With the increasing accumulation of plastic pollutants in various environments,research on microorganisms(including bacteria,fungi,and algae)with plastic degradation capabilities has gained significant attention.Howev...With the increasing accumulation of plastic pollutants in various environments,research on microorganisms(including bacteria,fungi,and algae)with plastic degradation capabilities has gained significant attention.However,only a limited number of microbial plastic-degrading enzymes have been identified to date.This highlights that the degradation mechanisms employed by many plastic-degrading microorganisms,particularly filamentous fungi,remain insufficiently explored.In this study,we utilized a versatile fungal plasmid(pCT74)to express green fluorescent protein(GFP)in a marine-derived fungus Alternaria alternata strain FB1 with plastic degradation capabilities.Upon evaluating the degradation effect of polyester-type polyurethane(PU)film,we observed that different transformants exhibited three kinds of activities(the same,reduced,or enhanced degradation capability)compared to the FB1 wild-type strain.Further analysis of the plasmid fragment insertion sites in different transformants revealed that pCT74 integrates randomly into the genome of the host fungus.Notably,a direct correlation was found between the plasmid insertion site and the degradation capability of the corresponding transformant.Our findings not only redefine the potential applications of plasmid pCT74 in filamentous fungi but also show a novel research approach to identifying key enzymes involved in plastic degradation by fungi.展开更多
OsMAPK6 plays a critical role in regulating rice growth,development,and stress responses.However,the embryonic lethality associated with loss-of-function mutations prevents the generation of homozygous mutant seeds,si...OsMAPK6 plays a critical role in regulating rice growth,development,and stress responses.However,the embryonic lethality associated with loss-of-function mutations prevents the generation of homozygous mutant seeds,significantly hindering functional studies of this gene.Although the weak mutant dsg1 has offered valuable insights into OsMAPK6 function,its extremely low seed-setting rate limits its use for detailed genetic analysis.Here,we employed prime editing to perform precise multi-site modifications at the C-terminus of OsMAPK6,generating a series of osmapk6 mutants with truncated proteins of varying lengths.Among these,the osmapk6(379)and osmapk6(383)mutants exhibited phenotypic defects similar to dsg1,while osmapk6(386)showed a significantly improved seed-setting rate despite persistent developmental defects.Through phenotypic characterization and protein functional analysis,we further clarified how different C-terminal deletion lengths affect plant growth,development,stress responses,and OsMAPK6 protein function.In summary,this study elucidates the importance of the OsMAPK6 C-terminus in rice biology and provides a fertile weak mutant with enhanced seed production,offering a valuable genetic resource for future research on OsMAPK6.展开更多

Fusarium crown rot(FCR),predominantly caused by Fusarium pseudograminearum,has been listed as a Category Ⅱ disease in six provinces of China,posing a significant threat to wheat production.The phenylpyrrole fungicide...Fusarium crown rot(FCR),predominantly caused by Fusarium pseudograminearum,has been listed as a Category Ⅱ disease in six provinces of China,posing a significant threat to wheat production.The phenylpyrrole fungicide fludioxonil is a key agent for FCR control.Previous studies indicated that resistance to fludioxonil in F.pseudograminearum is primarily associated with altered expression levels of the FpOS1 gene,which encodes a hybrid histidine kinase.However,the roles of mutations in other FpOS genes and the molecular interactions between FpOS proteins and fludioxonil remain elusive.To address these gaps,we generated 16 fludioxonil-resistant mutants with heritable resistance traits by in vitro selection of four sensitive F.pseudograminearum isolates.These mutants exhibited high resistance levels,with resistance factors(RF)ranging from 633.73 to 8617.07.Compared to their parental isolates,the resistant mutants showed significantly reduced mycelial growth rate,sporulation capacity,and pathogenicity.They were also more sensitive to ionic,osmotic,and oxidative stresses and displayed compromised cell wall and membrane integrity.Fludioxonil demonstrated no cross-resistance with tebuconazole or pydiflumetofen;however,it exhibited weak positive crossresistance to pyraclostrobin and moderate positive cross-resistance to iprodione.Fludioxonil treatment significantly promoted glycerol synthesis and inhibited deoxynivalenol(DON)production in parental isolates,whereas these regulatory effects were markedly attenuated in the resistant mutants.Mutation analysis identified mutation sites in FpOS1,FpOS4,and FpOS5 genes,with a lower mutation frequency in FpOS1 and no mutations detected in FpOS2.Molecular docking indicated that amino acid substitutions in FpOS4 and FpOS5 significantly reduced the binding affinity of fludioxonil to these target proteins.In conclusion,F.pseudograminearum poses a moderate risk of resistance to fludioxonil.Point mutations in FpOS4 and FpOS5 genes emerge as key molecular drivers of resistance,likely by diminishing the binding affinity between the fungicide and its proteins.This study clarifies the molecular basis of fludioxonil resistance in F.pseudograminearum and provides a scientific rationale for the judicious use of this fungicide in managing FCR.展开更多

