantly related to latitude and the type of geomagnetic storm.In contrast,the duration of negative ionospheric TEC disturbances is primarily influenced by latitude and is independent of the geomagnetic storm type.Notably,the duration of both positive-negative and negative-positive ionospheric TEC disturbance is not signifi cantly related to latitude or the type of geomagnetic storm.Furthermore,the study identifi es an asymmetry in the duration of ionospheric TEC disturbance under the same type of geomagnetic storm in the Northern and Southern Hemispheres with respect to latitude.For instance,during SB-type geomagnetic storms,the duration of positive ionospheric TEC disturbance at mid-latitudes(40°N,50°N,60°N)is mainly 0–12 hours,while at low-latitudes(10°S,0°,10°N,20°N,30°N)it ranges from 0–12 hours to 48–96 hours.These results provide valuable insights into the Sun-Earth energy coupling mechanism and off er a theoretical basis for the prediction and monitoring of ionospheric TEC disturbance,enhancing our understanding of the complex interactions between geomagnetic storms and ionospheric TEC disturbances.展开更多

The widespread use of antibiotics has led to their persistent accumulation in aquatic environments.Although flocculation is cost-effective,traditional flocculants exhibit limited removal efficiency for antibiotics.To ...The widespread use of antibiotics has led to their persistent accumulation in aquatic environments.Although flocculation is cost-effective,traditional flocculants exhibit limited removal efficiency for antibiotics.To address this problem,a novel cellulose-based flocculant(CE-DMC)was synthesized from paper pulp rejects.The methacryloxyethyltrimethyl ammonium chloride(DMC)monomer was successfully grafted onto the cellulose backbone with a grafting rate of 64%,endowing CE-DMC with high positive charge density(+71.1 mV at pH 6)and a branched architecture.Flocculation experiments demonstrated the superior performance of CE-DMC in removing four representative antibiotics.Notably distinct removal efficiencies were achieved:ofloxacin exhibited the highest removal(52.6%),followed by sulfamethoxazole(36.4%),norfloxacin(30.9%),and amoxicillin(29.8%),reflecting differential interaction mechanisms with the adsorbent.The flocculation mechanism of CE-DMC could be concluded as follows:(1)charge neutralization via quaternary ammonium groups of DMC chains,(2)n-π*electron donor-acceptor interactions andπ-π*conjugated interactions mediated by benzene rings and furan rings of cellulose skeleton,and(3)hydrogen bonding force provided by the hydroxyl group from the cellulose skeleton and the DMC chains.Notably,coexisting humic acid(HA)and kaolin enhanced antibiotic removal efficiency by forming large composite pollutants via n-π*electron donor-acceptor interactions,hydrogen bonding forces,andπ-π*conjugated interactions.CE-DMC synergistically removed antibiotics and co-pollutants,with its flocculation mechanism elucidated through structure-activity relationships and structural determinants.This work advances the rational design of natural polymer-based flocculants for multi-mechanistic contaminant removal.展开更多
Modeling and forecasting of the geomagnetic variation are important research topics concerning geomagnetic navigation and space environment monitoring.We propose a combined forecasting model using a dynamic recursive ...Modeling and forecasting of the geomagnetic variation are important research topics concerning geomagnetic navigation and space environment monitoring.We propose a combined forecasting model using a dynamic recursive neural network called echo state network(ESN),the method of complementary ensemble empirical mode decomposition(EEMD)and the complexity theory of sample entropy(SampEn).Firstly,we use EEMD-SampEn to decompose the geomagnetic variation time series into many series of geomagnetic variation subsequences whose complexity degrees are transparently different.Then,we use ESN to build a forecasting model for each subsequence,selecting the optimal model parameters.Finally,we use the real data collected from the geomagnetic observatory to conduct simulations.The results show that the forecasting value of the combined model can closely conform to the tendency of geomagnetic variation field,and is superior to the least square support vector machine(LSSVM)model.The mean absolute error of the model for three-hour forecasting is less than 1.40nT when Kp index is less than 3.展开更多

