Background:Acupuncture is recognized as an alternative therapy for rheumatoid arthritis(RA)pain,but its efficacy evaluations are often confounded by variability in sham acupuncture techniques.The accurate selection of sham acupuncture controls,which are administered at either therapeutic acupuncture points or non-acupuncture points,is crucial for the validity of assessment outcomes.Objective:To assess the efficacy of acupuncture in treating RA pain and identify the most effective acupuncture methods.Search strategy:Databases including MEDLINE,Embase,Pub Med,Cochrane Library,Scopus,Web of Science,China National Knowledge Infrastructure Database,Chinese Science and Technology Journal Database,and Wanfang Database were searched from inception to October 11,2024.Keywords included‘‘rheumatoid arthritis,”‘‘acupuncture,”‘‘electroacupuncture,”and‘‘pain.”Inclusion criteria:Randomized controlled trials(RCTs)in adults with RA that assessed pain using a visual analog scale and joint swelling by swollen joint count.Eligible trials compared electroacupuncture,conventional acupuncture,or sham acupuncture,against standard pain medication.Data extraction and analysis:Two reviewers independently extracted data on study design,participant characteristics,interventions and outcomes.Risk of bias was evaluated using the Cochrane Risk of Bias2 tool,and evidence certainty was assessed via the confidence in network meta-analysis framework.A frequentist network meta-analysis with random-effect models was conducted,and standardized mean difference(SMD)and 95%confidence interval(CI)were reported.Results:Ten RCTs involving 704 participants were analyzed.Electroacupuncture(SMD:–1.42;95%CI:[–1.87,–0.98])and conventional acupuncture(SMD:–1.11;95%CI:[–1.49,–0.73])outperformed conventional therapy and non-acupoint sham needling.Surface under cumulative ranking curve showed that electroacupuncture was most effective for pain reduction(97.7%),followed by conventional acupuncture(75.1%),non-acupoint sham(29.1%),same-acupoint sham(28.6%),and conventional therapy(19.5%).Conclusion:Electroacupuncture demonstrated the highest efficacy for RA pain relief.Same-acupoint sham acupuncture may underestimate acupuncture's true effect and is not recommended as a placebo control.Non-acupoint sham acupuncture is a more valid control for future trials. 展开更多

Objective To analyze the research status and hotspots,and evolutionary trend of publications on Jin’s three-needle therapy from 2000 to 2025,and to provide bibliometric evidence for the theoretical development and clinical expansion of this distinctive Lingnan(the area south of the Nanling Mountains in China)acupuncture-moxibustion therapy.Methods Relevant publications on Jin’s three-needle therapy were retrieved from China National Knowledge Infrastructure,VIP Database,Wanfang Data Knowledge Service Platform,and Chinese Biomedical Literature Database from January 1,2000 to July 9,2025.Two researchers independently screened and extracted data.Microsoft Excel was used for data management and disease spectrum analysis based on ICD-11 classification.CiteSpace software was employed to analyze publication trend,collaboration network,co-occurrence network,and keyword evolution.Results A total of 1295 publications were included,showing an overall upward trend in annual output.Research mainly focused on diseases of the nervous system(45.17%),those of the musculoskeletal system or connective tissue(28.88%),and mental,behavioral or neurodevelopmental disorders(9.11%),accounting for over 80%in total.The study types were predominantly clinical research,the proportion of mechanistic research was relatively low,and mechanistic research was weak across all core fields.Collaboration network analysis revealed low density between authors and institutions,exhibiting a“core-edge”structure with Professor Rui JIN’s inheritance team(Guangzhou University of Chinese Medicine and its affiliated hospitals)as the core and widespread in periphery,demonstrating a distinct geographical clustering characteristic in the Lingnan region.Keyword evolution indicated that the research focus had expanded from neurological rehabilitation to multisystem diseases,and the attention had been drawn in multidimensional outcomes such as quality of life and motor function.Conclusion The research scale of Jin’s three-needle therapy has significantly expanded,demonstrating a relatively concentrated application distribution across three major domains:neurological disorders,musculoskeletal or connective tissue diseases,and mental,behavioral,or neurodevelopmental disorders.However,there are some shortcomings in the research of diseases,such as insufficient exploration of mechanisms and limited cooperation between teams and institutions.Future efforts should focus on strengthening mechanism research,conducting multicenter trials,and expanding inter-institutional collaboration to establish a more comprehensive evidence-based foundation,thereby enhancing the clinical reproducibility and academic influence of Jin’s three-needle therapy. 展开更多

