This structure is involved in the regulation of food intake through extracellular blood glucose concentrations, and plays a crucial role in obesity onset. In the present study, obesity models established with high fat feeding were treated with electroacupuncture at Zusanli (ST36)/ tnner Court (ST44) on the left side and Tianshu (ST25) bilaterally. We found that electroacupuncture could effectively reduce body weight and the fat-weight ratio, and decrease serum leptin, resistin, tumor necrosis factor alpha, and neuropeptide Y levels, while increase serum adiponectin and cholecystokinin-8 levels. This treatment altered the electrical activity of glucose-inhibited neurons in the lateral hypothalamic area, with electroacupuncture at Zusanli/Inner Court exerting an inhibitory effect, while electroacupuncture at bilateral Tianshu exerting an excitatory effect. These data suggest that electroacupuncture at the lower limbs and abdominal cavity is an effective means for regulating the activity of glucose-inhibited neurons in the lateral hypothalamic area and for improving the secretory function of adipose tissue.展开更多
Enhancing remyelination after injury is of utmost importance for optimizing the recovery of nerve function.While the formation of myelin by Schwann cells(SCs)is critical for the function of the peripheral nervous syst...Enhancing remyelination after injury is of utmost importance for optimizing the recovery of nerve function.While the formation of myelin by Schwann cells(SCs)is critical for the function of the peripheral nervous system,the temporal dynamics and regulatory mechanisms that control the progress of the s lineage through myelination require further elucidation.Here,using in vitro co-culture models,gene expression profiling of laser capture-microdissected SCs at various stages of myelination,and multilevel bioinformatic analysis,we demonstrated that SCs exhibit three distinct transcriptional characteristics duringmyelination:the immature,promyelinating,and myelinating states.We showed that suppressor interacting 3a(Sin3A)and 16 other transcription factors and chromatin regulators play important roles in the progress of myelination.Sin3A knockdown in the sciatic nerve or specifically in SCs reduced or delayed the myelination of regenerating axons in a rat crushed sciatic nerve model,while overexpression of Sin3A greatly promoted the remyelination of axons.Further,in vitro experiments revealed that Sin3A silencing inhibited SC migration and differentiation at the promyelination stage and promoted SC proliferation at the immature stage.In addition,SC differentiation and maturation may be regulated by the Sin3A/histone deacetylase2(HDAC2)complex functionally cooperating with Sox10,as demonstrated by rescue assays.Together,these results complement the recent genome and proteome analyses of SCs during peripheral nerve myelin formation.The results also reveal a key role of Sin3A-dependent chromatin organization in promoting myelinogenic programs and SC differentiation to control peripheral myelination and repair.These findings may inform new treatments for enhancing remyelination and nerveregeneration.展开更多
Among the various pregnancy complications,gestational diabetes mellitus(GDM)is one of the most prevalent and damaging adverse conditions.GDM is characterised by dysregulated gut microbiota and dysglycaemia.There is a ...Among the various pregnancy complications,gestational diabetes mellitus(GDM)is one of the most prevalent and damaging adverse conditions.GDM is characterised by dysregulated gut microbiota and dysglycaemia.There is a keen search for active ingredients that can modify gut dysbiosis and glycaemic imbalances.In this study,the effects of dietary supplementation with high fibre and galacto-oligosaccharides(GOS)on GDM were evaluated.We found that GOS effectively alleviated blood glucose,hepatic inflammatory response,and lipid metabolism in high-fat-induced GDM mice.Furthermore,GOS redressed GDM-associated gut dysbiosis and altered glycerolipid metabolic pathways.GOS supplementation significantly downregulated the levels of fat synthesis genes and inflammatory factors in the liver and upregulated the expression of lipolytic catabolic genes(Cyp4a10).In dietary GOS supplementation,Lachnospiraceae bacteria strongly cooccurred with saturated lysophosphatidylcholine in the glycerolipid metabolic pathway.Metagenomic analyses illustrated that Lachnospiraceae bacteria-hostedα-galactosidase genes enriched upon GOS intake.GOS supplementation reduces lipopolysaccharide(LPS)entry into the systemic circulation by repairing intestinal mucosal damage,inhibits inflammatory cell production by reducing hepatic and intestinal Toll-like receptor-4(TLR-4)expression and potentially ameliorates blood glucose dysregulation in GDM by modulating glycerophospholipid metabolism.These results revealed that dietary supplementation of GOS during pregnancy may be a promising intervention against GDM.展开更多
