AIM: To review published clinical studies examining the effect of natamycin in the treatment of fungal keratitis.METHODS: We selected the publications in CENTRAL,MEDLINE, EMBASE, CNKI, and CBM. This study systematically reviewed published randomized controlled trials(RCTs) that compared natamycin to other antifungal agents, and conducted feasible Meta-analysis of efficacy results using Revman 5.2 software.RESULTS: We included seven trials which were mainly carried out in developing countries of Asia, with five trials conducted in India, one each in China and Bangladesh. A total of 804 participants were randomized to following comparisons: 2% econazole versus 5%natamycin showed little difference in the effects of treatment of fungal keratitis [RR =0.99, 95% confidence interval(CI), 0.8 to 1.21]; chlorhexidine gluconate versus5% natamycin indicated that the results on healing of the ulcer at 21 d was less conclusive(RR=0.77, 95% CI, 0.55 to 1.08; I2=0%); 1% voriconazole versus 5% natamycin suggested that natamycin treatment appeared to be significantly better outcomes than voriconazole(regression coefficient =-0.18 log MAR; 95% CI,-0.30 to-0.05; P =0.006), especially in Fusarium cases(regression coefficient=-0.41 log MAR; 95% CI,-0.61 to-0.20; P 【0.001);natamycin versus fluconazole showed a significant difference in cure rate(χ2=5.048, P 【0.05) and natamycin group was more effective than fluconazole in average period of therapy(t =7.94, P 【0.01).CONCLUSION: Natamycin was a preferable choice in the treatment of fungal keratitis, especially in the early period of Fusarium cases. 展开更多

AIM: To evaluate the efficacy of topical administration Natamycin, which is produced by China, in an experimental rabbit model of Fusarium solani keratitis, to provide experimental basis for the application of clinical safety. METHODS: Fusarium solani was induced in the right eye of 30 New Zealand rabbits. Forty-eight hours after inoculation, the animals were divided into 3 different treatment groups, 10 rabbit eyes of each group: Group 1 (Natamycin) treated with topical Natamycin, group 2 (Natacyn) treated with topical Natacyn, group 3 (control) treated with topical saline solution. The eyes of each group was examined clinically with slit lamp using ulcer scoring system on day 4, 10, 15, and 21 for status of healing, corneal vascularisation, iritis, hypopyon and macular nebula. The findings were recorded on day 10 and day 21. RESULTS: Ulcer score on day 10, day 15, day 21: The score of Natamycin group are 1.45 +/- 0.16, 1.08 +/- 0.11, 0.70 +/- 0.40. The score of Natacyn group are 1.35 +/- 0.12, 1.10 +/- 0.12, 0.65 +/- 0.35. the score of control group are 1.30 +/- 0.08, 3.63 +/- 0.28, 3.80 +/- 0.16. Natamycin group and Natacyn group were different from control group (P <0.01). There is no difference between Natamycin group and Natacyn group. Status of healing on day 10 and day 21: The cure rate of the Natamycin group is 90% on day 10, and 100% on day 21. The cure rate of the Natacyn group is 80% on day 10, and 100% on day 21.Natamycin group and Natacyn group were different from control group (P<0.01). There is no difference between Natamycin group and Natacyn group. Corneal vascularisation, iritis, hypopyon and macular nebula on day 10 and day 21: in Natamycin group, the number of the eyes which have Corner vascularisation, iritis, hypopyon and macular nebula are 2,0,0,2. In Natacyn group, the number of the eyes which have Corner vascularisation, iritis, hypopyon and macular nebula are 1,0,0,2. In control group, the number of the eyes which have Corner vascularisation, iritis, hypopyon and macular nebula are 9,9,8,9.Natamycin group and Natacyn group were different from control group (P<0.01). There is no difference between Natamycin group and Natacyn group. CONCLUSION: Natamycin was found to be effective in fungal keratitis, similar to Natacyn, and it can stop the corner vascularisation, iritis, hypopyon and macular nebula to happen. Natamyin manufactured in China is effective against fungal keratitis, with esay availability and low toxicity in its use. 展开更多

