Until recently, the functions of border-associated macrophages have been poorly understood and largely overlooked. However, a recent study reported that border-associated macrophages participate in stroke-induced inflammation, although many details and the underlying mechanisms remain unclear. In this study, we performed a comprehensive single-cell analysis of mouse border-associated macrophages using sequencing data obtained from the Gene Expression Omnibus(GEO) database(GSE174574 and GSE225948). Differentially expressed genes were identified, and enrichment analysis was performed to identify the transcription profile of border-associated macrophages. CellChat analysis was conducted to determine the cell communication network of border-associated macrophages. Transcription factors were predicted using the ‘pySCENIC' tool. We found that, in response to hypoxia, borderassociated macrophages underwent dynamic transcriptional changes and participated in the regulation of inflammatory-related pathways. Notably, the tumor necrosis factor pathway was activated by border-associated macrophages following ischemic stroke. The pySCENIC analysis indicated that the activity of signal transducer and activator of transcription 3(Stat3) was obviously upregulated in stroke, suggesting that Stat3 inhibition may be a promising strategy for treating border-associated macrophages-induced neuroinflammation. Finally, we constructed an animal model to investigate the effects of border-associated macrophages depletion following a stroke. Treatment with liposomes containing clodronate significantly reduced infarct volume in the animals and improved neurological scores compared with untreated animals. Taken together, our results demonstrate comprehensive changes in border-associated macrophages following a stroke, providing a theoretical basis for targeting border-associated macrophages-induced neuroinflammation in stroke treatment.展开更多

Interbody fusion is recognized as the golden standard of surgical intervention for degenerative disc disease(DDD).Interbody fusion cage made of polyetheretherketone(PEEK)is commonly used in lumbar interbody fusion sur...Interbody fusion is recognized as the golden standard of surgical intervention for degenerative disc disease(DDD).Interbody fusion cage made of polyetheretherketone(PEEK)is commonly used in lumbar interbody fusion surgery in the treatment of DDD worldwide.However,there are some limitations of PEEK including their bio-inert nature and impediment to host bone integration.This study aimed to evaluate the degradation profile and osteoinductive potential of biodegradable Mg-Zn-Nd-Zr cages with/without micro-arc oxidation(MAO)coatings.The Mg-Zn-Nd-Zr alloy cages,whether coated with MAO or not,demonstrated commendable biocompatibility and biomechanical properties.Immersion and electrochemical tests show better corrosion resistance of MAO coatings in vitro.mRNA sequencing,RT-qPCR and Western blotting revealed that Mg-Zn-Nd-Zr and Mg-Zn-Nd-Zr/MAO had a better effectiveness on osteoinductivity.In vivo evaluations in ovine models over 12 weeks and 24 weeks post-implantation revealed radiological and histological evidence of enhanced bone formation adjacent to the Mg-Zn-Nd-Zr alloy cages compared to PEEK counterparts.Moreover,the MAO-coated cages exhibited a reduced propensity for gas formation.The Mg-Zn-Nd-Zr alloy is as a superior osteoinductive material compared with PEEK,with the MAO coating offering an advantage in mitigating gas production.Nonetheless,further research is warranted to refine the alloy's composition or surface treatments,particularly to address the challenges associated with rapid gas evolution during the early post-implantation period.展开更多
In this study,the chemical freeze-out of hadrons,including light-and strange-flavor particles and light nuclei,produced in Au+Au collisions at the Relativistic Heavy Ion Collider(RHIC),was investigated.Using the Therm...In this study,the chemical freeze-out of hadrons,including light-and strange-flavor particles and light nuclei,produced in Au+Au collisions at the Relativistic Heavy Ion Collider(RHIC),was investigated.Using the Thermal-FIST thermodynamic statistical model,we analyzed various particle sets:those inclusive of light nuclei,those exclusive to light nuclei,and those solely comprising light nuclei.We determined the chemical freeze-out parameters at√SNN=7.7–200 Ge V and four different centralities.A significant finding was the decrease in the chemical freeze-out temperature Tchwith light-nuclei inclusion,with an even more pronounced reduction when considering light-nuclei yields exclusively.This suggests that light-nuclei formation occurs at a later stage in the system’s evolution at RHIC energies.We present parameterized formulas that describe the energy dependence of Tchand the baryon chemical potentialμB for three distinct particle sets in central Au+Au collisions at RHIC energies.Our results reveal at least three distinct Tchat RHIC energies correspond to different freeze-out hypersurfaces:a light-flavor freeze-out temperature of T_L=150.2±6 Me V,a strange-flavor freeze-out temperature T_s=165.1±2.7 Me V,and a light-nuclei freeze-out temperature Tln=141.7±1.4 Me V.Notably,at the Large Hadron Collider(LHC)Pb+Pb 2.76Te V,the expected lower freeze-out temperature for light nuclei was not observed;instead,the Tchfor light nuclei was found to be approximately 10 Me V higher than that for light-flavor hadrons.展开更多

