Background:Rodentibacter pneumotropicus and Rodentibacter heylii are important exclusion pathogens in specific pathogen-free(SPF)laboratory animals.However,routine detection methods such as traditional bacterial culture remain labor-intensive and time-consuming.The loop-mediated isothermal amplification-lateral flow dipstick(LAMP-LFD)assay provides a self-contained format for nucleic acid testing that combines speed,sensitivity,and specificity while minimizing aerosol contamination,making it well-suited for point-of-need applications.Objectives:This study aimed to develop a streamlined,on-site detection platform capable of rapidly identifying both pathogens with high analytical accuracy,operational simplicity,and suitability for routine laboratory surveillance.Methods:A LAMP-LFD system was established for the detection of the two pathogens using primers designed against the conserved 16S-23S rRNA ITS region.The assay's specificity,sensitivity,repeatability,and limit of detection(Lo D)were evaluated.Results:The assay achieved detection limits of 1×10-4 ng/μL for R.pneumotropicus and 1×10-5 ng/μL for R.heylii,demonstrating significantly higher sensitivity than PCR.No cross-reactivity was observed with 15 non-target bacteria.Among 28 infected samples,the method detected 19 positives,compared with 21 by multiplex PCR and 23 by q PCR.In 842 laboratory animal samples,culture-based,q PCR,and LAMP-LFD detected 2,41,and 37 positive samples,respectively.The Lo Ds were 10~2 and 10~1 CFU/mL in fecal samples and 10~1 and 10~0 CFU/mL in throat swab samples for R.pneumotropicus and R.heylii,respectively.Conclusions:This method provides an approach for monitoring the health status of laboratory mice by enabling the detection of these two target pathogens. 展开更多

This study describes a new bulk parameterization for sea salt aerosol(SSA)microphysics and investigates the impacts of SSA on an idealized tropical cyclone(TC)through numerical experiments.Results indicate that consideration of SSA microphysics enhances total precipitation and shifts its timing earlier,with opposite effects during the rapid intensification(RI)and mature(Mt)stages.During RI,SSA increases the maximum 10-m wind speed,strengthens eyewall updrafts,condensation,and cloudainwater content,while cooling mid-lower layers.In the Mt stage,SSA slightly reduces the wind speed,cools the warm core,elevates the latent heat release center to~500 m above the surface,and weakens the eyewall but enhances the outer rainband precipitation process.The findings reveal that SSA processes significantly strengthen TCs only during the RI stage,contrasting with previous studies showing continuous TC enhancement by SSA. 展开更多

Block copolymer(BCP) nanolithography offers potential beyond traditional photolithographic limits, yet reliably producing low-defect, perpendicular domains remains challenging. We introduce a microenvironmentdriven isothermal annealing method for directed self-assembly of BCP thin films. By annealing films at stable temperature in a quasi-sealed, inert-gas chamber, our approach promotes highly uniform perpendicular lamellar nanopatterns over large areas, effectively mitigating environmental fluctuations and emulating solvent-vapor annealing without solvent exposure. Resulting BCP structures demonstrate enhanced spatial coherence and notably low defect density. Furthermore, we successfully transfer these nanopatterns into precise metal nano-line arrays,confirming the method's capability for high-fidelity pattern replication. This scalable, solvent-free technique provides a robust, reliable route for high-resolution nanopatterning in advanced semiconductor manufacturing. 展开更多

Objective This study explored the job choice preferences of Center for Disease Prevention and Control(CDC)workers to provide CDC management information and recommendations for optimizing employee retention and motivation policies.Methods A discrete choice experiment was conducted in nine provinces across China.Seven key attributes were identified to analyze the job preferences of CDC workers.Mixed logit models,latent class models,and policy simulation tools were used.Results A valid sample of 5,944 cases was included in the analysis.All seven attributes significantly influenced the job choices of CDC workers.Heterogeneity analyses identified two main groups based on different levels of preference for attribute utility.Income-prioritizers were concerned with income and opportunities for career development,whereas bianzhi-prioritizers were concerned with bianzhi and welfare benefits.The policy simulation analysis revealed that income-prioritizers had a relatively higher sensitivity to multiple job preference incentives.Conclusion Income and bianzhi were the two key attributes influencing the job choices and retention preferences of CDC workers.Heterogeneity in job preferences was also identified.Based on the preference characteristics of different subgroups,policy content should be skewed to differentiate the importance of incentives. 展开更多