An improved mutant producing natamycin, S. gilvosporeus LK-45, was screened, which showed natamycin productivity of 1420mg·L^-1. A mutant resistant to 2-deoxy glucose, S.gilvosporeus LK-119, was further obtained using a'rational screening procedure. The natamycin productivity of 1940mg·L^-1 was achieved when glucose was used as the carbon source. 展开更多

Ground cherry(Physalis pubescens L.) is a kind of berry fruit favored by consumers in China; however, this fruit is particularly vulnerable to physiological senescence and pathogen attack during the traditional cold storage period. In order to maintain storage quality, a 1.5%(w/w) chitosan(CS) water solution containing 50 mg/L of natamycin(NA) was introduced. After all treatments were completed, the fruit was stored at 0°C and sampled every 10 d. At each sampling date, the following tests were performed: mold and yeast analyses; enzyme activity and content analyses which included superoxide dismutase(SOD), ascorbate peroxidase(APX), and malondialdehyde(MDA); and color analysis. In addition, a sensory evaluation was carried out for quality assessment at the end of the storage period. The results showed that the application of a chitosan coating combined with natamycin(CSNA) enhanced the activity of superoxide dismutase(SOD) and ascorbate peroxidase(APX), reduced the physiological rate of senescence, and inhibited pathogen attack. Thus, CSNA treatment can maintain ground cherries at an acceptable level of storage quality for 50 d. 展开更多

The aim was to research fresh-keeping effects of natamycin on cold-pre- served grape. Red globe grapes were processed with compound coating liquid of chitosan with mass fraction at 1% and natamycin with mass fractions at 0.20% (T1), 0.40% (T2) and 0.60% (T3), respectively. Grapes processed with water (CK3) and 1% chitosan (CK2) were taken as control groups. Rotten rate, seed shattering rate, mass loss rate, respiratory intensity and related physiological quality in test and control groups were compared. The results indicated that respiratory intensity, mass loss rate, rotten rate and seed shattering rate in CK1 were all higher than those in CK2. In addition, T1, T2 and T3 were lower in the indices than CK1 and CK2, but still kept at a high level in fruit hardness. Furthermore, mass fractions of Vc and titratable acid declined more slowly in T1, T2 and T3, compared with CK1 and CK2. Natamycin better preserved grapes and prolonged storage period. In general, natamycin with mass fraction at 0.4% proved best in fresh-keeping. 展开更多

A fluorescence immunochromatographic strip was developed in this study for natamycin detection in food. The results showed that the best amount of labeled antibody was 10 μg, for every 50 μl of fluorescent microspheres with a 2.5%(w/v) concentration. This labeled antibody was diluted for 10 times, and the diluted solution was dispensed into conjugate pad at the amount of 3 μl/cm. The concentrations of natamycin labeled BSA for test line and goat anti-mouse IgG for control line were 2.0 and 1 mg/ml, respectively, which performed best. With the best conditions, the limit of detection was 1 ng/ml, the linearity ranged from 2 to 100 ng/ml, the recovery was about 80% to 120%, and the CV was below 23%. 展开更多

Development and Characterisation of Natamycin Mini-Matrices Prepared by Hot-Melt Extrusion for Vaginal Delivery 认领 引用

In this study, bioadhesive mini-matrices of natamycin were prepared for vaginal application by hot-melt extrusion. In addition, melt viscosity measurements, thermogravimetric analysis, in vitro drug release studies and in vitro mucoadhesion test were performed. High molecular weight grades of KlucelTM hydroxypropylcellulose were used as a thermoplastic polymer. TEC and PEG 400 were chosen as plasticizer. According to the obtained results of melt viscosity measurements, the maximum torque of extrudates prepared using PEG 400 increased with increasing drug loading. The thermo-gravimetric analyses showed that natamycin is stable up to 198℃ and this result gives the opportunity to hot melt extrussion process at 90℃. In vitro drug release results showed that the release was extended up to 72 hours and drug release rate increased with increasing drug loading. In respect to the in vitro mucoadhesion test results, the values of work of mucoadhesion were found high as 771,977 mN.mm, 753,199 mN.mm, 686,356 mN.mm for the prepared hot melt extruded mini-matrices. Our results showed that the developed formulations were found worthy of further studies. 展开更多