目的探讨结直肠癌患者术后切口感染(incisional surgical site infection,I-SSI)的病原菌分布情况及危险因素。方法筛选并收集52例结直肠癌术后I-SSI患者(感染组)的临床资料,选取同期60例结直肠癌术后未发生感染的患者为对照组。采用Log...目的探讨结直肠癌患者术后切口感染(incisional surgical site infection,I-SSI)的病原菌分布情况及危险因素。方法筛选并收集52例结直肠癌术后I-SSI患者(感染组)的临床资料,选取同期60例结直肠癌术后未发生感染的患者为对照组。采用Logistic回归分析术后I-SSI的危险因素,分析感染组(n=52)病原菌分布情况及主要病原菌的耐药率。结果感染组(n=52)共分离出73株病原菌,其中,革兰阴性菌43株(58.9%,大肠埃希菌30株),革兰阳性菌27株(37.0%),真菌3株(4.1%);单因素分析显示,术后I-SSI与年龄≥60岁、BMI≥25 kg/m2、合并糖尿病、开腹手术、肿瘤部位、结肠梗阻、预防性造口有关(P<0.05);多因素分析结果显示,合并糖尿病、结肠梗阻、预防性造口为结直肠癌术后I-SSI的独立危险因素(P<0.05)。结论结直肠癌术后切口感染的主要病原菌为革兰阴性菌,其中,以大肠埃希菌多见。大肠埃希菌对左氧氟沙星、环丙沙星、氨苄西林的耐药率最高,对美罗培南的耐药率最低。合并糖尿病、结肠梗阻、预防性造口为结直肠癌术后I-SSI的独立危险因素。展开更多

To solve the problem of the lack of reference material(RM)for determination of allergenic ingredients in food,a RM of cashew nut powder was developed in the study.Cashew nut powder was prepared from cashew nut kernel ...To solve the problem of the lack of reference material(RM)for determination of allergenic ingredients in food,a RM of cashew nut powder was developed in the study.Cashew nut powder was prepared from cashew nut kernel by selecting,cleaning,crushing,n-hexane degreasing and sieving treatment.The reliability and traceability of RM was verified using real-time quantitative polymerase chain reaction(qPCR)and phylogenetic tree analysis.The cashew nut powder RM showed good homogeneity,and good stability under long-term storage at 4℃and short-term simulated transportation from-20 to 45℃.The RM was determined jointly by 8 collaborative laboratories,and the characteristic CT value was 24.732,and the extended uncertainty was 1.052%(k=2).The RM was applied to verify the amplification efficiency and the limit of detection for qPCR assay,and showed good applicability.The RM could be used for method validation and quality control,for the determination of allergenic ingredients in food mixed with trace amounts of cashew nut.展开更多
Transfer-based Adversarial Attacks(TAAs)can deceive a victim model even without prior knowledge.This is achieved by leveraging the property of adversarial examples.That is,when generated from a surrogate model,they re...Transfer-based Adversarial Attacks(TAAs)can deceive a victim model even without prior knowledge.This is achieved by leveraging the property of adversarial examples.That is,when generated from a surrogate model,they retain their features if applied to other models due to their good transferability.However,adversarial examples often exhibit overfitting,as they are tailored to exploit the particular architecture and feature representation of source models.Consequently,when attempting black-box transfer attacks on different target models,their effectiveness is decreased.To solve this problem,this study proposes an approach based on a Regularized Constrained Feature Layer(RCFL).The proposed method first uses regularization constraints to attenuate the initial examples of low-frequency components.Perturbations are then added to a pre-specified layer of the source model using the back-propagation technique,in order to modify the original adversarial examples.Afterward,a regularized loss function is used to enhance the black-box transferability between different target models.The proposed method is finally tested on the ImageNet,CIFAR-100,and Stanford Car datasets with various target models,The obtained results demonstrate that it achieves a significantly higher transfer-based adversarial attack success rate compared with baseline techniques.展开更多
Objective:This study aims to establish an economically viable and easily accessible adult animal model for optogenetic activation of auditory neurons using adeno-associated viruses(AAVs)carrying Ch R2(H134R)to explore...Objective:This study aims to establish an economically viable and easily accessible adult animal model for optogenetic activation of auditory neurons using adeno-associated viruses(AAVs)carrying Ch R2(H134R)to explore the potential of cochlear optogenetics as a hearing restoration technology.Methods:Healthy adult guinea pigs were used in the experiments.The viral vector AAV2/8-Ch R2(H134R)-h Syn-e YFP was administered to the right cochlea via the round window membrane.The confocal microscopy and reverse transcription polymerase chain reaction(RT-PCR)were utilized to analyze the Ch R2(H134R)expression localized to spiral ganglion neurons(SGNs).The auditory pathway activation was assessed by recording the optical compound action potential(oCAP)and acoustic compound action potential(a CAP)at various laser intensities.Results:The Ch R2(H134R)-e YFP expression was confirmed in 90%of the tested animals,localized to the SGNs of the injected ear.Higher m RNA levels of Ch R2(H134R)and e YFP were observed in the injected ear compared to the non-injected ear,while actin(Actb)m RNA levels were not significantly different.The o CAP was successfully elicited by a 470 nm blue light laser stimulus,with similar amplitudes and latency periods to those of a CAPs when the o CAP was evoked by 5.80 m W blue light and the a CAP was evoked by a 40 d B SPL click.The amplitudes of o CAPs increased with increasing laser intensity.Conclusion:This study demonstrates the viability of optogenetic activation of the auditory system in adult guinea pigs through the transduction of AAV-Ch R2(H134R)in SGNs.Cochlear optogenetics demonstrates potential as a hearing restoration technology,providing a basis for further clinical research and opening new avenues for investigation.展开更多

