Changes in border-associated macrophages after stroke: Single-cell sequencing analysis 认领 引用 被引量:2

Border-associated macrophages are located at the interface between the brain and the periphery, including the perivascular spaces, choroid plexus, and meninges. Until recently, the functions of border-associated macrophages have been poorly understood and largely overlooked. However, a recent study reported that border-associated macrophages participate in stroke-induced inflammation, although many details and the underlying mechanisms remain unclear. In this study, we performed a comprehensive single-cell analysis of mouse border-associated macrophages using sequencing data obtained from the Gene Expression Omnibus(GEO) database(GSE174574 and GSE225948). Differentially expressed genes were identified, and enrichment analysis was performed to identify the transcription profile of border-associated macrophages. CellChat analysis was conducted to determine the cell communication network of border-associated macrophages. Transcription factors were predicted using the ‘pySCENIC' tool. We found that, in response to hypoxia, borderassociated macrophages underwent dynamic transcriptional changes and participated in the regulation of inflammatory-related pathways. Notably, the tumor necrosis factor pathway was activated by border-associated macrophages following ischemic stroke. The pySCENIC analysis indicated that the activity of signal transducer and activator of transcription 3(Stat3) was obviously upregulated in stroke, suggesting that Stat3 inhibition may be a promising strategy for treating border-associated macrophages-induced neuroinflammation. Finally, we constructed an animal model to investigate the effects of border-associated macrophages depletion following a stroke. Treatment with liposomes containing clodronate significantly reduced infarct volume in the animals and improved neurological scores compared with untreated animals. Taken together, our results demonstrate comprehensive changes in border-associated macrophages following a stroke, providing a theoretical basis for targeting border-associated macrophages-induced neuroinflammation in stroke treatment. 展开更多

目的:揭示穴位埋线(AE)以P物质(SP)为刺激信号抑制鼻黏膜内质网应激(ERS),修复鼻腔黏膜屏障,纠正局部变应性鼻炎(LAR)的神经免疫功能失衡。方法:SD大鼠随机分为空白对照组、LAR组、AE组、假埋线(SAE)组、辣椒素组、AE+衣霉素组。以屋尘... 目的:揭示穴位埋线(AE)以P物质(SP)为刺激信号抑制鼻黏膜内质网应激(ERS),修复鼻腔黏膜屏障,纠正局部变应性鼻炎(LAR)的神经免疫功能失衡。方法:SD大鼠随机分为空白对照组、LAR组、AE组、假埋线(SAE)组、辣椒素组、AE+衣霉素组。以屋尘螨建立LAR模型,分别予以迎香穴AE、SAE、辣椒素滴鼻以及衣霉素共处理。取鼻黏膜进行检测:采用免疫荧光及RT-qPCR检测SP;采用免疫荧光及Western Blot检测BIP、CHOP、ZO-1、Claudin-1及Occludin;PERK-eIF2α-ATF4、IRE1α-XBP1、ATF6表达水平分析。结果:RT-qPCR示AE可以抑制LAR大鼠鼻腔SP mRNA表达(P<0.05);Western Blot示AE可以抑制ERS标记蛋白CHOP表达(P<0.05),促进ZO-1回调(P<0.05),修复黏膜屏障。此过程可能与p-eIF2α/eIF2α、ATF4以及XBP1调控有关。结论:基于“SP-ERS-黏膜屏障功能”的连续性调节可能是AE改善LAR效用路径之一。 展开更多

