Acupuncture therapy has demonstrated therapeutic effects on various systemic diseases through the use of specific acupoints along meridians. However, due to the multi-target effect of acupuncture and the complexity of meridian pathways, the underlying mechanisms of acupuncture have not been well understood.Tissue clearing and three-dimensional(3D)imaging is an emerging medical technology that provides high-resolution 3D molecular information while preserving the structural integrity of tissues. In relevant research articles,tissue-clearing and 3D imaging techniques stood out for their ability to visualize the structural characteristics of meridians and acupoints of intact tissues,in order to reveal their intricate connections by means of topological principles.Specifically, this technology has managed to show that acupuncture points like Guilai(ST29)and Sanyinjiao(SP6),which are commonly used in traditional Chinese medicine for treating gynecological disorders, promote angiogenesis in the ovaries and thus improve follicle numbers in polycystic ovary syndrome-like ovaries. This article summarizes recent progress on tissue-clearing and 3D imaging technologies and anticipates the prospects of utilizing this technology to further explore the effects of acupuncture. 展开更多

The increasing complexity of China’s electricity market creates substantial challenges for settlement automation, data consistency, and operational scalability.Existing provincial settlement systems are fragmented, lack a unified data structure, and depend heavily on manual intervention to process high-frequency and retroactive transactions. To address these limitations,a graph-based unified settlement framework is proposed to enhance automation, flexibility, and adaptability in electricity market settlements.A flexible attribute-graph model is employed to represent heterogeneousmulti-market data, enabling standardized integration, rapid querying, and seamless adaptation to evolving business requirements. An extensible operator library is designed to support configurable settlement rules, and a suite of modular tools—including dataset generation, formula configuration, billing templates, and task scheduling—facilitates end-to-end automated settlement processing.A robust refund-clearing mechanism is further incorporated, utilizing sandbox execution,data-version snapshots, dynamic lineage tracing, and real-time changecapture technologies to enable rapid and accurate recalculations under dynamic policy and data revisions.Case studies based on real-world data from regional Chinese markets validate the effectiveness of the proposed approach, demonstrating marked improvements in computational efficiency, system robustness, and automation. Moreover, enhanced settlement accuracy and high temporal granularity improve price-signal fidelity, promote cost-reflective tariffs, and incentivize energy-efficient and demand-responsive behavior among market participants. The method not only supports equitable and transparent market operations but also provides a generalizable, scalable foundation for modern electricity settlement platforms in increasingly complex and dynamic market environments. 展开更多

The construction of spot electricity markets plays a pivotal role in power system reforms, where market clearing systems profoundly influence market efficiency and security.Current clearing systems predominantly adopt a single-system architecture, with research focusing primarily on accelerating solution algorithms through techniques such as high-efficiency parallel solvers and staggered decomposition of mixed-integer programming models.Notably absent are systematic studies evaluating the adaptability of primary-backup clearing systems incontingency scenarios—a critical gap given redundant systems’expanding applications in operational environments. This paper proposes a comprehensive evaluation framework for analyzing dual-system adaptability, demonstrated through an in-depth case study of the Inner Mongolia power market.First,we establish the innovative“Dual-Active Heterogeneous”architecture that enables independent parallelized operation and fault-isolated redundancy.Subsequently, key performance indices are quantitatively evaluated across four critical dimensions:unit commitment decisions, generator output constraints, transmission section congestion patterns, and clearing price formation mechanisms. An integrated fuzzy evaluation methodology incorporating grey relational analysis is employed for objective indicator weighting, enabling systematic quantification of system superiority under specific grid operating states.Empirical results based on actual operational data from 200 generation units demonstrate the framework’s efficacy in guiding optimal system selection, with particularly strong performance observed during peak load periods. The proposed approach shows high generalization potential for other regional markets employing redundant clearing mechanisms—particularly those with increasing renewable penetration and associated uncertainty. 展开更多

Glycerophosphodiester phosphodiesterase 1(GDPD1)plays an important function in the abiotic stress responses and participates in the accumulation of sn-glycerol-3-phosphate(G3P)in plants, which is key to plant systemic acquired resistance(SAR).However, the role of GDPD1 in plant responses to biotic stress remains poorly understood. This study characterized the antivirus function of the GDPD1 gene(designated as ClGDPD1)from Eureka lemon.ClGDPD1 is located in the membrane and endoplasmic reticulum, where it interacts with the citrus yellow vein clearing virus(CYVCV)coat protein(CP).Compared to individually expressed ClGDPD1 or coexpressed ClGDPD1+CP140-326,transiently coexpressed ClGDPD1+CP or ClGDPD1+CP1-139 significantly upregulated the key substance contents and genes expression involved in glycerophospholipid metabolism.Over-expression of ClGDPD1 significantly facilitated the accumulation of G3P,upregulated the expression of SAR-related genes, and increased the resistance of transgenic Eureka lemon to CYVCV infection.Furthermore, exogenous glycerol treatment and over-expression of ClGPDH increased the G3P content and reduced CYVCV titers in plants or hairy roots. These results indicated that the enhanced resistance of ClGDPD1 transgenic Eureka lemon to CYVCV may be due to facilitating G3P accumulation through the interaction of ClGDPD1 with CP.Our findings provide novel insights into the role of ClGDPD1 as an important regulatory center in mediating the citrus defense response to viral infections. 展开更多