The classification of infrasound events has considerable importance in improving the capability to identify the types of natural disasters.The traditional infrasound classification mainly relies on machine learning al...The classification of infrasound events has considerable importance in improving the capability to identify the types of natural disasters.The traditional infrasound classification mainly relies on machine learning algorithms after artificial feature extraction.However,guaranteeing the effectiveness of the extracted features is difficult.The current trend focuses on using a convolution neural network to automatically extract features for classification.This method can be used to extract signal spatial features automatically through a convolution kernel;however,infrasound signals contain not only spatial information but also temporal information when used as a time series.These extracted temporal features are also crucial.If only a convolution neural network is used,then the time dependence of the infrasound sequence will be missed.Using long short-term memory networks can compensate for the missing time-series features but induces spatial feature information loss of the infrasound signal.A multiscale squeeze excitation–convolution neural network–bidirectional long short-term memory network infrasound event classification fusion model is proposed in this study to address these problems.This model automatically extracted temporal and spatial features,adaptively selected features,and also realized the fusion of the two types of features.Experimental results showed that the classification accuracy of the model was more than 98%,thus verifying the effectiveness and superiority of the proposed model.展开更多
Paleomagnetic results cannot be applied in global and regional tectonic reconstructions unless the paleosecular variation has been adequately averaged.However,how many sampling sites and samples are enough to calculat...Paleomagnetic results cannot be applied in global and regional tectonic reconstructions unless the paleosecular variation has been adequately averaged.However,how many sampling sites and samples are enough to calculate a reliable paleopole remains debated.Based on the relation among the sampling sites N,the precision parameter k,the virtual geomagnetic pole scatter s,and the confidence limit A95of the paleopole,we find that 20 sites(samples)or more are required to yield a paleopole with an A95≈5°based on a review of available paleomagnetic results from the Lhasa,Qiangtang and Tethyan Himalaya.Random samplings of Jurassic virtual geomagnetic poles from the Sangri area show that the Fisher mean pole with neglectable angle deviation can be obtained when sampling sites increase to 20.High-quality paleomagnetic results,with sites/samples number N≥~20–30,show that the Qiangtang,Lhasa,and Tethyan Himalaya moved northward in the Late Permian–Middle Triassic,Jurassic,and Cretaceous,respectively,and then accreted to Asia in the Late Triassic,Late Jurassic–Early Cretaceous and Paleocene–Early Eocene,respectively.展开更多

To elucidate the effect ofthe circHIAT1/miR-19a-3p/phosphatase and tensin homolog (PTEN) axis on regulating the malignant phenotype ofbreast cancer cells. Methods: The mRNA expression pattern of circHIAT1, miR-19a-3p, and PTEN was checked byreal-time quantitative polymerase chain reaction. Then, the knockdown assay was carried out to explore the effect ofcircHIAT1 and miR-19a-3p on breast cancer. The relative cell experiments, including MTT assay, scratch assay,transwell invasion assay, and flow cytometry analysis, were conducted to verify the influence of circHIAT1 and miR-19a-3p on breast cancer cells. Results: The levels of PTEN and circHIAT1 were reduced, while that of miR-19a-3pwas elevated in breast cancer tissues and cells. MiR-19a-3p was proved to be the target gene of circHIAT1 via a dualluciferase experiment, which could also modulate the PTEN mRNA level. Overexpression of circHIAT1 was able toundermine the growth, migratory ability, and invasiveness in breast cancer cells, which could be antagonized by miR-19a-3p mimic. The inhibition of miR-19a-3p in vitro also impaired the malignancy of breast cancer, which dependedon the modulation of PTEN expression. Conclusion: CircHIAT1 controls the PTEN expression level in cells of breastcancer by negatively regulating miR-19a-3p. This mechanism controls the growth, invasion, and migration of breastcancer.展开更多