This study has investigated cuproptosis caused by copper sulfate(CuSO4)in Escherichia coli O157:H7 and its mechanism.Meanwhile,the mechanism of CuSO4 combined with natamycin(Nata)against E.coli O157:H7 planktonic and biofilm cells were also investigated.Finally,we prepared an agar-based film,researched its properties,and used it for packaging cherry tomatoes and purple kale.The results indicated that CuSO4 could cause cuproptosis in E.coli O157:H7 and lead the genes related to Fe-S cluster protein(hyfA,sufB,xdhC,and narY)to down-regulate and cuproptosis-related proteins to change.Then,CuSO4 combined with Nata could synergistically decrease E.coli O157:H7 bacterial counts in planktonic condition and biofilm by synergistically affecting the membrane potential,cytoplasm leakage,permeability of outer membrane and inner membrane.After that,CuSO4 and Nata could change the mechanical properties,color and optical characteristics of agarbased film,and decrease the water vapor permeability and oxygen permeability value.Finally,CuSO4@Nata@Carboxymethylcellulose@Agar film effectively lowered the counts of E.coli O157:H7 in cherry tomatoes and purple kale during the storage. 展开更多

The combination of radio frequency(RF)heating and natamycin has been demonstrated to exert a synergistic fungicidal effect on Aspergillus niger spores in shiitake mushrooms.However,the underlying synergistic fungi-cidal mechanism remains unclear.This study investigated the synergistic fungicidal mechanism of RF heating combined with natamycin against A.niger spores in shiitake mushrooms at cellular and molecular levels.Results indicated that the combined treatment improved membrane permeability and facilitated intracellular substance leakage,with an increase in OD260 value(0.530±0.039)and OD280 value(0.562±0.007),while a decrease in the ATP concentration(0.22±0.03μmol/L).Additionally,the mitochondrial function of spores was damaged,and the fluorescence intensity of reactive oxygen species content increased to 36.2±0.34 AU.A total of 1391 differentially expressed genes and 288 differentially expressed proteins were identified after the combined treatment.The primary mechanism of the combined treatment was the significant inhibition of intracellular ATP synthesis,oxidoreductase activity,transcription,and translation processes at both the transcript and protein levels.Furthermore,the combined treatment exacerbated damage to the cell membrane by intensifying dis-ruptions in membrane transport functions.These alterations in gene and protein expression triggered diverse cellular dysfunctions,ultimately resulting in a synergistic fungicidal effect on A.niger spores.This study provides theoretical support for controlling fungal contamination in agricultural products via the combined application of RF heating and natamycin. 展开更多

There is increasing interest in the development of Natamycin(Nata)containing ingredients appropriate as substitutes for chemical food preservatives,due to its safe and efficient antimycotic effects.This work reported that gliadin(Gli)could effectively perform as nano-vehicles to enhance Nata’s water dispersibility and stability.Nata-Gli nanoparticles(NPs)were prepared by adjusting the solution pH and anti-solvent method using the amphiphilicity of Gli.The encapsulation rate of NPs could reach 85.5%,and the loading rate was 8.6%.Nata-Gli/CMC nanocomposite system was prepared using sodium carboxymethylcellulose(CMC)as an antisolvent.Compared with the Nata-Gli NPs,the spherical structure of the particles with CMC addition is more apparent and more homogeneous.The bacteriostatic properties of the Nata-Gli/CMC nanocomposite system against Penicillium expansum were tested in vitro and in vivo.The results in vitro results confirmed that the application of Nata-Gli/CMC nanocomposite system improved the water dispersibility of natamycin and thus enhanced the bacteriostatic activity of natamycin.The results in vivo indicated that Nata-Gli/CMC nanocomposite system reduced the occurrence of rot and mildew in inoculated kumquat.The findings provide an effective strategy to provide a new idea for applying Nata and provide a reference for developing the post-harvest colloidal delivery system. 展开更多

