BACKGROUND Icariin(ICA)is a natural compound derived from Epimedium that has shown promise in treating colorectal cancer(CRC)by reducing inflammation,inducing apoptosis,and limiting tumor progression.AIM To elucidate ...BACKGROUND Icariin(ICA)is a natural compound derived from Epimedium that has shown promise in treating colorectal cancer(CRC)by reducing inflammation,inducing apoptosis,and limiting tumor progression.AIM To elucidate the specific mechanisms underlying its therapeutic effects on this malignancy.METHODS We examined the impact of different ICA concentrations on the biological behavior of CRC cells in vitro.We also assessed the protein expression of SMAD4,CX-C motif chemokine ligand 1(CXCL1),and CXCL8,as well as epithelial-mesenchymal transition(EMT)-related signaling molecules,and the mRNA expression of proinflammatory factors.To further explore ICA’s mechanisms in CRC treatment,we introduced small interfering SMAD4 and its negative control group.In CRC cells,we evaluated proliferation,apoptosis,invasion,migration,EMT,and the recruitment of tumor-associated neutrophils(TANs).Moreover,we investigated the role of the CXCL8-C-X-C motif chemokine receptor 2(CXCR2)axis in the recruitment of ICA-affected TANs.In vivo,we preliminarily assessed the therapeutic efficacy of ICA against CRC.In ICA-treated mice,we measured tumor size,histological morphology,apoptosis,and neutrophil infiltration.Additionally,we analyzed the expression of SMAD4,CXCL1,CXCL8,CXCR2,EMT,and proteins related to migration.RESULTS Our findings indicate that ICA attenuated CRC cell proliferation,invasion,and migration,while suppressing TAN recruitment and promoting apoptosis in a concentration-dependent manner.When small interfering SMAD4 was introduced,the anticancer effects of ICA were diminished.Furthermore,disruption of the CXCL8-CXCR2 axis reduced neutrophil recruitment.We found that ICA inhibited tumor growth,improved tumor histology and morphology,alleviated CD66b+neutrophil infiltration,and inhibited EMT in mice with subcutaneous CRC tumors.In addition,ICA appeared to enhance SMAD4 protein expression while suppressing CXCL8 and CXCR2 expression in both in vivo and in vitro experiments.CONCLUSION ICA impedes the recruitment of TANs primarily through SMAD4-mediated modulation of the CXCL8-CXCR2 axis,thereby suppressing EMT and other pro-metastatic programs in CRC.展开更多
Objective:Multiple programmed cell death(PCD)pathways have been individually reported to be triggered by cisplatin,but whether and how they are co-regulated remains unclear.In this study,we comprehensively investigate...Objective:Multiple programmed cell death(PCD)pathways have been individually reported to be triggered by cisplatin,but whether and how they are co-regulated remains unclear.In this study,we comprehensively investigate the spectrum of cisplatin-induced PCD.Methods:We employed integrated in vitro and in vivo models including human cancer cell lines,a Cal27 xenograft mouse model,and paired clinical specimens from an oral squamous cell carcinoma patient receiving neoadjuvant cisplatin-based chemotherapy.A comprehensive methodological suite-encompassing cell death assays,Western blotting,Hematoxylin and eosin staining,immunofluorescence,Cyclic multiplexed tissue staining,and pathway-specific pharmacological inhibitors was utilized to dissect the activation of apoptosis,necroptosis,pyroptosis,and ferroptosis.Results:Cisplatin simultaneously upregulates markers of PCD pathways(including apoptosis,necroptosis,pyroptosis,and ferroptosis)in a dose-or time-dependent manner.Pharmacological inhibition or genetic knockdown of key genes in each pathway significantly reduced cytotoxicity.confirming their functional roles.Notably,indicators of key pro-inflammatory death modalities,pyroptosis and ferroptosis,were prominently co-upregulated in both xenograft tumors and clinical patient samples,suggesting that these two forms of PCD may represent the predominant death forms in cisplatin-induced tumor cell death.Conclusion:Cisplatin induces the coordinated activation of multiple cell death programs within a unified framework.Prominent engagement of immunogenic cell death pathways,particularly pyroptosis and ferroptosis,provides a mechanistic basis for the clinically observed synergy between cisplatin and immune checkpoint blockade therapy.展开更多

Background Liver cancer is among the top five causes of cancer death in 90 countries,with China accounting for a substantial proportion of the global burden.This study aimed to analyse the national liver cancer polici...Background Liver cancer is among the top five causes of cancer death in 90 countries,with China accounting for a substantial proportion of the global burden.This study aimed to analyse the national liver cancer policies and programs in China.Methods This study applied a documentary research method using the systematic READ approach.Six national official websites and one public policy database were searched.The document analysis was based on the WHO Health System's Six Building Blocks and the WHO's four modules of cancer control(prevention,early detection,diagnosis and treatment,and palliative care).Results A total of 74 liver cancer-related policies and 8 programs published from 1 January 1984 to 31 March 2025 were included in this study.The analysis revealed that liver cancer management in China mainly focused on Medical products and Technologies(n=46,62.16%),and Service Delivery(n=34,45.95%)within the WHO health system building blocks.When it came to WHO cancer control four modules,most policies(n=39,52.70%)targeted Diagnosis and Treatment of liver cancer,followed by Prevention of liver cancer(n=33,44.59%).Additionally,8 national programs were implemented to improve the prevention,diagnosis,and treatment of liver cancer.Before 2019,the emphasis of liver cancer prevention was primarily on hepatitis prevention and control.However,since 2020,the national-level programs aimed at preventing and controlling liver cancer emphasizing patient education and treatment for high-risk groups.Conclusions China's liver cancer control mainly focused heavily on prevention,diagnosis and treatment modules with special focus on medical products and technology,as well as service delivery.Currently,less attention has been given to the detection and palliative care of survivors.The control of liver cancer in China still requires further strengthening of the health system for implementation.Considering the continual increase in the burden of liver cancer,it is imperative for future efforts to develop a comprehensive national liver cancer strategy.展开更多