摘要To solve the problem of the lack of reference material(RM)for determination of allergenic ingredients in food,a RM of cashew nut powder was developed in the study.Cashew nut powder was prepared from cashew nut kernel by selecting,cleaning,crushing,n-hexane degreasing and sieving treatment.The reliability and traceability of RM was verified using real-time quantitative polymerase chain reaction(qPCR)and phylogenetic tree analysis.The cashew nut powder RM showed good homogeneity,and good stability under long-term storage at 4℃and short-term simulated transportation from-20 to 45℃.The RM was determined jointly by 8 collaborative laboratories,and the characteristic CT value was 24.732,and the extended uncertainty was 1.052%(k=2).The RM was applied to verify the amplification efficiency and the limit of detection for qPCR assay,and showed good applicability.The RM could be used for method validation and quality control,for the determination of allergenic ingredients in food mixed with trace amounts of cashew nut.

摘要Transfer-based Adversarial Attacks(TAAs)can deceive a victim model even without prior knowledge.This is achieved by leveraging the property of adversarial examples.That is,when generated from a surrogate model,they retain their features if applied to other models due to their good transferability.However,adversarial examples often exhibit overfitting,as they are tailored to exploit the particular architecture and feature representation of source models.Consequently,when attempting black-box transfer attacks on different target models,their effectiveness is decreased.To solve this problem,this study proposes an approach based on a Regularized Constrained Feature Layer(RCFL).The proposed method first uses regularization constraints to attenuate the initial examples of low-frequency components.Perturbations are then added to a pre-specified layer of the source model using the back-propagation technique,in order to modify the original adversarial examples.Afterward,a regularized loss function is used to enhance the black-box transferability between different target models.The proposed method is finally tested on the ImageNet,CIFAR-100,and Stanford Car datasets with various target models,The obtained results demonstrate that it achieves a significantly higher transfer-based adversarial attack success rate compared with baseline techniques.

摘要Objective:This study aims to establish an economically viable and easily accessible adult animal model for optogenetic activation of auditory neurons using adeno-associated viruses(AAVs)carrying Ch R2(H134R)to explore the potential of cochlear optogenetics as a hearing restoration technology.Methods:Healthy adult guinea pigs were used in the experiments.The viral vector AAV2/8-Ch R2(H134R)-h Syn-e YFP was administered to the right cochlea via the round window membrane.The confocal microscopy and reverse transcription polymerase chain reaction(RT-PCR)were utilized to analyze the Ch R2(H134R)expression localized to spiral ganglion neurons(SGNs).The auditory pathway activation was assessed by recording the optical compound action potential(oCAP)and acoustic compound action potential(a CAP)at various laser intensities.Results:The Ch R2(H134R)-e YFP expression was confirmed in 90%of the tested animals,localized to the SGNs of the injected ear.Higher m RNA levels of Ch R2(H134R)and e YFP were observed in the injected ear compared to the non-injected ear,while actin(Actb)m RNA levels were not significantly different.The o CAP was successfully elicited by a 470 nm blue light laser stimulus,with similar amplitudes and latency periods to those of a CAPs when the o CAP was evoked by 5.80 m W blue light and the a CAP was evoked by a 40 d B SPL click.The amplitudes of o CAPs increased with increasing laser intensity.Conclusion:This study demonstrates the viability of optogenetic activation of the auditory system in adult guinea pigs through the transduction of AAV-Ch R2(H134R)in SGNs.Cochlear optogenetics demonstrates potential as a hearing restoration technology,providing a basis for further clinical research and opening new avenues for investigation.