Postoperative ileus remains a significant challenge in surgical recovery.Increasing evidence highlights the gut-brain axis as a promising target for managing gastrointestinal motility disorders.Acupuncture,particularly at Zusanli(ST36),has shown considerable potential to regulate gut function,although its mechanisms warrant further investigation.This editorial discusses a clinical study demonstrating that electroacupuncture at Zusanli(ST36)accelerates gastrointestinal recovery in patients with postoperative ileus by activating the vagus nervecholinergic anti-inflammatory pathway,suppressing pro-inflammatory cytokines,and rebalancing gastrointestinal hormones.We further summarize how acupuncture modulates the gut-brain axis through neural,immune,endocrine,microbial,and central pathways,supporting integration with modern neuroimmunology.These findings support incorporating acupuncture into enhanced recovery after surgery protocols.Future studies should prioritize mechanistic investigations using multi-omics technologies,large-scale clinical trials to enable standardization,and exploration of acupuncture-microbiota-gut-brain interactions to broaden clinical applications in gastrointestinal disorders. 展开更多

Diabetes is a major global metabolic disorder,with the type 2 diabetes mellitus(T2DM)population in China expected to reach 168 million by 2050.Electroacupuncture(EA)and auricular acupuncture represent safe,accessible,and multitargeted strategies for glycemic control with strong translational potential.Clinical studies indicate that EA,targeting trunk and limb acupoints,can significantly reduce fasting plasma glucose and glycated hemoglobin.Mechanistically,EA modulates the neuro-endocrine-immune axis,regulates gut microbiota,and activates insulin signaling pathways.Auricular acupuncture,through vagal innervation,may also exert rapid effects on glucose homeostasis via autonomic modulation.The review’s objective is to synthesize and critically evaluate current clinical trials and animal studies on EA and auricular acupuncture for glycemic control in diabetes,particularly T2DM,and to highlight translational implications,mechanistic insights,and evidence gaps.This review also emphasizes translational considerations by highlighting species differences between rodents and humans-an underappreciated yet critical factor influencing the clinical applicability of preclinical findings.Through the integration of recent research advances,the present review not only consolidates clinical and preclinical evidence but also advances a deeper mechanistic framework and underscores the importance of species-specific factors in bridging experimental research and patient care.Future progress requires rigorously designed,adequately powered,multicenter randomized controlled trials with standardized protocols to validate efficacy and define their role in precision diabetes management. 展开更多

Chronic pain is often accompanied by depressive emotions,and they are interacted with each other,which is one of the crucial factors to affect the clinical efficacy on pain.Acupuncture therapy has the advantages of multi-dimensional and multi-target regulation,and it is remarkably effective on chronic pain and pain-related depressive emotions.This paper reviews the mechanism researches of acupuncture in treatment of chronic pain with depression.Research indicates that the primary molecular mechanism by which acupuncture exerts analgesic and anti-depressive effects involves the regulation of central nervous system neurotransmitters,including 5-hydroxytryptamine,glutamic acid,gamma-aminobutyric acid,and dopamine.Additionally,acupuncture inhibits inflammatory responses within the central nervous system,modulates the expression of neurotrophic factors,enhances neuronal synaptic plasticity,and regulates the BDNF/TrkB/CREB and ERK signaling pathways.These findings underscore the multifaceted and multi-targe ted regulatory advantages of acupuncture. 展开更多

OBJECTIVE:To observe the effect of acupuncture on astrocyte activation following cerebral ischemiareperfusion injury(CIRI)via the p38 mitogen-activated protein kinase(p38 MAPK)/mitogen-and stress-activated protein kinase 1(MSK1)signaling pathway.METHODS:Transcriptome and single-cell sequencing were used to analyse gene expression in middle cerebral artery occlusion(MCAO)-induced rats.Acupuncture was applied at Baihui(GV20)and Dazhui(GV14)for 7 d.The infarct volume was assessed via 2,3,5-triphenyltetrazolium chloride staining and T2-weighted imaging.Serum interleukin-1 beta(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-alpha levels were measured via enzyme-linked immunosorbent assay.The expression of glial fibrillary acidic protein(GFAP),complement component 3(C3),and S100 calciumbinding protein A10(S100A10),as well as components of the p38 MAPK/MSK1 pathway,was examined by Western blotting and immunohistochemistry.RESULTS:CIRI activated p38 MAPK signaling,increased the expression of A1 markers(GFAP and C3),and proinflammatory cytokines,and decreased the expression of the A2 marker S100A10.Acupuncture inhibited p38 phosphorylation,upregulated MSK1,reduced C3 and inflammatory cytokines,increased S100A10 expression,decreased infarct volume,and improved neurological function.CONCLUSION:Acupuncture protects against ischemic stroke by targeting the p38 MAPK/MSK1 pathway.It inhibits p38 MAPK phosphorylation and activates MSK1,thereby promoting a shift in astrocyte polarization from the pro-inflammatory A1 to the reparative A2 type,as reflected by decreased C3 and increased S100A10 expression.This shift reduces pro-inflammatory cytokines,alleviates cerebral inflammation,and promotes neural repair,ultimately diminishing infarct volume,restoring neurons,and improving behavior.These findings elucidate the mechanism of acupuncture and support its clinical use. 展开更多