Papaya(Carica papaya L.)is an important tropical species popular for highly nutritious fruit as well as medicinal value.In addition,non-commercial cultivation of papaya trees has resulted in dual-purpose cultivars grown for both fruit and ornamental value in residential areas.Petiole color is a key ornamental trait in papaya that varies amongst cultivars depending on anthocyanin accumulation resulting in purple or green pigmentation.Although inherited as a simple trait,genetic characterization and genomic loci responsible for the purple petiole color in papaya is unknown.In this study,F1 and F2 populations generated from two breeding lines PR-2043(green petiole)and T5-2562(purple petiole)were used to evaluate the inheritance patterns of petiole color as well as determine genetic loci and genes involved in petiole pigmentation in papaya through bulk segregant analysis(BSA)and transcriptome sequencing.The segregation of purple petiole color followed a single dominant gene inheritance model(3:1).BSA-seq analysis indicated key genes influencing petiole color are mainly located in chromosome 1(0.01 to 5.96 Mb)of the papaya genome.Four major genes,including CHS,MYB20,MYB315-like,and MYB75-like within this region exhibited significant differential expression in a comparison between purple and green petiole papaya plants.A relatively high abundance of CHS transcripts was observed in purple petioles and may signify a major involvement in regulating anthocyanins accumulation in papaya petioles.The findings of this study facilitate the future efforts of breeding papaya cultivars with higher economical value in residential landscapes. 展开更多

Engineered probiotics can serve as therapeutics based on their ability of produce recombinant immune-stimulating properties.In this study,we built the recombinant Bacillus subtilis WB800 expressing antimicrobial peptide KR32(WB800-KR32)using genetic engineering methods and investigated its protective effects of nuclear factor-E2-related factor 2(Nrf2)-Kelch-like ECH-associated protein 1(Keap1)pathway activation in intestinal oxidative disturbance induced by enterotoxigenic Escherichia coli(ETEC)K88 in weaned piglets.Twenty-eight weaned piglets were randomly distributed into four treatment groups with seven replicates fed with a basal diet.The feed of the control group(CON)was infused with normal sterilized saline;meanwhile,the ETEC,ETEC+WB800,and ETEC+WB800-KR32 groups were orally administered normal sterilized saline,5×1010CFU(CFU:colony forming units)WB800,and 5×1010CFU WB800-KR32,respectively,on Days 1-14 and all infused with ETEC K881×1010CFU on Days 15-17.The results showed that pretreatment with WB800-KR32 attenuated ETEC-induced intestinal disturbance,improved the mucosal activity of antioxidant enzyme(catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GPx))and decreased the content of malondialdehyde(MDA).More importantly,WB800-KR32 downregulated genes involved in antioxidant defense(GPx and SOD1).Interestingly,WB800-KR32 upregulated the protein expression of Nrf2 and downregulated the protein expression of Keap1 in the ileum.WB800-KR32 markedly changed the richness estimators(Ace and Chao)of gut microbiota and increased the abundance of Eubacterium_rectale_ATCC_33656 in the feces.The results suggested that WB800-KR32 may alleviate ETEC-induced intestinal oxidative injury through the Nrf2-Keap1 pathway,providing a new perspective for WB800-KR32 as potential therapeutics to regulate intestinal oxidative disturbance in ETEC K88 infection. 展开更多

Stroke is caused by an acute focal disruption of the vasculature in the central nervous system.Neurological-related functional deficits are the most devastating consequences for stroke survi-vors.Neural signals from stroke patients can reflect the functional statuses of patients and provide insights into the neuronal recovery mechanism for functioning,which could be used as the basis for designing optimal treatment strategies.Near-infrared spectroscopy(NIRS)is a low-cost,noninvasive,easily operated neuroimage method and it is compatible with various rehabilitative programs.These advantages make NIRS an excellent candidate in research for stroke recovery.Here,we focused on the brain functions and recovery for stroke patients at stable status,conducted a systematic literature review about NIRS applications in stroke since 2000 and identified a total of 72 references through ScienceDirect and PubMed database retrieval.The NIRS studies in stroke include resting-state function and its recovery,motor function and itsrecovery,motor and cognition interference,cognitive function and its recovery,language function and its recovery,emotional function and its recovery and other applications.Based on the results of the quality assessment,we identified some study gaps from the previous research and provided suggestions for some methodological improvement in the future.The trend of NIRS gives a boost to its application in stroke,and the potential research directions for NIRS in stroke are pros-pected,including multi-center clinical research,treatment efficacy prediction research and brain-muscle coupling research.Finally,limitations of NIRS are discussed. 展开更多

