Objective:To investigate the modulatory effects of a Morus alba leaf extract(FBCC-EP1619)on melanogenesis using enzymatic,cellular,and zebrafish models.Methods:Phytochemical profiling was conducted using UPLC-QTOF-MS/...Objective:To investigate the modulatory effects of a Morus alba leaf extract(FBCC-EP1619)on melanogenesis using enzymatic,cellular,and zebrafish models.Methods:Phytochemical profiling was conducted using UPLC-QTOF-MS/MS in negative ion mode.Mushroom tyrosinase inhibition was assessed in vitro.Cytotoxicity and melanin content were measured in B16F10 melanoma cells under basal andα-melanocyte-stimulating hormone(α-MSH)-stimulated conditions.An in vivo pigmentation model was established usingα-MSH-treated zebrafish larvae,and pigmentation was quantified.Expression of melanogenesis-related genes(mitfa and tyr)was analyzed by RT-qPCR.Results:FBCC-EP1619 contained diverse phenolic and lipid-derived metabolites.The extract significantly inhibited mushroom tyrosinase activity in a concentration-dependent manner.In B16F10 cells,it did not induce cytotoxicity but increased melanin production.In contrast,inα-MSH-stimulated zebrafish larvae,FBCC-EP1619 attenuated hyperpigmentation and significantly downregulated mitfa and tyr expression,indicating system-and stimulus-dependent regulation.Conclusions:FBCC-EP1619 differentially modulates melanogenesis depending on the biological system,enhancing basal melanin production in vitro while suppressingα-MSH-induced pigmentation in vivo.These findings provide pharmacological support for the ethnopharmacological relevance of Morus alba leaves and warrant further investigation into the underlying molecular mechanisms.展开更多
OBJECTIVE:To determine direct targeting of localized adiposity through Morus alba Linne bark injection based on pharmacology network analysis.METHODS:Male C57BL/6J mice were fed a high-fat diet(HFD)to induce obesity.A...OBJECTIVE:To determine direct targeting of localized adiposity through Morus alba Linne bark injection based on pharmacology network analysis.METHODS:Male C57BL/6J mice were fed a high-fat diet(HFD)to induce obesity.After 6 weeks on HFD,the water extract of Morus alba L.bark(MAB,2 mg/mL)was locally injected into one inguinal fat pad,while saline was injected into the other side,3 times/week for 6 weeks(n=6/group).The water extract of MAB was freeze-dried and then diluted in saline before use.RESULTS:HFD-fed mice treated with local MAB topical injection showed reduced adipocyte weight and size in inguinal fat pads by dual-energy X-ray absorptiometry.No toxicity changes seen in liver,spleen,kidney tissue,or alanine aminotransferase/aspartate aminotransferase levels in serum by MAB injection.Protein levels of phosphorylated insulin receptor substrate-1 and glucose transporter type 4,and mRNA expression of adiponectin,were increased in inguinal adipose tissue injected with MAB locally.Locally MAB injection led to a decrease in glucose-6-phosphatase and phosphoenolpyruvate carboxykinase,linked to gluconeogenesis,while forkhead box protein O1,which regulates these factors,was increased.Moreover,there was an increase in adenosine 5‘-monophosphate-activated protein kinase,related to lipogenesis,as well as elevated levels of hormone-sensitive lipase and fatty acid synthase,both associated with lipolysis.These results support the'insulin signaling pathway'and'regulation of lipolysis in adipocytes'identified in the Kyoto Encyclopedia of Genes and Genomes pathway through network analysis.CONCLUSION:This study suggests that MAB topical injection exhibits localized fat reduction by inhibiting insulin resistance,gluconeogenesis and lipogenesis mediator,while activating lipolysis enzymes within targeted adipose site.展开更多