It is believed that the chalcone compounds of the plant play an important role in the treatment since they may activate tyrosinase and improve melanin production. In this study, twenty-one chalcones and nine analogues were synthesized in view of three different components of chalcone(A, B ring and a,b-unsaturated carbonyl). After biological evaluation of their activity on tyrosinase in cell-free systems,the result showed that most compounds(except polyhydroxy chalcones) possess activator effect on the tyrosinase, especially for 13a–15a, 20 a and 1b, which bearing a comparable activity to the positive control8-MOP. SAR of these tyrosinase activator was summed up for the first time as well. Finally, compound 13 a was found to increase melanin contents and tyrosinase activity 1.75 and 1.3 fold, respectively, compared with that of untreated murine B16 cells at the concentration of 40 mg/m L.展开更多
Ammonia nitrogen is one of the most common environmental stress factors in aquaculture.The aim of this experiment was to evaluate the effects of ammonia nitrogen stress on ammonia metabolism,digestion and immune funct...Ammonia nitrogen is one of the most common environmental stress factors in aquaculture.The aim of this experiment was to evaluate the effects of ammonia nitrogen stress on ammonia metabolism,digestion and immune function as well as the protective effects of dietary N-carbamylglutamate(NCG)in the Chinese mitten crab(Eriocheir sinensis).Two feeds were used,namely a control feed(regular commercial feed)and an NCG feed(NCG(302.96±4.07)mg/kg added).Subsequently,animals fed by these two feeds were exposed to 30 mg/L or 60 mg/L ammonia nitrogen stress(named A30 or A60)for 21 days,respectively.The rate of hemolymph ammonia excretion was significantly increased in NCG group compared to C(control)group(P<0.05).Hemolymph ammonia levels were significantly lower in NCG A30(NCG feed and A30)and NCG A60(NCG feed and A60)groups compared to A30(control feed and A30)and A60(control feed and A60)groups(P<0.05).Glutamate dehydrogenase and glutamine synthetase activities in the gills and hepatopancreas of NCG A60 group were significantly increased compared with the A60 group,which promoted glutamine synthesis(P<0.05).The cumulative mortality rate of Eriocheir sinensis in NCG A30 and NCG A60 groups were effectively reduced(P<0.05).The arginine levels of hepatopancreas were significantly increased in NCG A60 group(P<0.05).Under ammonia nitrogen stress,NCG significantly increased the activities of trypsin,pepsin,lipase andα-Amylase in NCG feed group,and promoted feeding(P<0.05).In hemolymph,hepatopancreas and intestine,NCG significantly increased nitric oxide synthase activity,lysozyme and phenol oxidase activities compared to the ammonia nitrogen stress group(P<0.05).The results indicated that,NCG added in feed could decelerate the toxic effects of ammonia on Eriocheir sinensis by promoting ammonia excretion and accelerating glutamine and endogenous arginine synthesis by metabolism.Moreover,the activity of digestive enzymes and immune-related enzymes is improved.展开更多
Multi-target analgesics with minimal side effects and high efficacy are a key research focus in addressing the global pain crisis.Using a molecular networking approach,five pairs of potent analgesic alkaloid enantiome...Multi-target analgesics with minimal side effects and high efficacy are a key research focus in addressing the global pain crisis.Using a molecular networking approach,five pairs of potent analgesic alkaloid enantiomers were isolated from the roots of Anacyclus pyrethrum(A.pyrethrum).Their structures were elucidated by comprehensive spectroscopic data analysis,including LR-HSQMBC and 1H–15N HMBC,quantum 13C NMR DP4+and ECD calculations,and single-crystal X-ray diffraction analysis.Anacyphrethines A(1)and B(2)are highly conjugated and polymethylated 6/6/6/6/5/7/5/5-fused octacyclic tetraazabic alkaloids possessing an unprecedented 8,14,18,24-tetraaza-octacyclo[16.8.2.11,23.04,28.05,17.09,16.011,15.021,27]nonacosane motif.Their biosynthetic pathways are proposed involving key aldol,hydroamination,and Schiff base reactions.All isolates showed potent analgesic effects in vivo.Even at a lower dose of 0.2 mg/kg,(±)-1 and(+)-1 still exhibited more potent analgesic activities than morphine.Interestingly,the racemic mixture(±)-1 showed stronger analgesic effect than either pure enantiomer alone at higher doses of 5 and 1 mg/kg;while,(±)-1 showed significant analgesic activities comparable to(+)-1 at lower doses of 0.2 and 0.04 mg/kg.(+)-1 had stronger analgesic effect than(−)-1 at five tested does.Further tests on 44 analgesic-related targets demonstrated that(+)-1 showed significant inhibitory effects against many ion channels such as TRPM8,Kv1.2,Kv1.3,and Cav2.1 with IC50 values of 1.10±0.26,4.20±0.07,2.20±0.24,and 10.40±0.69μmol/L,respectively,while(−)-1 primarily inhibited TRPC6,Kv1.2,and Kv1.3 ion channels with IC50 values of 0.81±0.05,0.91±0.04,and 1.50±0.13μmol/L,respectively,without affecting the opioid receptors,suggesting their non-opioid analgesic potentials.The molecular dockings provided structural guidance to develop potent non-opioid analgesics.展开更多
Platelets act as a crucial indicator for monitoring hypercoagulability and thrombosis and a key target for pharmacological intervention.Genotype-phenotype association studies have confirmed that platelet traits are qu...Platelets act as a crucial indicator for monitoring hypercoagulability and thrombosis and a key target for pharmacological intervention.Genotype-phenotype association studies have confirmed that platelet traits are quantitatively regulated by multiple genes.However,there is currently a lack of genetic studies on the heterogeneity of platelet traits in β-thalassemia under a hypercoagulable state.Here,we studied the phenotypic heterogeneity of platelet count(PLT)and mean platelet volume(MPV)in a cohort of 1020β-thalassemia patients.We further performed a functionally informed whole-genome sequencing(WGS)association analysis of common variants and rare variants for PLT and MPV in 916 patients through integrative analysis of WGS data and functional annotation data.Extreme phenotypic heterogeneity of platelet traits was observed in β-thalassemia patients.Additionally,the common variant-based gene-level analysis identified RNF144B as a novel gene associated with MPV.The rare variant analysis identified several novel associations in both coding and noncoding regions,including missense rare variants of PPP2R5C associated with PLT and missense rare variants of TSSK1B associated with MPV.In conclusion,this comprehensive and systematic whole-genome scan of platelet traits in the β-thalassemia cohort reveals the specific genetic regulation of platelet traits in the context of β-thalassemia,providing potential targets for intervention.展开更多
This study evaluated the effects of feeding a microbial-derived antioxidant diet(MA)for 56 days on the growth performance of Eriocheir sinensis,as well as on digestive and metabolic enzyme activity and antioxidant cap...This study evaluated the effects of feeding a microbial-derived antioxidant diet(MA)for 56 days on the growth performance of Eriocheir sinensis,as well as on digestive and metabolic enzyme activity and antioxidant capacity under ammonia stress(400 mg/L ammonia nitrogen for 48 h).The weight gain at 28d and the specific growth rate at 56ds for crabs in the MA group were significantly higher than those in the control diet(C)group(p<0.05).Compared with the C group,the color of the hepatopancreas in the MA group was significantly improved(p<0.05).A significant decrease in the level of Hemolymph ammonia was found in the microbial antioxidant ammonia-N(MA_A)group(p<0.05),which may be related to the decrease of transaminase(AST,ALT,GDH)activities caused by MA diet(p<0.05).There is a significant increase in the digestive enzyme activities(Trypsin and Lipase),which reduced the accumulation of triglycerides in the hepatopancreas in the(MA_A)group(p<0.05).In addition,the antioxidant damage of crabs in hepatopancreas caused by ammonia nitrogen toxicity could also significantly be improved by MA fed in the(MA_A)group(p<0.05).microbe-derived antioxidants(supplemented with 0.2%in feeds)can protect the health of hepatopancreas,promote growth,and improve the ability to resist ammonia nitrogen stress for E.sinensis.展开更多