Severe aortic stenosis has increased exponentially as a result of the aging of the population.Transcatheter aortic valve replacement(TAVR)procedures increase by 20%-30%per year in Spain.Referred patients are increasin...Severe aortic stenosis has increased exponentially as a result of the aging of the population.Transcatheter aortic valve replacement(TAVR)procedures increase by 20%-30%per year in Spain.Referred patients are increasingly older,have comorbidities,and frailty.TAVR cardiac rehabilitation(CR)programs significantly improve both functional capacity and quality of life of patients.However,there are no guidelines on how to carry out them and few elderly patients are referred to CR.Furthermore,mortality in women who undergo TAVR is higher than in men and even fewer are referred to CR programs.Multidisciplinary patient care,including comorbidities and frailty,is essential.It is necessary to ensure exquisite continuity of care during TAVR,CR,and the rest of the patient's life.Telerehabilitation could be an option in some lower-risk patients to include more patients in CR programs after TAVR,given the high demand.This article reviews the evidence on why CR should be performed in post-TAVR patients and proposes a practical and novel approach to the care process and the recommended aspects and components of the CR program.展开更多
Background Supplementation of choline chloride in culture medium programs the preimplantation bovine embryo to increase weaning weight of the resultant calf.Here,it was hypothesized that similar programming actions of...Background Supplementation of choline chloride in culture medium programs the preimplantation bovine embryo to increase weaning weight of the resultant calf.Here,it was hypothesized that similar programming actions of choline can be induced by feeding rumen-protected choline(RPC)to beef cows during the periconceptional period.Results A preliminary experiment was conducted to determine changes in circulating concentrations of choline and its metabolites after RPC supplementation.Suckled beef cows were individually fed 0,30,60,and 90 g of RPC(i.e.,0,8.6,17.3 and 25.9 g choline chloride)and blood samples were collected at random times after feeding.There were no differences in plasma concentrations of choline or its metabolites between groups.In the second experiment,effects of feeding 60 g/d RPC from d-1 to 7 relative to timed artificial insemination were examined for suckled beef cows.Feeding RPC did not affect pregnancy or calving rates,pregnancy losses,plasma concentrations of pregnancy-associated glycoproteins,gestation length or calf birth weight.Calves from RPC fed dams were lighter than control calves at~118 days of age(range 75–150;age included in the statistical model)and at weaning at~248 days of age.There was no effect of treatment on hip height at~118 days of age although there was a trend for RPC calves to be shorter at weaning.Weight/height ratio was lower for RPC than control at both 118 and 248 days of age.Treatment did not affect testis weight at~118 days of age.Conclusions Supplementation of RPC during the periconceptional period programmed development to alter calf phenotype in the postnatal period.The net result,reduced body weight,was the opposite of the phenotype caused by the addition of choline to embryo culture medium.展开更多
BACKGROUND Cluster of differentiation 24(CD24)serves as a liver cancer stem cell marker,and its upregulation is related to chronic liver disease malignancy.However,the exact relationship between CD24 expression and he...BACKGROUND Cluster of differentiation 24(CD24)serves as a liver cancer stem cell marker,and its upregulation is related to chronic liver disease malignancy.However,the exact relationship between CD24 expression and hepatocarcinogenesis remains unknown.AIM To investigate CD24 levels among individuals with chronic liver diseases and confirm the alterations in CD24 and programmed death-ligand 1(PD-L1)expression in a dynamic model of rat hepatocarcinogenesis.METHODS Approved by the ethics committee,CD24 levels were detected in the serum of 129 patients with hepatocellular carcinoma(HCC),72 patients with chronic hepatitis(CH),60 patients with liver cirrhosis(LC)and 111 normal control(NC).Receiver operating characteristic curves and clinicopathological characteristics of CD24 were used to evaluate the diagnostic or prognostic value for HCC,and the CD24+T lymphocyte ratio and dynamic alterations in CD24 and PD-L1 expression were confirmed in a rat HCC model.RESULTS Compared with those in the CH,LC and NC groups,the average CD24 level in the HCC group was significantly greater(P<0.01).The CD24+T-cell ratio in the HCC group was greater than that in the CH or NC group.Clinicopathological characteristics of high CD24 in HCC patients included hepatitis B virus infection,single/multicenter status,tumor size,lymph node or extrahepatic metastasis,differentiation degree,tumor-node-metastasis grade,Child-Pugh score,portal vein tumor thrombus and poor prognosis.Mechanistically,CD24 is dynamically upregulated during hepatocarcinogenesis and closely positively correlated with PD-L1 for immune escape,metastasis(CD44),and with respect to HCC markers(Wnt3a,GPC-3 and alpha-fetoprotein).CONCLUSION Activated CD24 promoted HCC formation through programmed death-ligand 1 signaling and could be a valuable biomarker for monitoring chronic liver disease malignancy.展开更多
Progressive photoreceptor cell death is one of the main pathological features of age-related macular degeneration and eventually leads to vision loss.Ferroptosis has been demonstrated to be associated with retinal deg...Progressive photoreceptor cell death is one of the main pathological features of age-related macular degeneration and eventually leads to vision loss.Ferroptosis has been demonstrated to be associated with retinal degenerative diseases.However,the molecular mechanisms underlying ferroptosis and photoreceptor cell death in age-related macular degeneration remain largely unexplored.Bioinformatics and biochemical analyses in this study revealed xC–,solute carrier family 7 member 11-regulated ferroptosis as the predominant pathological process of photoreceptor cell degeneration in a light-induced dry age-related macular degeneration mouse model.This process involves the nuclear factor-erythroid factor 2-related factor 2-solute carrier family 7 member 11-glutathione peroxidase 4 signaling pathway,through which cystine depletion,iron ion accumulation,and enhanced lipid peroxidation ultimately lead to photoreceptor cell death and subsequent visual function impairment.We demonstrated that solute carrier family 7 member 11 overexpression blocked this process by inhibiting oxidative stress in vitro and in vivo.Conversely,solute carrier family 7 member 11 knockdown or the solute carrier family 7 member 11 inhibitor sulfasalazine and ferroptosis-inducing agent erastin aggravated H2O2-induced ferroptosis of 661W cells.These findings indicate solute carrier family 7 member 11 may be a potential therapeutic target for patients with retinal degenerative diseases including age-related macular degeneration.展开更多