Nerve regeneration in adult mammalian spinal cord is poor because of the lack of intrinsic regeneration of neurons and extrinsic factors–the glial scar is triggered by injury and inhibits or promotes regeneration.Rec...Nerve regeneration in adult mammalian spinal cord is poor because of the lack of intrinsic regeneration of neurons and extrinsic factors–the glial scar is triggered by injury and inhibits or promotes regeneration.Recent technological advances in spatial transcriptomics(ST)provide a unique opportunity to decipher most genes systematically throughout scar formation,which remains poorly understood.Here,we frst constructed the tissue-wide gene expression patterns of mouse spinal cords over the course of scar formation using ST after spinal cord injury from 32 samples.Locally,we profled gene expression gradients from the leading edge to the core of the scar areas to further understand the scar microenvironment,such as neurotransmitter disorders,activation of the pro-infammatory response,neurotoxic saturated lipids,angiogenesis,obstructed axon extension,and extracellular structure re-organization.In addition,we described 21 cell transcriptional states during scar formation and delineated the origins,functional diversity,and possible trajectories of subpopulations of fbroblasts,glia,and immune cells.Specifcally,we found some regulators in special cell types,such as Thbs1 and Col1a2 in macrophages,CD36 and Postn in fbroblasts,Plxnb2 and Nxpe3 in microglia,Clu in astrocytes,and CD74 in oligodendrocytes.Furthermore,salvianolic acid B,a blood–brain barrier permeation and CD36 inhibitor,was administered after surgery and found to remedy fbrosis.Subsequently,we described the extent of the scar boundary and profled the bidirectional ligand-receptor interactions at the neighboring cluster boundary,contributing to maintain scar architecture during gliosis and fbrosis,and found that GPR37L1_PSAP,and GPR37_PSAP were the most signifcant gene-pairs among microglia,fbroblasts,and astrocytes.Last,we quantifed the fraction of scar-resident cells and proposed four possible phases of scar formation:macrophage infltration,proliferation and diferentiation of scar-resident cells,scar emergence,and scar stationary.Together,these profles delineated the spatial heterogeneity of the scar,confrmed the previous concepts about scar architecture,provided some new clues for scar formation,and served as a valuable resource for the treatment of central nervous system injury.展开更多

Spinal cord injury(SCI)is an incurable trauma that frequently results in partial or complete loss of motor and sensory function.Massive neurons are damaged after the initial mechanical insult.Secondary injuries,which ...Spinal cord injury(SCI)is an incurable trauma that frequently results in partial or complete loss of motor and sensory function.Massive neurons are damaged after the initial mechanical insult.Secondary injuries,which are triggered by immunological and inflammatory responses,also result in neuronal loss and axon retraction.This results in defects in the neural circuit and a deficiency in the processing of information.Although inflammatory responses are necessary for spinal cord recovery,conflicting evidence of their contributions to specific biological processes have made it difficult to define the specific role of inflammation in SCI.This review summarizes our understanding of the complex role of inflammation in neural circuit events following SCI,such as cell death,axon regeneration and neural remodeling.We also review the drugs that regulate immune responses and inflammation in the treatment of SCI and discuss the roles of these drugs in the modulation of neural circuits.Finally,we provide evidence about the critical role of inflammation in facilitating spinal cord neural circuit regeneration in zebrafish,an animal model with robust regenerative capacity,to provide insights into the regeneration of the mammalian central nervous system.展开更多

A central question in neural tissue engineering is how the tissue-engineered nerve(TEN)translates detailed transcriptional signals associated with peripheral nerve regeneration into meaningful biological processes.Her...A central question in neural tissue engineering is how the tissue-engineered nerve(TEN)translates detailed transcriptional signals associated with peripheral nerve regeneration into meaningful biological processes.Here,we report a skin-derived precursor-induced Schwann cell(SKP-SC)-mediated chitosan/silk fibroin-fabricated tissue-engineered nerve graft(SKP-SCs-TEN)that can promote sciatic nerve regeneration and functional restoration nearly to the levels achieved by autologous nerve grafts according to behavioral,histological,and electrophysiological evidence.For achieving better effect of neuroregeneration,this is the first time to jointly apply a dynamic perfusion bioreactor and the ascorbic acid to