摘要This study investigates the relationship between geomagnetic storms and ionospheric total electron content(TEC)disturbances during interplanetary coronal mass ejections(ICME)events from 2000 to 2017.The research employed statistical analysis methods to examine data from 56 ICME events and their corresponding ionospheric TEC disturbances.The results reveal that TEC disturbances preceded geomagnetic storms in 68%of SB-type events,particularly in mid-latitude regions compared to low-latitude regions.Specifi cally,the duration of positive ionospheric TEC disturbances is signifi cantly related to latitude and the type of geomagnetic storm.In contrast,the duration of negative ionospheric TEC disturbances is primarily influenced by latitude and is independent of the geomagnetic storm type.Notably,the duration of both positive-negative and negative-positive ionospheric TEC disturbance is not signifi cantly related to latitude or the type of geomagnetic storm.Furthermore,the study identifi es an asymmetry in the duration of ionospheric TEC disturbance under the same type of geomagnetic storm in the Northern and Southern Hemispheres with respect to latitude.For instance,during SB-type geomagnetic storms,the duration of positive ionospheric TEC disturbance at mid-latitudes(40°N,50°N,60°N)is mainly 0–12 hours,while at low-latitudes(10°S,0°,10°N,20°N,30°N)it ranges from 0–12 hours to 48–96 hours.These results provide valuable insights into the Sun-Earth energy coupling mechanism and off er a theoretical basis for the prediction and monitoring of ionospheric TEC disturbance,enhancing our understanding of the complex interactions between geomagnetic storms and ionospheric TEC disturbances.

摘要The technology of locating magnetic anomaly targets via geomagnetic eld measurements has been increasingly widely applied,with multiple magnetic anomaly target localization emerging as a critical research direction.However,when two magnetic anomaly targets are horizontally close but vertically separated,traditional clustering-based localization methods tend to omit the deeper target.To address this issue,we propose an improved clustering-based localization method for multiple magnetic anomaly targets,which integrates two core innovations:the introduction of a reference target to establish a benchmark for normal magnetic moment distribution,and the utilization of spatial distribution characteristics of magnetic moment estimates to judge the presence of omitted targets.Simulation results demonstrate that the proposed method not only achieves accurate localization of conventional targets but also eectively identies the omission of deeper targets,providing a reliable basis for determining whether supplementary localization steps are required.

