BACKGROUND Prediabetes mellitus(PDM)is receiving increasing attention as a precursor to type 2 diabetes mellitus.Lifestyle and traditional Chinese medicine(TCM)inter-ventions are effective for PDM prevention and treat...BACKGROUND Prediabetes mellitus(PDM)is receiving increasing attention as a precursor to type 2 diabetes mellitus.Lifestyle and traditional Chinese medicine(TCM)inter-ventions are effective for PDM prevention and treatment.Therefore,we con-ducted a preliminary investigation and an exploratory randomised controlled trial to assess the effects of a combined lifestyle and TCM intervention on PDM indicators.AIM To study the effectiveness of Xiaokeqing granules(XQG)and lifestyle inter-ventions in PDM participants while using metabolomics to identify potential markers.METHODS Forty PDM participants with yin deficiency syndrome with excessive heat were recruited and randomly allocated to the control(Con)group or the XQG group(20 per group).The Con group underwent lifestyle interventions,whereas the XQG group underwent lifestyle and XQG interventions.The follow-up duration was 2 months.Fasting blood glucose,2-hour postprandial glucose(2hPG),gly-cated haemoglobin A1c,fasting insulin,homeostasis model assessment-insulin resistance levels,and serum metabolomics characteristics were compared via liquid chromatography-tandem mass spectrometry analysis.RESULTS There were significant differences in 2hPG between the two groups(P0.05).Groenlandicine,kaempferol,isomangiferin,etc.,are the XQG constituents absorbed in the blood.N-Nervonoyl methionine and 5-hydroxy-L-tryptophan are core potential metabolomic biomarkers for the effectiveness of XQG and lifestyle interventions.HTR1A,HTR2C,SLC6A4,etc.,are the core targets of XQG and lifestyle interventions,as well as the reason for their clinical efficacy.Possible mechanistic pathways include tryptophan metabolism,pantothenate and certificate of analysis biosynthesis,lysine degradation and biosynthesis of cofactors.CONCLUSION This pilot study provides evidence that a combined XQG and lifestyle intervention can improve 2hPG in par-ticipants with PDM.The mechanism of action is related to multiple constituents,targets and pathways.展开更多
BACKGROUND Large number of decompensated liver cirrhosis patients in China have been diagnosed with hepatitis B virus(HBV).Human umbilical cord-derived mesenchymal stem cells(hUC-MSCs)can possibly cure decompensated l...BACKGROUND Large number of decompensated liver cirrhosis patients in China have been diagnosed with hepatitis B virus(HBV).Human umbilical cord-derived mesenchymal stem cells(hUC-MSCs)can possibly cure decompensated liver cirrhosis because of their self-renewal and multidirectional differentiation potential.AIM To explore the safety and effect of treating liver cirrhosis with HBV by hUC-MSCs.METHODS Twenty-four participants were recruited,divided into 3 groups,and injected with different amounts of hUC-MSCs via the peripheral vein.Therapy was administered 3 times.A 24-week follow-up examination of each patient’s liver function,coagulation function,general condition,and immune system was performed.Adverse events were also recorded.A 2-year survival assessment was subsequently performed.RESULTS Infusion therapy rapidly improved liver function.Serum albumin transiently increased on days 57 and 85 but returned to baseline by day 169,while prothrombin time activity demonstrated sustained improvement from day 29 through day 169.Interleukin-8 levels decreased persistently throughout treatment.All dosage groups achieved 100%6-month survival;2-year survival rates were 66.7%(low-dose),100%(medium-dose),and 87.5%(high-dose).The interaction between dosage and efficacy was weak.Notably,the improvement in liver function was statistically significant and sustained for almost 3 months,suggesting clinically meaningful therapeutic durability.CONCLUSION hUC-MSCs can be considered a safe treatment for patients with decompensated liver cirrhosis associated with HBV.However,larger-scale randomized controlled trials are needed to prove its therapeutic effect.展开更多
