目的探究miR-944靶向调控MARCH7对非小细胞肺癌细胞增殖、凋亡及顺铂耐药的机制研究。方法将肺癌A549细胞常规培养后用DDP浓度梯度法建立DDP耐药细胞株,完成后设立miR-944 mimics组(将miR-944 mimics转染至细胞株中)、MARCH7 siRNA组(将... 目的探究miR-944靶向调控MARCH7对非小细胞肺癌细胞增殖、凋亡及顺铂耐药的机制研究。方法将肺癌A549细胞常规培养后用DDP浓度梯度法建立DDP耐药细胞株,完成后设立miR-944 mimics组(将miR-944 mimics转染至细胞株中)、MARCH7 siRNA组(将MARCH7 siRNA转染至细胞株中)、miR-944 mimics+MARCH7 siRNA组(将miR-944 mimics与MARCH7 siRNA共同转染至细胞株中)。采用CCK-8法检测细胞增殖情况,采用TUNEL法检测细胞凋亡,对顺铂耐药机制进行检测并用线性回归法计算药物半抑制浓度(IC50)值,采用荧光素酶测定系统检测miR-944与MARCH7靶向关系,采用PCR检测miR-944与MARCH7表达。结果与对照组比较,miR-944 mimics组、MARCH7 siRNA组及miR-944 mimics+MARCH7 siRNA组细胞中的miR-944、细胞凋亡率升高,MARCH7、IC50及24、48、72 h增殖率降低(P0.05)。双荧光素酶报告实验结果显示,miR-944 mimics转染MARCH7 siRNA后MARCH7明显降低(P0.05),提示MARCH7与miR-944可能存在靶向以及负调控的关系。结论在非小细胞肺癌中,升高miR-944及敲低MARCH7可能有助于抑制癌细胞增殖、促进其凋亡,同时还可能对改善耐药状态具有一定积极作用。 展开更多

Dentin,the main component of dental hard tissues,is produced by differentiated odontoblasts.How odontoblast differentiation is regulated remains understudied.Here,we screen that the expression of membrane-associated RING finger protein 2(March2) is the highest among all March family members,with an increasing trend during odontoblast differentiation.In mouse incisors and molars,MARCH2 is moderately expressed in the undifferentiated dental papilla cells and strongly expressed in the odontoblasts.Knockdown and overexpression experiments demonstrate that MARCH2 inhibits odontoblastic differentiation of mouse dental papilla cells(mDPCs).Additionally,both March2 deficient mice and mice with odontoblast specific knockdown of March2 exhibit the phenotype of increased dentin thickness,accelerated dentin deposition as well as elevated expression levels of odontoblast markers compared with control littermates.Therefore,MARCH2 plays an inhibitory role in odontoblast differentiation.Mechanistically,MARCH2 interacts with protein tyrosine phosphatase receptor delta(PTPRD) and facilitates its K27-linked polyubiquitination and subsequent degradation,which is dependent on the ligase activity of MARCH2.The presence of MARCH2promotes the translocation of PTPRD from the cell membrane to the lysosome,thereby enhancing its degradation via the lysosomal pathway.Further experiments show that knockdown of endogenous Ptprd impairs odontoblastic differentiation of mDPCs.Ptprd and March2 double knockdown in mDPCs apparently reversed the enhanced odontoblastic differentiation by knockdown of March2 alone,indicating that MARCH2 inhibits odontoblastic differentiation by promoting PTPRD degradation.This study unveils a novel mechanism where an E3 ubiquitin ligase regulates odontoblast differentiation through post-translational modification of a membrane protein,highlighting a promising direction for future exploration. 展开更多

The literature analysis,data collection,and on-site exploration are conducted on 128 heritage nodes in the three historical event routes with linear characteristics at the Jiangxi section of the red army's long march.Combining their characteristics,protection status,heritage value,etc.,a system framework for evaluating heritage corridor resources is constructed by integrating the analytic hierarchy process and Delphi method.On this basis,the weight values corresponding to each evaluation factor are accurately determined,and the comprehensive score of heritage resources is calculated based on the Rosenberg-Fishbein mathematical model formula.The login route,distribution area,protection level,and protection status are analyzed,providing a solid theoretical basis and scientific support for the scientific construction and effective protection of the red cultural heritage corridor in the Jiangxi section of the red army’s long march. 展开更多