目的:探讨壮骨胶囊(ZGC)调控Ras蛋白(RAS)/Raf蛋白激酶(RAF)丝裂原激活蛋白激酶(MEK)/细胞外信号调节激酶(ERK)通路对骨质疏松症(OP)大鼠骨代谢的影响。方法:将60只大鼠随机分为正常组、模型组、壮骨胶囊低剂量(ZGC-L)组(0.5 g/kg)、壮... 目的:探讨壮骨胶囊(ZGC)调控Ras蛋白(RAS)/Raf蛋白激酶(RAF)丝裂原激活蛋白激酶(MEK)/细胞外信号调节激酶(ERK)通路对骨质疏松症(OP)大鼠骨代谢的影响。方法:将60只大鼠随机分为正常组、模型组、壮骨胶囊低剂量(ZGC-L)组(0.5 g/kg)、壮骨胶囊高剂量(ZGC-H)组(2.0 g/kg)、ZGC-H(2.0 g/kg)+ML-099(11.7 mg/kg)组,每组12只。给药12周后测量各组大鼠股骨的骨密度(BMD);HE染色检测股骨组织病理变化;TUNEL染色检测股骨细胞凋亡;酶联免疫吸附试验(ELISA)检测血清中骨特异性碱性磷酸酶(BALP)、骨钙素(OC)、核心结合因子-α1(CBF-α1)、Ⅰ型原胶原N端前肽(PINP)水平,蛋白质印迹法(Western blotting)检测RAS/RAF/MEK/ERK信号通路相关蛋白的表达。结果:正常组大鼠骨小梁排列整齐、致密,间隙均匀,骨组织结构完整且形态规整;模型组大鼠骨小梁间隙较大,股骨组织稀疏,排列紊乱,股骨组织损伤明显;ZGC-L组、ZGC-H组大鼠骨小梁间距较窄,股骨组织相对紧密,形态结构规整,股骨组织损伤降低;ZGC-H+ML-099组大鼠股骨组织损伤程度高于ZGC-H组。模型组大鼠股骨细胞凋亡率、股骨RAS、RAF蛋白相对表达量及p-MEK1/2/MEK1/2、p-ERK1/2/ERK1/2高于正常组(P<0.05),股骨BMD及血清BALP、OC、CBF-α1、PINP水平低于正常组(P<0.05);ZGC-L组、ZGC-H组大鼠股骨细胞凋亡率、股骨RAS、RAF蛋白相对表达量及p-MEK1/2/MEK1/2、p-ERK1/2/ERK1/2低于模型组(P<0.05),股骨BMD及血清BALP、OC、CBF-α1、PINP水平高于模型组(P<0.05);ZGC-H+ML-099组大鼠股骨细胞凋亡率、股骨RAS、RAF蛋白相对表达量及p-MEK1/2/MEK1/2、p-ERK1/2/ERK1/2高于ZGC-H组,股骨BMD及血清BALP、OC、CBF-α1、PINP水平低于ZGC-H组(P<0.05)。结论:壮骨胶囊可能通过抑制RAS/RAF/MEK/ERK通路调节OP大鼠骨代谢。 展开更多

目的:通过HPLC法建立山楂、炒山楂及焦山楂的指纹图谱,建立5-羟甲基糠醛、绿原酸、金丝桃苷、异槲皮苷含量测定方法,探讨3种炮制品化学成分变化规律。方法:采用C18色谱柱,以乙腈-0.1%乙酸水为流动相,流速1.0 mL/min,检测波长325 nm,... 目的:通过HPLC法建立山楂、炒山楂及焦山楂的指纹图谱,建立5-羟甲基糠醛、绿原酸、金丝桃苷、异槲皮苷含量测定方法,探讨3种炮制品化学成分变化规律。方法:采用C18色谱柱,以乙腈-0.1%乙酸水为流动相,流速1.0 mL/min,检测波长325 nm,柱温30℃,进样量10μL;通过“中药色谱指纹图谱相似度评价系统”软件进行相似度分析;结合聚类分析(HCA)、主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)对山楂不同炮制品进行多模式识别研究。结果:指纹图谱共标定了14个峰作为共有峰,指认共有峰4个(5-羟甲基糠醛、绿原酸、金丝桃苷、异槲皮苷);山楂、炒山楂及焦山楂14批样品的相似度均在0.95以上,多模式识别可有效区分3种炮制品,对上述4个成分进行含量测定,且线性关系良好(r>0.992),平均加样回收率为98.61%~101.61%,相对标准偏差(RSD)为1.44%~2.33%。结论:建立的HPLC指纹图谱及多组分含量测定方法,能客观、真实反映山楂、炒山楂及焦山楂质量,为山楂及其炮制品的质量控制及临床应用提供科学依据。 展开更多