Naturally occurring yellow leaf mutants are an important resource for studying pigment content and biosynthesis,as well as related gene expression.In our ongoing cultivation of Rehmannia chingii H.L.Li,we found an off...Naturally occurring yellow leaf mutants are an important resource for studying pigment content and biosynthesis,as well as related gene expression.In our ongoing cultivation of Rehmannia chingii H.L.Li,we found an off-type yellow plant.The yellowing started with the new leaves and gradually spread downward until the entire plant exhibited a stable shade of yellow.We studied the differences in the chlorophyll and carotenoid content,carotenoid profile,and transcriptome of this yellow-leaf mutant(P2).Compared to the wild-type R.chingii plant(P1),P2 leaves had significantly lower chlorophyll and carotenoid content.LC-MS/MS analysis revealed that P2 had higher quantities of severalmetabolites in the carotenoid biosynthesis pathway.Transcriptome sequencing results showed that genes involved in porphyrin metabolism,carbon fixation,photosynthesis and antenna proteins,terpenoid backbone biosynthesis,and carotenoid biosynthesis were differentially expressed between P1 and P2.Large-scale expression differences were observed in the phytohormone and MAPK signaling pathways,as well as in 15 transcription factor families.We discuss possible mechanisms responsible for the yellowleaf color in P2.These preliminary data are valuable for further exploring the molecular mechanisms of leaf color formation and associated pathways.展开更多
Lilium leichtlinii var.Maximowiczii is a new type of edible lily with important nutritional and economic value.However,it is prone to lodging,and its bulb often follows the roots.To obtain better lily germplasms,diffe...Lilium leichtlinii var.Maximowiczii is a new type of edible lily with important nutritional and economic value.However,it is prone to lodging,and its bulb often follows the roots.To obtain better lily germplasms,different concentrations of colchicine were used to induce the growth of L.leichtlinii var.Maximowiczii seeds.The identification was conducted via plant morphology,chromosomal ploidy analysis,and ISSR molecular markers.The best induction effect was achieved by treating seeds with 0.15%colchicine for 48 h,with a seedling variation rate of 31.67%.After 17 months of cultivation,11 mutant plants(Nos.1-11)survived.The No.2 plant grew rapidly and flowered earlier than the control plants.The No.3 plant was significantly dwarfed,and its leaves became wider.Flow cytometry and root tip squash analysis revealed that the No.3 plant was aneuploid,with 38 chromosomes in the root tip cells;the other plants were diploids,with 24 chromosomes.Microscopic observation revealed that the size of leaf epidermal cells and guard cells increased in the No.3 plant,and the stomatal density significantly decreased,consistent with the characteristics of the polyploid plants.Additionally,ISSR marker analysis revealed significant differences in the No.2 plant at the molecular genetic level compared with the control,with a variation rate of 18.99%.These results provide a reference for lily mutant breeding using the seeds by colchicine induction and the basis for further research on the early flowering mechanism in lily.展开更多
AIM: To conduct a retrospective study in 400 chronic hepatitis B patients in order to identify hepatitis B viral factors associated with complications of liver disease or development of hepatocellular carcinoma. METH...AIM: To conduct a retrospective study in 400 chronic hepatitis B patients in order to identify hepatitis B viral factors associated with complications of liver disease or development of hepatocellular carcinoma. METHODS: The mean follow-up time was 83.6 ± 39.6 mo. Alpha-fetoprotein test and abdominal ultrasound were used for cancer surveillance. Hepatitis B basal core promoter mutants, precore mutants, genotypes, hepatitis B viral DNA (HBV DNA) level and hepatitis B e antigen (HBeAg) were measured. Univariate analysis and logistic regression were used to assess odds ratios for viral factors related to liver deaths and hepatocellular carcinoma development. RESULTS: During follow-up, 38 patients had liver deaths not related to hepatocellular carcinoma. On multivariate analysis, older age [odds ratio: 95.74 (12.13-891.31), P 〈 0.0001], male sex [odds ratio: 7.61 (2.20-47.95); P = 0.006], and higher Iogzo HBV DNA [odds ratio: 4.69 (1.16-20.43); P 〈 0.0001] were independently predictive for these liver related deaths. Also, 31 patients developed hepatocellular carcinoma. Multivariate analysis showed that older age [odds ratio: 26.51 (2.36-381.47); P = 0.007], presence of precore mutants [odds ratio: 4.23 (1.53-19.58), P = 0.02] and presence of basal core promoter mutants [odds ratio: 2.93 (1.24-7.57); P = 0.02] were independent predictors for progression to hepatocellular carcinoma. CONCLUSION: Our results show that high levels of baseline serum HBV DNA are associated with non- hepatocellular carcinoma-related deaths of liver failure, while genetic mutations in the basal core promoter and precore regions are predictive for development of hepatocellular carcinoma.展开更多