Acupuncture research increasingly involves heterogeneous and multimodal data that are difficult to analyze using conventional methods.This review summarizes data-driven approaches in acupuncture research within a framework encompassing intervention,response,and contextual data.We discuss causal inference,artificial intelligence,text mining,and integrative analysis,along with their applications in efficacy evaluation,outcome prediction,mechanistic investigation,and clinical decision support.These approaches shift the focus of acupuncture research from population-level average effects toward individualized clinical decision-making by enabling the analysis of treatment heterogeneity and underlying mechanisms.However,current research remains limited by inadequate data standardization,insufficient external validation,and limited model interpretability.Despite these challenges,data-driven approaches offer substantial promise for advancing more rigorous and personalized acupuncture research. 展开更多

Post-stroke cognitive impairment(PSCI)is a common disabling complication after stroke.Acupuncture can effectively improve learning and memory function in patients with PSCI and enhance their quality of life in long term.Research on its effect mechanisms has evolved from phenomenological observation to multidimensional exploration progressively.In this paper,recent studies on effect mechanisms of acupuncture for PSCI were collected,which revealed that acupuncture can alleviate PSCI through multi-target synergism.The therapeutic mechanism of acupuncture primarily involves reducing immune damage in the brain by suppressing neuroinflammation,and promoting synaptic and memory circuit reconstruction through enhancing neural plasticity,egulating autophagy and maintaining the stability of neurovascular units as the critical auxiliary pathways,which can synergistically remove the damage products and preserve the neuronal microenvironment.Furthermore,emerging research directions such as modulating gut microbiota,influencing epigenetics,and improving brain metabolism expand the mechanistic understanding.However,existing studies predominantly isolate and describe individual mechanisms without integrating their intrinsic correlations,nor do they adequately elucidate the corresponding relationships between different acupuncture techniques and their therapeutic advantages.Therefore,this review integrates the fragmented mechanistic evidences,and emphasizes that future research need multi-omics approaches,so as to explore the synergistic relationships among mechanisms,and to strengthen the correlative research between the specificity of acupuncture regimens and clinical effect,in order to facilitate bench-to-bedside translation. 展开更多

Objective To systemically evaluate the effectiveness and safety of acupuncture and moxibustion for chronic eczema,and to provide evidence for acupuncture and moxibustion in the treatment of chronic eczema.Methods Randomized controlled trials(RCTs)were retrieved from PubMed,Cochrane Library,China National Knowledge Infrastructure(CNKI),VIP database,Wanfang database and China Biology Medicine disc(CBM),from the inception of the database to April 23,2025.The control group were sham acupuncture and moxibustion,no treatment,or conventional western medication.The intervention group were acupuncture and moxibustion therapy or acupuncture and moxibustion on the basis of the interventions as the control group.Meta-analysis was performed by using RevMan5.4.0 software according to Cochrane systematic evaluation method.Results Ultimately,53 RCTs were included.The results of the meta-analysis showed that compared with sham acupuncture and moxibustion,no treatment,or conventional western medication,acupuncture and moxibustion treatment for eczema area and severity index(EASI)score(mean difference[MD]=−2.23,95%confidence interval[CI]−2.61 to−1.85;P<0.00001),visual analogue scale(VAS)score(standardized mean difference[SMD]=−1.45,95%CI−2.04 to−0.87;P<0.00001),dermatology life quality index(DLQI)score(MD=−2.36,95%CI−3.25 to−1.48;P<0.00001),immunoglobulin E(IgE)(SMD=−1.26,95%CI−2.46 to−0.05;P=0.04),the overall response rate(odds ratio[OR]=4.58,95%CI 3.84 to 5.46;P<0.00001)and the recurrence rate(OR=0.14,95%CI 0.09 to 0.21;P<0.00001)were all better than those in the control group,and there were no severe adverse events reported.Conclusion Acupuncture and moxibustion can reduce the lesion area and severity of the patients with chronic eczema,effectively alleviate pruritus,improve the quality of life and decrease serum IgE.This therapy owns high overall response rate with low recurrence rate,and without severe adverse events,which is associated with a favorable long-term prognosis. 展开更多