Non-precious metal cobalt-based oxide inevitably dissolves for acid oxygen evolution reaction(OER).Designing an efficient deposition channel for leaching cobalt species is a promising approach.The dissolution-deposition equilibrium of Co is achieved by doping Mn in the lattice of LaCo1-xMnxO3,prolonging the lifespan in acidic conditions by 14 times.The lattice doping of Mn produces a strain that enhances the adsorption capacity of OH-.The self-catalysis of Mn causes the leaching Co to be deposited in the form of CoO2,which ensures that the long-term stability of LaCo1-xMnxO3is 70 h instead of 5 h for LaCoO3.Mn doping enhances the deprotonation of*OOH→O2in acidic environments.Notably,the over-potential of optimized LaCo1-xMnxO3is 345 mV at 10 mA cm-2for acidic OER.This work presents a promising method for developing noble metal-free catalysts that enhance the acidic OER activity and stability. 展开更多

Interbody fusion is recognized as the golden standard of surgical intervention for degenerative disc disease(DDD).Interbody fusion cage made of polyetheretherketone(PEEK)is commonly used in lumbar interbody fusion surgery in the treatment of DDD worldwide.However,there are some limitations of PEEK including their bio-inert nature and impediment to host bone integration.This study aimed to evaluate the degradation profile and osteoinductive potential of biodegradable Mg-Zn-Nd-Zr cages with/without micro-arc oxidation(MAO)coatings.The Mg-Zn-Nd-Zr alloy cages,whether coated with MAO or not,demonstrated commendable biocompatibility and biomechanical properties.Immersion and electrochemical tests show better corrosion resistance of MAO coatings in vitro.mRNA sequencing,RT-qPCR and Western blotting revealed that Mg-Zn-Nd-Zr and Mg-Zn-Nd-Zr/MAO had a better effectiveness on osteoinductivity.In vivo evaluations in ovine models over 12 weeks and 24 weeks post-implantation revealed radiological and histological evidence of enhanced bone formation adjacent to the Mg-Zn-Nd-Zr alloy cages compared to PEEK counterparts.Moreover,the MAO-coated cages exhibited a reduced propensity for gas formation.The Mg-Zn-Nd-Zr alloy is as a superior osteoinductive material compared with PEEK,with the MAO coating offering an advantage in mitigating gas production.Nonetheless,further research is warranted to refine the alloy's composition or surface treatments,particularly to address the challenges associated with rapid gas evolution during the early post-implantation period. 展开更多

In this study,the chemical freeze-out of hadrons,including light-and strange-flavor particles and light nuclei,produced in Au+Au collisions at the Relativistic Heavy Ion Collider(RHIC),was investigated.Using the Thermal-FIST thermodynamic statistical model,we analyzed various particle sets:those inclusive of light nuclei,those exclusive to light nuclei,and those solely comprising light nuclei.We determined the chemical freeze-out parameters at√SNN=7.7–200 Ge V and four different centralities.A significant finding was the decrease in the chemical freeze-out temperature Tchwith light-nuclei inclusion,with an even more pronounced reduction when considering light-nuclei yields exclusively.This suggests that light-nuclei formation occurs at a later stage in the system’s evolution at RHIC energies.We present parameterized formulas that describe the energy dependence of Tchand the baryon chemical potentialμB for three distinct particle sets in central Au+Au collisions at RHIC energies.Our results reveal at least three distinct Tchat RHIC energies correspond to different freeze-out hypersurfaces:a light-flavor freeze-out temperature of T_L=150.2±6 Me V,a strange-flavor freeze-out temperature T_s=165.1±2.7 Me V,and a light-nuclei freeze-out temperature Tln=141.7±1.4 Me V.Notably,at the Large Hadron Collider(LHC)Pb+Pb 2.76Te V,the expected lower freeze-out temperature for light nuclei was not observed;instead,the Tchfor light nuclei was found to be approximately 10 Me V higher than that for light-flavor hadrons. 展开更多