Tissue optical clearing by use of optical clearing agents(OCAs)has been proven to have potential to reduce the highly scattering effect of biological tissues in optical techniques. However, the difference in tissue samples could lead to unreliable results, making it difficult to quantitatively control the dose of OCAs during the course of tissue optical clearing. In this work,in order to study the effects of optical clearing,we customized tissue-like phantoms with optical properties of some biological tissue.Diffuse reflectance and total transmittance of tissue-like phantoms with different OCAs(DMSO or glycerol)and porcine skin tissues were measured. Then optical property parameters were calculated by inverse adding-doubling(IAD)algorithm.Results showed that OCAs could lead to a reduction in scattering of tissue-like phantoms as it did to porcine skin tissue in vitro.Furthermore,a series of relational expressions could be fit to quantitatively describe the relationship between the doses of OCAs and the reduction of scattering effects. Therefore, proper tissue-like phantom could facilitate optical clearing to be used in quantitative control of tissue optical properties, and further promote the application potential of optical clearing to light-based noninvasive diagnostic and therapeutic techniques. 展开更多

Morphological analyses are key outcome assessments for nerve regeneration studies but are historically limited to tissue sections.Novel optical tissue clearing techniques enabling three-dimensional imaging of entire organs at a subcellular resolution have revolutionized morphological studies of the brain. To extend their applicability to experimental nerve repair studies we adapted these techniques to nerves and their motor and sensory targets in rats. The solvent-based protocols rendered harvested peripheral nerves and their target organs transparent within 24 hours while preserving tissue architecture and fluorescence. The optical clearing was compatible with conventional laboratory techniques, including retrograde labeling studies, and computational image segmentation, providing fast and precise cell quantitation.Further, optically cleared organs enabled three-dimensional morphometry at an unprecedented scale including dermatome-wide innervation studies, tracing of intramuscular nerve branches or mapping of neurovascular networks.Given their wide-ranging applicability, rapid processing times, and low costs, tissue clearing techniques are likely to be a key technology for next-generation nerve repair studies. All procedures were approved by the Hospital for Sick Children’s Laboratory Animal Services Committee(49871/9)on November 9,2019. 展开更多

Citrus yellow vein clearing virus (CYVCV) is considered as the causal agent of Citrus yellow vein clearing disease and belongs to the genus Mandarivirus in the family Alphaflexiviridae. Capsid protein (CP) of CYVCV Chongqing isolate (CYVCV- CQ) was produced using a prokaryotic expression system and used as the immunogen for monoclonal antibody (MAb) production. Four highly specific and sensitive murine MAbs and one polyclonal antibody were prepared in this study. Titers of the four MAbs in ascites fluids ranged from 10-6 to 10-7 as determined by indirect enzyme-linked immunosorbent assay (ELISA). Three serological assays, including dot enzyme-linked immunosorbent assay (dot-ELISA), tissue blot-ELISA, and double-antibody sandwich (DAS)-ELISA, were developed for quick and reliable detections of CYVCV in citrus samples. The developed dot-ELISA and DAS-ELISA methods could detect CYVCV in the infected citrus leaf crude extracts diluted at 1:2 560 and 1:10 240 (w/v, g mL^-1), respectively. The detection result of 125 citrus leaf samples collected from citrus groves in Yunnan Province and Chongqing Municipality of China showed that approximately 36% samples were positive for CYVCV. This virus was, however, not'detected in any sample collected from Zhejiang or Jiangxi Province, China. 展开更多

