Diabetic retinopathy,a prevalent and vision-threatening microvascular complication of diabetes mellitus,is the leading cause of blindness among middle-aged and elderly individuals.Natural diterpenoids isolated from Siegesbeckia pubescens demonstrate potent anti-inflammatory properties.This study aimed to identify novel bioactive diterpenoids from S.pubescens and investigate their effects on oxidative stress and inflammatory responses in diabetic retinopathy,both in vitro and in vivo.Three new ent-pimarane-type diterpenoids(1–3)and six known compounds(4–9)were isolated from the aerial parts of S.pubescens.Their structures were elucidated through spectroscopic data interpretation,and absolute configurations were determined by comparing calculated and experimental electronic circular dichroism(ECD)spectra.Among these compounds,14β,16-epoxy-ent-3β,15α,19-trihydroxypimar-7-ene(5)exhibited the most potent protective effect against high glucose and interleukin-1β(IL-1β)-stimulated human retinal endothelial cells.Mechanistically,compound 5 promoted endothelial cell survival while ameliorating oxidative stress and inflammatory response in diabetic retinopathy,both in vivo and in vitro.These findings not only suggest that diterpenoids such as compound 5 are important anti-inflammatory constituents in S.pubescens,but also indicate that compound 5 may serve as a lead compound for preventing or treating vascular complications associated with diabetic retinopathy. 展开更多

Epstein-Barr virus(EBV)is very common,with the infection rate in adults over 90%worldwide.Infectious mononucleosis(IM)is caused by primary infection with EBV.Most IM patients are generally considered to have a favorable prognosis,but a few patients will also develop complications.Children with severe symptoms will require hospitalization.However,the disease burden of children hospitalized with IM in China has been rarely described.In this study,we included the Face sheets of discharge medical records from 27 member children's hospitals of Futang Research Center of Pediatric Development from Jan 1st,2016 to Dec 31st,2020,and medical information such as gender,age,region,time of admission,length of stay and expenditure were extracted.There were 24,120 IM cases,which accounted for 0.42%(24,120/5,693,262)of all hospitalized cases during this period.The ratio of male to female was 1.48:1.Hospitalization for IM in the 4-6 years age group was the highest among inpatients of all age groups.Case numbers increased year by year between 2016 and 2020,and the monthly hospitalization was generally high from Jul to Sep but reduced from Jan to Feb per year.Bronchitis/pneumonia and hepatic dysfunction were two common complications in hospitalized IM patients.The median length of stay was 8 days,and the median cost of hospitalization was 970.59 US dollars.This study will help understand the epidemiological characteristics and disease burden of hospitalized children with IM in China. 展开更多

Quantum dots(QDs)are promising candidates for the next-generation optical and electronic devices due to the outstanding photoluminance efficiency,tunable bandgap and facile solution synthesis.Nevertheless,the limited optoelectronic performance and poor lifetime of QDs devices hinder their further applications.As a gas-phase surface treatment method,atomic layer deposition(ALD)has shown the potential in QDs surface modification and device construction owing to the atomic-level control and excellent uniformity/conformality.In this perspective,the attempts to utilize ALD techniques in QDs modification to improve the photoluminance efficiency,stability,carrier mobility,as well as interfacial carrier utilization are introduced.ALD proves to be successful in the photoluminance quantum yield(PLQY)enhancement due to the elimination of QDs surface dangling bonds and defects.The QDs stability and devices lifetime are improved greatly through the introduction of ALD barrier layers.Furthermore,the carrier transport is ameliorated efficiently by infilling interstitial spaces during ALD process.Attributed to the ultra-thin and dense coating on the interface,the improvement on optoelectronic performance is achieved.Finally,the challenges of ALD applications in QDs at present and several prospects including ALD process optimization,in-situ characterization and computational simulations are proposed. 展开更多

Fluorescence(FL)active 8-aryl guanosine derivatives were prepared and applied for cation mediated self-assembly to form the H-bonded G8-quadruplexes.The p-cyano(p-CN)and 8-anthracene(8-An)substituted guanosines were identified to give the strongest fluorescence with the formation of G8-octamers(G8)both in solution(NMR)and solid state(X-ray).This well-defined G8-octamer system has provided the first direct evidence on the self-assembled G-quadruplex fluorescence emission with aggregation-induced emission(AIE),which could be applied as the foundation for FL molecular probe design toward G-quadruplex recognition. 展开更多

Introduction:Infectious mononucleosis(IM)and Kawasaki disease(KD)can present with overlapping clinical features,occasionally complicating the differential diagnosis.Given the significant differences in their management,accurate distinction is critical.It is exceptionally rare for a single patient to simultaneously suffer from both IM and KD in clinical practice.Case presentation:We report two cases of children over five years of age who initially presented with clinical manifestations suggestive of IM and serological evidence consistent with primary Epstein-Barr virus(EBV)infection,supporting an initial diagnosis of IM.Subsequently,both patients developed clinical features meeting the diagnostic criteria for KD.The final diagnosis was co-occurrence of IM and KD.Conclusion:We describe two cases of concurrent IM and KD,offering further insight into a possible association between primary EBV infection and KD.Clinicians should be mindful of the potential co-occurrence of these disorders,strive to avoid conflicts in treatment strategies,and take proactive measures to prevent complications. 展开更多

Background Epilepsy is a common neurological disorder.The homologous to E6-AP carboxy terminus(HECT)E3 ligase is associated with epilepsy.NEDD4-like ubiquitin protein ligase-1(NEDL1)is a HECT E3 ligase that is highly expressed in the brain.This study aimed to investigate the involvement of NEDL1 in epilepsy and the potential effect of NEDL1 on the cognitive ability.Methods The pilocarpine-induced epileptic mouse model was used to assess cognitive functions in Barnes maze,the pathological changes,and the activation of astrocytes and microglia in wild-type(Nedl1+/+)and Nedl1 knockout(Nedl1−/−)mice.The RNA-seq method was used to analyze differentially expressed genes and explore the brain pathophysiology after epilepsy development.Results Nedl1 knockout resulted in a protective effect against epilepsy.The Nedl1−/−mice showed improved spatial learning and memory,alleviation of pathological damage in the hippocampus induced by epilepsy,and reduced microglial activation in the hippocampus.Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis of differentially expressed genes also revealed several prominently enriched T-cell-related pathways.Conclusions Nedl1 knockout reduces seizures and alleviates neuroinflammation.The potential functional link between NEDL1 and epilepsy provides a new approach to the treatment and intervention of epilepsy. 展开更多