BACKGROUND The prognostic value of programmed death ligand-1(PD-L1)expression in patients with gastric or gastroesophageal junction cancer(G/GEJC)at different stages remains unclear.We hypothesized that high PD-L1 exp...BACKGROUND The prognostic value of programmed death ligand-1(PD-L1)expression in patients with gastric or gastroesophageal junction cancer(G/GEJC)at different stages remains unclear.We hypothesized that high PD-L1 expression is associated with poor survival in patients with pathological stageⅢ(pⅢ)G/GEJC.AIM To investigate the relation between PD-L1 expression and survival outcomes in patients with pathological stageⅡ(pⅡ)-pⅢG/GEJC.METHODS In this retrospective cohort study,we collected the clinicopathological data of 388 consecutive patients with pⅡ/pⅢG/GEJC who underwent gastrectomy without neoadjuvant therapy at Nanfang Hospital,Southern Medical University.Postoperative tissue samples were collected to evaluate PD-L1 expression.Combined positive score(CPS)≥1 was defined as PD-L1-positive,and CPS≥5 as high PD-L1 expression.Survival outcomes were analyzed using Kaplan-Meier and Cox proportional hazards models.RESULTS In pⅡG/GEJC,no significant differences were found in 3-year disease-free survival(DFS;88.5%vs 87.9%,P=0.939)or 3-year overall survival(OS;92.3%vs 89.9%,P=0.621)between PD-L1-positive and PD-L1-negative groups or between high and low PD-L1 expression groups(3-year DFS:91.8%vs 84.2%,P=0.138;3-year OS:94.2%vs 88.4%,P=0.233).In pⅢG/GEJC,PD-L1-positive patients had poorer 3-year DFS(52.2%vs 67.8%,P=0.030)and 3-year OS(65.1%vs 78.2%,P=0.007)than PD-L1-negative patients.High PD-L1 expression was associated with significantly inferior 3-year DFS(50.2%vs 64.8%,P=0.023)and 3-year OS(64.0%vs 75.1%,P=0.036)compared to low PD-L1 expression.Multivariate analysis revealed that high PD-L1 expression was independently associated with shorter DFS(HR=1.624,P=0.027)and OS(HR=1.653,P=0.043)in patients with pⅢG/GEJC.Sensitivity analyses confirmed the robustness of the OS findings.CONCLUSION High PD-L1 expression serves as an independent adverse prognostic biomarker in stageⅢbut not stageⅡG/GEJC.These findings indicate a stage-dependent prognostic value of PD-L1 expression in G/GEJC.展开更多

Objectives:The objective of this study was to examine communication effectiveness(CE)in conjunction with participant satisfaction in a multicultural fitness program.Methods:Adults from a fitness program site with part...Objectives:The objective of this study was to examine communication effectiveness(CE)in conjunction with participant satisfaction in a multicultural fitness program.Methods:Adults from a fitness program site with participant diversity and multicultural backgrounds were selected and asked to complete a three-part Fitness CE Evaluation Scale questionnaire:CE Questionnaire and Customer Satisfaction(CS)Scale plus a short demographic survey.A sample of 95 participants was recruited from a Midwest Functional 45 Fitness Training Program site in the U.S.A.Multivariate Analysis of Variance(MANOVA)tests were conducted for CE and CS variable vectors.Results:The results showed that there were significant main effects on CE and CS for nationality(p=0.001;p<0.001)and language(p=0.006;p<0.001),but no effect for participation frequency,respectively;there were also significant main effects on CS for gender(p=0.006)and membership type(p=0.019).Univariate tests revealed that domestics and/or first language participants scored significantly higher on reactionto strangers(p=0.001;p=0.001),uncertainty(p<0.001;p<0.001),facilities/services(p<0.001;p<0.001),individual/psychological factor(p=0.009;p=0.034),relaxation(p=0.018;p=0.021),social factor(p<0.001;p=0.002),and health/fitness(p<0.001;p=0.001),respectively.Conclusions:Being highly aware of fitness participants’uncertainty and reaction to strangers can help multicultural fitness program managers or trainers improve their communication effectiveness,which will continue improving their customers’satisfaction.展开更多