BACKGROUND Endothelial injury and inflammation are the main pathological changes in hyperuricemic nephropathy(HN);however,they have not been assessed in patients in the early,middle,and late phases of HN.AIM To invest...BACKGROUND Endothelial injury and inflammation are the main pathological changes in hyperuricemic nephropathy(HN);however,they have not been assessed in patients in the early,middle,and late phases of HN.AIM To investigate endothelial injury and inflammatory conditions between patients with HN at chronic kidney disease(CKD)stages 3-4 and CKD 1-2.METHODS This study enrolled 80 patients(49 and 31 with HN at CKD stage 1-2 and 3-4,respectively)from the Department of Nephrology,Shuguang Hospital Affiliated to Shanghai University of Traditional Chinese Medicine between July 2021 and January 2022.Plasma levels of heparan sulfate,endocan,oxidized low-density lipoprotein(Ox-LDL),E-selectin,soluble intercellular adhesion molecule-1(slCAM1),interleukin(IL)-1β,and IL-6 and urine levels of lipocalin-type prostaglandin D synthase(L-PGDS),IL-1β,and IL-6 were measured using enzyme-linked immunosorbnent assay.RESULTS Comparison between patients with HN at CKD 1-2 and those with HN at CKD 3-4 showed that age and disease course were significant factors(P<0.001 and P<0.010,respectively).There were no statistical differences in sex,heart rate,body mass index,and systolic and diastolic blood pressures.The incidence of hypertension was also significant(P=0.03).Plasma levels of heparin sulfate(P<0.001),endocan(P=0.034),E-selectin(P<0.001),slCAM1(P<0.001),IL-1β(P=0.006),and IL-6(P=0.004)and the urine levels of L-PGDS(P<0.001),IL-1β(P=0.003),and IL-6(P<0.001)were high in patients with HN at CKD 3-4 than in those with HN at CKD 1-2.The difference in plasma Ox-LDL levels was not significant(P=0.078).CONCLUSION Vascular endothelial injury and inflammation were higher in patients with HN at CKD3-4 than at CKD 1-2.Plasma heparin sulfate and slCAM1 levels are synergistic factors for CKD staging in HN.展开更多
Solvent extraction based on ionic liquids is generally considered to be an environmentally benign and effective technology for gold(Ⅰ)recovery.The aim of this work is to study gold(Ⅰ)extraction from aurocyanide solu...Solvent extraction based on ionic liquids is generally considered to be an environmentally benign and effective technology for gold(Ⅰ)recovery.The aim of this work is to study gold(Ⅰ)extraction from aurocyanide solution using fluorine-free ionic liquids[A336][SCN],[A336][MTBA]and[A336][Mal].Various factors that affect gold(Ⅰ)extraction(including concentration of ionic liquids,equilibrium pH,concentration of the modifier tributyl phosphate(TBP),reaction time and initial concentration of gold in an aqueous solution)were studied and optimized.The results indicate that the three Aliquat336-based ionic liquids all exhibit excellent behaviors for gold(Ⅰ)extraction.More than 99.8%of gold(Ⅰ)can be extracted from the aqueous phase into the ionic liquid phase.The gold-loaded ionic liquids were characterized using infrared spectroscopy and mass spectrometry to study the extraction mechanism of gold(Ⅰ).The results revealed that extraction of gold(Ⅰ)into the ionic liquid phase was based on an exchange reaction between the anion Au(CN)2-in aqueous solution and the anion SCN-in ionic liquid[A336][SCN].The logarithmic relationship between distribution coefficient and TBP concentration indicates that two TBP molecules are involved in the formation of the extracted complex.The extracted complex was determined to be A336+·Au(CN)2-·2 TBP.In addition,the gold(Ⅰ)-loaded ionic liquids can be efficiently stripped using NH4 SCN,2-methylthiobenzoic acid and methyl maltol.The results establish that Aliquat 336-based ionic liquids have potential application prospects in gold(Ⅰ)recovery from cyanide solutions.展开更多
BACKGROUND Juvenile dermatomyositis(JDM)is an idiopathic inflammatory myopathy that occurs in childhood.It is characterized by muscle weakness and a characteristic rash.Previous literature reports have rarely describe...BACKGROUND Juvenile dermatomyositis(JDM)is an idiopathic inflammatory myopathy that occurs in childhood.It is characterized by muscle weakness and a characteristic rash.Previous literature reports have rarely described JDM with severe skin ulcers and infections.CASE SUMMARY Herein,we describe a case of a 2-year-old female patient who suffered from JDM,whose myositis-specific autoantibodies were positive for anti-nuclear matrix protein 2 antibody,with progressively worsening skin ulcers and severe infections.The patient was treated with glucocorticoids and various immunosuppressants.Nevertheless,further progression of the disease and the combination of primary disease and severe infection in the later period were fatal.CONCLUSION In children,anti-nuclear matrix protein 2+JDM combined with skin ulcers often indicates severe disease.In such cases,personalized treatment for the primary disease and infection prevention and control are essential.展开更多