摘要The widespread use of antibiotics has led to their persistent accumulation in aquatic environments.Although flocculation is cost-effective,traditional flocculants exhibit limited removal efficiency for antibiotics.To address this problem,a novel cellulose-based flocculant(CE-DMC)was synthesized from paper pulp rejects.The methacryloxyethyltrimethyl ammonium chloride(DMC)monomer was successfully grafted onto the cellulose backbone with a grafting rate of 64%,endowing CE-DMC with high positive charge density(+71.1 mV at pH 6)and a branched architecture.Flocculation experiments demonstrated the superior performance of CE-DMC in removing four representative antibiotics.Notably distinct removal efficiencies were achieved:ofloxacin exhibited the highest removal(52.6%),followed by sulfamethoxazole(36.4%),norfloxacin(30.9%),and amoxicillin(29.8%),reflecting differential interaction mechanisms with the adsorbent.The flocculation mechanism of CE-DMC could be concluded as follows:(1)charge neutralization via quaternary ammonium groups of DMC chains,(2)n-π*electron donor-acceptor interactions andπ-π*conjugated interactions mediated by benzene rings and furan rings of cellulose skeleton,and(3)hydrogen bonding force provided by the hydroxyl group from the cellulose skeleton and the DMC chains.Notably,coexisting humic acid(HA)and kaolin enhanced antibiotic removal efficiency by forming large composite pollutants via n-π*electron donor-acceptor interactions,hydrogen bonding forces,andπ-π*conjugated interactions.CE-DMC synergistically removed antibiotics and co-pollutants,with its flocculation mechanism elucidated through structure-activity relationships and structural determinants.This work advances the rational design of natural polymer-based flocculants for multi-mechanistic contaminant removal.

摘要Modeling and forecasting of the geomagnetic variation are important research topics concerning geomagnetic navigation and space environment monitoring.We propose a combined forecasting model using a dynamic recursive neural network called echo state network(ESN),the method of complementary ensemble empirical mode decomposition(EEMD)and the complexity theory of sample entropy(SampEn).Firstly,we use EEMD-SampEn to decompose the geomagnetic variation time series into many series of geomagnetic variation subsequences whose complexity degrees are transparently different.Then,we use ESN to build a forecasting model for each subsequence,selecting the optimal model parameters.Finally,we use the real data collected from the geomagnetic observatory to conduct simulations.The results show that the forecasting value of the combined model can closely conform to the tendency of geomagnetic variation field,and is superior to the least square support vector machine(LSSVM)model.The mean absolute error of the model for three-hour forecasting is less than 1.40nT when Kp index is less than 3.

摘要Programmed silencing ofγ-globin genes in adult erythropoiesis is mediated by several chromatin remodeling complexes,which determine the stage-specific genome architecture in this region.Identification of cis-or trans-acting mutations contributing to the diverse extent of fetal hemoglobin(Hb F)might illustrate the underlying mechanism ofγ-β-globin switching.Here,we recruit a cohort of 1142β-thalassemia patients and dissect the natural variants in the wholeβ-globin gene cluster through a targeted next-generation sequencing panel.A previously unreported SNP rs7948668,predicted to disrupt the binding motif of IKAROS as a key component of chromatin remodeling complexes,is identified to be significantly associated with higher levels of Hb F and age at onset.Gene-editing on this SNP leads to the elevation of Hb F in both HUDEP-2 and primary CD34+cells while the extent of elevation is amplified in the context ofβ-thalassemia mutations,indicating epistasis effects of the SNP in the regulation of Hb F.Finally,we perform ChIP-qPCR and 4C assays to prove that this variant disrupts the binding motif of IKAROS,leading to enhanced competitiveness of HBG promoters to locus control regions.This study highlights the significance of common regulatory SNPs and provides potential targets for treatingβ-hemoglobinopathy.

摘要Vector magnetic measurement is increasingly widely used.In order to improve the accuracy of vector magnetic measurement system on board a vehicle,researchers have proposed various calibration methods.Most of them require altering the magnetic vector in the vehicle coordinate system.Exploring the use of geomagnetic variation to change the geomagnetic vector in the vehicle coordinate system,this paper proposes a novel vector magnetic measurement calibration method.In this method,a vector magnetometer mounted on a vehicle and an accurate vector magnetometer separately measure the geomagnetic field at diff erent locations within the same area.Based on the physical principle that the geomagnetic variation at two nearby locations is equal,the calibration parameters of the magnetometer on the vehicle can be determined through a set of equations containing the measurements from the two magnetometers.The theoretical derivation and simulation experiment results demonstrate the feasibility of this method.Therefore,it can serve as a new alternative calibration method,especially in scenarios where a high degree of accuracy in the estimation of calibration parameters is not required.

