Differentiation of oligodendrocyte progenitor cells into mature myelin-forming oligodendrocytes contributes to remyelination.Failure of remyelination due to oligodendrocyte progenitor cell death can result in severe nerve damage.Ferroptosis is an iron-dependent form of regulated cell death caused by membrane rupture induced by lipid peroxidation,and plays an important role in the pathological process of ischemic stroke.However,there are few studies on oligodendrocyte progenitor cell ferroptosis.We analyzed transcriptome sequencing data from GEO databases and identified a role of ferroptosis in oligodendrocyte progenitor cell death and myelin injury after cerebral ischemia.Bioinformatics analysis suggested that perilipin-2(PLIN2)was involved in oligodendrocyte progenitor cell ferroptosis.PLIN2 is a lipid storage protein and a marker of hypoxia-sensitive lipid droplet accumulation.For further investigation,we established a mouse model of cerebral ischemiaeperfusion.We found significant myelin damage after cerebral ischemia,as well as oligodendrocyte progenitor cell death and increased lipid peroxidation levels around the infarct area.The ferroptosis inhibitor,ferrostatin-1,rescued oligodendrocyte progenitor cell death and subsequent myelin injury.We also found increased PLIN2 levels in the peri-infarct area that co-localized with oligodendrocyte progenitor cells.Plin2 knockdown rescued demyelination and improved neurological deficits.Our findings suggest that targeting PLIN2 to regulate oligodendrocyte progenitor cell ferroptosis may be a potential therapeutic strategy for rescuing myelin damage after cerebral ischemia. 展开更多

Aim:Glioma,the most common primary brain tumor,is known for its poor prognosis,limited treatment success,and high level of aggressiveness.Although cuproptosis-related genes have been linked to outcomes in other cancers,their role in glioma is still not well understood.Methods:By leveraging the Cancer Genome Atlas(TCGA)and additional databases,we conducted Cox regression and Kaplan-Meier analysis to determine the predictive importance of cuproptosis-related genes in individuals with glioma.By utilizing data from Gene Expression Omnibus(GEO)and other relevant databases,we examined how the expression of cuproptosis-associated genes correlates with immune cell infiltration,immunological checkpoint status,pathological stage,and histological grade.Additionally,we analyzed the connection between gene expression linked to cuproptosis and the prognosis in glioma patients.Results:Our newly developed cuproptosis-based glioma predictive model demonstrated promising prediction performance.Additionally,we identified glutaminase(GLS)in glioblastoma as a potential novel diagnostic indicator for glioma patients.Conclusion:GLS holds the potential to provide novel perspectives on cancer management and serve as a valuable diagnostic predictor for glioma patients. 展开更多

Lactoferrin is a member of the transferrin family of multifunctional iron binding glycoproteins.While numerous physiological functions have been described for lactoferrin,the mechanisms underlying these functions are not clear.To further study the functions and mechanisms of lactoferrin,we modified the lactoferrin promoter of mice using the CRISPR/Cas9 system to reduce or eliminate lactoferrin expression.Seven mice with lactoferrin promoter mutations were obtained with an efficiency of 24%(7/29)by injecting the plasmid pX330,expressing a small guide RNA and human codonoptimized SpCas9,into fertilized eggs of mice.Plasmid integration and off-targeting of pX330 were not detected.These results confirmed that pronuclear injection of a circular plasmid is a feasible and efficient method for targeted mutagenesis in mice. 展开更多