[Result] Our results showed that GR and SSR were 45.2% and 40.2% respectively at 1.0% EMS, close to LD50, with GI (17.6) and seed Ⅵ (19.7) being half of that of control; 562 M4 mutants were identified in 2015, and the mutation could be characterized according 11 major categories and 32 subcategories; Simultaneously, we found that plant height, plant width, diameter of mainstem, length of main-stem, the number of main-stem nodes and branch of lines E29, E58, E142 and E312 were all significantly different from that of the control. The mutation of lines E29, E58 and E312 was all controlled by a single recessive gene. AIM:To investigate precore/basal core promoter(PC/BCP) mutants throughout hepatitis B virus(HBV) infection and to determine their relationship to hepatitis B early antigen(HBeA g) titers.METHODS:We enrolled 191 patien...AIM:To investigate precore/basal core promoter(PC/BCP) mutants throughout hepatitis B virus(HBV) infection and to determine their relationship to hepatitis B early antigen(HBeA g) titers.METHODS:We enrolled 191 patients in various stages of HBV infection at the Huashan Hospital and the Taizhou Municipal Hospital from 2010 to 2012.None of the patients received antiviral therapy.HBV DNA from serum,was quantified by real-time PCR.The HBV genotype was determined by direct sequencing of the S gene.We used the Simpleprobe ultrasensitivequantitative method to detect PC/BCP mutants in each patient.We compared the strain number,percentage,and the changes in PC/BCP mutants in different phases,and analyzed the relationship between PC/BCP mutants and HBe Ag by multiple linear regression and logistic regression.RESULTS:Patients with HBV infection(n = 191) were assigned to groups by phase:Immune tolerance(IT) = 55,Immune clearance(IC) = 67,Low-replicative(LR) = 49,and HBeA g-negative hepatitis(ENH) = 20.Of the patients(male,112; female,79) enrolled,122 were HBe Ag-positive and 69 were HBe Ag-negative.The median age was 33 years(range:18-78 years).PC and BCP mutation detection rates were 84.82%(162/191) and 96.86%(185/191),respectively.In five HBe Ag-negative cases,we detected double mutation G1896A/G1899 A.The logarithm value of PC mutant quantities(log10 PC) significantly differed in IT,IC,and LR phases,as well as in the ENH phase(F = 49.350,P < 0.001).The logarithm value of BCP mutant quantities(log10 BCP) also differed during the four phases(F = 25.530,P < 0.001).Log10 PC and log10 BCP values were high in the IT and IC phases,decreased in the LR phase,and increased in the ENH phase,although the absolute value at this point remained lower than that in the IT and IC phases.PC mutant quantity per total viral load(PC%) and BCP mutant quantity per total viral load(BCP%) differed between phases(F = 20.040,P < 0.001; F = 10.830,P < 0.001),with PC% and BCP% gradually increasing in successive phases.HBeA g titers negatively correlated with PC%(Spearman's rho =-0.354,P < 0.001) and BCP%(Spearman's rho =-0.395,P < 0.001).The negative correlation between PC% and HBeA g status was significant(B =-5.281,P = 0.001),but there was no such correlation between BCP% and HBeA g status(B =-0.523,P = 0.552).CONCLUSION:PC/BCP mutants become predominant in a dynamic and continuous process.Log10 PC,log10 BCP,PC% and BCP% might be combined to evaluate disease progression.PC% determines HBeA g status.展开更多

However, the genomic resources and tools for functional genomics studies in cucumber are still limited. In this paper, we conducted ethyl methyl sulfone(EMS) mutagenesis in the northern China ecotype cucumber inbred line 406 to construct a mutant library. We optimized the conditions of EMS mutagenesis on inbred line 406 which included treatment of seeds at 1.5% EMS for 12h. We obtained a number of mutant lines showing inheritable morphological changes in plant architecture, leaves, floral organs, fruits and other traits through M1, M2 and M3 generations. The F2 segregating populations were constructed and analyzed.We found that a short fruit mutant and a yellow-green fruit peel mutant were both under the control of a single recessive gene, respectively. These results provide valuable germplasm resources for the improvement of cucumber genetics and functional genomic research.展开更多

Here we report a novel rice mutant low temperature albino I (ltal) that showed albino leaves before 4-leaf stage when grown under temperature lower than 20℃, but developed normal green leaves under temperature higher than 24℃ or similar morphological phenotypes in dark as did the wild-type (WT). Our analysis showed that the contents of chlorophylls and chlo- rophyll precursors were remarkably decreased in the Ital mutant under low temperature compared to WT. Transmission electron microscope observation revealed that chloroplasts were defectively developed in the albino ltal leaves, which lacked of well-stacked granum and contained less stroma lamellae. These results suggested that the ltal mutation may delay the light-induced thylakoid assembly under low temperature. Genetic analysis indicated that the albino phenotype was controlled by a single recessive locus. Through map-based approach, we finally located the Ltal gene to a region of 40.3 kb on the short arm of chromosome 11. There are 8 predicted open reading frames (ORFs) in this region and two of them were deleted in ltal genome compared with the WT genome. The further characterization of the Ltal gene would provide a good approach to uncover the novel molecular mechanisms involved in chloroplast development under low temperature stress.展开更多

The mutant exhibited phenotypes of dwarfism and withered leaf tip (dwll). Based on the intemode length of dwll, this mutant belongs to the dm type of dwarfing. Analysis of elongation of the second sheath and m-amylase activity in endosperm showed that the phenotype caused by dwll was insensitive to gibberellin acid treatment. Using a large F2 population derived from a cross between the dwll and an indica rice variety, TN1, the DWL1 gene was mapped to the terminal region of the long arm of chromosome 3. Fine-mapping delimited it into a 46 kb physical distance between two STS markers, HL921 and HL944, where 6 open reading frames were predicted. Cloning of DWL1 will contribute to dissecting molecular mechanism that regulates plant height in rice, which will be beneficial to molecular assisted selection of this important trait.展开更多