Successful establishment of reconnection between retinal ganglion cells and retinorecipient regions in the brain is critical to optic nerve regeneration. However, morphological assessments of retinorecipient regions are limited by the opacity of brain tissue. In this study,we used an innovative tissue cleaning technique combined with retrograde trans-synaptic viral tracing to observe changes in retinorecipient regions connected to retinal ganglion cells in mice after optic nerve injury.Specifically,we performed light-sheet imaging of whole brain tissue after a clearing process. We found that pseudorabies virus 724(PRV724)mostly infected retinal ganglion cells, and that we could use it to retrogradely trace the retinorecipient regions in whole tissue-cleared brains.Unexpectedly,PRV724-traced neurons were more widely distributed compared with data from previous studies. We found that optic nerve injury could selectively modify projections from retinal ganglion cells in the hypothalamic paraventricular nucleus, intergeniculate leaflet, ventral lateral geniculate nucleus, central amygdala, basolateral amygdala,Edinger-Westphal nucleus, and oculomotor nucleus, but not the superior vestibular nucleus, red nucleus, locus coeruleus, gigantocellular reticular nucleus,or facial nerve nucleus. Our findings demonstrate that the tissue clearing technique, combined with retrograde trans-synaptic viral tracing, can be used to objectively and comprehensively evaluate changes in mouse retinorecipient regions that receive projections from retinal ganglion cells after optic nerve injury. Thus, our approach may be useful for future estimations of optic nerve injury and regeneration. 展开更多

To identity the potential pathogen associated with the yellow vein clearing symptom on lemon trees, the profiles of virus-de- rived small interfering RNAs from citrus samples were obtained and analyzed by deep sequencing method in this study. Twenty-seven contigs almost cover the full length genome of Citrus yellow vein clearing virus (CYVCV) isolate YN were obtained using the small RNA deep sequencing technology. Analysis showed that this isolate CQ shared the highest nucleotide identity with isolate Y1 (JX040635) and YN (KP313242), both of which belong to the genus Mandarivirus in the family Alphaflexiviridae. Mapping analysis of viral-derived siRNA (vsiRNA) profiles revealed an uneven distribution pattern of their generations along both positive and negative genome strands, and 22- and 21-nt vsiRNAs ranked the majority. BLAST against viroids and other viral databases confirmed that this sample was single-infected only by CYVCV, which indicated that CYVCV was the exact causal agent for the yellow clearing symptom occurring on lemon. This is the first CYVCV isolate detected in Chongqing and the second in China. This result could provide a molecular basis for the investigation of citrus viral diseases to protect citrus health in this region. 展开更多

Laser Speckle Contrast Imaging(LSCI)plays an important role in studying blood flow, but suffers from limited penetration depth of light in turbid tissue. The strong scattering of tissue obviously reduces the image contrast which decreases the sensitivity to flow velocity. Some image processing or optical clearing methods have been proposed to lessen the deficiency, but quantitative assessment of improvement is seldom given. In this study,LSCI was applied to monitor the blood flow through a capillary embedded within various tissue phantoms at depths of 0.25,0.45,0.65,0.85 and 1.05 mm,and the flow velocity in capillary was controllable from 0 to 4mm/s.Here,glycerol,a common optical clearing agent, was mixed with Intralipid at different volume ratio to make the reduced scattering coefficient of tissue phantom decrease from 13.00 to 0.50 cm−1.The quantitative analysis demonstrates that the optical clearing method can obviously enhance the image contrast, imaging depth, and sensitivity to blood flow velocity.Comparing the Laser Speckle Contrast Analysis methods and the optical clearing method,we find that for typical turbid tissue, the sensitivity to velocity estimated by the Laser Speckle Temporal Contrast Analysis(LSTCA)is twice of that by the Laser Speckle Spatial Contrast Analysis(LSSCA);while the sensitivity to velocity estimated by using the two analysis methods has a 10-fold increase, respectively,if addition of glycerol makes the reduced scattering coefficient of tissue phantom decrease by 30%.Combining the LSTCA and the optical clearing method, the sensitivity to flow velocity will be further enhanced. 展开更多

In order to enhance the optical clearing effect of topically applied optical clearing agents(OCAs),we evaluated the effect of propylene glycol(PG)as a chemical penetration enhancer(PE)on optical clearing of skin in vitro by observation and measurement of optical-transmittance and diffuse-reflectance spectra.Three OCAs,i.e.,glycerol,D-sorbitol and PEG400,and two other penetration enhancers,Azone and Thiazone, were used in this study. The results indicated that the decrease of reduced scattering coefficient caused by OCA/PG was larger than that by pure OCA,and the change by OCA/water was the least after the same treatment time. There were significant differences for the reduced scattering coefficient at 630 nm after 120 min application of agents between OCA and OCA/PG.The efficacy of optical clearing caused by OCA/PG depended on the OCA itself. When PEG400 was mixed with three different PEs,we found the optical clearing were different. The penetration enhancing ability of PG was much better compared to Azone, and suboptimal to Thiazone. Also, this study provides evidence for the use of PG as a PE in order to improve skin optical clearing. 展开更多