Sesame is one of the eight major allergens that cause food allergies.Study of the epitopes of sesame allergens is important for understanding their sensitization mechanisms.Currently,less information is available on the epitope studies of sesame allergens.In this study,we analyzed the molecular characteristics,structure and homology of Ses i 3,one of the important sesame allergens.We predicted the B-cell linear epitopes of Ses i 3 using bioinformatics tools and characterized them by slot blot immuno-microarrays technology.Eight peptides as B-cell linear epitopes of Ses i 3 were identified,in addition,key amino acids in these epitopes were predicted and leucine 422 was identified as a key amino acid.The present work will contribute to further understanding of the sesame allergen and provide some help in the prevention and treatment of sesame allergy. 展开更多

酵母交配型转换/蔗糖不发酵(SWI/SNF)相关基质关联肌动蛋白依赖性染色质亚家族B成员1(SMARCB1)/整合酶相互作用分子1(INI1)(SMARCB1/INI1)缺失型未分化胰腺癌(UPC)是一种极为罕见的胰腺癌特殊类型。本文作者报道1例SMARCB1/INI1缺失型UP... 酵母交配型转换/蔗糖不发酵(SWI/SNF)相关基质关联肌动蛋白依赖性染色质亚家族B成员1(SMARCB1)/整合酶相互作用分子1(INI1)(SMARCB1/INI1)缺失型未分化胰腺癌(UPC)是一种极为罕见的胰腺癌特殊类型。本文作者报道1例SMARCB1/INI1缺失型UPC患者的临床表现、辅助检查和诊治经过,并进行相关文献复习。患者,女性,65岁,因间断性上腹痛1个月余,加重伴恶心、呕吐9 d入院。曾于外院行全腹部CT提示胰腺占位性病变。入院查体显示上腹部压痛,无反跳痛或肌紧张。实验室检查,糖类抗原199(CA199)水平为50.58 U·mL-1,空腹血糖为9.98 mmol·L-1。腹部MRI显示胰腺体尾部见囊实混合异常信号影,呈不规则向胰腺外隆起;T1加权成像(T1WI)呈稍低信号,瘤体出血部分呈高信号;T2加权成像(T2WI)呈高信号,边界不清,大小为2.0~5.6 cm,致胃壁受压、受侵;增强扫描示肿瘤囊壁呈明显环形强化,脾动脉和静脉受侵,门静脉栓子形成。胃镜见胃体大弯侧有4.0 cm×5.0 cm的黏膜隆起,考虑胰腺占位压迫所致。临床诊断为胰腺占位性病变,恶性肿瘤可能性大,手术切除为首选治疗方案。术后病理诊断为SMARCB1/INI1缺失型UPC,伴少量中分化鳞状细胞癌成分。患者术后接受化疗,已随访5个月,生活质量较术前改善,无明显不适,仍在密切随访中。SMARCB1/INI1缺失型UPC较为罕见,临床表现缺乏特异性,通常进展迅速且预后不良,临床中应争取尽早诊治。 展开更多

糖尿病动脉粥样硬化(DM-AS)是糖尿病大血管病变的病理基础,也是糖尿病患者发生心血管事件的主要病因。持续高血糖可损伤血管内皮并加剧脂质代谢紊乱与炎症反应,推动斑块由形成向易损演变,从而增加破裂与血栓风险,因此维持斑块稳定性对... 糖尿病动脉粥样硬化(DM-AS)是糖尿病大血管病变的病理基础,也是糖尿病患者发生心血管事件的主要病因。持续高血糖可损伤血管内皮并加剧脂质代谢紊乱与炎症反应,推动斑块由形成向易损演变,从而增加破裂与血栓风险,因此维持斑块稳定性对预防心血管事件具有重要意义。巨噬细胞泡沫化程度与巨噬细胞促炎型(M1)/抗炎修复型(M2)极化失衡直接影响斑块稳定性。自噬是维持巨噬细胞内稳态的关键通路,可通过调控脂质代谢与炎症相关信号、维持自噬流通量,影响斑块炎性负荷与坏死核心扩展,从而参与维持斑块稳定性。中医药具有多成分、多靶点、整体调节的优势,已有研究表明多种中药活性成分及复方对调控自噬网络、干预DM-AS斑块进展有明显优势。该文围绕巨噬细胞自噬,系统梳理腺苷酸活化蛋白激酶/哺乳动物雷帕霉素靶蛋白(AMPK/mTOR)、磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)、过氧化物酶体增殖物激活受体(PPAR)、晚期糖基化终产物受体/核转录因子-κB(RAGE/NF-κB)等关键通路靶向调控巨噬细胞自噬介导极化平衡在DM-AS不同阶段发挥作用的机制,对中药化合物、中药复方调控自噬进而抑制斑块演进的研究现状进行总结,以期为DM-AS机制研究及临床转化提供新的研究思路。 展开更多