AIM:To investigate the role of FAT10 and mutant p53 in the pathogenesis,severity and prognosis of gastric cancer.METHODS:FAT10,mutant p53 mRNA and protein levels were measured by reverse transcription(RT)-PCR and immu...AIM:To investigate the role of FAT10 and mutant p53 in the pathogenesis,severity and prognosis of gastric cancer.METHODS:FAT10,mutant p53 mRNA and protein levels were measured by reverse transcription(RT)-PCR and immunohistochemistry in gastric cancer tissue(n=62),tumoradjacent tissue(n=62)and normal gastric tissue(n=62).Relation of FAT10 and mutant p53 expression with clinicopathological features and clinical outcomes of gastric cancer patients were analyzed.RESULTS:The FAT10,mutant p53 mRNA and protein levels were signif icantly higher in gastric cancer than in its adjacent and normal tissue.The FAT10 and mutant p53 levels in gastric cancer tissue were significantly correlated with lymph node metastasis and tumor,nodes,metastasis(TNM)staging.Moreover,the high FAT10 level was associated with the overall survival rate of patients.Multivariate Cox-proportional hazards model analysis showed that mRNA and protein levels of FAT10 and mutant p53,lymph node metastasis,distant metastasis and TNM stage were the independent prognostic factors for gastric cancer.CONCLUSION:FAT10 may be involved in gastric carcinogenesis,and is a potential marker for the prognosis of gastric cancer patients.FAT10 and mutant p53 may play a common role in the carcinogenesis of gastric cancer.展开更多
The anthocyanin content in apple skin determines its red coloration,as seen in a Fuji apple mutant.Comparative RNA-seq analysis was performed to determine differentially expressed genes at different fruit development ...The anthocyanin content in apple skin determines its red coloration,as seen in a Fuji apple mutant.Comparative RNA-seq analysis was performed to determine differentially expressed genes at different fruit development stages between the wild-type and the skin color mutant.A novel R2R3-MYB transcription factor,MdMYB90-like,was uncovered as the key regulatory gene for enhanced coloration in the mutant.The expression of MdMYB90-like was 21.3 times higher in the mutant.MdMYB90-like regulates anthocyanin biosynthesis directly through the activation of anthocyanin biosynthesis genes and indirectly through the activation of other transcription factors that activate anthocyanin biosynthesis.MdMYB90-like bound to the promoters of both structural genes(MdCHS and MdUFGT)and other transcription factor genes(MdMYB1 and MdbHLH3)in the yeast one-hybrid system,electrophoretic mobility shift assay,and dual-luciferase assay.Transgenic analysis showed that MdMYB90-like was localized in the nucleus,and its overexpression induced the expression of other anthocyanin-related genes,including MdCHS,MdCHI,MdANS,MdUFGT,MdbHLH3,and MdMYB1.The mutant had reduced levels of DNA methylation in two regions(−1183 to−988 and−2018 to−1778)of the MdMYB90-like gene promoter,which might explain the enhanced expression of the gene and the increased anthocyanin content in the mutant apple skin.展开更多

It showed whole green-yellow plant from the seedling stage, reduced number of tillers and longer growth duration. The contents of chlorophyll, chlorophyll a, chlorophyll b and net photosynthetic rate in leaves of the mutant obviously decreased, as well as the number of spikelets per panicle, seed setting rate and 1000-grain weight compared with its wild-type parent. Genetic analyses on F1 and F2 generations of 824ys crossed with three normal green varieties showed that the chlorophyll-deficit mutant character was controlled by a pair of recessive nuclear gene. Genetic mapping of the mutant gene was conducted by using microsatellite markers and F2 mapping population of 495R/824ys, and the mutant gene of 824ys was mapped on the short arm of rice chromosome 3. The genetic distances from the target gene to the markers RM218, RM282 and RM6959 were 25.6 cM, 5.2 cM and 21.8 cM, respectively. It was considered to be a new chlorophyll-deficit mutant gene and tentatively named as chill(t).展开更多

摘要With the increasing accumulation of plastic pollutants in various environments,research on microorganisms(including bacteria,fungi,and algae)with plastic degradation capabilities has gained significant attention.However,only a limited number of microbial plastic-degrading enzymes have been identified to date.This highlights that the degradation mechanisms employed by many plastic-degrading microorganisms,particularly filamentous fungi,remain insufficiently explored.In this study,we utilized a versatile fungal plasmid(pCT74)to express green fluorescent protein(GFP)in a marine-derived fungus Alternaria alternata strain FB1 with plastic degradation capabilities.Upon evaluating the degradation effect of polyester-type polyurethane(PU)film,we observed that different transformants exhibited three kinds of activities(the same,reduced,or enhanced degradation capability)compared to the FB1 wild-type strain.Further analysis of the plasmid fragment insertion sites in different transformants revealed that pCT74 integrates randomly into the genome of the host fungus.Notably,a direct correlation was found between the plasmid insertion site and the degradation capability of the corresponding transformant.Our findings not only redefine the potential applications of plasmid pCT74 in filamentous fungi but also show a novel research approach to identifying key enzymes involved in plastic degradation by fungi.