stimulate the SKP-SCs secretion of extracellular matrix(ECM).To overcome the limitation of traditional tissue-engineered nerve grafts,jointly utilizing SKP-SCs and their ECM components were motivated by the thought of prolongating the effect of support cells and their bioactive cues that promote peripheral nerve regeneration.To further explore the regulatory model of gene expression and the related molecular mechanisms involved in tissue engineering-aided peripheral nerve regeneration,we performed a cDNA microarray analysis of gene expression profiling,a comprehensive bioinformatics analysis and a validation study on the grafted segments and dorsal root ganglia tissues.A wealth of transcriptomic and bioinformatics data has revealed complex molecular networks and orchestrated functional regulation that may be responsible for the effects of SKP-SCs-TEN on promoting peripheral nerve regeneration.Our work provides new insights into transcriptomic features and patterns of molecular regulation in nerve functional recovery aided by SKP-SCs-TEN that sheds light on the broader possibilities for novel repair strategies of peripheral nerve injury.展开更多

摘要Spinal cord injury(SCI)is an incurable trauma that frequently results in partial or complete loss of motor and sensory function.Massive neurons are damaged after the initial mechanical insult.Secondary injuries,which are triggered by immunological and inflammatory responses,also result in neuronal loss and axon retraction.This results in defects in the neural circuit and a deficiency in the processing of information.Although inflammatory responses are necessary for spinal cord recovery,conflicting evidence of their contributions to specific biological processes have made it difficult to define the specific role of inflammation in SCI.This review summarizes our understanding of the complex role of inflammation in neural circuit events following SCI,such as cell death,axon regeneration and neural remodeling.We also review the drugs that regulate immune responses and inflammation in the treatment of SCI and discuss the roles of these drugs in the modulation of neural circuits.Finally,we provide evidence about the critical role of inflammation in facilitating spinal cord neural circuit regeneration in zebrafish,an animal model with robust regenerative capacity,to provide insights into the regeneration of the mammalian central nervous system.
摘要In this manuscript,we study fracture of prestressed cylindrical concrete pipes.Such concrete pipes play a major role in tunneling and underground engineering.The structure is modelled fully in 3D using three-dimensional continuum elements for the concrete structure which beam elements are employed to model the reinforcement.This allows the method to capture important phenomena compared to a pure shell model of concrete.A continuous approach to fracture is chosen when concrete is subjected to compressive loading while a combined continuous-discrete fracture method is employed in tension.The model is validated through comparisons with experimental data.

摘要A central question in neural tissue engineering is how the tissue-engineered nerve(TEN)translates detailed transcriptional signals associated with peripheral nerve regeneration into meaningful biological processes.Here,we report a skin-derived precursor-induced Schwann cell(SKP-SC)-mediated chitosan/silk fibroin-fabricated tissue-engineered nerve graft(SKP-SCs-TEN)that can promote sciatic nerve regeneration and functional restoration nearly to the levels achieved by autologous nerve grafts according to behavioral,histological,and electrophysiological evidence.For achieving better effect of neuroregeneration,this is the first time to jointly apply a dynamic perfusion bioreactor and the ascorbic acid to stimulate the SKP-SCs secretion of extracellular matrix(ECM).To overcome the limitation of traditional tissue-engineered nerve grafts,jointly utilizing SKP-SCs and their ECM components were motivated by the thought of prolongating the effect of support cells and their bioactive cues that promote peripheral nerve regeneration.To further explore the regulatory model of gene expression and the related molecular mechanisms involved in tissue engineering-aided peripheral nerve regeneration,we performed a cDNA microarray analysis of gene expression profiling,a comprehensive bioinformatics analysis and a validation study on the grafted segments and dorsal root ganglia tissues.A wealth of transcriptomic and bioinformatics data has revealed complex molecular networks and orchestrated functional regulation that may be responsible for the effects of SKP-SCs-TEN on promoting peripheral nerve regeneration.Our work provides new insights into transcriptomic features and patterns of molecular regulation in nerve functional recovery aided by SKP-SCs-TEN that sheds light on the broader possibilities for novel repair strategies of peripheral nerve injury.