摘要The classification of infrasound events has considerable importance in improving the capability to identify the types of natural disasters.The traditional infrasound classification mainly relies on machine learning algorithms after artificial feature extraction.However,guaranteeing the effectiveness of the extracted features is difficult.The current trend focuses on using a convolution neural network to automatically extract features for classification.This method can be used to extract signal spatial features automatically through a convolution kernel;however,infrasound signals contain not only spatial information but also temporal information when used as a time series.These extracted temporal features are also crucial.If only a convolution neural network is used,then the time dependence of the infrasound sequence will be missed.Using long short-term memory networks can compensate for the missing time-series features but induces spatial feature information loss of the infrasound signal.A multiscale squeeze excitation–convolution neural network–bidirectional long short-term memory network infrasound event classification fusion model is proposed in this study to address these problems.This model automatically extracted temporal and spatial features,adaptively selected features,and also realized the fusion of the two types of features.Experimental results showed that the classification accuracy of the model was more than 98%,thus verifying the effectiveness and superiority of the proposed model.

摘要Paleomagnetic results cannot be applied in global and regional tectonic reconstructions unless the paleosecular variation has been adequately averaged.However,how many sampling sites and samples are enough to calculate a reliable paleopole remains debated.Based on the relation among the sampling sites N,the precision parameter k,the virtual geomagnetic pole scatter s,and the confidence limit A95of the paleopole,we find that 20 sites(samples)or more are required to yield a paleopole with an A95≈5°based on a review of available paleomagnetic results from the Lhasa,Qiangtang and Tethyan Himalaya.Random samplings of Jurassic virtual geomagnetic poles from the Sangri area show that the Fisher mean pole with neglectable angle deviation can be obtained when sampling sites increase to 20.High-quality paleomagnetic results,with sites/samples number N≥~20–30,show that the Qiangtang,Lhasa,and Tethyan Himalaya moved northward in the Late Permian–Middle Triassic,Jurassic,and Cretaceous,respectively,and then accreted to Asia in the Late Triassic,Late Jurassic–Early Cretaceous and Paleocene–Early Eocene,respectively.

摘要Objective: Breast cancer is a major cancer threatening the health of women globally. To elucidate the effect ofthe circHIAT1/miR-19a-3p/phosphatase and tensin homolog (PTEN) axis on regulating the malignant phenotype ofbreast cancer cells. Methods: The mRNA expression pattern of circHIAT1, miR-19a-3p, and PTEN was checked byreal-time quantitative polymerase chain reaction. Then, the knockdown assay was carried out to explore the effect ofcircHIAT1 and miR-19a-3p on breast cancer. The relative cell experiments, including MTT assay, scratch assay,transwell invasion assay, and flow cytometry analysis, were conducted to verify the influence of circHIAT1 and miR-19a-3p on breast cancer cells. Results: The levels of PTEN and circHIAT1 were reduced, while that of miR-19a-3pwas elevated in breast cancer tissues and cells. MiR-19a-3p was proved to be the target gene of circHIAT1 via a dualluciferase experiment, which could also modulate the PTEN mRNA level. Overexpression of circHIAT1 was able toundermine the growth, migratory ability, and invasiveness in breast cancer cells, which could be antagonized by miR-19a-3p mimic. The inhibition of miR-19a-3p in vitro also impaired the malignancy of breast cancer, which dependedon the modulation of PTEN expression. Conclusion: CircHIAT1 controls the PTEN expression level in cells of breastcancer by negatively regulating miR-19a-3p. This mechanism controls the growth, invasion, and migration of breastcancer.

摘要The Vernohia anthelmintica L.'s extract is one of the most popular Uygur medicines used for vitiligo. It is believed that the chalcone compounds of the plant play an important role in the treatment since they may activate tyrosinase and improve melanin production. In this study, twenty-one chalcones and nine analogues were synthesized in view of three different components of chalcone(A, B ring and a,b-unsaturated carbonyl). After biological evaluation of their activity on tyrosinase in cell-free systems,the result showed that most compounds(except polyhydroxy chalcones) possess activator effect on the tyrosinase, especially for 13a–15a, 20 a and 1b, which bearing a comparable activity to the positive control8-MOP. SAR of these tyrosinase activator was summed up for the first time as well. Finally, compound 13 a was found to increase melanin contents and tyrosinase activity 1.75 and 1.3 fold, respectively, compared with that of untreated murine B16 cells at the concentration of 40 mg/m L.