Cloud microphysical processes occur at the smallest end of scales among cloud-related processes and thus must be parameterized not only in large-scale global circulation models(GCMs)but also in various higher-resolution limited-area models such as cloud-resolving models(CRMs)and large-eddy simulation(LES)models.Instead of giving a comprehensive review of existing microphysical parameterizations that have been developed over the years,this study concentrates purposely on several topics that we believe are understudied but hold great potential for further advancing bulk microphysics parameterizations:multi-moment bulk microphysics parameterizations and the role of the spectral shape of hydrometeor size distributions;discrete vs“continuous”representation of hydrometeor types;turbulence-microphysics interactions including turbulent entrainment-mixing processes and stochastic condensation;theoretical foundations for the mathematical expressions used to describe hydrometeor size distributions and hydrometeor morphology;and approaches for developing bulk microphysics parameterizations.Also presented are the spectral bin scheme and particle-based scheme(especially,super-droplet method)for representing explicit microphysics.Their advantages and disadvantages are elucidated for constructing cloud models with detailed microphysics that are essential to developing processes understanding and bulk microphysics parameterizations.Particle-resolved direct numerical simulation(DNS)models are described as an emerging technique to investigate turbulence-microphysics interactions at the most fundamental level by tracking individual particles and resolving the smallest turbulent eddies in turbulent clouds.Outstanding challenges and future research directions are explored as well. 展开更多

Effects of sludge age on volatile fatty acids (VFAs) production and Phosphorus (P) release during anaerobic acidification of waste activated sludge (WAS) were investigated.Sequencing batch reactors (SBR) fed with simulating domestic sewage were applied to produce WAS of different sludge ages,and batch tests were used for anaerobic acidification.The maximum dissolved total organic carbon,release of PO43+-P,and accumulation of acetate (C2),propionate (C3),butyrate (C4),and valerate (C5) decreased by 56.2%,55.8%,52.6%,43.7%,82.4%and 84.8%,respectively,as the sludge age of WAS increased from 5 to 40 days.Limited degradation of protein played a dominating role in decreasing DTOC and VFAs production.Moreover,the increase in molecular weight of organics and organic nitrogen content in the supernatant after acidification suggested that the refractory protein in WAS increased as sludge age extended.Although the production of C2,C3,C4,and C5 from WAS decreased as the sludge age increased,the proportions of C2 and C3 in VFAs increased,which might be due to the declined production of C5 from protein and the faded genus Dechlorobacter.Keeping sludge age of WAS at a relatively low level (<10 days) is more appropriate for anaerobic acidification of WAS as internal carbon sources and P resource. 展开更多

The dysregulation of exosomal microRNAs(miRNAs)plays a crucial role in the development and progression of cancer.This study investigated the role of a newly identified serum exosomal miRNA miR-4256 in gastric cancer(GC)and the underlying mechanisms.The differentially expressed miRNAs were firstly identified in serum exosomes of GC patients and healthy individuals using next-generation sequencing and bioinformatics.Next,the expression of serum exosomal miR-4256 was analyzed in GC cells and GC tissues,and the role of miR-4256 in GC was investigated by in vitro and in vivo experiments.Then,the effect of miR-4256 on its downstream target genes HDAC5/p16INK4a was studied in GC cells,and the underlying mechanisms were evaluated using dual luciferase reporter assay and Chromatin Immunoprecipitation(ChIP).Additionally,the role of the miR-4256/HDAC5/p16INK4a axis in GC was studied using in vitro and in vivo experiments.Finally,the upstream regulators SMAD2/p300 that regulate miR-4256 expression and their role in GC were explored using in vitro experiments.miR-4256 was the most significantly upregulated miRNA and was overexpressed in GC cell lines and GC tissues;in vitro and in vivo results showed that miR-4256 promoted GC growth and progression.Mechanistically,miR-4256 enhanced HDAC5 expression by targeting the promoter of the HDAC5 gene in GC cells,and then restrained the expression of p16INK4a through the epigenetic modulation of HDAC5 at the p16INK4a promoter.Furthermore,miR-4256 overexpression was positively regulated by the SMAD2/p300 complex in GC cells.Our data indicate that miR-4256 functions as an oncogene in GC via the SMAD2/miR-4256/HDAC5/p16INK4a axis,which participates in GC progression and provides novel therapeutic and prognostic biomarkers for GC. 展开更多

