摘要BACKGROUND Three-dimensional(3D)culture systems closely recapitulate the tumor microenvironment, allowing the assessment of drug effects on cell-cell interactions and metabolic heterogeneity.Colorectal adenocarcinoma(CRAC)cells exhibit epithelial and mesenchymal features, reflecting tumor heterogeneity, which may influence drug sensitivity. However, the morphological alterations and functional states of the residual CRAC spheres after chemotherapy remain unclear.Understanding these morphological and metabolic adaptations may help identify cancer stem cell(CSC)-like and epithelial-mesenchymal transition(EMT)-related phenotypes that contribute to therapeutic resistance and recurrence.AIM To elucidate the morphological and functional responses of 3D-cultured CRAC spheres to anticancer drugs and to characterize the residual CSC/EMT phenotypes.METHODS Five CRAC cell lines were cultured as 3D spheres, treated with 5-fluorouracil,irinotecan,or oxaliplatin, and analyzed morphologically and molecularly.RESULTS All drugs significantly suppressed sphere growth, but the morphological and metabolic responses varied.Scanning electron microscopy revealed cell surface flattening or detachment in epithelial-type spheres, whereas mesenchymal-type SW480 spheres exhibited a bowl-shaped structure with depressed centers.ATP levels decreased in all lines, except for 5-fluorouracil-treated SW480 spheres, which retained ATP but showed reduced water-soluble tetrazolium 8 activity, suggesting metabolic dormancy.Caspase 3/7 activity markedly increased in irinotecan-treated HCT-15 cells.Quantitative reverse transcriptase PCR revealed the upregulation of CD44,ALDH1A1,and PROM1 in HCT-15 cells, and increased CD44/ALDH1A1 with reduced PROM1 in SW480 cells.CDH1,VIM,and ZEB1 levels increased in HCT-15 cells, whereas SW480 cells upregulated CDH1 and VIM but downregulated ZEB1,indicating partial EMT and mesenchymal-epithelial reverting transitions.CONCLUSION Anticancer drugs induce distinct morphological and molecular adaptations, and residual CRAC spheres acquire hybrid epithelial-mesenchymal phenotypes associated with chemoresistance.

摘要Pollen viability plays a pivotal role in determining fertilization success, directly influencing fruit and seed production,as well as the genetic improvement and conservation of flowering plants. This study sought to establish the optimal culture medium and environmental conditions for in vitro pollen germination and pollen tube growth in Hibiscus mutabilis Linn.,a species of notable ornamental and ecological significance. We employed Brewbaker and Kwack medium(BK)supplemented with varying concentrations of sucrose,H3BO3,CaCl2,MgSO4⋅7H2O,and KNO2to systematically evaluate the effects of these components on pollen viability.Additionally,we investigated the impact of environmental factors including temperature, light intensity, humidity, and pH.The optimal culture medium consisted of 300 g/L sucrose,50 mg/L H3BO3,and 150 mg/L CaCl2,yielding maximum germination rates(69.3%for red pollen,65.7%for white pollen)and tube lengths(467.5μm and 428.6μm,respectively).Temperature was identified as a crucial factor, with 20◦C being optimal.High humidity(100%)and slightly acidic pH(6.1)further enhanced pollen viability.Pollen germination was most successful under dark conditions, while moderate light intensity(6000–12,000 lux)facilitated optimal pollen tube elongation. This study provides a systematic investigation of pollen germination in H.mutabilis, offering critical insights for hybrid breeding programs and conservation efforts.
摘要This paper looks back to the development of plant tissue culture in China in the last century. Since 1934, tissue culture studies in China has kept up with the international development in the fields. Progress has been made by Chinese in nearly every branches of tissue culture, including in vitro organogenesis, shoot tip culture, anther culture, ovary culture, endosperm culture, protoplast culture as well as mass cell culture. On the basis of reviewing the articles written by Chinese on plant tissue culture, the internationally recognized contributions are specially mentioned. The applications of plant tissue culture to agriculture and industry in China are also introduced.
摘要We read with great interest the study by Yadav et al published in the World Journal of Experimental Medicine, which postulated a nomogram including patient’s critical factors, other than urine sample.European Association of Urology(EAU)published the guidelines on urological infection 2025.The EAU guidelines 2025 of urinary infections(UIs)has classified in two distanced categories:Localized UTs and systemic UTs according to specific patient’s symptoms and clinical signs, this new practical classification replaced previous concept of non-complicated urinary tract infection(UTI)against complicated UTI.The new EAU classification categorizes UIs as either localized or systemic, according to the presence of specific clinical signs and symptoms, this new practical classification replaced previous concept of non-complicated UTI against complicated UTI,irrespective of the results of bacteriological findings. In the new classification of UIs,the classification is based on clinical set-up on which the practitioner or urologist will manage the patient.Management of UIs is crucial to consider the urinary and gut microbiota. It was established recently that antibiotic use affects microbiota homeostasis in the gut and urinary tract that will initiate dysbiosis.