鉴于移动立方体算法在医学成像、计算机图形学、工程建模等领域被广泛应用,顾及激光雷达检验法具有诸多优点,提出了一种新的利用MarchingCubes算法的塔身垂直度激光雷达检验方法。实验选取施工现场某型号塔式起重机塔身进行测试,采用激... 鉴于移动立方体算法在医学成像、计算机图形学、工程建模等领域被广泛应用,顾及激光雷达检验法具有诸多优点,提出了一种新的利用MarchingCubes算法的塔身垂直度激光雷达检验方法。实验选取施工现场某型号塔式起重机塔身进行测试,采用激光雷达扫描采集塔身标准节原始点云数据,经预处理后获取塔身标准节有效高精度点云数据;给出塔身标准节横向切段立方体点云分割策略,利用MarchingCubes算法提取塔身标准节横向切段立方体点云轮廓线,并求解其体中心点坐标;运用最小二乘法拟合体中心点所在空间直线,计算其方向向量,并在站心空间坐标系下分别与x轴和z轴进行向量运算,获得塔身轴心线倾斜方位角、倾斜角和塔身垂直度。结果表明:采用本文方法设计5种策略方案获得的塔身垂直度中数值为1.80‰,与传统的经纬仪检验法结果为3.02‰基本吻合。本文为塔身垂直度无损检验提供一种高精度激光雷达算法。 展开更多

目的·探讨E3泛素连接酶膜相关锌指蛋白9(membrane-associated RING-CH 9,MARCH9)调控受体型蛋白质酪氨酸磷酸酶α(receptor protein tyrosine phosphatase alpha,RPTPα)泛素化修饰的分子机制及其生物学功能。方法·采用Wester... 目的·探讨E3泛素连接酶膜相关锌指蛋白9(membrane-associated RING-CH 9,MARCH9)调控受体型蛋白质酪氨酸磷酸酶α(receptor protein tyrosine phosphatase alpha,RPTPα)泛素化修饰的分子机制及其生物学功能。方法·采用Western blotting鉴定RPTPα的泛素化修饰类型及MARCH9对其泛素化水平的调控作用;比较MARCH9野生型、MARCH9酶活突变体(MARCH9 S198A或MARCH9-HC/CC),以及shRNA介导的内源MARCH9敲低对RPTPα蛋白稳定性的影响;利用蛋白酶体抑制剂MG132、自噬抑制剂3-MA和溶酶体抑制剂氯喹(chloroquine,CQ)鉴定MARCH9介导的RPTPα泛素化降解类型;探究43℃热激促进RPTPα蛋白降解的可能机制;利用慢病毒载体构建MARCH9单敲低(H1299-sh MARCH9)及MARCH9/RPTPα双敲低(H1299-sh MARCH9-sh RPTPα)的肺癌稳转细胞株,通过CCK-8细胞增殖、平板克隆形成和软琼脂克隆形成实验,评估MARCH9或RPTPα对肺癌细胞增殖与集落形成能力的影响;通过血管拟态形成实验和划痕实验检测MARCH9或RPTPα对肺癌细胞侵袭、迁移能力的影响;建立裸鼠皮下移植瘤模型评价MARCH9或RPTPα对肺癌细胞体内成瘤能力的影响;通过生物信息学方法比较MARCH9和RPTPα在肺癌患者中的表达差异与预后相关性。结果·RPTPα主要发生K63连接型多聚泛素化修饰,E3泛素连接酶MARCH9过表达显著增强其泛素化水平;过表达野生型MARCH9而非酶活突变体能显著降低RPTPα蛋白稳定性,而shRNA敲低内源MARCH9后RPTPα蛋白水平又会明显上升;CQ而非MG132或3-MA处理能增加RPTPα蛋白稳定性,提示MARCH9主要介导RPTPα的泛素化-溶酶体降解途径;43℃热激可以特异增强MARCH9-RPTPα相互作用并促进RPTPα蛋白降解。功能实验显示:与对照H1299细胞相比,H1299-sh MARCH9细胞的RPTPα蛋白水平升高,细胞增殖速率加快,克隆形成和细胞侵袭能力均明显增强,裸鼠中成瘤能力也明显增强,而双敲低MARCH9/RPTPα的稳转细胞株又能逆转上述表型(均P<0.05)。生物信息学分析肺癌患者相关数据,显示RPTPα高表达与肺癌患者不良预后及肿瘤转移呈正相关,而MARCH9则为负相关。结论·MARCH9介导了磷酸酶RPTPα的K63型泛素化依赖的溶酶体降解过程。这一发现为开发靶向RPTPα的肿瘤治疗策略提供了新的依据。 展开更多