目的:探讨督脉电针调控HMGB1/TLR4/NF-κB通路减轻脊髓损伤(SCI)后额叶皮层神经炎症并改善认知功能的作用机制。方法:36只雄性SD大鼠随机分为假手术组(Sham)、SCI模型组(SCI)和电针治疗组(EA),每组12只。采用Allen s打击法制备T 10节段... 目的:探讨督脉电针调控HMGB1/TLR4/NF-κB通路减轻脊髓损伤(SCI)后额叶皮层神经炎症并改善认知功能的作用机制。方法:36只雄性SD大鼠随机分为假手术组(Sham)、SCI模型组(SCI)和电针治疗组(EA),每组12只。采用Allen s打击法制备T 10节段SCI模型,EA组于术后第2天开始接受大椎穴(GV14)和命门穴(GV4)电针治疗,1次/d,连续28 d。通过BBB评分、斜板实验评估运动功能,采用Y迷宫和新物体识别实验评估认知功能。运用免疫荧光染色和Western blot技术检测额叶皮层HMGB1、TLR4、NF-κB p65及TNF-α蛋白表达及小胶质细胞活化情况。结果:与Sham组比较,SCI组BBB评分和斜板角度显著降低,差异具有统计学意义(P<0.01),Y迷宫交替率和新物体识别指数显著下降,差异具有统计学意义(P<0.01),额叶皮层HMGB1、TLR4、NF-κB p65及TNF-α蛋白表达显著增加,差异具有统计学意义(P<0.01),Iba-1阳性小胶质细胞数量显著增多,差异具有统计学意义(P<0.01)。EA治疗后,运动功能(BBB评分和斜板角度)和认知功能(Y迷宫交替率和新物体识别指数)均显著改善,差异具有统计学意义(P<0.01),同时HMGB1、TLR4、NF-κB p65及TNF-α蛋白表达显著降低,差异具有统计学意义(P<0.01),小胶质细胞活化明显减轻,差异具有统计学意义(P<0.05)。结论:督脉电针可同时改善SCI大鼠的运动以及认知功能,其机制可能在于跨节段抑制额叶皮层HMGB1/TLR4/NF-κB炎症通路激活,进而减轻小胶质细胞介导的神经炎症。 展开更多

目的:观察督脉电针对脊髓损伤(SCI)大鼠小胶质细胞M1型和M2型表型极化及炎症因子表达的影响,探讨其改善运动功能的作用机制。方法:将雄性SD大鼠随机分为空白组、假手术组、模型组和督脉电针组,每组12只,采用改良Allen's方法建立T_(9... 目的:观察督脉电针对脊髓损伤(SCI)大鼠小胶质细胞M1型和M2型表型极化及炎症因子表达的影响,探讨其改善运动功能的作用机制。方法:将雄性SD大鼠随机分为空白组、假手术组、模型组和督脉电针组,每组12只,采用改良Allen's方法建立T9~11节SCI模型。督脉电针组选用“大椎”(GV14)与“命门”(GV4)穴位介入。电针治疗1次/d,20 min/次,连续干预28 d。采用BBB评分评估大鼠后肢运动功能,HE染色观察脊髓组织形态结构变化,免疫荧光染色观察小胶质细胞标志物离子化钙结合适配蛋白1(Iba-1)与诱导型一氧化氮合酶(iNOS)或精氨酸酶-1(Arg-1)的共表达情况,统计Iba-1阳性细胞数量,并计算M1型和M2型小胶质细胞的比例,Western blot检测iNOS与Arg-1蛋白表达水平,ELISA检测脊髓组织中肿瘤坏死因子-α(TNF-α)及白细胞介素-10(IL-10)水平。结果:与空白组和假手术组比较,模型组术后各时点BBB评分显著降低,差异具有统计学意义(P<0.01),脊髓组织结构紊乱,炎性细胞浸润严重,M1型小胶质细胞比例显著升高,M2型比例显著下降,差异具有统计学意义(P<0.01),TNF-α水平显著升高,IL-10水平升高,差异具有统计学意义(P<0.01)。与模型组比较,督脉电针组自术后7 d起BBB评分显著升高,差异具有统计学意义(P<0.05),脊髓组织结构损伤和炎性细胞浸润程度减轻,M1型小胶质细胞比例显著降低,M2型比例显著升高,差异具有统计学意义(P<0.01),TNF-α水平显著降低,差异具有统计学意义(P<0.01),IL-10水平进一步升高,差异具有统计学意义(P<0.01)。结论:督脉电针能够抑制小胶质细胞向促炎性M1型极化,促进其向抗炎修复性M2型极化,进而调节炎症因子的表达,减轻SCI后的继发性炎症反应,这可能是其促进SCI大鼠运动功能恢复的潜在作用机制。 展开更多