Objective: Electroacupuncture (EA) in the treating principle of “soothing the liver and regulating the kidney” was applied to intervion the rats with post-traumatic stress disorder (PTSD), and its effect on anxiety-...Objective: Electroacupuncture (EA) in the treating principle of “soothing the liver and regulating the kidney” was applied to intervion the rats with post-traumatic stress disorder (PTSD), and its effect on anxiety-like behavior, spatial learning and memory ability, and expressions of synaptophysin (SYN) and postsynaptic dense 95 (PSD95) in hippocampus of rats were observed to explore the underlying mechanism.Methods: Twenty-four male SD rats were randomly divided into control group, model group, and EA group, with 8 rats in each group. There was no intervention in the control group. The rest 16 rats were prepared for modeling. The single-prolonged stress & shock (SPS&S) method was used to establish the PTSD models. There was no intervention in the model group after modeling. In the EA group, “Bǎihuì (百会GV20)”“ Shéntíng (神庭GV24)”“Gānshū (肝俞BL18)”“Shènshū (肾俞BL23)” were manipulated with EA stimulation, intensity 1 mA, frequency 2/100 Hz, disperse-dense wave, and treatment was performed once a day, 20 min each time, for a total of 21 days. Open field test, elevated plus maze and Morris water maze tests were used to observe the behavioral differences of rats in each group. Immunohistochemical method was used to observe the differences of positive expressions of proteins SYN and PSD95 in hippocampus.Results: In the open field test, compared with the control group, the total traveling distance, the percentage of the time spent in the central cell and the numbers of the central cells crossing in the model group were all decreased (all P < 0.05). Compared with the model group, these indicators in the EA group were significantly all increased (all P < 0.05). In the elevated plus maze test, compared with the control group, the percentages of open arm staying time and entering times in the model group were both decreased (both P < 0.05). Compared with the model group, these indicators in the EA group were both significantly increased (both P < 0.05). The results of Morris water maze test showed that compared with the control group, the escape latency time and travelled distance of rats in the model group were all increased from day1 to day 4 (all P < 0.05), and the percentage of staying time in the target quadrant was decreased (P < 0.05). Compared with the model group, the escape latency time and travelled distance of EA group were both significantly shortened (both P < 0.05), and the percentage of staying time in the target quadrant was significantly increased (P < 0.05). The immunohistochemical results showed that, compared with the control group, the positive expressions of SYN and PSD95 in the hippocampus of the model group were significantly down-regulated (both P < 0.05). Compared with the model group, the positive expressions of SYN and PSD95 in the hippocampus of the EA group were significantly up-regulated (both P < 0.05).Conclusions: EA in the treating principle of “soothing the liver and regulating the kidney” can effectively relieve anxiety-like behavior and improve spatial learning and memory ability of rats with PTSD, and the mechanism is related to the up-regulation of the expressions of SYN and PSD95 in the hippocampus.展开更多
Depression is a prevalent and severe mental health condition that is a primary cause of disability and suicide.Ginsenoside Rg1,an active ingredient derived from Panax ginseng C.A.Mey,has recently received considerable...Depression is a prevalent and severe mental health condition that is a primary cause of disability and suicide.Ginsenoside Rg1,an active ingredient derived from Panax ginseng C.A.Mey,has recently received considerable attention for its remarkable potential in treating depression.To review the existing evidence from preclinical studies on Rg1 and uncover its multi-target antidepressant mechanisms and pharmacological properties through a thorough search of relevant electronic databases.We searched digital repositories from inception until December 2024,e.g.,EMBASE,MEDLINE,Cochrane Library,and PubMed.We reviewed the antidepressant mechanism of Rg1,e.g.,inhabitation of inflammation,regulation of neurotransmitters,mediation of gap junction function of astrocytes,anti-oxidative stress,and anti-apoptosis.In addition to its potential implications for the clinical treatment of depression,we also analyzed its current limitations and put forward several novel research hypotheses as well as prospective research directions.It is hoped that Rg1 will be developed as a new treatment for clinical depression.展开更多