Depression is a prevalent mental disorder characterized by persistent disinterest and a depressed mood,with severe cases potentially leading to suicide.In recent years,the incidence of depression has steadily increase...Depression is a prevalent mental disorder characterized by persistent disinterest and a depressed mood,with severe cases potentially leading to suicide.In recent years,the incidence of depression has steadily increased,making it the second-largest global health burden.The pathogenesis of depression involves a series of complex pathological mechanisms,although the key underlying causes remain unclear.Programmed cell death(PCD),including apoptosis,autophagy,pyroptosis,ferroptosis,and necroptosis,involves highly organized gene expression processes that may influence the occurrence and development of depression by regulating cellular fate.Furthermore,numerous studies have shown that natural products can modulate PCDs through various signaling pathways,presenting significant potential for managing depression.Natural products offer benefits such as cost-effectiveness,fewer side effects,and other advantages,making them viable supplements or alternatives to traditional antidepressant drugs.To explore this potential,we reviewed studies demonstrating the antidepressant effects of natural products through multi-target modulation of PCDs.In addition,we discussed the toxicity and clinical applications of these natural products.This study highlights that diverse core biological pathways and targets are involved in determining the fate of depression-associated brain cells,including the PI3K/Akt signaling pathway,caspase-8,GSDMD,and others.In conclusion,the multi-target mechanisms of PCD regulation by natural products may provide a promising foundation for the future development of novel antidepressant medications.展开更多
It is important for a lunar lander to possess a large divert capability during the final landing phase,as this can enhance the tolerance for flight deviations in the early phase or improve the obstacle avoidance perfo...It is important for a lunar lander to possess a large divert capability during the final landing phase,as this can enhance the tolerance for flight deviations in the early phase or improve the obstacle avoidance performance.Therefore,when designing the powered descent trajectory,sufficient final phase divert capability should be reserved at the minimum propellant cost.To this end,a multi-phase trajectory programming(MPTP)method for powered descent with approaching phase divert capability is proposed.First,the entire powered descent trajectory is divided into the main braking phase and the approaching phase.The main braking phase is responsible for dissipating the majority of the initial velocity.The approaching phase is responsible for safely and precisely flying toward the landing site.It is nominally a vertical descent trajectory and possesses equal divert capability in all horizontal directions.Then,a constant-thrust linear tangent guidance(LTG)accounting for the lunar curvature is designed for the main braking phase.For the approaching phase,a variable-thrust lossless convex programming(LCP)guidance considering the constraints of tilt angle and glide-slope angle is developed.Subsequently,to connect the two phases and further optimize the propellant consumption throughout the entire trajectory,a method for determining the phase switching condition is proposed.The originally difficult-to-solve two-parameter optimization problem is decomposed into two more easily solvable subproblems,which are solved iteratively via a bilevel optimization framework.Finally,the divert capability of the proposed method is verified through numerical simulation.The programmed trajectory is basically consistent with the results of the pseudospectral method,with the difference in propellant consumption being only 0.006%.This method is suitable for the rapid iterative design of nominal trajectories for lunar lander powered descent in engineering applications.展开更多
This study introduces FTCSEM,a FORTRAN-based,parallelized one-dimensional controlledsource electromagnetic(CSEM)forward modeling and inversion software capable of accommodating arbitrary source-receiver confi guration...This study introduces FTCSEM,a FORTRAN-based,parallelized one-dimensional controlledsource electromagnetic(CSEM)forward modeling and inversion software capable of accommodating arbitrary source-receiver confi gurations.In comparison to existing one-dimensional CSEM tools,FTCSEM incorporates several signifi cant enhancements:it supports transmitters of diverse shapes,quantities,and spatial locations;permits receivers to be positioned flexibly on the surface,subsurface,or in the atmosphere;facilitates simulations and inversions in both frequency and time domains;integrates an adaptive regularized inversion algorithm with multiple model constraints;and leverages GPU-accelerated parallel computing to attain high computational efficiency.Validation through numerical experiments and field data inversion confirms the program’s accuracy and practical applicability.The findings indicate that FTCSEM performs robustly in complex geoelectric environments,multi-source and multi-receiver arrangements,as well as multi-component joint inversion scenarios,thereby offering a versatile and powerful tool for advancing CSEM research and applications.展开更多
BACKGROUND High-frequency irreversible electroporation(H-FIRE),a second-generation tumor ablation therapy,has demonstrated efficacy and safety in treating liver cancer.However,its clinical application is hindered by t...BACKGROUND High-frequency irreversible electroporation(H-FIRE),a second-generation tumor ablation therapy,has demonstrated efficacy and safety in treating liver cancer.However,its clinical application is hindered by the risk of residual disease and metastasis resulting from incomplete ablation,while H-FIRE’s inherent capacity to induce immunogenic cell death(ICD)provides a potential direction for optimizing its efficacy.AIM To leverage H-FIRE’s ICD capacity,and investigate the efficacy and safety of H-FIRE combined with immunotherapy in the treatment of liver cancer.METHODS A murine subcutaneous hepatocellular carcinoma model was established using hepatoma 22 cells.Mice were randomized into eight groups:Control,BMS-1,R848,BMS-1+R848,H-FIRE,H-FIRE+BMS-1,H-FIRE+R848,and H-FIRE+BMS-1+R848.Treatments comprised:Intraperitoneal BMS-1 and intratumoral R848 administered every 2 days(4 total doses);H-FIRE ablation performed once when tumors reached 7-8 mm in long diameter.Efficacy was assessed by monitoring survival,primary tumor volume,primary tumor cell proliferation and damageassociated molecular patterns expression,immune microenvironment alterations in primary tumors and spleens,distant secondary tumor apoptosis,and treatment safety.RESULTS H-FIRE robustly induced ICD in primary liver tumors.Notably,the triple-combination therapy(H-FIRE+BMS-1+R848)exerted the most potent antitumor efficacy:It significantly reduced primary tumor burden relative to all other monotherapy or dual-combination groups,with a mean tumor volume of 305.3±35.0 mm3 in the triplecombination group.This therapeutic benefit was further confirmed by markedly extended overall survival,with median survival times of 17,24.5,25,28.5,28.5,33.5,37.5,and 53 days,respectively.Mechanistically,the triplecombination strategy was accompanied by enhanced tumor cell apoptosis and suppressed cell proliferation,underscoring its multi-faceted antitumor effects.In primary tumors and spleens,this combination markedly increased proportions of CD3+CD4+T cells,CD3+CD8+T cells,CD11c+CD80+CD86+dendritic cells,F4/80+CD11b+CD86+M1 macrophages,and the M1/M2 macrophage ratio.Conversely,it reduced proportions of CD4+CD25+FOXP3+regulatory T cells and F4/80+CD11b+CD206+M2 macrophages,with no significant toxicity observed.Furthermore,the triple therapy effectively promoted apoptosis and inhibited growth in distant secondary tumors.CONCLUSION The triple therapy synergistically reverses local and systemic immune tolerance,eliciting robust anti-tumor immunity.This strategy not only effectively inhibits primary and secondary tumor progression but also exhibits favorable safety,underscoring its potential as a novel combinatorial cancer treatment.展开更多