基于冠豪猪优化算法-变分模态分解的原位红外光谱去噪方法 认领 引用

原位红外光谱信号常受到仪器热噪声、环境扰动、样品散射及光路波动等多源噪声的叠加干扰,传统去噪方法往往存在参数依赖性强、模态混叠、高频细节丢失或过度平滑等问题。针对这些问题,本研究提出一种基于冠豪猪优化算法(Crested Porcup... 原位红外光谱信号常受到仪器热噪声、环境扰动、样品散射及光路波动等多源噪声的叠加干扰,传统去噪方法往往存在参数依赖性强、模态混叠、高频细节丢失或过度平滑等问题。针对这些问题,本研究提出一种基于冠豪猪优化算法(Crested Porcupine Optimizer,CPO)-变分模态分解(Variational Mode Decomposition,VMD)的去噪方法。该方法引入CPO算法,以分解后模态分量的最小能量熵为适应度函数,实现对VMD参数模态数K与惩罚因子α的自动全局寻优,从而克服了传统VMD参数依赖经验设置的局限。优化后的VMD将光谱信号自适应分解为一系列具有不同中心频率的模态分量,其中代表随机噪声的高频模态被舍弃,相反,低频模态包含了基本化学信息和特征光谱,对它们求和以重构最终的去噪光谱。通过1个模拟原位红外光谱信号和2个实测的原位红外光谱信号来验证所提出方法的有效性,其中模拟信号通过三个叠加了高斯噪声的高斯峰构建,实测信号是镍铁层状双氢氧化物(NiFe Layered Double Hydroxide,NiFe LDH)催化剂和镀金硅晶体材料的傅里叶变换红外光谱。与S-G平滑、经验模态分解(Empirical Mode Decomposition,EMD)、离散小波变换(Discrete Wavelet Transform,DWT)、VMD等传统去噪方法对比,结果表明:对于模拟信号,CPO-VMD方法取得了24.12的最高信噪比,其重构信号与真实信号的相关系数达0.9948,均优于其他方法;对于NiFe LDH催化剂光谱,该方法在去除噪声的同时能保留2400~2300 cm-1的峰值信息,而其他方法在此区域要么引入了畸变,要么残留了噪声;对于镀金硅晶体光谱,该方法在1200~1050 cm-1主峰区域能有效保留峰形与强度,而其他技术则存在过度平滑或噪声去除不足的问题。这表明了该方法在有效抑制噪声的同时,能更完整地保留特征峰的峰位、峰形与强度信息,提升了光谱质量与分析可靠性。本研究为原位红外光谱提供了一种高效、稳健的去噪方案,也为VMD参数优化问题提供了新思路,具有重要的方法论意义与应用价值。 展开更多

