Utilizing small molecules as markers for specific cells or organs within biosystems is a crucial approach for studying and regulating physiological processes. However, current tagging strategies, due to the presence of exposed highly reactive groups, suffer from drawbacks such as low tagging efficiency or insufficient spatial specificity, thereby diminishing their expected effectiveness. Consequently, there is a pressing need to develop a strategy capable of in situ labeling of active groups in response to cellular or in vivo stimuli, ensuring both high tagging efficiency and spatial specificity. In this work, we devised a strategy for releasing aldehyde groups activated by hypochlorous acid(HOCl). Compounds synthesized through this strategy can release the fiuorophore methylene blue(MB) and aldehyde-based compounds upon HOCl activation. Given high reactivity of the released aldehyde group, it can effectively interact with macromolecules in biological systems, facilitating tagging and enabling prolonged imaging. To validate this concept, we further incorporated a naphthalimide structure with stable light emission to create SW-110. SW-110 can specifically respond to in vitro and endogenous HOCl, when release MB, it also releases naphthalimide fiuorophore with highly reactive aldehyde group for tagging within cells. This strategy provides a simple but efficient strategy for proximity tagging in situ. 展开更多

The cylindrical virtual cathode reflex triode is a new type of pulsed hard X-ray load,which has the advantages of simple structure,high radiation conversion efficiency,and simplicity in seriesparallel operation.This paper presents a method to reduce the impedance of the triode using a multiring cathode.The average electric field on the ring-cathode emission surface is enhanced due to edge effect,and the beam intensity is greatly increased in proportion to the square of the electric field strength.Multi-ring cathode is used to enlarge the emission area.Therefore,the reflex triode can work at lower impedance and generate a stronger beam under the same anode-cathode gap.In addition,the electric field enhancement of the cathode reduces the cathode emission stabilization time and enhances the operation stability of the triode.The effects of parameters such as ring width and ring gap on the triode impedance are simulated and studied.The cathode emission stabilization time and the X-ray conversion efficiency are compared.The design basis of cathode structure parameters and the impedance control method of the cylindrical virtual cathode reflex triode are given according to the simulations. 展开更多

Data-driven control has rapidly matured into a powerful complement to classical model-based paradigms.Among the most successful approaches are nonparametric,trajectory-centric methods built on Willems et al.’s fundamen-tal lemma and model-free reinforcement learning(RL)schemes.In this survey,we reverse the usual presentation order to reflect their differing assumptions and scopes.First,this paper provides a unified treatment of fundamen-tal lemma-based methods,which require knowledge of the system structure(LTI or suitably lifted nonlinear)but offer rigorous guarantees by replacing explicit model identification with input-output trajectory representations.This paper covers state-feedback synthesis for both linear and nonlinear systems,extensions to model predic-tive control that directly leverage data,and recent advances in data-driven state estimation under noise-free and noisy measurements.This paper then turns to RL-based control,which lifts the structural constraints entirely-trading formal stability proofs for broad applicability to complex and uncertain cyber-physical energy systems(CPESs)dynamics.This paper reviews key RL architectures,policy-and value-based algorithms,and their prac-tical challenges in energy applications.Recognizing the network-induced vulnerabilities of modern grids,this paper also surveys data-driven defense strategies against Denial-of-Service and false-data-injection attacks,from anomaly detection to resilient control.To illustrate practical impact,this paper steps through case studies ranging from canonical generator benchmarks to high-fidelity,grid-connected wind-turbine simulations,demonstrating both trajectory-based controllers and RL agents.This paper concludes by outlining future directions:integrating learning-augmented control with fundamental lemma frameworks,enhancing robustness to network effects,and embedding data-driven models in large-scale optimization for next-generation CPESs. 展开更多