The LAB strai...The objective of this study was to isolate lactic acid bacteria(LAB) strains from different origins and to select the best strains for ensiling Robinia pseudoacacia(RB) and Morus alba L.(MB)leaves. The LAB strains were inoculated into the extracted liquid obtained from RB and MB leaves to evaluate the fermentation products. 11 LAB strains were selected for further experiments based on the highest products of lactic or acetic acid, including 1 strain of Weissella confusa, 2 of Lactobacillus reuteri and 8 of Lactobacillus plantarum.The API 50 CH fermentation experiment indicated that all of the selected 11 LAB strains utilised most of the carbohydrates. All the strains grew at temperatures between 10 and 45℃ and at a p H of 3.5 to 4.5; however, L. reuteri F7 and F8 tolerated a p H as low as 3.0. All 11 LAB strains showed antibacterial activity against Listeria monocytogens, Escherichia coil, Salmonella sp. and Acetobacter pasteurianus; however, after excluding the effect of organic acids, only F7 and F8 still exhibited antibacterial activity. The present study indicated that the selected 11 LAB strains could be used to prepare silages of RB and MB leaves, especially L. reuteri F7 and F8.展开更多
[Objective]The aim was to explore the active compound of Morus alba leaves with the analysis of active components of hexane extractives of M.alba leaves.[Method]Antibacterial assays of hexane extractives of M.alba lea...[Objective]The aim was to explore the active compound of Morus alba leaves with the analysis of active components of hexane extractives of M.alba leaves.[Method]Antibacterial assays of hexane extractives of M.alba leaves were done and the volatile components of hexane extractives of M.alba leaves with the strongest antibacterial activity were analyzed by gas chromatography-mass spectrometry(GC-MS).[Result]The results of inhibitory effect of hexane extractives of M.alba leaves on Staphylococcus aureus and Escherichia coli showed that hexane extractives of M.alba leaves collected in June,July and August had antibacterial activity against S.aureus and had no antibacterial effect against E.coli.Furthermore,hexane extractives of M.alba leaves collected in June had the strongest antibacterial activity against S.aureus(inhibition diameter 10.95 mm).The hexane extractives of M.alba leaves collected in June was analyzed by GC-MS,the main volatile components were tetradecane(16.76%),dodecane(13.20%),diisobutyl phthalate(10.26%),decane(9.10%),hexadecane(8.71%),linolenyl alcohol(7.25%),octadecane(5.88%),eicosane(3.26%),dibutyl phthalate(2.59%).[Conclusion]Linolenyl alcohol was the potential antibacterial compound.展开更多

[Conclusion] The essential oils of M. alba leaves and C. pisifera outer heartwood with hydrodistillation have medicinal value.

摘要The objective of this study was to isolate lactic acid bacteria(LAB) strains from different origins and to select the best strains for ensiling Robinia pseudoacacia(RB) and Morus alba L.(MB)leaves. The LAB strains were inoculated into the extracted liquid obtained from RB and MB leaves to evaluate the fermentation products. 11 LAB strains were selected for further experiments based on the highest products of lactic or acetic acid, including 1 strain of Weissella confusa, 2 of Lactobacillus reuteri and 8 of Lactobacillus plantarum.The API 50 CH fermentation experiment indicated that all of the selected 11 LAB strains utilised most of the carbohydrates. All the strains grew at temperatures between 10 and 45℃ and at a p H of 3.5 to 4.5; however, L. reuteri F7 and F8 tolerated a p H as low as 3.0. All 11 LAB strains showed antibacterial activity against Listeria monocytogens, Escherichia coil, Salmonella sp. and Acetobacter pasteurianus; however, after excluding the effect of organic acids, only F7 and F8 still exhibited antibacterial activity. The present study indicated that the selected 11 LAB strains could be used to prepare silages of RB and MB leaves, especially L. reuteri F7 and F8.

摘要[Objective]The aim was to explore the active compound of Morus alba leaves with the analysis of active components of hexane extractives of M.alba leaves.[Method]Antibacterial assays of hexane extractives of M.alba leaves were done and the volatile components of hexane extractives of M.alba leaves with the strongest antibacterial activity were analyzed by gas chromatography-mass spectrometry(GC-MS).[Result]The results of inhibitory effect of hexane extractives of M.alba leaves on Staphylococcus aureus and Escherichia coli showed that hexane extractives of M.alba leaves collected in June,July and August had antibacterial activity against S.aureus and had no antibacterial effect against E.coli.Furthermore,hexane extractives of M.alba leaves collected in June had the strongest antibacterial activity against S.aureus(inhibition diameter 10.95 mm).The hexane extractives of M.alba leaves collected in June was analyzed by GC-MS,the main volatile components were tetradecane(16.76%),dodecane(13.20%),diisobutyl phthalate(10.26%),decane(9.10%),hexadecane(8.71%),linolenyl alcohol(7.25%),octadecane(5.88%),eicosane(3.26%),dibutyl phthalate(2.59%).[Conclusion]Linolenyl alcohol was the potential antibacterial compound.

摘要A new Diels-Alder type adduct mongolicin G (1) and a new flavone 5'-(1", 1"-dimethylallyl)-5,7,2',4'-tetrahydroxyflavone (2) were isolated from the stem and root bark of Morus mongolica. Their structures were determined by spectroscopic analysis and chiroptical methods.