摘要OsMAPK6 plays a critical role in regulating rice growth,development,and stress responses.However,the embryonic lethality associated with loss-of-function mutations prevents the generation of homozygous mutant seeds,significantly hindering functional studies of this gene.Although the weak mutant dsg1 has offered valuable insights into OsMAPK6 function,its extremely low seed-setting rate limits its use for detailed genetic analysis.Here,we employed prime editing to perform precise multi-site modifications at the C-terminus of OsMAPK6,generating a series of osmapk6 mutants with truncated proteins of varying lengths.Among these,the osmapk6(379)and osmapk6(383)mutants exhibited phenotypic defects similar to dsg1,while osmapk6(386)showed a significantly improved seed-setting rate despite persistent developmental defects.Through phenotypic characterization and protein functional analysis,we further clarified how different C-terminal deletion lengths affect plant growth,development,stress responses,and OsMAPK6 protein function.In summary,this study elucidates the importance of the OsMAPK6 C-terminus in rice biology and provides a fertile weak mutant with enhanced seed production,offering a valuable genetic resource for future research on OsMAPK6.

摘要KRAS is a critical proto-oncogene and molecular switch that is frequently mutated in human cancers.Oncogenic mutations,primarily at codons 12,13,and 61,lock KRAS into a GTPbound active state,thus resulting in constitutive signaling through downstream effectors such as RAF and phosphoinositide 3-kinase(PI3K)1.These alterations are highly prevalent in pancreatic cancer,colorectal cancer(CRC),and non-small cell lung cancer(NSCLC),and they drive tumor initiation,progression,and therapy resistance.

摘要Fusarium crown rot(FCR),predominantly caused by Fusarium pseudograminearum,has been listed as a Category Ⅱ disease in six provinces of China,posing a significant threat to wheat production.The phenylpyrrole fungicide fludioxonil is a key agent for FCR control.Previous studies indicated that resistance to fludioxonil in F.pseudograminearum is primarily associated with altered expression levels of the FpOS1 gene,which encodes a hybrid histidine kinase.However,the roles of mutations in other FpOS genes and the molecular interactions between FpOS proteins and fludioxonil remain elusive.To address these gaps,we generated 16 fludioxonil-resistant mutants with heritable resistance traits by in vitro selection of four sensitive F.pseudograminearum isolates.These mutants exhibited high resistance levels,with resistance factors(RF)ranging from 633.73 to 8617.07.Compared to their parental isolates,the resistant mutants showed significantly reduced mycelial growth rate,sporulation capacity,and pathogenicity.They were also more sensitive to ionic,osmotic,and oxidative stresses and displayed compromised cell wall and membrane integrity.Fludioxonil demonstrated no cross-resistance with tebuconazole or pydiflumetofen;however,it exhibited weak positive crossresistance to pyraclostrobin and moderate positive cross-resistance to iprodione.Fludioxonil treatment significantly promoted glycerol synthesis and inhibited deoxynivalenol(DON)production in parental isolates,whereas these regulatory effects were markedly attenuated in the resistant mutants.Mutation analysis identified mutation sites in FpOS1,FpOS4,and FpOS5 genes,with a lower mutation frequency in FpOS1 and no mutations detected in FpOS2.Molecular docking indicated that amino acid substitutions in FpOS4 and FpOS5 significantly reduced the binding affinity of fludioxonil to these target proteins.In conclusion,F.pseudograminearum poses a moderate risk of resistance to fludioxonil.Point mutations in FpOS4 and FpOS5 genes emerge as key molecular drivers of resistance,likely by diminishing the binding affinity between the fungicide and its proteins.This study clarifies the molecular basis of fludioxonil resistance in F.pseudograminearum and provides a scientific rationale for the judicious use of this fungicide in managing FCR.

摘要Huntington's disease(HD)is an inherited neurodegenerative disorder caused by poly-glutamine expansion in the mutant huntingtin(mHTT)protein.While the pathogenesis involves both cell-autonomous and non-cell-autonomous mechanisms,the role of specific intercellular crosstalk in HD remains unclear.The PLP-150Q mouse model,which expresses mHTT selectively in oligodendrocytes,serves as an excellent platform for studying the progression of HD in these cells.RNA sequencing of PLP-150Q mouse brains revealed significant alterations in immune-inflammatory pathways and glial dysfunction,particularly in the corpus callosum and striatum.Notably,we observed an age-dependent upregulation of key inflammatory factors specifically within the corpus callosum.Western blot and immunohistochemical analyses further demonstrated reactive gliosis,characterized by elevated Iba1+and CD68+microglia,as well as GFAP⁺and S100β+astrocytes,alongside decreased myelin protein levels.Our findings suggest that mHTT in oligodendrocytes triggers age-dependent inflammation,contributing to HD progression and revealing new mechanisms in its pathogenesis.