However, many cathode interfacial materials(CIMs) are unsuitable for printing manufacturing due to theirsensitivity to thickness. To tackle this issue, researchers havedeveloped a series of CIMs with n-type conjugated frame-works, particularly the naphthalene diimide (NDI) unit, owingto its high electron mobility and complementary absorptionwith commonly used active layers. Despite this, individualNDI molecules have a strong tendency to form large crystallinedomains, which can lead to interfacial defects in CILs. In thiswork, a different approach from other NDI-based CIMs wasadopted by substituting amino polar groups at the core posi-tion of NDI and polymerizing them into ionene-type CIMs.We designed and synthesized three self-doped polymer CIMsnamed PN-Pi, PN-Pe and PN-Eh. Among them, PN-Pi notablyreduces the work function of the Ag electrode, aligns inter-facial energies appropriately, smooths the active layer film andsuppresses carrier injection. This results in an impressivepower conversion efficiency (PCE) of 18.33% in the PM6:L8-BO system and maintains 90.4% PCE even at 127 nm thick-ness, ranking among the top film-thickness tolerance in theOSC field. This work demonstrates that combining conjugatedbackbone substitution with ionic polymerization is a promis-ing strategy for designing high-performance CIMs for OSCs.展开更多
Apoplastic iron(Fe)in roots represents an essential Fe storage pool.Reallocation of apoplastic Fe is of great importance to plants experiencing Fe deprivation,but how this reallocation process is regulated remains elu...Apoplastic iron(Fe)in roots represents an essential Fe storage pool.Reallocation of apoplastic Fe is of great importance to plants experiencing Fe deprivation,but how this reallocation process is regulated remains elusive,likely because of the highly complex cell wall structure and the limited knowledge about cell wall biosynthesis and modulation.Here,we present genetic and biochemical evidence to demonstrate that the Cdi-mediated galactosylation of rhamnogalacturonan-II(RG-II)is required for apoplastic Fe reallocation.Cdi is expressed in roots and up-regulated in response to Fe deficiency.It encodes a putative glycosyltransferase localized to the Golgi apparatus.Biochemical and mass spectrometry assays showed that Cdi catalyzes the transfer of GDP-L-galactose to the terminus of side chain A on RG-II.Disruption of Cdi essentially decreased RG-II dimerization and hence disrupted cell wall formation,as well as the reallocation of apoplastic Fe from roots to shoots.Further transcriptomic,Fourier transform infrared spectroscopy,and Fe desorption kinetic analyses coincidently suggested that Cdi mediates apoplastic Fe reallocation through extensive modulation of cell wall components and consequently the Fe adsorption capacity of the cell wall.Our study provides direct evidence demonstrating a link between cell wall biosynthesis and apoplastic Fe reallocation,thus indicating that the structure of the cell wall is important for efficient usage of the cell wall Fe pool.展开更多
Acetic acid is a common inhibitor present in lignocellulose hydrolysate,which inhibits the ethanol production by yeast strains.Therefore,the cellulosic ethanol industry requires yeast strains that can tolerate acetic ...Acetic acid is a common inhibitor present in lignocellulose hydrolysate,which inhibits the ethanol production by yeast strains.Therefore,the cellulosic ethanol industry requires yeast strains that can tolerate acetic acid stress.Here we demonstrate that overexpressing a yeast native arginase-encoding gene,CAR1,renders Saccharomyces cerevisiae acetic acid tolerance.Specifically,ethanol yield increased by 27.3%in the CAR1-overexpressing strain compared to the control strain under 5.0 g/L acetic acid stress.The global intracellular amino acid level and compositions were further analyzed,and we found that CAR1 overexpression reduced the total amino acid content in response to acetic acid stress.Moreover,the CAR1 overexpressing strain showed increased ATP level and improved cell membrane integrity.Notably,we demonstrated that the effect of CAR1 overexpression was independent of the spermidine and proline metabolism,which indicates novel mechanisms for enhancing yeast stress tolerance.Our studies also suggest that CAR1 is a novel genetic element to be used in synthetic biology of yeast for efficient production of fuel ethanol.展开更多