摘要BACKGROUND Icariin(ICA)is a natural compound derived from Epimedium that has shown promise in treating colorectal cancer(CRC)by reducing inflammation,inducing apoptosis,and limiting tumor progression.AIM To elucidate the specific mechanisms underlying its therapeutic effects on this malignancy.METHODS We examined the impact of different ICA concentrations on the biological behavior of CRC cells in vitro.We also assessed the protein expression of SMAD4,CX-C motif chemokine ligand 1(CXCL1),and CXCL8,as well as epithelial-mesenchymal transition(EMT)-related signaling molecules,and the mRNA expression of proinflammatory factors.To further explore ICA’s mechanisms in CRC treatment,we introduced small interfering SMAD4 and its negative control group.In CRC cells,we evaluated proliferation,apoptosis,invasion,migration,EMT,and the recruitment of tumor-associated neutrophils(TANs).Moreover,we investigated the role of the CXCL8-C-X-C motif chemokine receptor 2(CXCR2)axis in the recruitment of ICA-affected TANs.In vivo,we preliminarily assessed the therapeutic efficacy of ICA against CRC.In ICA-treated mice,we measured tumor size,histological morphology,apoptosis,and neutrophil infiltration.Additionally,we analyzed the expression of SMAD4,CXCL1,CXCL8,CXCR2,EMT,and proteins related to migration.RESULTS Our findings indicate that ICA attenuated CRC cell proliferation,invasion,and migration,while suppressing TAN recruitment and promoting apoptosis in a concentration-dependent manner.When small interfering SMAD4 was introduced,the anticancer effects of ICA were diminished.Furthermore,disruption of the CXCL8-CXCR2 axis reduced neutrophil recruitment.We found that ICA inhibited tumor growth,improved tumor histology and morphology,alleviated CD66b+neutrophil infiltration,and inhibited EMT in mice with subcutaneous CRC tumors.In addition,ICA appeared to enhance SMAD4 protein expression while suppressing CXCL8 and CXCR2 expression in both in vivo and in vitro experiments.CONCLUSION ICA impedes the recruitment of TANs primarily through SMAD4-mediated modulation of the CXCL8-CXCR2 axis,thereby suppressing EMT and other pro-metastatic programs in CRC.

摘要Objective:Multiple programmed cell death(PCD)pathways have been individually reported to be triggered by cisplatin,but whether and how they are co-regulated remains unclear.In this study,we comprehensively investigate the spectrum of cisplatin-induced PCD.Methods:We employed integrated in vitro and in vivo models including human cancer cell lines,a Cal27 xenograft mouse model,and paired clinical specimens from an oral squamous cell carcinoma patient receiving neoadjuvant cisplatin-based chemotherapy.A comprehensive methodological suite-encompassing cell death assays,Western blotting,Hematoxylin and eosin staining,immunofluorescence,Cyclic multiplexed tissue staining,and pathway-specific pharmacological inhibitors was utilized to dissect the activation of apoptosis,necroptosis,pyroptosis,and ferroptosis.Results:Cisplatin simultaneously upregulates markers of PCD pathways(including apoptosis,necroptosis,pyroptosis,and ferroptosis)in a dose-or time-dependent manner.Pharmacological inhibition or genetic knockdown of key genes in each pathway significantly reduced cytotoxicity.confirming their functional roles.Notably,indicators of key pro-inflammatory death modalities,pyroptosis and ferroptosis,were prominently co-upregulated in both xenograft tumors and clinical patient samples,suggesting that these two forms of PCD may represent the predominant death forms in cisplatin-induced tumor cell death.Conclusion:Cisplatin induces the coordinated activation of multiple cell death programs within a unified framework.Prominent engagement of immunogenic cell death pathways,particularly pyroptosis and ferroptosis,provides a mechanistic basis for the clinically observed synergy between cisplatin and immune checkpoint blockade therapy.
摘要Instrumental and expert assessments were used to assess the effect of massage on facial skin blood perfusion,as well as the effects of the long-term use of the two skin health programs on the water content of the stratum corneum,F4,L*,dermal and epidermal thickness and density,facial wrinkles,and so on.Sixty healthy Chinese women were selected as the study subjects,which were subsequently divided into G1 non-massage group and G2 massage group.The results show that massage can increase the blood perfusion of facial skin.Group G2 solution is found to be more effective in improving the skin structural barrier(epidermal density),skin elasticity,firmness(dermal thickness)and forehead wrinkles.