Breast cancer is the leading cause of cancer-related deaths in women worldwide,with Hormone Receptor(HR)+being the predominant subtype.Tamoxifen(TAM)serves as the primary treatment for HR+breast cancer.However,drug resistance often leads to recurrence,underscoring the need to develop new therapies to enhance patient quality of life and reduce recurrence rates.Artemisinin(ART)has demonstrated efficacy in inhibiting the growth of drug-resistant cells,positioning art as a viable option for counteracting endocrine resistance.This study explored the interaction between artemisinin and tamoxifen through a combined approach of bioinformatics analysis and experimental validation.Five characterized genes(ar,cdkn1a,erbb2,esr1,hsp90aa1)and seven drug-disease crossover genes(cyp2e1,rorc,mapk10,glp1r,egfr,pgr,mgll)were identified using WGCNA crossover analysis.Subsequent functional enrichment analyses were conducted.Our findings confirm a significant correlation between key cluster gene expression and immune cell infiltration in tamoxifen-resistant and-sensitized patients.scRNA-seq analysis revealed high expression of key cluster genes in epithelial cells,suggesting artemisinin’s specific impact on tumor cells in estrogen receptor(ER)-positive BC tissues.Molecular target docking and in vitro experiments with artemisinin on LCC9 cells demonstrated a reversal effect in reducing migratory and drug resistance of drug-resistant cells by modulating relevant drug resistance genes.These results indicate that artemisinin could potentially reverse tamoxifen resistance in ER-positive breast cancer. 展开更多

河南省地处中原,地理环境复杂多样,为各种类型植物的生长提供了多种多样的自然条件。历年来,河南省丰富的植物多样性受到有关部门和广大学者的关注。前人采集的大量植物标本为《河南植物志》1–4册的出版提供了有力支撑。然而,《河南植... 河南省地处中原,地理环境复杂多样,为各种类型植物的生长提供了多种多样的自然条件。历年来,河南省丰富的植物多样性受到有关部门和广大学者的关注。前人采集的大量植物标本为《河南植物志》1–4册的出版提供了有力支撑。然而,《河南植物志(补修编)》缺少大量野外调查,遗漏了之前发表的一些新记录。本研究通过收集已公开发表的论著,全面清查全国植物标本馆馆藏的河南植物标本,并于2016–2020年对河南省林木种质资源进行了全面普查。最后,本研究利用近年来国内外最新研究成果,确定了河南省木本植物的有效性和分类归属,对河南省野生木本植物名录进行了系统的整理与修订。结果表明:河南省有野生木本植物85科288属984种32亚种148变种11变型,包括《河南植物志》新分布类群168个。另外,我们对名录中的所有类群进行了文献考证,属、种和种下分类单元的分类学处理以Flora of China为准,发现《中国植物志》属异名5个、种异名102个、亚种异名21个、变种异名24个,还发现《河南植物志》属异名6个、种异名194个、亚种异名20个、变种异名35个和变型异名1个。此外,对50个类群是否分布于河南省产生质疑。本文还对出现的异名、存疑类群和新记录的主客观原因进行了分析,提出应持续开展野外考察工作,尤其加强对河南省边界市县,如商城县、新县、桐柏县、淅川县、卢氏县和灵宝市等进行重点植物普查,彻底摸清河南省植物资源家底。 展开更多

In this study,headspace solid phase microextraction(SPME),solvent-assisted flavor evaporation(SaFE)and gas chromatography-mass spectrometry were used to detect the changes in aroma components of mandarin orange juice before and after sterilization,the characteristic flavor substances were also analyzed using odor activity value,principal component analysis,and heatmap.The results showed that 149 volatile components were detected,mainly including terpenes,esters,alcohols,aldehydes,and ketones.Compared with unpasteurized juice,the SPME method measured that 38 new volatile components were produced after pasteurization,while 25 volatile components disappeared;the SAFE method measured the production of 34 volatile components after pasteurization,while 18 volatile components disappeared.β-Myrcene,D-limonene,linalool,and p-cymene were important volatile substances in mandarin orange juice.These results provide a reference for further control and detection of mandarin orange juice processing flavors. 展开更多

The development of efficient transition metal oxygen reduction reaction(ORR)catalysts as substitutes for platinumbased catalysts has served as a key strategy in advancing the commercialization of fuel cells.Herein,an effective Fe-N-C catalyst for the ORR was fabricated with hemin as the iron source of Fe-NX active sites through a simple precipitation-pyrolysis method.With the aid of hemin,the leaf-like ZIF-8 synthesized in an aqueous environment can be driven to transform into a stable three-dimensional dodecahedral structure,resulting in a substantial increase in the yield of Fe-N-C materials.Meanwhile,the influence of hemin loading in the precursors and pyrolysis temperature on the oxygen reduction catalytic activity of the final product Fe-N-C was investigated.Concretely,the optimal Fe100-N-C950 displays an onset potential of 1.054 V(vs.RHE)and a half-wave potential of 0.847 V(vs.RHE).Additionally,the direct ethanol fuel cell(DEFC)with Fe100-N-C950 as the cathode catalyst demonstrated a higher power density than the one with the commercial Pt-C catalyst. 展开更多