“Prospection”refers to the ability to mentally simulate one’s future episodes.Negative bias in prospection,which includes both reduced positivity and enhanced negativity,is considered to be closely related to the d...“Prospection”refers to the ability to mentally simulate one’s future episodes.Negative bias in prospection,which includes both reduced positivity and enhanced negativity,is considered to be closely related to the development and maintenance of depression.However,there is a relative paucity of measures that adequately assess negative bias in prospection in depressed people.The current study developed and initially validated the Negative Bias in Prospection Scale(NBPS)using a nonclinical sample(n=959).The validity and reliability of the NBPS were investigated in several ways.Results suggested the 14 NBPS items loaded onto three factors,namely the“Increased negativity,”the“Reduced positivity,”and the“Overgeneralization.”The NBPS demonstrated decent internal consistency and test-retest reliability.Further,the NBPS was strongly associated with severity of depressive symptoms.Moreover,the NBPS was correlated in a theoretically meaningful way with other measures of future-oriented cognitions or negative biases.In conclusion,the NBPS has promising preliminary psychometric properties.It will be an efficient tool for exploring dysfunctional prospection in depression and provides a novel measure for mechanism of change in prospection-based interventions.展开更多

摘要Solvent extraction based on ionic liquids is generally considered to be an environmentally benign and effective technology for gold(Ⅰ)recovery.The aim of this work is to study gold(Ⅰ)extraction from aurocyanide solution using fluorine-free ionic liquids[A336][SCN],[A336][MTBA]and[A336][Mal].Various factors that affect gold(Ⅰ)extraction(including concentration of ionic liquids,equilibrium pH,concentration of the modifier tributyl phosphate(TBP),reaction time and initial concentration of gold in an aqueous solution)were studied and optimized.The results indicate that the three Aliquat336-based ionic liquids all exhibit excellent behaviors for gold(Ⅰ)extraction.More than 99.8%of gold(Ⅰ)can be extracted from the aqueous phase into the ionic liquid phase.The gold-loaded ionic liquids were characterized using infrared spectroscopy and mass spectrometry to study the extraction mechanism of gold(Ⅰ).The results revealed that extraction of gold(Ⅰ)into the ionic liquid phase was based on an exchange reaction between the anion Au(CN)2-in aqueous solution and the anion SCN-in ionic liquid[A336][SCN].The logarithmic relationship between distribution coefficient and TBP concentration indicates that two TBP molecules are involved in the formation of the extracted complex.The extracted complex was determined to be A336+·Au(CN)2-·2 TBP.In addition,the gold(Ⅰ)-loaded ionic liquids can be efficiently stripped using NH4 SCN,2-methylthiobenzoic acid and methyl maltol.The results establish that Aliquat 336-based ionic liquids have potential application prospects in gold(Ⅰ)recovery from cyanide solutions.
摘要BACKGROUND Juvenile dermatomyositis(JDM)is an idiopathic inflammatory myopathy that occurs in childhood.It is characterized by muscle weakness and a characteristic rash.Previous literature reports have rarely described JDM with severe skin ulcers and infections.CASE SUMMARY Herein,we describe a case of a 2-year-old female patient who suffered from JDM,whose myositis-specific autoantibodies were positive for anti-nuclear matrix protein 2 antibody,with progressively worsening skin ulcers and severe infections.The patient was treated with glucocorticoids and various immunosuppressants.Nevertheless,further progression of the disease and the combination of primary disease and severe infection in the later period were fatal.CONCLUSION In children,anti-nuclear matrix protein 2+JDM combined with skin ulcers often indicates severe disease.In such cases,personalized treatment for the primary disease and infection prevention and control are essential.