It is well known that exposure to environmental stresses could enhance the adaptability of bacteria and up-regulate the expression of a variety of oxidative stress-related genes and antioxidant enzymes.It is unclear whether the adaptability of microorganisms formed naturally in special environments could transfer to other organisms.The study aimed to evaluate the effects of untreated and ethanol-induced Lactococcus lactis intracellular extracts(U-IE and E-IE)on alcohol metabolism in mice.The positive effects of E-IE on alcohol metabolism in mice were revealed by the enhanced latency of loss of righting reflex(LORR),the reduced duration of LORR,the decrease of blood alcohol concentration,as well as the elevation of alcohol dehydrogenase(ADH)activities in the stomach and liver tissues.Furthermore,the potential benefits of E-IE on the liver were evaluated by biochemical parameters including the activities of serum transaminase,the levels of antioxidant enzymes,and the pathological changes of liver tissue.The present work put forward a new point that appropriate ethanol stress could enhance the intracellular ADH activity of L.lactis,and its intracellular extracts could continue to enhance alcohol metabolism in mice. 展开更多

Anaerobic digestion and incineration are widely used sewage sludge(SS)treatment and disposal approaches to recovering energy from SS,but it is difficult to select a suitable technical process from the various technologies.In this study,life-cycle assessments were adopted to compare the energy-and greenhouse gas-(GHG)emission footprints of two sludge-to-energy systems.One system uses a combination of AD with incineration(the AI system),whereas the other was simplified by direct incineration(the DI system).Comparison between three SS feedstocks(VS/TS:57.61-73.1 ds.%)revealed that the AI system consistently outperformed the DI system.The results of sensitivity analyses showed that the energy and GHG emission performances were mainly affected by VS content of the SS,AD conversion efficiency,and the energy consumption of sludge drying.Furthermore,the energy and GHG emission credit of the two systems increased remarkably with the increase in the VS content of the SS.For the high-organic-content sludge(VS/TS:55%–80%),the energy and GHG emission credit of the AI system increase with the increase of AD conversion efficiency.However,for the low organic content sludge(VS/TS:30%–55%),it has the opposite effect.In terms of energy efficiency and GHG performance,the AI system is a good choice for the treatment of high-organic-content sludge(VS/TS>55%),but DI shows superiority over AI when dealing with low organic content sludge(VS/TS<55%). 展开更多

To improve the sludge conditioning efficiency without increasing the ozone dose,an in-situ sludge reduction process based on Mn2+-catalytic ozonation conditioning was proposed.Using ozone conditioning alone as a control,a lab-scale sequencing batch reactor coupled with ozonated sludge recycle was evaluated for its operating performance at an ozone dose of 75 mg O3/g VSS and 1.5 mmol/L Mn2+addition.The results showed a 39.4%reduction in MLSS and an observed sludge yield of 0.236 kg MLSS/kg COD for the O3+Mn2+group compared to the O3group (15.3%and 0.292 kg MLSS/kg COD),accompanied by better COD,NH4+-N,TN and TP removal,improved effluent SS and limited impact on excess sludge properties.Subsequently,activity tests,BIOLOG ECO microplates and 16S rRNA sequencing were applied to elucidate the changing mechanisms of Mn2+-catalytic ozonation related to microbial action:(1) Dehydrogenase activity reached a higher peak.(2) Microbial utilization of total carbon sources had an elevated effect,up to approximately 18%,and metabolic levels of six carbon sources were also increased,especially for sugars and amino acids most pronounced.(3) The abundance of Defluviicoccus under the phylum Proteobacteria was enhanced to 12.0%and dominated in the sludge,they had strong hydrolytic activity and metabolic capacity.Denitrifying bacteria of the genus Ferruginibacter also showed an abundance of 7.6%,they contributed to the solubilization and reduction of sludge biomass.These results could guide researchers to further reduce ozonation conditioning costs,improve sludge management and provide theoretical support. 展开更多