Malignant melanoma of the skin is prone to metastasis and recurrence.Photodynamic therapy(PDT)excited by near-infrared(NIR)light,as a non-invasive method,is very suitable for treating malignant melanoma of the skin.However,because of the high expression of melanin that absorbs red or NIR light and consumes reactive oxygen species in malignant melanoma,achieving good PDT efficiency remains a challenge.In this study,we report a prodrug SW-111 with a simple structure that can be activated by HOCl and ONOO-,and simultaneously release the photosensitizer methylene blue(MB)for PDT and the inhibitor 4-hydroxybenzaldehyde(4-HBA)that can inhibit tyrosinase activity and reduce melanin expression to enhance the PDT effect.SW-111 can not only reduce melanin production in B16F10 cells but also exhibits excellent anti-tumor effects in a melanoma mouse model.The tumors are significantly suppressed at an inhibition rate of 98.8%according to the excised tumor weights at the end of the treatment and no recurrence happens after the mice are treated with both SW-111 and laser. 展开更多

The sputtering mode diagram(SMD)provides a powerful tool for comprehensive structure engineering of functional films in developing advanced electronic devices,but has not reported due to the complexity of dynamic process and multi-parameters.Here,we report the SMD of superconductor niobium nitride(NbN)films with reactive magnetron sputtering.Poisoned mode,competing mode,and metallic mode are drawn by the boundaries identified by the current‒voltage curves of the sputtering system in the SMD,by which the phase structures and electronic properties of the NbN films can be precisely engineered.Typically,9-nm-thick NbN films grown in the optimal poisoned and competing modes are applied for superconducting nanowire single-photon detectors(SNSPDs).The as-fabricated SNSPDs have flexible performances with saturated quantum efficiency and small kinetic inductance,which enables precise manipulation of the sensitivity and speed from the SMD.This work is also providing guidance for the research of other functional films and electronic devices,which can undoubtedly promote its practical application such as dark matter detection and high-speed quantum communication. 展开更多

This randomized,prospective,multicenter(12 centers in China)phase III trial(Chinese Clinical Trial Registry#ChiCTR2000041170)compared drug-eluting bead transarterial chemoembolization(DEB-TACE)combined with apatinib and DEB-TACE monotherapy for patients with unresectable hepatocellular carcinoma(uHCC).Progression-free survival(PFS)was the primary endpoint.Overall survival(OS),mRECIST-based objective response rates(ORR)and disease control rates(DCR),and treatment-related adverse events(TRAEs)were secondary endpoints.Totally 243 cases were randomized,with 122 and 121 in the DEB-TACE+apatinib and DEBTACE groups,respectively.Cases administered DEB-TACE+apatinib displayed markedly improved median PFS(7.1 months[95%CI 6.6-8.3]vs.5.2 months[95%CI 5.0-5.9])and OS(23.3 months[95%CI 20.7-29.6]vs.18.9 months[95%CI 17.9-20.1]compared with those treated with DEB-TACE(both p<0.001).Additionally,patients administered DEB-TACE+apatinib had elevated ORR(56.6%vs.38.8%)and DCR(89.3%vs.80.2%)versus the DEB-TACE group(both p<0.001).Majority of TRAEs were mild and manageable.Regarding DEB-TACE-related TRAEs,the rates of hepatic artery thinning and spasms were elevated during the second DEB-TACE in cases administered DEB-TACE+apatinib vs.DEB-TACE.The commonest apatinib-related TRAEs in the DEB-TACE+apatinib group included hypertension,hand-foot syndrome,fatigue,and diarrhea.In conclusion,DEB-TACE plus apatinib demonstrates superior PFS versus DEB-TACE monotherapy in uHCC cases,maintaining a favorable safety profile with similar occurrences of AEs. 展开更多