摘要In recent years, artificial intelligence visual technology has achieved rapid development. With the technical advantages of non-contact collection, high-precision identification, and real-time data analysis, AI visual motion capture creates new possibilities for the innovation of physical education in higher vocational colleges and lays a foundation for constructing career-oriented personalized training programs. Teachers can build career-oriented personalized physical training programs relying on this technology, completely breaking the limitations of traditional training and realizing the transformation of physical training from “unified and extensive mode” to “targeted and personalized mode,” and from general physical fitness training to career-specific physical training. It helps physical education in higher vocational colleges truly serve students’ professional competence development. In view of this, this paper first elaborates the significance of constructing personalized physical training programs in higher vocational colleges empowered by AI visual motion capture under career orientation, and then puts forward a series of feasible construction strategies, so as to provide certain references for relevant researchers.

摘要Background Liver cancer is among the top five causes of cancer death in 90 countries,with China accounting for a substantial proportion of the global burden.This study aimed to analyse the national liver cancer policies and programs in China.Methods This study applied a documentary research method using the systematic READ approach.Six national official websites and one public policy database were searched.The document analysis was based on the WHO Health System's Six Building Blocks and the WHO's four modules of cancer control(prevention,early detection,diagnosis and treatment,and palliative care).Results A total of 74 liver cancer-related policies and 8 programs published from 1 January 1984 to 31 March 2025 were included in this study.The analysis revealed that liver cancer management in China mainly focused on Medical products and Technologies(n=46,62.16%),and Service Delivery(n=34,45.95%)within the WHO health system building blocks.When it came to WHO cancer control four modules,most policies(n=39,52.70%)targeted Diagnosis and Treatment of liver cancer,followed by Prevention of liver cancer(n=33,44.59%).Additionally,8 national programs were implemented to improve the prevention,diagnosis,and treatment of liver cancer.Before 2019,the emphasis of liver cancer prevention was primarily on hepatitis prevention and control.However,since 2020,the national-level programs aimed at preventing and controlling liver cancer emphasizing patient education and treatment for high-risk groups.Conclusions China's liver cancer control mainly focused heavily on prevention,diagnosis and treatment modules with special focus on medical products and technology,as well as service delivery.Currently,less attention has been given to the detection and palliative care of survivors.The control of liver cancer in China still requires further strengthening of the health system for implementation.Considering the continual increase in the burden of liver cancer,it is imperative for future efforts to develop a comprehensive national liver cancer strategy.

摘要Severe aortic stenosis has increased exponentially as a result of the aging of the population.Transcatheter aortic valve replacement(TAVR)procedures increase by 20%-30%per year in Spain.Referred patients are increasingly older,have comorbidities,and frailty.TAVR cardiac rehabilitation(CR)programs significantly improve both functional capacity and quality of life of patients.However,there are no guidelines on how to carry out them and few elderly patients are referred to CR.Furthermore,mortality in women who undergo TAVR is higher than in men and even fewer are referred to CR programs.Multidisciplinary patient care,including comorbidities and frailty,is essential.It is necessary to ensure exquisite continuity of care during TAVR,CR,and the rest of the patient's life.Telerehabilitation could be an option in some lower-risk patients to include more patients in CR programs after TAVR,given the high demand.This article reviews the evidence on why CR should be performed in post-TAVR patients and proposes a practical and novel approach to the care process and the recommended aspects and components of the CR program.

摘要Background Supplementation of choline chloride in culture medium programs the preimplantation bovine embryo to increase weaning weight of the resultant calf.Here,it was hypothesized that similar programming actions of choline can be induced by feeding rumen-protected choline(RPC)to beef cows during the periconceptional period.Results A preliminary experiment was conducted to determine changes in circulating concentrations of choline and its metabolites after RPC supplementation.Suckled beef cows were individually fed 0,30,60,and 90 g of RPC(i.e.,0,8.6,17.3 and 25.9 g choline chloride)and blood samples were collected at random times after feeding.There were no differences in plasma concentrations of choline or its metabolites between groups.In the second experiment,effects of feeding 60 g/d RPC from d-1 to 7 relative to timed artificial insemination were examined for suckled beef cows.Feeding RPC did not affect pregnancy or calving rates,pregnancy losses,plasma concentrations of pregnancy-associated glycoproteins,gestation length or calf birth weight.Calves from RPC fed dams were lighter than control calves at~118 days of age(range 75–150;age included in the statistical model)and at weaning at~248 days of age.There was no effect of treatment on hip height at~118 days of age although there was a trend for RPC calves to be shorter at weaning.Weight/height ratio was lower for RPC than control at both 118 and 248 days of age.Treatment did not affect testis weight at~118 days of age.Conclusions Supplementation of RPC during the periconceptional period programmed development to alter calf phenotype in the postnatal period.The net result,reduced body weight,was the opposite of the phenotype caused by the addition of choline to embryo culture medium.

摘要BACKGROUND Cluster of differentiation 24(CD24)serves as a liver cancer stem cell marker,and its upregulation is related to chronic liver disease malignancy.However,the exact relationship between CD24 expression and hepatocarcinogenesis remains unknown.AIM To investigate CD24 levels among individuals with chronic liver diseases and confirm the alterations in CD24 and programmed death-ligand 1(PD-L1)expression in a dynamic model of rat hepatocarcinogenesis.METHODS Approved by the ethics committee,CD24 levels were detected in the serum of 129 patients with hepatocellular carcinoma(HCC),72 patients with chronic hepatitis(CH),60 patients with liver cirrhosis(LC)and 111 normal control(NC).Receiver operating characteristic curves and clinicopathological characteristics of CD24 were used to evaluate the diagnostic or prognostic value for HCC,and the CD24+T lymphocyte ratio and dynamic alterations in CD24 and PD-L1 expression were confirmed in a rat HCC model.RESULTS Compared with those in the CH,LC and NC groups,the average CD24 level in the HCC group was significantly greater(P<0.01).The CD24+T-cell ratio in the HCC group was greater than that in the CH or NC group.Clinicopathological characteristics of high CD24 in HCC patients included hepatitis B virus infection,single/multicenter status,tumor size,lymph node or extrahepatic metastasis,differentiation degree,tumor-node-metastasis grade,Child-Pugh score,portal vein tumor thrombus and poor prognosis.Mechanistically,CD24 is dynamically upregulated during hepatocarcinogenesis and closely positively correlated with PD-L1 for immune escape,metastasis(CD44),and with respect to HCC markers(Wnt3a,GPC-3 and alpha-fetoprotein).CONCLUSION Activated CD24 promoted HCC formation through programmed death-ligand 1 signaling and could be a valuable biomarker for monitoring chronic liver disease malignancy.