结直肠癌患者术后切口感染病原菌分布及危险因素 认领 引用

目的探讨结直肠癌患者术后切口感染(incisional surgical site infection,I-SSI)的病原菌分布情况及危险因素。方法筛选并收集52例结直肠癌术后I-SSI患者(感染组)的临床资料,选取同期60例结直肠癌术后未发生感染的患者为对照组。采用Log... 目的探讨结直肠癌患者术后切口感染(incisional surgical site infection,I-SSI)的病原菌分布情况及危险因素。方法筛选并收集52例结直肠癌术后I-SSI患者(感染组)的临床资料,选取同期60例结直肠癌术后未发生感染的患者为对照组。采用Logistic回归分析术后I-SSI的危险因素,分析感染组(n=52)病原菌分布情况及主要病原菌的耐药率。结果感染组(n=52)共分离出73株病原菌,其中,革兰阴性菌43株(58.9%,大肠埃希菌30株),革兰阳性菌27株(37.0%),真菌3株(4.1%);单因素分析显示,术后I-SSI与年龄≥60岁、BMI≥25 kg/m2、合并糖尿病、开腹手术、肿瘤部位、结肠梗阻、预防性造口有关(P<0.05);多因素分析结果显示,合并糖尿病、结肠梗阻、预防性造口为结直肠癌术后I-SSI的独立危险因素(P<0.05)。结论结直肠癌术后切口感染的主要病原菌为革兰阴性菌,其中,以大肠埃希菌多见。大肠埃希菌对左氧氟沙星、环丙沙星、氨苄西林的耐药率最高,对美罗培南的耐药率最低。合并糖尿病、结肠梗阻、预防性造口为结直肠癌术后I-SSI的独立危险因素。 展开更多

目的探讨多发性腔隙性脑梗死(multi-lacunar infarction,MLI)患者继发血管性帕金森综合征(vascular Parkinsonism,VP)的相关危险因素,评估血液学指标与神经影像学特征在VP发生风险预测中的应用价值。方法纳入2021年5月至2024年12月吉林... 目的探讨多发性腔隙性脑梗死(multi-lacunar infarction,MLI)患者继发血管性帕金森综合征(vascular Parkinsonism,VP)的相关危险因素,评估血液学指标与神经影像学特征在VP发生风险预测中的应用价值。方法纳入2021年5月至2024年12月吉林省敦化市医院收治的MLI患者169例,根据是否继发VP分为MLI组和多发性腔隙性脑梗死后帕金森综合征(vascular Parkinsonism after multi-lacunar infarction,MLI-VP)组。采用随机分配方法,将患者按1∶1分为模型构建集(n=84)和内部评估集(n=85)。比较两组患者的临床资料及影像学特征,筛选MLI患者发生VP的危险因素,进而构建风险预测模型,并对其性能和临床应用价值进行评价。结果中性粒细胞计数、高密度脂蛋白胆固醇(high density lipoprotein-cholesterol,HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein-cholesterol,LDL-C)、基底节受累与MLI-VP的发生显著相关(P0.05),决策曲线提示模型具有一定的临床获益。结论中性粒细胞计数、HDL-C、LDL-C和基底节受累与MLI-VP密切相关,是独立危险因素。基于血液学指标和影像学特征的预测模型在本研究患者中表现出较好的判别能力,可用于早期高风险患者,为实施个体化精准干预提供依据。 展开更多

To solve the problem of the lack of reference material(RM)for determination of allergenic ingredients in food,a RM of cashew nut powder was developed in the study.Cashew nut powder was prepared from cashew nut kernel by selecting,cleaning,crushing,n-hexane degreasing and sieving treatment.The reliability and traceability of RM was verified using real-time quantitative polymerase chain reaction(qPCR)and phylogenetic tree analysis.The cashew nut powder RM showed good homogeneity,and good stability under long-term storage at 4℃and short-term simulated transportation from-20 to 45℃.The RM was determined jointly by 8 collaborative laboratories,and the characteristic CT value was 24.732,and the extended uncertainty was 1.052%(k=2).The RM was applied to verify the amplification efficiency and the limit of detection for qPCR assay,and showed good applicability.The RM could be used for method validation and quality control,for the determination of allergenic ingredients in food mixed with trace amounts of cashew nut. 展开更多