OBJECTIVE:To assess the impact of body mass index(BMI)on the effectiveness of acupuncture in treating postprandial distress syndrome(PDS).METHODS:Data from 278 patients enrolled in a prior multicenter,randomized,sham-controlled trial were analyzed in this secondary study.Patients were classified into three groups according to baseline BMI:low,normal,and high.The primary outcome was the response rate after 4 weeks of treatment.The χ2 test was used to examine the effect of BMI on acupuncture efficacy.After multiple imputation for missing data,logistic regression analysis was performed to evaluate this association.A sensitivity analysis was conducted by combining the low and normal BMI groups.In addition,BMI was analyzed as a continuous variable using logistic regression to explore its relationship with treatment efficacy in the acupuncture group.RESULTS:The χ2 test revealed a statistically significant difference,with the acupuncture group demonstrating a markedly higher response rate than the sham group(82.91%vs 51.79%,χ2=25.34,P<0.001).In both the normal and high BMI categories,the response rate in the acupuncture group(82.91%)was higher than that in the sham acupuncture group(48.21%)(χ2=25.34,P<0.001).Following multiple imputation,logistic regression analysis demonstrated a significant overall association between BMI category and treatment response(P<0.001).Acupuncture showed superior efficacy compared with sham acupuncture[OR=1.97,95%CI(1.34,2.90),P<0.001].At week 4,response rates were 68.4%in the low BMI group,81.7%in the normal BMI group,and 96.3%in the high BMI group(χ2=6.303,P=0.043).A significant difference was observed only between the low and high BMI groups(χ2=6.717,P=0.010).Sensitivity analysis showed that the high BMI group achieved significantly better outcomes than the non-high BMI group in elimination rates at week 8 and week 12(χ2=4.571,P=0.033;χ2=4.589,P=0.032).Logistic regression analysis indicated that BMI was an independent predictor of response rate at week 4[OR=1.186,95%CI(1.010,1.394),P=0.038]and elimination rate at week 12[OR=1.126,95%CI(1.002,1.265),P=0.047],whereas no significant associations were observed at other follow-up points.CONCLUSION:This subgroup analysis indicated that acupuncture improved PDS symptoms across different BMI categories.Acupuncture was suggested as a viable therapeutic option for patients with PDS,particularly those with a higher BMI. 展开更多

Objective:To systematically analyze the research hotspots and developmental trajectory of acupuncture ophthalmology using bibliometric methods,with the aim of clarifying research trends,collaborative networks,the evolution of thematic focuses,and predicting future directions.Methods:Literature on acupuncture ophthalmology published between January 1,1985,and May 31,2025,was retrieved from the Web of Science Core Collection.Bibliometric analyses were conducted using CiteSpace 6.2.R3,VOSviewer 1.6.20,and SCImago Graphics to visualize publication characteristics,country and regional contributions,authorship patterns,journals,institutions,keywords,and cited references.Results:A total of 349 publications were included.(1)Annual publications:The number of studies on acupuncture in ophthalmology showed a generally increasing trend from 1985 to 2025.(2)Countriesegions:Research in this field spanned 33 countries and regions,with China(203 papers),the United States(55 papers),and South Korea(25 papers)as the top three contributors.(3)Institutions and authors:A total of 321 institutions and 621 authors were involved.Authors with more than three publications formed seven collaborative research teams,comprising three major collaboration networks.Xiao-peng MA was the most prolific author(12 papers),whereas Tae-Hun Kim was the most influential author(41 citations).(4)Journals:The 349 articles appeared in 152 journals,with Medicine(32 articles)and Evidence-Based Complementary and Alternative Medicine(18 articles)being the most common publication venues.(5)Keyword and reference analyses identified several major thematic areas,including dry eye syndrome,systematic reviews,etiology identification,secondary glaucoma,visual symptoms,retinal pigment research,and myopia control.Conclusion:This bibliometric analysis demonstrates that current research in acupuncture ophthalmology is primarily focused on disorders in which acupuncture shows notable clinical benefits,incorporating multimodal therapies and multidisciplinary approaches.The field is progressing toward greater protocol standardization and clearer visualization of underlying mechanisms.Acupuncture has become an important complementary and alternative therapeutic modality within ophthalmic care,offering unique advantages in symptom improvement,enhancement of quality of life,and reduction of medication dependence for conditions such as dry eye disease,asthenopia,and myopia.Future research priorities include:(1)deepening mechanistic research on acupuncture for ophthalmic diseases;(2)advancing integration with modern ophthalmology in chronic disease management and prevention;and(3)generating high-level clinical evidence to establish standardized acupuncture regimens for eye health. 展开更多