The effect of optical cleaning method combined with laser speckle imaging(LSI)was discussed to improve the detection depth of LSI due to high scattering characteristics of skin, which limit its clinical application.A double-layer skin tissue model embedded with a single blood vessel was established, and the Monte Carlo method was used to simulate photon propagation under the action of light-permeating agent.808 nm semiconductor and 632.8 nm He–Ne lasers were selected to study the e®ect of optical clearing agents(OCAs)on photon deposition in tissues. Results show that the photon energy deposition density in the epidermis increases with the amount of tissue°uid replaced by OCA.Compared with glucose solution, polyethylene glycol 400(PEG 400)and glycerol can considerably increase the average penetration depth of photons in the skin tissue, thereby raising the sampling depth of the LSI.After the action of glycerol,PEG 400,and glucose, the average photon penetration depth is increased by 51.78%,51.06%,and 21.51%for 808nm,68.93%,67.94%,and 26.67%for 632.8 nm lasers, respectively. In vivo experiment by dorsal skin chamber proves that glycerol can cause a substantial decrease in blood°ow rate, whereas PEG 400 can signicantly improve the capability of light penetration without a®ecting blood velocity, which exhibits considerable potential in the monitoring of blood°ow in skin tissues. 展开更多

Objective:Wallerian degeneration is a pathological process closely related to peripheral nerve regeneration following injury, and includes the disintegration and phagocytosis of peripheral nervous system cells.Traditionally, morphological changes are observed by performing immunofluorescence staining after sectioning, which results in the loss of some histological information. The purpose of this study was to explore a new, nondestmetive, and systematic method for observing axonal histological changes during Wallerian degeneration. Methods:Thirty male Thy1-YFP-16 mice(SPF grade,6 weeks old,20±5 g)were randomly selected and divided into clear, unobstructed brain imaging cocktails and computational analysis(CUBIC)optical clearing(n=15)and traditional method groups(n=15).Five mice in each group were sacrificed at 1st,3rd,and 5th day following a crush operation. The histological axon changes were observed by CUBIC light optical clearing treatment, direct tissue section imaging, and HE staining. Results:The results revealed that, compared with traditional imaging methods, there was no physical damage to the samples, which allowed for three-dimensional and deep-seated tissue imaging through CUBIC.Local image information could be nicely obtained by direct fluorescence imaging and HE staining, but it was difficult to obtain image information of the entire sample. At the same time, the image information obtained by fluorescence imaging and HE staining was partially lost. Conclusion:The combining of CUBIC and Thy1-YFP transgenic mice allowed for a clear and comprehensive observation of histological changes of axons in Wallerian degeneration. 展开更多

Organisms are composed of a vast number of highly specialized cells with characteristic physiological functions determined by their structure. Therefore, understanding physiological and pathological mechanisms requires obtaining 3-dimensional(3D)structural information at the cellular level across tissues, organs, and even the whole body. However, the scattering and absorption of tissue limits the penetration depth of light and thus extremely reduces the imaging performance in deep tissue. It is well known that 3D imaging outperforms 2D imaging in various scenarios for 3D reconstruction of tissue structures. This includes tracing convoluted 3D structures like vasculature, neurons, and prostate glands,as well as elucidating intricate distributions of cells or lesions. 展开更多

Objective: To observe the therapeutic effect of acupuncture in the treatment of post-stroke depression (PSD) with the method of activating the Du Meridian and clearing the mind. Methods: Two hundred and thirty one cases of PSD were randomly divided into two groups, acupuncture and control groups. In acupuncture group, Baihui (百会 GV 20), Dazhui (大椎 GV 14), Neiguan (内关 PC 6), Taichong (太冲 LR 3) and Shenmen (神门 HT 7) were selected as the main points to invigorate flow of qi in the Du Meridian and clear the mind. Patients in the control group received Fluoxetine orally, 20 mg once per-day. In addition, the same routine treatments were given to both groups, strengthened with rehabilitation exercises as early as possible; 8 weeks constituted one therapeutic course. HAMD and severity of neuro-functional defect of each case were evaluated before and 8-week after the treatment. Results: The total effective rate of acupuncture group was 92.37%, the control group, 72.88%. According to χ2 test, the difference between the two groups was remarkably significant (P<0.05). Conclusion: Acupuncture therapy of activating the Du Meridian and clearing the mind in the treatment of PSD was safe and effective. 展开更多

Tissue Optical Clearing Devices(TOCDs)have been shown to increase light transmission through mechanically compressed regions of naturally turbid biological tissues. We hypothesize that zones of high compressive strain induced by TOCD pins produce localized water displacement and reversible changes in tissue optical properties. In this paper,we demonstrate a novel combined mechanical finite element model and optical Monte Carlo model which simulates TOCD pin compression of an ex vivo porcine skin sample and modified spatial photon fluence distributions within the tissue. Results of this simulation qualitatively suggest that light transmission through the skin can be significantly affected by changes in compressed tissue geometry as well as concurrent changes in tissue optical properties. The development of a comprehensive multi-domain model of TOCD application to tissues such as skin could ultimately be used as a framework for optimizing future design of TOCDs. 展开更多