摘要Cost stickiness reflects the corporate resource allocation efficiency, such as capital, labor, and technology.Based on financial data of the listed manufacturing companies over the 2010-2020 period, this paper introduces machine learning methods, uses the word embedding model of machine learning methods to measure the development of intelligent manufacturing at the micro level, and investigates the impact of intelligent manufacturing on corporate cost stickiness and its consequences. The results show that intelligent manufacturing helps to alleviate cost stickiness and that this effect becomes more significant in an enterprise with a strong innovation culture. After replacing the major variables and testing methods, adding control variables for robustness tests, and addressing endogeneity concerns using methods such as fixed effects, instrumental variables, and propensity score matching, the above conclusions still hold. The mechanism testing reveals that technological empowerment and information empowerment are important mechanisms for intelligent manufacturing to alleviate cost stickiness.Heterogeneity tests reveal that the effect of intelligent manufacturing in alleviating corporate cost stickiness is stronger in environments characterized by high economic policy uncertainty and fierce industry competition.Drawing on informal institutional factors, this paper reveals the consequences and process through which intelligent manufacturing improves corporate resource allocation efficiency, providing insights for the transformation and upgrading of the manufacturing industry through intelligent manufacturing.

摘要Background Under high-concentrate feeding conditions, ruminants often experience rumen microecological imbalance and dysfunction, which can impair growth performance and increase the risk of antibiotic resistance gene(ARG)dissemination. Results To evaluate the ameliorative effects of Aspergillus niger(A.niger)cultures, fattening sheep were randomly allocated into the following five groups:a control group(CON),a control diet supplemented with 250,500,or 1,000 mg/kg A.niger cultures(designated as LA,MA,and HA,respectively);and an antibiotic group supplemented with 5,000 mg/kg chlortetracycline premix(AN).Microbial community analysis indicated that several bacterial taxa, including Succinivibrio sp900317105,Prevotella sp002353485,Quinella sp017515635,Quinella sp015206805,and Prevotella sp900320255,were significantly enriched in the A.niger culture-supplemented groups(P<0.05).ARG profiling showed that the abundance of tetracycline resistance genes was significantly lower in all A.niger groups compared with the CON and AN groups(P<0.05),whileβ-lactam resistance genes were significantly reduced in the HA group(P<0.05).Furthermore,the abundances of Rank I and Rank II ARGs were significantly higher in the AN group than in the other groups, whereas the abundances of Rank II and Rank IV ARGs were significantly lower in the A.niger culture groups than in the CON and AN groups.Metabolomic analysis further demonstrated that supplementation with A.niger cultures significantly decreased the concentration of N-decanoyl-L-homoserine lactone(P<0.05)while increasing the levels of N-3-oxotetradec-7Z-enoyl-L-homoserine lactone,indole-3-methyl acetate, and indole-3-propionic acid(P<0.05).Conclusions These findings suggest that A.niger cultures can reduce the abundance of ARGs and mitigate the risk of ARG dissemination by modulating the rumen microbial community and associated metabolites.