摘要Progressive photoreceptor cell death is one of the main pathological features of age-related macular degeneration and eventually leads to vision loss.Ferroptosis has been demonstrated to be associated with retinal degenerative diseases.However,the molecular mechanisms underlying ferroptosis and photoreceptor cell death in age-related macular degeneration remain largely unexplored.Bioinformatics and biochemical analyses in this study revealed xC–,solute carrier family 7 member 11-regulated ferroptosis as the predominant pathological process of photoreceptor cell degeneration in a light-induced dry age-related macular degeneration mouse model.This process involves the nuclear factor-erythroid factor 2-related factor 2-solute carrier family 7 member 11-glutathione peroxidase 4 signaling pathway,through which cystine depletion,iron ion accumulation,and enhanced lipid peroxidation ultimately lead to photoreceptor cell death and subsequent visual function impairment.We demonstrated that solute carrier family 7 member 11 overexpression blocked this process by inhibiting oxidative stress in vitro and in vivo.Conversely,solute carrier family 7 member 11 knockdown or the solute carrier family 7 member 11 inhibitor sulfasalazine and ferroptosis-inducing agent erastin aggravated H2O2-induced ferroptosis of 661W cells.These findings indicate solute carrier family 7 member 11 may be a potential therapeutic target for patients with retinal degenerative diseases including age-related macular degeneration.

摘要During the use of robotics in applications such as antiterrorism or combat,a motion-constrained pursuer vehicle,such as a Dubins unmanned surface vehicle(USV),must get close enough(within a prescribed zero or positive distance)to a moving target as quickly as possible,resulting in the extended minimum-time intercept problem(EMTIP).Existing research has primarily focused on the zero-distance intercept problem,MTIP,establishing the necessary or sufficient conditions for MTIP optimality,and utilizing analytic algorithms,such as root-finding algorithms,to calculate the optimal solutions.However,these approaches depend heavily on the properties of the analytic algorithm,making them inapplicable when problem settings change,such as in the case of a positive effective range or complicated target motions outside uniform rectilinear motion.In this study,an approach employing a high-accuracy and quality-guaranteed mixed-integer piecewise-linear program(QG-PWL)is proposed for the EMTIP.This program can accommodate different effective interception ranges and complicated target motions(variable velocity or complicated trajectories).The high accuracy and quality guarantees of QG-PWL originate from elegant strategies such as piecewise linearization and other developed operation strategies.The approximate error in the intercept path length is proved to be bounded to h2/(4√2),where h is the piecewise length.

摘要BACKGROUND The prognostic value of programmed death ligand-1(PD-L1)expression in patients with gastric or gastroesophageal junction cancer(G/GEJC)at different stages remains unclear.We hypothesized that high PD-L1 expression is associated with poor survival in patients with pathological stageⅢ(pⅢ)G/GEJC.AIM To investigate the relation between PD-L1 expression and survival outcomes in patients with pathological stageⅡ(pⅡ)-pⅢG/GEJC.METHODS In this retrospective cohort study,we collected the clinicopathological data of 388 consecutive patients with pⅡ/pⅢG/GEJC who underwent gastrectomy without neoadjuvant therapy at Nanfang Hospital,Southern Medical University.Postoperative tissue samples were collected to evaluate PD-L1 expression.Combined positive score(CPS)≥1 was defined as PD-L1-positive,and CPS≥5 as high PD-L1 expression.Survival outcomes were analyzed using Kaplan-Meier and Cox proportional hazards models.RESULTS In pⅡG/GEJC,no significant differences were found in 3-year disease-free survival(DFS;88.5%vs 87.9%,P=0.939)or 3-year overall survival(OS;92.3%vs 89.9%,P=0.621)between PD-L1-positive and PD-L1-negative groups or between high and low PD-L1 expression groups(3-year DFS:91.8%vs 84.2%,P=0.138;3-year OS:94.2%vs 88.4%,P=0.233).In pⅢG/GEJC,PD-L1-positive patients had poorer 3-year DFS(52.2%vs 67.8%,P=0.030)and 3-year OS(65.1%vs 78.2%,P=0.007)than PD-L1-negative patients.High PD-L1 expression was associated with significantly inferior 3-year DFS(50.2%vs 64.8%,P=0.023)and 3-year OS(64.0%vs 75.1%,P=0.036)compared to low PD-L1 expression.Multivariate analysis revealed that high PD-L1 expression was independently associated with shorter DFS(HR=1.624,P=0.027)and OS(HR=1.653,P=0.043)in patients with pⅢG/GEJC.Sensitivity analyses confirmed the robustness of the OS findings.CONCLUSION High PD-L1 expression serves as an independent adverse prognostic biomarker in stageⅢbut not stageⅡG/GEJC.These findings indicate a stage-dependent prognostic value of PD-L1 expression in G/GEJC.