目的:明确膜相关的RING-CH型E3泛素连接酶6(membrane-associated RING finger protein 6, March6)在成牙本质细胞分化以及牙本质生成中的作用。方法:采用免疫组织化学染色(immunohistochemistry, IHC)分析不同发育阶段小鼠牙齿切片,分析... 目的:明确膜相关的RING-CH型E3泛素连接酶6(membrane-associated RING finger protein 6, March6)在成牙本质细胞分化以及牙本质生成中的作用。方法:采用免疫组织化学染色(immunohistochemistry, IHC)分析不同发育阶段小鼠牙齿切片,分析March6的时空表达特征。体外敲低小鼠牙乳头细胞(mouse dental papilla cells, mDPCs)中的March6,并进行矿化诱导,分别使用实时定量PCR、Western bolt、碱性磷酸酶(alkaline phosphatase, ALP)染色以及茜素红染色,评估March6对mDPCs成牙本质细胞向分化潜能的影响。将Prx1cre与March6flox/flox小鼠配繁,获得牙齿间充质中特异性敲除March6的小鼠(March6 cKO,Prx1cre;March6flox/flox)。通过IHC验证March6被特异性敲除。随后,对出生后10 d的March6 cKO及其同窝对照小鼠的下颌行切片苏木素-伊红染色、显微CT(micro-computed tomography, Micro-CT)观察,以确定March6在体内对牙本质生成的影响。结果:March6在胚胎时期小鼠牙齿间充质中表达水平较低,而在成牙本质细胞形成后,March6在成牙本质细胞中高表达。体外敲低mDPCs中的March6,并诱导其向成牙本质细胞分化,牙本质基质蛋白1(dentin matrix protein 1,Dmp1)和牙本质涎磷蛋白(dentin sialophosphoprotein, Dspp)的mRNA以及蛋白水平相较于对照显著上调。同时,细胞ALP活性和钙化结节生成能力在敲低组中也明显增强。组织学形态观察以及Micro-CT分析结果显示,March6 cKO小鼠的牙本质层相较于同窝对照明显增厚。结论:E3泛素连接酶March6对成牙本质细胞分化以及牙本质生成发挥抑制作用。 展开更多

Methods: MTS and EDU were used to detect the growth of MARCH2 overexpressing hepatocellular carcinoma HUH7 cells. Flow cytometry was used to detect apoptosis and cell cycle in MARCH2 overexpressing hepatocellular carcinoma HUH7 cells. Western blot and laser confocal microscopy were used to detect the autophagy levels in MARCH2 overexpressing HUH7 hepatocellular carcinoma cells. Results: In this study, MTS and EDU assays showed that the overexpression of MARCH2 promoted the growth of hepatocellular carcinoma HUH7 cells. Flow cytometry detection illustrated that MARCH2 overexpression had no obvious effect on the hepatocellular carcinoma HUH7 cell apoptosis and cell cycle. Further experiments displayed that MARCH2 overexpression impaired the autophagy of hepatocellular carcinoma HUH7 cells. Rescued autophagy with the common autophagy inducer rapamycin can bring the proliferation rate of hepatocellular carcinoma HUH7 cells back to the control level. Conclusions: Our data indicates that MARCH2 plays an essential role in the progression of tumor development through inhibiting autophagy. Therefore, MARCH2 is an attractive novel target for tumor therapy. 展开更多

The idea of cultural route heritage protection expands the protection theory for world cultural heritage.The Long March route carries the prominent characteristic of cultural route heritage.By actively building the Long March National Cultural Park,China has strengthened the overall protection and integrated utilization of the Long March cultural heritage,upgraded the hardware facilities for exhibition,and investigated ways to protect cultural route heritage with Chinese characteristics.During the construction of the Park,it is necessary to draw on the concept and norms of the cultural route heritage protection,optimize the construction approach,and make the construction of the Park more in line with the linear cultural characteristics of Long March heritage.Specifically,it is proposed to improve the Long March cultural route heritage protection system oriented towards“authenticity”protection,to deeply tap and interpret the prominent universal worth of the Long March route,augmenting the international dissemination of the Long March culture in an innovative way,and realizing a more scientific and effective protection and utilization of Long March cultural resources. 展开更多

Microseismic(MS)event locations are vital aspect of MS monitoring technology used to delineate the damage zone inside the surrounding rock mass.However,complex geological conditions can impose significantly adverse effects on the final location results.To achieve a high-accuracy location in a complex cavern-containing structure,this study develops an MS location method using the fast marching method(FMM)with a second-order difference approach(FMM2).Based on the established velocity model with three-dimensional(3D)discrete grids,the realization of the MS location can be achieved by searching the minimum residual between the theoretical and actual first arrival times.Moreover,based on the calculation results of FMM2,the propagation paths from the MS sources to MS sensors can be obtained using the linear interpolation approach and the Runge–Kutta method.These methods were validated through a series of numerical experiments.In addition,our proposed method was applied to locate the recorded blasting and MS events that occurred during the excavation period of the underground caverns at the Houziyan hydropower station.The location results of the blasting activities show that our method can effectively reduce the location error compared with the results based on the uniform velocity model.Furthermore,the obtained MS location was verified through the occurrence of shotcrete fractures and spalling,and the monitoring results of the in-situ multipoint extensometer.Our proposed method can offer a more accurate rock fracture location and facilitate the delineation of damage zones inside the surrounding rock mass. 展开更多