摘要Naturally occurring yellow leaf mutants are an important resource for studying pigment content and biosynthesis,as well as related gene expression.In our ongoing cultivation of Rehmannia chingii H.L.Li,we found an off-type yellow plant.The yellowing started with the new leaves and gradually spread downward until the entire plant exhibited a stable shade of yellow.We studied the differences in the chlorophyll and carotenoid content,carotenoid profile,and transcriptome of this yellow-leaf mutant(P2).Compared to the wild-type R.chingii plant(P1),P2 leaves had significantly lower chlorophyll and carotenoid content.LC-MS/MS analysis revealed that P2 had higher quantities of severalmetabolites in the carotenoid biosynthesis pathway.Transcriptome sequencing results showed that genes involved in porphyrin metabolism,carbon fixation,photosynthesis and antenna proteins,terpenoid backbone biosynthesis,and carotenoid biosynthesis were differentially expressed between P1 and P2.Large-scale expression differences were observed in the phytohormone and MAPK signaling pathways,as well as in 15 transcription factor families.We discuss possible mechanisms responsible for the yellowleaf color in P2.These preliminary data are valuable for further exploring the molecular mechanisms of leaf color formation and associated pathways.

摘要Lilium leichtlinii var.Maximowiczii is a new type of edible lily with important nutritional and economic value.However,it is prone to lodging,and its bulb often follows the roots.To obtain better lily germplasms,different concentrations of colchicine were used to induce the growth of L.leichtlinii var.Maximowiczii seeds.The identification was conducted via plant morphology,chromosomal ploidy analysis,and ISSR molecular markers.The best induction effect was achieved by treating seeds with 0.15%colchicine for 48 h,with a seedling variation rate of 31.67%.After 17 months of cultivation,11 mutant plants(Nos.1-11)survived.The No.2 plant grew rapidly and flowered earlier than the control plants.The No.3 plant was significantly dwarfed,and its leaves became wider.Flow cytometry and root tip squash analysis revealed that the No.3 plant was aneuploid,with 38 chromosomes in the root tip cells;the other plants were diploids,with 24 chromosomes.Microscopic observation revealed that the size of leaf epidermal cells and guard cells increased in the No.3 plant,and the stomatal density significantly decreased,consistent with the characteristics of the polyploid plants.Additionally,ISSR marker analysis revealed significant differences in the No.2 plant at the molecular genetic level compared with the control,with a variation rate of 18.99%.These results provide a reference for lily mutant breeding using the seeds by colchicine induction and the basis for further research on the early flowering mechanism in lily.
摘要AIM: To conduct a retrospective study in 400 chronic hepatitis B patients in order to identify hepatitis B viral factors associated with complications of liver disease or development of hepatocellular carcinoma. METHODS: The mean follow-up time was 83.6 ± 39.6 mo. Alpha-fetoprotein test and abdominal ultrasound were used for cancer surveillance. Hepatitis B basal core promoter mutants, precore mutants, genotypes, hepatitis B viral DNA (HBV DNA) level and hepatitis B e antigen (HBeAg) were measured. Univariate analysis and logistic regression were used to assess odds ratios for viral factors related to liver deaths and hepatocellular carcinoma development. RESULTS: During follow-up, 38 patients had liver deaths not related to hepatocellular carcinoma. On multivariate analysis, older age [odds ratio: 95.74 (12.13-891.31), P 〈 0.0001], male sex [odds ratio: 7.61 (2.20-47.95); P = 0.006], and higher Iogzo HBV DNA [odds ratio: 4.69 (1.16-20.43); P 〈 0.0001] were independently predictive for these liver related deaths. Also, 31 patients developed hepatocellular carcinoma. Multivariate analysis showed that older age [odds ratio: 26.51 (2.36-381.47); P = 0.007], presence of precore mutants [odds ratio: 4.23 (1.53-19.58), P = 0.02] and presence of basal core promoter mutants [odds ratio: 2.93 (1.24-7.57); P = 0.02] were independent predictors for progression to hepatocellular carcinoma. CONCLUSION: Our results show that high levels of baseline serum HBV DNA are associated with non- hepatocellular carcinoma-related deaths of liver failure, while genetic mutations in the basal core promoter and precore regions are predictive for development of hepatocellular carcinoma.

摘要[Objective] The aim was to improve the genetic property of peppers, the mutant population of Capsicum annuum L cultivar "6421" was constructed. [Method] The seeds of "6421" were treated with 0.2% to 1.2% ethyl methane sulfonate to identified LD50, and then 10 000 LD^o of treated seeds were sowed to construct mutant population. The agronomic characters and genetic regularity of dwarf mutants in M4 generation were analyzed. [Result] Our results showed that GR and SSR were 45.2% and 40.2% respectively at 1.0% EMS, close to LD50, with GI (17.6) and seed Ⅵ (19.7) being half of that of control; 562 M4 mutants were identified in 2015, and the mutation could be characterized according 11 major categories and 32 subcategories; Simultaneously, we found that plant height, plant width, diameter of mainstem, length of main-stem, the number of main-stem nodes and branch of lines E29, E58, E142 and E312 were all significantly different from that of the control. The mutation of lines E29, E58 and E312 was all controlled by a single recessive gene. [Conclusionl The study first created a pepper mutant population, which provides not only the germplasm resources for further breeding but also direct and effective materials for genomic study of the pepper.