Objective:To observe the potential mechanisms of Tongdu Tiaoshen(unblocking the governor vessel and regulating the mind)acupuncture therapy in protecting against cerebral ischemia-reperfusion injury(CIRI).Methods:SD rats were divided into a sham-operated group,a model group,and a Tongdu Tiaoshen acupuncture group(acupuncture group)randomly,with 12 rats in each group.Middle cerebral artery occlusion models were prepared using the Longa suture-occluded method.In the acupuncture group,acupuncture was operated at“Dazhu(GV14)”,“Shenting(GV24)”and“Baihui(GV20)”for 20 min in each intervention,once daily and for 12 consecutive days.In the sham-operated group and the model group,the rats were grasped and fixed for the same duration as those in the acupuncture group,but without acupuncture intervention.The degree of neurological impairment in rats after intervention was assessed using the Zea Longa method.HE staining was employed to observe the pathological changes in the rat cerebral cortex.Western blotting was used to detect the protein expression levels of apoptosis-associated speck-like protein containing a CARD(ASC),pro-caspase-1,and P2×7 receptor(P2×7R)in the rat cerebral cortex.ELISA was performed to measure the levels of interleukin(IL)-1βand IL-18 in the rat cerebral cortex.Immunofluorescence was utilized to observe the positive expression of ASC,pro-caspase-1,and P2×7R in the rat cerebral cortex.Results:Compared to the sham-operated group,the model group showed the increase in neurological deficit score(P<0.01),the cells surrounding the ischemic foci were sparse and loosely distributed under the microscope;the protein expression of ASC,pro-caspase-1 and P2×7R were elevated(P<0.01),their positive expression increased;and the levels of IL-1βand IL-18 rose(P<0.01).Compared to the model group,in the acupuncture group,the neurological deficit score decreased(P<0.05),and cellular morphological changes were mild around the ischemic foci observed under microscopy;the protein expression of ASC,pro-caspase-1 and P2×7R was reduced(P<0.01)and their positive expression was declined;and the expression of IL-1βand IL-18 decreased(P<0.01).Conclusion:The mechanisms of Tongdu Tiaoshen acupuncture in protecting against CIRI may be associated with down-regulating the expression of P2×7R and attenuating inflammatory response. 展开更多

The field of acupuncture is currently facing paradigmatic issues,including the difficulty of turning theoretical strengths into technical discourse power,a gap between mechanistic research and clinical imperatives,and the fragmentation of research activities.Therefore,constructing a research paradigm grounded in traditional Chinese medicine theory and validated through contemporary scientific methods is essential.Hand Twelve Jing-Well Points(HTWP)acupuncture,an ancient therapy known for its rapid efficacy and practical application,offers an optimal solution for these challenges.In this study,the theoretical foundations,clinical effectiveness,biological explanations,and Translational applications of HTWP acupuncture were consolidated by integrating a four-step research framework.Initially,we developed the“Well point-Brain Connection”framework rooted in classical theories and informed by the“Root-Knot”concept.Subsequent clinical practice provided evidence of its effectiveness in modulating awareness and facilitating motor-cognitive rehabilitation for patients with central nervous system issues.Moreover,mechanistic research provides scientific verification of a“Sensation-Transmission-Effect”cascade that encompasses the activation of arousal circuits and the repair of the blood-brain barrier.Ultimately,the promotion of translation,standardization of systems,and the use of wearable technologies have aided the transition from passive therapy to proactive health management.This closed-loop concept offers compelling evidence for the“Well point-Brain Connection”and provides an applicable framework for scientific innovation and global distribution of innovative acupuncture hypotheses. 展开更多