为探明食品保鲜剂脱氢乙酸钠(sodium dehydroacetate,SD)和纳他霉素(natamycin,Nata)在植物病害防治中的潜在应用价值及其抑菌作用,以水稻稻瘟病菌为对象,测定SD和Nata对其菌丝平皿生长和致病性的影响。通过分析SD和Nata处理后稻瘟病菌... 为探明食品保鲜剂脱氢乙酸钠(sodium dehydroacetate,SD)和纳他霉素(natamycin,Nata)在植物病害防治中的潜在应用价值及其抑菌作用,以水稻稻瘟病菌为对象,测定SD和Nata对其菌丝平皿生长和致病性的影响。通过分析SD和Nata处理后稻瘟病菌孢子萌发率、附着胞形成、菌体超微结构变化、细胞膜通透性、液泡和线粒体完整性等指标,揭示SD和Nata的潜在抑菌机制。结果表明,SD和Nata对稻瘟病菌菌丝生长具有明显抑制作用,SD的有效中浓度(EC50)为47.40 mg·L-1,Nata的EC50为4.49 mg·L-1。采用4倍EC50浓度的SD和Nata喷雾处理可有效减轻稻瘟病的发生,病害严重度分别降低31.65和24.18百分点。SD和Nata处理随浓度升高显著抑制孢子萌发和附着胞形成。经SD和Nata处理后,稻瘟病菌细胞结构受损,菌丝体出现变形和皱缩;细胞膜通透性改变,导致胞内核酸、蛋白质等大分子物质外渗。碘化丙啶(propidium iodide,PI)和伊文思蓝(Evans blue,EB)染色后,菌丝体荧光强度显著增强。此外,SD和Nata处理使菌丝体液泡无法正常形成,SD处理还显著降低胞内线粒体活性。以上结果表明,SD和Nata主要通过破坏真菌细胞结构、干扰菌体物质代谢和能量代谢实现其抑菌效果。 展开更多

为研究纳他霉素复合壳聚糖处理对采后黄皮(25±1)℃货架期品质的影响,以“贵妃”黄皮为试验材料,以蒸馏水处理为对照组,采用不同浓度纳他霉素复合壳聚糖涂膜处理黄皮,比较其色泽、营养成分及其他生理生化指标的变化。结果表明,1000 ... 为研究纳他霉素复合壳聚糖处理对采后黄皮(25±1)℃货架期品质的影响,以“贵妃”黄皮为试验材料,以蒸馏水处理为对照组,采用不同浓度纳他霉素复合壳聚糖涂膜处理黄皮,比较其色泽、营养成分及其他生理生化指标的变化。结果表明,1000 mg/L纳他霉素+1.0%壳聚糖处理组保鲜效果最佳,能有效延长采后黄皮货架期(贮藏8 d时,好果率达80%以上),延缓果实失重和褐变进程。与对照组相比,该复合保鲜剂处理有助于保持果实硬度和可溶性固形物含量,同时可减缓可滴定酸含量和维生素C(vitamin C,VC)含量的下降。进一步研究发现,该复合保鲜剂处理显著抑制黄皮果皮和果肉中丙二醛含量的上升,其含量分别比对照组低6.69 nmol/g和4.97 nmol/g(P<0.05)。此外,复合保鲜剂处理还可以抑制多酚氧化酶的活性,使过氧化氢酶、过氧化物酶、超氧化物歧化酶的活性维持在较高水平。因此,适宜浓度的纳他霉素复合壳聚糖处理能有效抑制黄皮采后品质的下降,可作为一种绿色环保的方法应用于黄皮贮藏保鲜。 展开更多