摘要AIM:To investigate precore/basal core promoter(PC/BCP) mutants throughout hepatitis B virus(HBV) infection and to determine their relationship to hepatitis B early antigen(HBeA g) titers.METHODS:We enrolled 191 patients in various stages of HBV infection at the Huashan Hospital and the Taizhou Municipal Hospital from 2010 to 2012.None of the patients received antiviral therapy.HBV DNA from serum,was quantified by real-time PCR.The HBV genotype was determined by direct sequencing of the S gene.We used the Simpleprobe ultrasensitivequantitative method to detect PC/BCP mutants in each patient.We compared the strain number,percentage,and the changes in PC/BCP mutants in different phases,and analyzed the relationship between PC/BCP mutants and HBe Ag by multiple linear regression and logistic regression.RESULTS:Patients with HBV infection(n = 191) were assigned to groups by phase:Immune tolerance(IT) = 55,Immune clearance(IC) = 67,Low-replicative(LR) = 49,and HBeA g-negative hepatitis(ENH) = 20.Of the patients(male,112; female,79) enrolled,122 were HBe Ag-positive and 69 were HBe Ag-negative.The median age was 33 years(range:18-78 years).PC and BCP mutation detection rates were 84.82%(162/191) and 96.86%(185/191),respectively.In five HBe Ag-negative cases,we detected double mutation G1896A/G1899 A.The logarithm value of PC mutant quantities(log10 PC) significantly differed in IT,IC,and LR phases,as well as in the ENH phase(F = 49.350,P < 0.001).The logarithm value of BCP mutant quantities(log10 BCP) also differed during the four phases(F = 25.530,P < 0.001).Log10 PC and log10 BCP values were high in the IT and IC phases,decreased in the LR phase,and increased in the ENH phase,although the absolute value at this point remained lower than that in the IT and IC phases.PC mutant quantity per total viral load(PC%) and BCP mutant quantity per total viral load(BCP%) differed between phases(F = 20.040,P < 0.001; F = 10.830,P < 0.001),with PC% and BCP% gradually increasing in successive phases.HBeA g titers negatively correlated with PC%(Spearman's rho =-0.354,P < 0.001) and BCP%(Spearman's rho =-0.395,P < 0.001).The negative correlation between PC% and HBeA g status was significant(B =-5.281,P = 0.001),but there was no such correlation between BCP% and HBeA g status(B =-0.523,P = 0.552).CONCLUSION:PC/BCP mutants become predominant in a dynamic and continuous process.Log10 PC,log10 BCP,PC% and BCP% might be combined to evaluate disease progression.PC% determines HBeA g status.

摘要Cucumber is an important vegetable crop and a model crop for the study of sex expression in plants. However, the genomic resources and tools for functional genomics studies in cucumber are still limited. In this paper, we conducted ethyl methyl sulfone(EMS) mutagenesis in the northern China ecotype cucumber inbred line 406 to construct a mutant library. We optimized the conditions of EMS mutagenesis on inbred line 406 which included treatment of seeds at 1.5% EMS for 12h. We obtained a number of mutant lines showing inheritable morphological changes in plant architecture, leaves, floral organs, fruits and other traits through M1, M2 and M3 generations. The F2 segregating populations were constructed and analyzed.We found that a short fruit mutant and a yellow-green fruit peel mutant were both under the control of a single recessive gene, respectively. These results provide valuable germplasm resources for the improvement of cucumber genetics and functional genomic research.

摘要Albino mutants are useful genetic resource for studying chlorophyll biosynthesis and chloroplast development and cloning genes involved in these processes in plants. Here we report a novel rice mutant low temperature albino I (ltal) that showed albino leaves before 4-leaf stage when grown under temperature lower than 20℃, but developed normal green leaves under temperature higher than 24℃ or similar morphological phenotypes in dark as did the wild-type (WT). Our analysis showed that the contents of chlorophylls and chlo- rophyll precursors were remarkably decreased in the Ital mutant under low temperature compared to WT. Transmission electron microscope observation revealed that chloroplasts were defectively developed in the albino ltal leaves, which lacked of well-stacked granum and contained less stroma lamellae. These results suggested that the ltal mutation may delay the light-induced thylakoid assembly under low temperature. Genetic analysis indicated that the albino phenotype was controlled by a single recessive locus. Through map-based approach, we finally located the Ltal gene to a region of 40.3 kb on the short arm of chromosome 11. There are 8 predicted open reading frames (ORFs) in this region and two of them were deleted in ltal genome compared with the WT genome. The further characterization of the Ltal gene would provide a good approach to uncover the novel molecular mechanisms involved in chloroplast development under low temperature stress.

摘要A green-revertible albino mutant-Qiufeng M was found from the japonica rice(Oryza sativa L.ssp.japonica)Qiufeng in the field.The first three leaves of the mutant were albino with some green.The leaf color became pale green since the fourth leaf and the glume had the same phenomenon as the first three leaves.The measuring data of the pigment content confirmed the visually observed results.It truly had a remarkable changing process in the leaf color in Qiufeng M.Comparison of the main agronomic characters between Qiufeng and Qiufeng M indicated that the neck length and grain weight showed significant difference at the 1%level,and other characters were not different.Genetic analysis showed that the green-revertible albino trait was controlled by a single recessive nucleic gene.Using 209 recessive mutant individuals in the F2 population derived from the cross Pei'ai 64S×Qiufeng M,a gene,tentatively named gra(t),was located between the SSR markers of RM475 and RM2-22 on the long arm of chromosome 2.The genetic distance were 17.3 cM and 2.9 cM respectively.