摘要Ammonia nitrogen is one of the most common environmental stress factors in aquaculture.The aim of this experiment was to evaluate the effects of ammonia nitrogen stress on ammonia metabolism,digestion and immune function as well as the protective effects of dietary N-carbamylglutamate(NCG)in the Chinese mitten crab(Eriocheir sinensis).Two feeds were used,namely a control feed(regular commercial feed)and an NCG feed(NCG(302.96±4.07)mg/kg added).Subsequently,animals fed by these two feeds were exposed to 30 mg/L or 60 mg/L ammonia nitrogen stress(named A30 or A60)for 21 days,respectively.The rate of hemolymph ammonia excretion was significantly increased in NCG group compared to C(control)group(P<0.05).Hemolymph ammonia levels were significantly lower in NCG A30(NCG feed and A30)and NCG A60(NCG feed and A60)groups compared to A30(control feed and A30)and A60(control feed and A60)groups(P<0.05).Glutamate dehydrogenase and glutamine synthetase activities in the gills and hepatopancreas of NCG A60 group were significantly increased compared with the A60 group,which promoted glutamine synthesis(P<0.05).The cumulative mortality rate of Eriocheir sinensis in NCG A30 and NCG A60 groups were effectively reduced(P<0.05).The arginine levels of hepatopancreas were significantly increased in NCG A60 group(P<0.05).Under ammonia nitrogen stress,NCG significantly increased the activities of trypsin,pepsin,lipase andα-Amylase in NCG feed group,and promoted feeding(P<0.05).In hemolymph,hepatopancreas and intestine,NCG significantly increased nitric oxide synthase activity,lysozyme and phenol oxidase activities compared to the ammonia nitrogen stress group(P<0.05).The results indicated that,NCG added in feed could decelerate the toxic effects of ammonia on Eriocheir sinensis by promoting ammonia excretion and accelerating glutamine and endogenous arginine synthesis by metabolism.Moreover,the activity of digestive enzymes and immune-related enzymes is improved.

摘要Multi-target analgesics with minimal side effects and high efficacy are a key research focus in addressing the global pain crisis.Using a molecular networking approach,five pairs of potent analgesic alkaloid enantiomers were isolated from the roots of Anacyclus pyrethrum(A.pyrethrum).Their structures were elucidated by comprehensive spectroscopic data analysis,including LR-HSQMBC and 1H–15N HMBC,quantum 13C NMR DP4+and ECD calculations,and single-crystal X-ray diffraction analysis.Anacyphrethines A(1)and B(2)are highly conjugated and polymethylated 6/6/6/6/5/7/5/5-fused octacyclic tetraazabic alkaloids possessing an unprecedented 8,14,18,24-tetraaza-octacyclo[16.8.2.11,23.04,28.05,17.09,16.011,15.021,27]nonacosane motif.Their biosynthetic pathways are proposed involving key aldol,hydroamination,and Schiff base reactions.All isolates showed potent analgesic effects in vivo.Even at a lower dose of 0.2 mg/kg,(±)-1 and(+)-1 still exhibited more potent analgesic activities than morphine.Interestingly,the racemic mixture(±)-1 showed stronger analgesic effect than either pure enantiomer alone at higher doses of 5 and 1 mg/kg;while,(±)-1 showed significant analgesic activities comparable to(+)-1 at lower doses of 0.2 and 0.04 mg/kg.(+)-1 had stronger analgesic effect than(−)-1 at five tested does.Further tests on 44 analgesic-related targets demonstrated that(+)-1 showed significant inhibitory effects against many ion channels such as TRPM8,Kv1.2,Kv1.3,and Cav2.1 with IC50 values of 1.10±0.26,4.20±0.07,2.20±0.24,and 10.40±0.69μmol/L,respectively,while(−)-1 primarily inhibited TRPC6,Kv1.2,and Kv1.3 ion channels with IC50 values of 0.81±0.05,0.91±0.04,and 1.50±0.13μmol/L,respectively,without affecting the opioid receptors,suggesting their non-opioid analgesic potentials.The molecular dockings provided structural guidance to develop potent non-opioid analgesics.

摘要Platelets act as a crucial indicator for monitoring hypercoagulability and thrombosis and a key target for pharmacological intervention.Genotype-phenotype association studies have confirmed that platelet traits are quantitatively regulated by multiple genes.However,there is currently a lack of genetic studies on the heterogeneity of platelet traits in β-thalassemia under a hypercoagulable state.Here,we studied the phenotypic heterogeneity of platelet count(PLT)and mean platelet volume(MPV)in a cohort of 1020β-thalassemia patients.We further performed a functionally informed whole-genome sequencing(WGS)association analysis of common variants and rare variants for PLT and MPV in 916 patients through integrative analysis of WGS data and functional annotation data.Extreme phenotypic heterogeneity of platelet traits was observed in β-thalassemia patients.Additionally,the common variant-based gene-level analysis identified RNF144B as a novel gene associated with MPV.The rare variant analysis identified several novel associations in both coding and noncoding regions,including missense rare variants of PPP2R5C associated with PLT and missense rare variants of TSSK1B associated with MPV.In conclusion,this comprehensive and systematic whole-genome scan of platelet traits in the β-thalassemia cohort reveals the specific genetic regulation of platelet traits in the context of β-thalassemia,providing potential targets for intervention.

