Short-chain fatty acids(SCFAs)are key colonic metabolic regulators,yet microbiota endogenous biosynthesis often falls short of therapeutic requirements.To address this limitation,a colon-targeting delivery system was ...Short-chain fatty acids(SCFAs)are key colonic metabolic regulators,yet microbiota endogenous biosynthesis often falls short of therapeutic requirements.To address this limitation,a colon-targeting delivery system was developed by covalently coupling SCFAs with resistant dextrin(RD).FTIR and moderate molecular-weight distribution confirmed the esterification success and homogeneity.1H NMR verified preservation of the indi-gestible glycosidic bond,a structural prerequisite for resistance to upper gastrointestinal digestion.The esterified RD(ERD)demonstrated remarkable stability under simulated digestion.In vitro fermentation assays revealed high-substitution ERD induced approximately three-fold increase in total SCFAs production compared to RD.In vivo microbiome analysis showed ERD increased beneficial bacteria like Bacteroidota,Firmicutes,and Verruco-microbiota.Non-targeted metabolomics further demonstrated that distinct ERDs modulate host metabolism by regulating specific metabolic pathways.This study introduces ERD as a novel SCFA-delivery material to the colon,providing a novel strategy for the development of precision functional foods aimed at intestinal health.展开更多