Transfer-based Adversarial Attacks(TAAs)can deceive a victim model even without prior knowledge.This is achieved by leveraging the property of adversarial examples.That is,when generated from a surrogate model,they retain their features if applied to other models due to their good transferability.However,adversarial examples often exhibit overfitting,as they are tailored to exploit the particular architecture and feature representation of source models.Consequently,when attempting black-box transfer attacks on different target models,their effectiveness is decreased.To solve this problem,this study proposes an approach based on a Regularized Constrained Feature Layer(RCFL).The proposed method first uses regularization constraints to attenuate the initial examples of low-frequency components.Perturbations are then added to a pre-specified layer of the source model using the back-propagation technique,in order to modify the original adversarial examples.Afterward,a regularized loss function is used to enhance the black-box transferability between different target models.The proposed method is finally tested on the ImageNet,CIFAR-100,and Stanford Car datasets with various target models,The obtained results demonstrate that it achieves a significantly higher transfer-based adversarial attack success rate compared with baseline techniques. 展开更多

Objective:This study aims to establish an economically viable and easily accessible adult animal model for optogenetic activation of auditory neurons using adeno-associated viruses(AAVs)carrying Ch R2(H134R)to explore the potential of cochlear optogenetics as a hearing restoration technology.Methods:Healthy adult guinea pigs were used in the experiments.The viral vector AAV2/8-Ch R2(H134R)-h Syn-e YFP was administered to the right cochlea via the round window membrane.The confocal microscopy and reverse transcription polymerase chain reaction(RT-PCR)were utilized to analyze the Ch R2(H134R)expression localized to spiral ganglion neurons(SGNs).The auditory pathway activation was assessed by recording the optical compound action potential(oCAP)and acoustic compound action potential(a CAP)at various laser intensities.Results:The Ch R2(H134R)-e YFP expression was confirmed in 90%of the tested animals,localized to the SGNs of the injected ear.Higher m RNA levels of Ch R2(H134R)and e YFP were observed in the injected ear compared to the non-injected ear,while actin(Actb)m RNA levels were not significantly different.The o CAP was successfully elicited by a 470 nm blue light laser stimulus,with similar amplitudes and latency periods to those of a CAPs when the o CAP was evoked by 5.80 m W blue light and the a CAP was evoked by a 40 d B SPL click.The amplitudes of o CAPs increased with increasing laser intensity.Conclusion:This study demonstrates the viability of optogenetic activation of the auditory system in adult guinea pigs through the transduction of AAV-Ch R2(H134R)in SGNs.Cochlear optogenetics demonstrates potential as a hearing restoration technology,providing a basis for further clinical research and opening new avenues for investigation. 展开更多

【目的】为了提高水库底泥的高值化利用率,拟使用水库底泥来制备泡沫混凝土,以水胶比、底泥掺量、泡沫掺量为变量【方法】采用Design-Expert 13.0软件中响应曲面法对水库底泥泡沫混凝土的配合比进行优化设计,分析变量对水库底泥泡沫混... 【目的】为了提高水库底泥的高值化利用率,拟使用水库底泥来制备泡沫混凝土,以水胶比、底泥掺量、泡沫掺量为变量【方法】采用Design-Expert 13.0软件中响应曲面法对水库底泥泡沫混凝土的配合比进行优化设计,分析变量对水库底泥泡沫混凝土抗压强度和导热系数的影响。采用SEM和XRD探究了底泥泡沫混凝土微观结构。【结果】研究结果显示,对水库底泥泡沫混凝土28 d抗压强度影响顺序为:水胶比>泡沫掺量>底泥掺量,而导热系数的影响顺序为:水胶比>底泥掺量>泡沫掺量。随着水胶比增加,抗压强度先升高后下降,导热系数逐渐降低。增加泡沫和底泥掺量会导致抗压强度和导热系数均下降。孔隙率与导热系数呈现负相关,符合指数模型。【结论】经优化配比,水库底泥泡沫混凝土在水胶比0.4、底泥掺量30%、泡沫掺量4.2%条件下制备,其28 d抗压强度为18.19 MPa,导热系数为0.1214W/(m·K),表观密度为701.2 kg/m3。微观结果分析表明,底泥掺入量为30%条件下,试块内部物相结合紧密,孔隙分布均匀。研究成果为水库底泥在建筑材料领域的利用提供了新途径。 展开更多