摘要Objective: Electroacupuncture (EA) in the treating principle of “soothing the liver and regulating the kidney” was applied to intervion the rats with post-traumatic stress disorder (PTSD), and its effect on anxiety-like behavior, spatial learning and memory ability, and expressions of synaptophysin (SYN) and postsynaptic dense 95 (PSD95) in hippocampus of rats were observed to explore the underlying mechanism.Methods: Twenty-four male SD rats were randomly divided into control group, model group, and EA group, with 8 rats in each group. There was no intervention in the control group. The rest 16 rats were prepared for modeling. The single-prolonged stress & shock (SPS&S) method was used to establish the PTSD models. There was no intervention in the model group after modeling. In the EA group, “Bǎihuì (百会GV20)”“ Shéntíng (神庭GV24)”“Gānshū (肝俞BL18)”“Shènshū (肾俞BL23)” were manipulated with EA stimulation, intensity 1 mA, frequency 2/100 Hz, disperse-dense wave, and treatment was performed once a day, 20 min each time, for a total of 21 days. Open field test, elevated plus maze and Morris water maze tests were used to observe the behavioral differences of rats in each group. Immunohistochemical method was used to observe the differences of positive expressions of proteins SYN and PSD95 in hippocampus.Results: In the open field test, compared with the control group, the total traveling distance, the percentage of the time spent in the central cell and the numbers of the central cells crossing in the model group were all decreased (all P < 0.05). Compared with the model group, these indicators in the EA group were significantly all increased (all P < 0.05). In the elevated plus maze test, compared with the control group, the percentages of open arm staying time and entering times in the model group were both decreased (both P < 0.05). Compared with the model group, these indicators in the EA group were both significantly increased (both P < 0.05). The results of Morris water maze test showed that compared with the control group, the escape latency time and travelled distance of rats in the model group were all increased from day1 to day 4 (all P < 0.05), and the percentage of staying time in the target quadrant was decreased (P < 0.05). Compared with the model group, the escape latency time and travelled distance of EA group were both significantly shortened (both P < 0.05), and the percentage of staying time in the target quadrant was significantly increased (P < 0.05). The immunohistochemical results showed that, compared with the control group, the positive expressions of SYN and PSD95 in the hippocampus of the model group were significantly down-regulated (both P < 0.05). Compared with the model group, the positive expressions of SYN and PSD95 in the hippocampus of the EA group were significantly up-regulated (both P < 0.05).Conclusions: EA in the treating principle of “soothing the liver and regulating the kidney” can effectively relieve anxiety-like behavior and improve spatial learning and memory ability of rats with PTSD, and the mechanism is related to the up-regulation of the expressions of SYN and PSD95 in the hippocampus.

摘要Depression is a prevalent and severe mental health condition that is a primary cause of disability and suicide.Ginsenoside Rg1,an active ingredient derived from Panax ginseng C.A.Mey,has recently received considerable attention for its remarkable potential in treating depression.To review the existing evidence from preclinical studies on Rg1 and uncover its multi-target antidepressant mechanisms and pharmacological properties through a thorough search of relevant electronic databases.We searched digital repositories from inception until December 2024,e.g.,EMBASE,MEDLINE,Cochrane Library,and PubMed.We reviewed the antidepressant mechanism of Rg1,e.g.,inhabitation of inflammation,regulation of neurotransmitters,mediation of gap junction function of astrocytes,anti-oxidative stress,and anti-apoptosis.In addition to its potential implications for the clinical treatment of depression,we also analyzed its current limitations and put forward several novel research hypotheses as well as prospective research directions.It is hoped that Rg1 will be developed as a new treatment for clinical depression.