摘要This study evaluated the effects of feeding a microbial-derived antioxidant diet(MA)for 56 days on the growth performance of Eriocheir sinensis,as well as on digestive and metabolic enzyme activity and antioxidant capacity under ammonia stress(400 mg/L ammonia nitrogen for 48 h).The weight gain at 28d and the specific growth rate at 56ds for crabs in the MA group were significantly higher than those in the control diet(C)group(p<0.05).Compared with the C group,the color of the hepatopancreas in the MA group was significantly improved(p<0.05).A significant decrease in the level of Hemolymph ammonia was found in the microbial antioxidant ammonia-N(MA_A)group(p<0.05),which may be related to the decrease of transaminase(AST,ALT,GDH)activities caused by MA diet(p<0.05).There is a significant increase in the digestive enzyme activities(Trypsin and Lipase),which reduced the accumulation of triglycerides in the hepatopancreas in the(MA_A)group(p<0.05).In addition,the antioxidant damage of crabs in hepatopancreas caused by ammonia nitrogen toxicity could also significantly be improved by MA fed in the(MA_A)group(p<0.05).microbe-derived antioxidants(supplemented with 0.2%in feeds)can protect the health of hepatopancreas,promote growth,and improve the ability to resist ammonia nitrogen stress for E.sinensis.
摘要Conventional dendritic cells(cDCs)are categorized into two main subsets,cDC1 and cDC2,distinguished by their differential expression of cell surface markers,and the development of these two cDC subsets is dependent on interferon regulatory factor 8(IRF8)and IRF4,respectively.1 Kim et al.2 recently reported that cDC1 can be distinguished from cDC2 based on an AP1–IRF composite element(AICE)-dependent gene program.IRF8 at a high concentration engages AICEs at enhancer regions to direct a program toward establishing the cDC1 identity.These findings provide a molecular basis to explain the distinct transcriptional signatures of cDC1s and cDC2s.

摘要A new series of(E)-1-(4-((1-benzyl-1H-1,2,3-triazol-4-yl)methoxy)phenyl)-3-phenylprop-2-en-1-one 1a-15a and(E)-3-(4-((1-benzyl-1H-1,2,3-triazol-4-yl)methoxy)phenyl)-1-phenylprop-2-en-1-one 1b-15b were designed,synthesized based on click chemistry,and biologically evaluated for their activity on tyrosinase.The result showed that most of prepared compounds 1a-15a have potent activating effect on tyrosinase,especially for 3a,8a-10a and 14a-15a.Among them,compounds 10a and 14a demonstrated the best activity with EC50=1.71 and 5.60μmol•L−1 respectively,even better than the positive control 8-MOP(EC50=14.8μmol•L−1).Conversely,com-pounds 3b,5b-6b,9b-10b,and 15b induced enzymatic inhibition on tyrosinase.

摘要We study the linear instability and nonlinear dynamical evolution of the Reissner-Nordstrom(RN)black hole in the Einstein-Maxwell-scalar theory in asymptotic flat spacetime.We focus on the coupling function f(φ)=e-bφ^(2),which facilitates both scalar-free RN and scalarized black hole solutions.We first present the evolution of system parameters during dynamic scalarization.For parameter regions in which spontaneous scalarization occurs,we observe that the evolution of the scalar field at the horizon is dominated by the fundamental unstable mode from linear analysis at early times.At late times,the nonlinear evolution can be considered to be the perturbation of scalarized black holes.

摘要We study the linear instability of the charged massless scalar perturbation in regularized 4D charged Einstein-Gauss-Bonnet-AdS black holes by exploring their quasinormal modes.We find that the linear instability is triggered by superradiance.The charged massless scalar perturbation becomes more unstable with increasing Gauss-Bonnet coupling constant or black hole charge.Decreasing the AdS radius,on the other hand,will make the charged massless scalar perturbation more stable.The stable region in parameter space(α,Q,Λ)is given.Moreover,we find that the charged massless scalar perturbation is more unstable for larger scalar charge.The modes of multipoles are more stable than that of the monopole.

摘要We studied the instability of regularized 4D charged Einstein-Gauss-Bonnet de-Sitter black holes under charged scalar perturbations.The unstable modes satisfy the superradiant condition,but not all of the modes satisfying the superradiant condition are unstable.The instability occurs when the cosmological constant is small and the black hole charge is not too large.The Gauss-Bonnet coupling constant further destabilizes black holes when both the black hole charge and the cosmological constant are small and further stabilizes black holes when the black hole charge is large.