In the present study,a simple,rapid,sensitive,robust,convenient,economical and environmentally friendly Capillary Zone Electrophoresis(CZE)method based on the use of non-toxic chemicals was developed to monitor the concentrations of natamycin,which is used to prevent mold and yeast that may occur in cheese crust,cheese crumb,yogurt and ayran.Additionally,a LC-ESI-MS method was developed and used for confirmation as well.The validated methods produced excellent results in terms of linearity(R2>0.999)and recovery values(ranging from 99.06%to 100.54%)and successfully applied for the analysis of natamycin,according to European regulations and Turkish Food Codex in 14 different brands of ayran,17 different brands of yogurt and 12 different brands of Kashar cheese collected from local markets.CZE demonstrated flexibility,sensitivity and selectivity,accuracy,robustness,and low cost.The developed CZE method is very advantageous because it is environmentally friendly and economical due to the analysis time and the absence of organic solvents.In addition,the reliability of the study was increased by using the internal standard and possible errors were prevented.Both CZE and LC-MS methods provided good results for natamycin analysis and these methods can be used for natamycin analysis in dairy products and possible pharmaceuticals.Finally,embracing the primary concepts of green chemistry,the proposed technique can be taken into consideration friendly to the nature,sustainable and“reagent-free”.These assays are easily applicable as quality control methods for natural products and pharmaceuticals containing natamycin. 展开更多

目的:对比分析伏立康唑滴眼液与那他霉素滴眼液分别联合角膜溃疡清创术治疗真菌性角膜炎(FK)患者的临床疗效及对菌丝浸润深度的影响。方法:回顾性分析2019年6月~2024年6月本院收治的80例接受角膜溃疡清创术的FK患者病历资料,根据不同治... 目的:对比分析伏立康唑滴眼液与那他霉素滴眼液分别联合角膜溃疡清创术治疗真菌性角膜炎(FK)患者的临床疗效及对菌丝浸润深度的影响。方法:回顾性分析2019年6月~2024年6月本院收治的80例接受角膜溃疡清创术的FK患者病历资料,根据不同治疗方法分为对照组(n=38)和观察组(n=42)。两组患者术后同时给予左氧氟沙星滴眼液进行基础治疗,对照组给予那他霉素滴眼液治疗,观察组给予伏立康唑滴眼液治疗,两组均连续治疗4周。比较两组临床疗效、菌丝浸润深度、眼部总评分、视力情况及不良反应发生情况。结果:治疗4周后,观察组临床治疗总有效率(90.48%)高于对照组(73.68%,P<0.05);菌丝浸润深度及眼部总评分均低于对照组(P<0.05);视力情况优于对照组(P<0.05)。两组不良反应总发生率比较无统计学差异(χ2=0.339,P=0.560)。结论:在角膜溃疡清创术治疗基础上,给予伏立康唑滴眼液可有效提高FK患者临床疗效,改善菌丝浸润深度、眼部总评分及视力。 展开更多

目的 探析真菌性角膜炎患者治疗期间联合那他霉素滴眼液进行睑缘擦拭的作用。方法 真菌性角膜炎患者90例,按照随机数表法划分为空白组、对照组、试验组,每组30例。空白组予以常规抗真菌治疗及相应眼部干预;对照组在空白组治疗干预基础... 目的 探析真菌性角膜炎患者治疗期间联合那他霉素滴眼液进行睑缘擦拭的作用。方法 真菌性角膜炎患者90例,按照随机数表法划分为空白组、对照组、试验组,每组30例。空白组予以常规抗真菌治疗及相应眼部干预;对照组在空白组治疗干预基础上加生理盐水进行睑缘擦拭;试验组在空白组治疗干预基础上加那他霉素滴眼液与生理盐水按1∶1比例稀释后的混合液进行睑缘擦拭。比较三组患者临床疗效、治疗期间时间指标、转阴率及复发率。结果 三组患者临床总有效率比较,差异显著(P0.05)。试验组角膜溃疡愈合时间及眼痛、畏光、异物感缓解时间分别为(20.23±3.10)、(6.48±1.95)、(8.28±2.01)、(5.62±1.75)d,均短于对照组的(24.83±3.47)、(8.12±2.35)、(10.32±2.64)、(7.98±2.14)d与空白组的(28.51±4.22)、(10.22±2.81)、(12.51±3.15)、(9.85±2.55)d,且对照组短于空白组,差异显著(P0.05)。结论 真菌性角膜炎患者基于常规抗真菌治疗及相应眼部干预,联合那他霉素滴眼液进行睑缘擦拭可增强临床治疗效果,加快相关症状的缓解速度及溃疡愈合速度,提升真菌消除率,降低疾病复发率,值得推广应用。 展开更多