摘要The cathode interfacial layer (CIL) in organicsolar cells (OSCs) is crucial for the transport and collection ofcharge carriers. However, many cathode interfacial materials(CIMs) are unsuitable for printing manufacturing due to theirsensitivity to thickness. To tackle this issue, researchers havedeveloped a series of CIMs with n-type conjugated frame-works, particularly the naphthalene diimide (NDI) unit, owingto its high electron mobility and complementary absorptionwith commonly used active layers. Despite this, individualNDI molecules have a strong tendency to form large crystallinedomains, which can lead to interfacial defects in CILs. In thiswork, a different approach from other NDI-based CIMs wasadopted by substituting amino polar groups at the core posi-tion of NDI and polymerizing them into ionene-type CIMs.We designed and synthesized three self-doped polymer CIMsnamed PN-Pi, PN-Pe and PN-Eh. Among them, PN-Pi notablyreduces the work function of the Ag electrode, aligns inter-facial energies appropriately, smooths the active layer film andsuppresses carrier injection. This results in an impressivepower conversion efficiency (PCE) of 18.33% in the PM6:L8-BO system and maintains 90.4% PCE even at 127 nm thick-ness, ranking among the top film-thickness tolerance in theOSC field. This work demonstrates that combining conjugatedbackbone substitution with ionic polymerization is a promis-ing strategy for designing high-performance CIMs for OSCs.

摘要Apoplastic iron(Fe)in roots represents an essential Fe storage pool.Reallocation of apoplastic Fe is of great importance to plants experiencing Fe deprivation,but how this reallocation process is regulated remains elusive,likely because of the highly complex cell wall structure and the limited knowledge about cell wall biosynthesis and modulation.Here,we present genetic and biochemical evidence to demonstrate that the Cdi-mediated galactosylation of rhamnogalacturonan-II(RG-II)is required for apoplastic Fe reallocation.Cdi is expressed in roots and up-regulated in response to Fe deficiency.It encodes a putative glycosyltransferase localized to the Golgi apparatus.Biochemical and mass spectrometry assays showed that Cdi catalyzes the transfer of GDP-L-galactose to the terminus of side chain A on RG-II.Disruption of Cdi essentially decreased RG-II dimerization and hence disrupted cell wall formation,as well as the reallocation of apoplastic Fe from roots to shoots.Further transcriptomic,Fourier transform infrared spectroscopy,and Fe desorption kinetic analyses coincidently suggested that Cdi mediates apoplastic Fe reallocation through extensive modulation of cell wall components and consequently the Fe adsorption capacity of the cell wall.Our study provides direct evidence demonstrating a link between cell wall biosynthesis and apoplastic Fe reallocation,thus indicating that the structure of the cell wall is important for efficient usage of the cell wall Fe pool.

摘要Acetic acid is a common inhibitor present in lignocellulose hydrolysate,which inhibits the ethanol production by yeast strains.Therefore,the cellulosic ethanol industry requires yeast strains that can tolerate acetic acid stress.Here we demonstrate that overexpressing a yeast native arginase-encoding gene,CAR1,renders Saccharomyces cerevisiae acetic acid tolerance.Specifically,ethanol yield increased by 27.3%in the CAR1-overexpressing strain compared to the control strain under 5.0 g/L acetic acid stress.The global intracellular amino acid level and compositions were further analyzed,and we found that CAR1 overexpression reduced the total amino acid content in response to acetic acid stress.Moreover,the CAR1 overexpressing strain showed increased ATP level and improved cell membrane integrity.Notably,we demonstrated that the effect of CAR1 overexpression was independent of the spermidine and proline metabolism,which indicates novel mechanisms for enhancing yeast stress tolerance.Our studies also suggest that CAR1 is a novel genetic element to be used in synthetic biology of yeast for efficient production of fuel ethanol.

摘要“Prospection”refers to the ability to mentally simulate one’s future episodes.Negative bias in prospection,which includes both reduced positivity and enhanced negativity,is considered to be closely related to the development and maintenance of depression.However,there is a relative paucity of measures that adequately assess negative bias in prospection in depressed people.The current study developed and initially validated the Negative Bias in Prospection Scale(NBPS)using a nonclinical sample(n=959).The validity and reliability of the NBPS were investigated in several ways.Results suggested the 14 NBPS items loaded onto three factors,namely the“Increased negativity,”the“Reduced positivity,”and the“Overgeneralization.”The NBPS demonstrated decent internal consistency and test-retest reliability.Further,the NBPS was strongly associated with severity of depressive symptoms.Moreover,the NBPS was correlated in a theoretically meaningful way with other measures of future-oriented cognitions or negative biases.In conclusion,the NBPS has promising preliminary psychometric properties.It will be an efficient tool for exploring dysfunctional prospection in depression and provides a novel measure for mechanism of change in prospection-based interventions.