Functional near-infrared spectroscopy (fNIRS) is predominantly employed to investigate the atypical response characteristics of the cerebral cortex in neurological disorders. Recently, this technology has been applied to examine the neural mechanisms underlying the use of acupuncture in the treatment of insomnia. This article provides a comprehensive review and analysis of the current applications of fNIRS in the contexts of sleep-wake cycles, insomnia, and acupuncture-based interventions for insomnia. The findings indicate that existing research predominantly focuses on the prefrontal cortex, with limited studies addressing the temporal, occipital, and parietal lobes. fNIRS can be integrated with various data analysis methodologies or multimodal brain functional imaging techniques to investigate the regulatory effects of acupuncture on functional connectivity both among and within brain regions, as well as on the efficiency of information transmission. This approach may elucidate the neural mechanisms of acupuncture in combating insomnia from diverse perspectives and offer an enhanced framework for the diagnosis and treatment of insomnia. 展开更多

BACKGROUND Depression is a prevalent global mental health issue with substantial disease burden.Although pharmacotherapy remains the first-line treatment,its efficacy and tolerability are limited.Acupuncture,a core modality in traditional Chinese medicine,has shown promise in improving depressive symptoms via neurotransmitter modulation.However,prior meta-analyses often considered manual and electroacupuncture together,increasing heterogeneity.This review focuses solely on traditional acupuncture to clarify its therapeutic role and provide evidence for standardized clinical application.AIM To evaluate the efficacy of acupuncture in patients with depression and explore its feasibility as an adjunctive treatment.METHODS Twenty randomized controlled trials on traditional acupuncture for depression—whose quality was assessed using the Cochrane tool—were included in this meta-analysis.Primary outcomes were the Hamilton Depression Rating Scale(HAM-D),Beck Depression Inventory-II(BDI-II),Self-Rating Depression Scale(SDS),and MontgomeryÅsberg Depression Rating Scale(MADRS).Secondary outcomes were the Pittsburgh Sleep Quality Index(PSQI),Hamilton Anxiety Rating Scale(HAMA),Self-Rating Scale for the Side Effects(SERS),and serum 5-hydroxytryptamine(5-HT)levels.RevMan 5.4 software was employed for the meta-analysis.RESULTS Results revealed that acupuncture significantly reduced HAM-D,SDS,and MADRS scores.Although BDI-II scores improved in some studies,its overall effect was not statistically significant.Acupuncture also significantly improved HAMA and PSQI scores and increased serum 5-HT levels.Notably,SERS analysis indicated that acupuncture can significantly alleviate adverse reactions associated with antidepressants.However,most pooled results revealed substantial heterogeneity.CONCLUSION The efficacy of traditional acupuncture improving symptoms,sleep,and anxiety and alleviating antidepressant side effects is highly encouraging.However,the low quality of the included studies warrants cautious interpretation. 展开更多

BACKGROUND Neural stem cells(NSCs)transplantation is a promising clinical therapy for Alzheimer’s disease(AD).The Notch and Wnt signaling pathways play important roles in the biological functions of NSCs,and microRNA-124(miR-124)regulates these pathways through its regulatory effects.AIM To explore the mechanism of acupuncture in enhancing the function of transplanted NSCs and their therapeutic potential in AD.METHODS This study utilized enzyme-linked immunosorbent assay,western blotting,and real-time fluorescent quantitative polymerase chain reaction,to investigate the effects of acupuncture on the role of miR-124 in regulating the Notch and Wnt signaling pathways,in NSCs transplantation therapy in a mouse model of AD-senescence-accelerated mouse prone 8 mice.An in vitro coculture model of mouse hippocampal brain slices and NSCs was established,and flow cytometry was used to examine the effects of acupuncture on the regulation of cyclin D1,an interactive protein in the Notch and Wnt signaling pathways,and on NSCs proliferation and differentiation.RESULTS Acupuncture significantly improved cognitive impairment in AD mice after NSCs transplantation(P<0.05);inhibited expression of characteristic pathological biomarkers of AD(P<0.05);and upregulated expression of NSCs-specific neuroproliferation and differentiation biomarkers(P<0.05).Upregulation of miR-124 modulated the key target genes Notch homolog 1,hairy and enhancer of split 5,and glycogen synthase kinase 3βin the Notch and Wnt signaling pathways(P<0.05);regulated the Notch and Wnt dual signaling pathways and achieved interaction(P<0.05);promoted NSCs proliferation and differentiation(P<0.05);restored damaged cells;and slowed the progression of AD.CONCLUSION Acupuncture may improve the hippocampal microenvironment by upregulating miR-124 to regulate the Notch and Wnt dual signaling pathways,promote NSCs proliferation and differentiation,facilitate the repair of damaged neurons,integrate neural circuits,restore biological functions,and improve cognitive impairment in AD mice. 展开更多