Evaluating anaerobic biodegradability of sludge and then identifying the stabilization of digestate is necessary in sludge treatment and disposal.48 sludge samples from 24 typical waste water treatment plants(WWTPs)in 11 provinces in China were selected to investigate the relationship between Biochemical Acidogenic Potential(BAP)test and Biochemical Methane Potential(BMP)test.The volatile fatty acid(VFA)production obtained from BAP tests was found linearly related to the ultimate methane production from corresponding BMP tests.Satisfying results were obtained with Pearson correlation coefficient as 0.929 and R^2 value as 0.76.Furthermore,the physio-chemical characteristics(FCI,SUVA254,E 4/E 6)of supernatant,which were associated with humic-like substances(HS),were investigated before and after BMP tests.Through which a new criterion(FCI>1.50,SUVA 254>1.10,E 4/E 6<4.0)was proposed to evaluate the stabilization level of anaerobic digested sludge. 展开更多

Based on 362 consumer questionnaire data, this study builds a structural equation model to discuss the driving factors of residents’ choice of online car-hailing and whether the development of online car-hailing will have a certain substitution impact on the sales of private cars. From the perspective of consumers’ purchase intention, the research results show that consumers’ price consciousness, convenience consciousness, environmental protection consciousness and possession tendency will affect their choice of travel mode, and the use of online car-hailing is positively correlated with consumers’ willingness to replace private car ownership with online car-hailing. 展开更多

In order to predict the remaining service life of brake pads accurately and efficiently,and to achieve intelligent warning,this paper proposes a CNN-LSTM brake pad remaining life prediction model based on an attention mechanism.The model constructs a non-linear relationship between brake pad features such as brake temperature,brake oil pressure and brake speed and brake pad wear data through convolutional neural network(CNN)and long and short term memory network(LSTM),as well as capturing the time dependence that exists in the brake pad wear sequence.The attention mechanism is also introduced to assign different weight values to the features output from multiple historical moments,highlighting the features with high saliency and avoiding the influence of invalid features,so as to improve the prediction effect of the remaining brake pad life.The results show that the proposed CNN-LSTM-Attention model can effectively predict the remaining life of brake pads,with the mean absolute error MAE value of 0.0048,root mean square error RMSE value of 0.0059 and coefficient of determination R2 value of 0.9636;and compared with the BP model,CNN model,LSTM model and CNN-LSTM model,the coefficient of determination R2 values are closest to 1,with an improvement of 8.26%,5.25%,3.99%and 1.85%respectively,enabling more effective monitoring and intelligent warning of the remaining brake pad life. 展开更多

Triple-negative breast cancer(TNBC)is one of the most aggressive and metastatic forms of breast cancer,for which there are currently no satisfactory therapeutic agents.Here,we reported for the first time that boholamide A,a naturally occurring macrocyclic depsipeptide,exhibited hypoxia-selective anti-TNBC activity against in MDA-MB-231 cells.However,its structure-activity relationships and target had not yet been elucidated.A series of boholamide A analogues were chemically synthesized and evaluated for anti-TNBC potency.The most promising compound 1j was prepared in 13 linear steps with an overall yield of 7.92%,and exhibited high potency against MDA-MB-231 cells with an IC50 value of 0.15μmol/L under hypoxic condition.Moreover,1j significantly inhibited proliferation and migration,and induced apoptosis in MDA-MB-231 cells.Compound 16,a prodrug of 1j,significantly inhibited the tumor volume and tumor weight in xenografts.Furthermore,we identified that 1j covalently targeted eukaryotic translation elongation factor 1alpha 1(eEF1A1)which might underlie the anticancer activity and hypoxia selectivity of boholamide A analogues.These results suggested that boholamide A analogues represented a promising scaffold for discovering hypoxia-selective anti-TNBC agents,and compound 16 deserved further investigation as a candidate for TNBC treatment. 展开更多