Accurate extraction of crop row is very important for automation of agricultural production.Crop rows are required for accurate machine guidance in agricultural production such as fertilization,plant protection,weeding and harvesting.In this study,an efficient crop row detection algorithm called Crop-BiSeNet V2 was proposed,which combined BiSeNet V2 with a spatial convolutional neural network.The proposed Crop-BiSeNet V2 detected crop rows in color images without the use of threshold and other pre-information such as number of rows.A data set had 2697 maize crop images was constructed in challenging field trial conditions such as variable light,shadows,presence of weeds,and irregular crop shape.The proposed system was experimentally determined to overcome the interference of different complex scenes.And it can be applied to crop rows of different numbers,straight lines and curves.Different analyses were performed to check the robustness of the algorithm.Comparing this algorithm with the Fully Convolutional Networks(FCN)algorithm,it exhibited superior performance and saved 84.85 ms.The accuracy rate reached 0.9811,and the detection speed reached 65.54 ms/frame.The Crop-BiSeNet V2 algorithm proposed in this study show strong generalization performance for seedling crop row recognition.It provides high-reliability technical support for crop row detection research and assists in the study of intelligent field operation machinery navigation. 展开更多

Background Activation of hepatic stellate cells(HSC)leads to initiation and progression of hepatic fibrosis.HSC senescence is inversely correlated with HSC proliferation and activation.Therefore,induction of HSC senescence may be a strategy to treat hepatic fibrosis.Coumarin-derivatives like esculetin have been suggested to inhibit proliferation of fibrogenic cells.Therefore,in this study we aimed to investigate the effect of esculetin on HSC activation and senescence.Methods Primary rat HSC were used in all experiments.Real-time cell analyzer and BrdU incorporation assay were used to determine HSC proliferation.Gene expression of the senescence-associated genes Cdkn1a(p21),P53,activation markers Acta2,Col1a1 and quiescence markers Pparg and Lrat were measured by RT-qPCR.Senescence associatedβ-Galactosidase(SA-β-Gal)staining was used to identify senescent HSC.Results Our results demonstrate that esculetin increased markers of senescence and decreased proliferation and activation markers in HSC.Protein expression of P21Cip1,accompanied by phosphorylation of Ser473 Akt and Ser9 GSK3βwas increased by esculetin.The effect of esculetin was dependent on PI3K-Akt signaling.Conclusion We conclude that esculetin induces HSC senescence and reverses the activated phenotype of HSC.The induction of senescence depends on PI3K-Akt-GSK3βsignaling.Esculetin could be a potential therapeutic drug for the treatment of hepatic fibrosis by inducing stellate cell senescence. 展开更多

Understanding the structure-property relationships in polycyclic conjugated hydrocarbons(PCHs)is crucial in controlling their electronic properties and developing new optically functional materials.Aromaticity is a fundamentally important and intriguing property of numerous organic chemical structures and has stimulated a myriad of experimental and theoretical investigations.Exploiting aromaticity rules for the rational design of optoelectronic materials with the desired photophysical characteristics is a challenging yet fascinating task.Herein we present an in-depth computational and spectroscopic study on the structure-property relationships of dinaphthopentalenes(DNPs).Results highlight that the different fusion patterns between 4nπand 4n+2πunits endow these PCHs with the tunable aromaticity in the ground state/excited state,which leads to the diverse electronic structures and consequently the distinctive excited state photophysics.Accordingly,we propose a combined aromaticity design strategy for rationally modulating and tailoring electronic and optical properties of PCH skeletons.These outcomes not only present a full picture of the excited state dynamics of the DNP system and afford a new class of efficient singlet fission-active materials but also provide some basic guidelines for exploiting aromaticity rules to design and develop new optical function materials. 展开更多

The practical efficiency of singlet fission(SF)-based photovoltaic devices is still far from satisfactory due to the limited scope of SF materials suitable for device application and the scarcity of schemes available for triplet utilization.Most SF materials identi-fied to date are typically electron donors while acceptor-type SF materials remain largely unexplored.Basically,the combination of a conventional electron donor and SF-active electron acceptor could circumvent the competitive energy transfer channel and better play the unique advantages of the SF process,which might be an adequate alternative for practical application.In this work,we presented a new acceptor-type SF material based on a tetracyanothienoquinoid skeleton.Such a quinoid skeleton exhibited strong absorption,ultrafast SF process,and excellent stability.Using transient spectroscopy and multireference calculations(XDWCASPT2),the SF dynamics were examined featuring the rapid generation and subsequent annihilation and/or partial dissociation of multiexciton states.Therefore,our results not only provide a robust acceptor-type SF material but also suggest an adequate donor–acceptor alternative for SF-based solar cells,which could pave the way for the practical application of such a potential process. 展开更多