摘要A dwarf mutant of rice (Oryza sativa L.) by mutagenesis of ethylene methylsulfonate (EMS) treatment from Nipponbare was identified. The mutant exhibited phenotypes of dwarfism and withered leaf tip (dwll). Based on the intemode length of dwll, this mutant belongs to the dm type of dwarfing. Analysis of elongation of the second sheath and m-amylase activity in endosperm showed that the phenotype caused by dwll was insensitive to gibberellin acid treatment. Using a large F2 population derived from a cross between the dwll and an indica rice variety, TN1, the DWL1 gene was mapped to the terminal region of the long arm of chromosome 3. Fine-mapping delimited it into a 46 kb physical distance between two STS markers, HL921 and HL944, where 6 open reading frames were predicted. Cloning of DWL1 will contribute to dissecting molecular mechanism that regulates plant height in rice, which will be beneficial to molecular assisted selection of this important trait.

摘要Plant lesion mimic mutants(lmms)generally possess autoimmunity and hypersensitive response(HR)-like cell death in the absence of biotic or abiotic stress.They have attracted much attention because they are useful tools for deciphering the interaction between defense signaling and growth.Recent studies have identified more than 30 lmms involved in the plant immune response and cell death in rice.Genes underlying these lmms,coding for diverse types of proteins,mainly regulate transcription,protein translation and modification,vesicular trafficking and catalyzation of metabolism.Here,we presented an overview of the most recent advances on the study of lmms in rice and proposed a perspective on potential utilization of LMM genes in agriculture.
摘要AIM:To investigate the role of FAT10 and mutant p53 in the pathogenesis,severity and prognosis of gastric cancer.METHODS:FAT10,mutant p53 mRNA and protein levels were measured by reverse transcription(RT)-PCR and immunohistochemistry in gastric cancer tissue(n=62),tumoradjacent tissue(n=62)and normal gastric tissue(n=62).Relation of FAT10 and mutant p53 expression with clinicopathological features and clinical outcomes of gastric cancer patients were analyzed.RESULTS:The FAT10,mutant p53 mRNA and protein levels were signif icantly higher in gastric cancer than in its adjacent and normal tissue.The FAT10 and mutant p53 levels in gastric cancer tissue were significantly correlated with lymph node metastasis and tumor,nodes,metastasis(TNM)staging.Moreover,the high FAT10 level was associated with the overall survival rate of patients.Multivariate Cox-proportional hazards model analysis showed that mRNA and protein levels of FAT10 and mutant p53,lymph node metastasis,distant metastasis and TNM stage were the independent prognostic factors for gastric cancer.CONCLUSION:FAT10 may be involved in gastric carcinogenesis,and is a potential marker for the prognosis of gastric cancer patients.FAT10 and mutant p53 may play a common role in the carcinogenesis of gastric cancer.

摘要The anthocyanin content in apple skin determines its red coloration,as seen in a Fuji apple mutant.Comparative RNA-seq analysis was performed to determine differentially expressed genes at different fruit development stages between the wild-type and the skin color mutant.A novel R2R3-MYB transcription factor,MdMYB90-like,was uncovered as the key regulatory gene for enhanced coloration in the mutant.The expression of MdMYB90-like was 21.3 times higher in the mutant.MdMYB90-like regulates anthocyanin biosynthesis directly through the activation of anthocyanin biosynthesis genes and indirectly through the activation of other transcription factors that activate anthocyanin biosynthesis.MdMYB90-like bound to the promoters of both structural genes(MdCHS and MdUFGT)and other transcription factor genes(MdMYB1 and MdbHLH3)in the yeast one-hybrid system,electrophoretic mobility shift assay,and dual-luciferase assay.Transgenic analysis showed that MdMYB90-like was localized in the nucleus,and its overexpression induced the expression of other anthocyanin-related genes,including MdCHS,MdCHI,MdANS,MdUFGT,MdbHLH3,and MdMYB1.The mutant had reduced levels of DNA methylation in two regions(−1183 to−988 and−2018 to−1778)of the MdMYB90-like gene promoter,which might explain the enhanced expression of the gene and the increased anthocyanin content in the mutant apple skin.
摘要A rice etiolation mutant 824ys featured with chlorophyll deficiency was identified from a normal green rice variety 824B. It showed whole green-yellow plant from the seedling stage, reduced number of tillers and longer growth duration. The contents of chlorophyll, chlorophyll a, chlorophyll b and net photosynthetic rate in leaves of the mutant obviously decreased, as well as the number of spikelets per panicle, seed setting rate and 1000-grain weight compared with its wild-type parent. Genetic analyses on F1 and F2 generations of 824ys crossed with three normal green varieties showed that the chlorophyll-deficit mutant character was controlled by a pair of recessive nuclear gene. Genetic mapping of the mutant gene was conducted by using microsatellite markers and F2 mapping population of 495R/824ys, and the mutant gene of 824ys was mapped on the short arm of rice chromosome 3. The genetic distances from the target gene to the markers RM218, RM282 and RM6959 were 25.6 cM, 5.2 cM and 21.8 cM, respectively. It was considered to be a new chlorophyll-deficit mutant gene and tentatively named as chill(t).