摘要Objectives:The objective of this study was to examine communication effectiveness(CE)in conjunction with participant satisfaction in a multicultural fitness program.Methods:Adults from a fitness program site with participant diversity and multicultural backgrounds were selected and asked to complete a three-part Fitness CE Evaluation Scale questionnaire:CE Questionnaire and Customer Satisfaction(CS)Scale plus a short demographic survey.A sample of 95 participants was recruited from a Midwest Functional 45 Fitness Training Program site in the U.S.A.Multivariate Analysis of Variance(MANOVA)tests were conducted for CE and CS variable vectors.Results:The results showed that there were significant main effects on CE and CS for nationality(p=0.001;p<0.001)and language(p=0.006;p<0.001),but no effect for participation frequency,respectively;there were also significant main effects on CS for gender(p=0.006)and membership type(p=0.019).Univariate tests revealed that domestics and/or first language participants scored significantly higher on reactionto strangers(p=0.001;p=0.001),uncertainty(p<0.001;p<0.001),facilities/services(p<0.001;p<0.001),individual/psychological factor(p=0.009;p=0.034),relaxation(p=0.018;p=0.021),social factor(p<0.001;p=0.002),and health/fitness(p<0.001;p=0.001),respectively.Conclusions:Being highly aware of fitness participants’uncertainty and reaction to strangers can help multicultural fitness program managers or trainers improve their communication effectiveness,which will continue improving their customers’satisfaction.

摘要Mathematical programming solvers are software tools designed to solve real‑world problems using mathematical programming algorithms.This survey explores the evolution of optimization technologies,from traditional methods such as the simplex algorithm and branch‑and‑bound techniques to modern advancements that are facilitated by parallel computing,GPU acceleration,and AI algorithms.We also emphasize the recent emergence of mathematical programming solvers developed by research institutes and companies headquartered in China as major players,who have achieved remarkable success in benchmarks when compared to established solvers.This article provides a comprehensive overview of the theoretical foundations,historical progress,and emerging trends in mathematical programming solvers,offering valuable insights for both researchers and practitioners in the field.

摘要Depression is a prevalent mental disorder characterized by persistent disinterest and a depressed mood,with severe cases potentially leading to suicide.In recent years,the incidence of depression has steadily increased,making it the second-largest global health burden.The pathogenesis of depression involves a series of complex pathological mechanisms,although the key underlying causes remain unclear.Programmed cell death(PCD),including apoptosis,autophagy,pyroptosis,ferroptosis,and necroptosis,involves highly organized gene expression processes that may influence the occurrence and development of depression by regulating cellular fate.Furthermore,numerous studies have shown that natural products can modulate PCDs through various signaling pathways,presenting significant potential for managing depression.Natural products offer benefits such as cost-effectiveness,fewer side effects,and other advantages,making them viable supplements or alternatives to traditional antidepressant drugs.To explore this potential,we reviewed studies demonstrating the antidepressant effects of natural products through multi-target modulation of PCDs.In addition,we discussed the toxicity and clinical applications of these natural products.This study highlights that diverse core biological pathways and targets are involved in determining the fate of depression-associated brain cells,including the PI3K/Akt signaling pathway,caspase-8,GSDMD,and others.In conclusion,the multi-target mechanisms of PCD regulation by natural products may provide a promising foundation for the future development of novel antidepressant medications.

摘要It is important for a lunar lander to possess a large divert capability during the final landing phase,as this can enhance the tolerance for flight deviations in the early phase or improve the obstacle avoidance performance.Therefore,when designing the powered descent trajectory,sufficient final phase divert capability should be reserved at the minimum propellant cost.To this end,a multi-phase trajectory programming(MPTP)method for powered descent with approaching phase divert capability is proposed.First,the entire powered descent trajectory is divided into the main braking phase and the approaching phase.The main braking phase is responsible for dissipating the majority of the initial velocity.The approaching phase is responsible for safely and precisely flying toward the landing site.It is nominally a vertical descent trajectory and possesses equal divert capability in all horizontal directions.Then,a constant-thrust linear tangent guidance(LTG)accounting for the lunar curvature is designed for the main braking phase.For the approaching phase,a variable-thrust lossless convex programming(LCP)guidance considering the constraints of tilt angle and glide-slope angle is developed.Subsequently,to connect the two phases and further optimize the propellant consumption throughout the entire trajectory,a method for determining the phase switching condition is proposed.The originally difficult-to-solve two-parameter optimization problem is decomposed into two more easily solvable subproblems,which are solved iteratively via a bilevel optimization framework.Finally,the divert capability of the proposed method is verified through numerical simulation.The programmed trajectory is basically consistent with the results of the pseudospectral method,with the difference in propellant consumption being only 0.006%.This method is suitable for the rapid iterative design of nominal trajectories for lunar lander powered descent in engineering applications.

摘要This study introduces FTCSEM,a FORTRAN-based,parallelized one-dimensional controlledsource electromagnetic(CSEM)forward modeling and inversion software capable of accommodating arbitrary source-receiver confi gurations.In comparison to existing one-dimensional CSEM tools,FTCSEM incorporates several signifi cant enhancements:it supports transmitters of diverse shapes,quantities,and spatial locations;permits receivers to be positioned flexibly on the surface,subsurface,or in the atmosphere;facilitates simulations and inversions in both frequency and time domains;integrates an adaptive regularized inversion algorithm with multiple model constraints;and leverages GPU-accelerated parallel computing to attain high computational efficiency.Validation through numerical experiments and field data inversion confirms the program’s accuracy and practical applicability.The findings indicate that FTCSEM performs robustly in complex geoelectric environments,multi-source and multi-receiver arrangements,as well as multi-component joint inversion scenarios,thereby offering a versatile and powerful tool for advancing CSEM research and applications.
摘要BACKGROUND High-frequency irreversible electroporation(H-FIRE),a second-generation tumor ablation therapy,has demonstrated efficacy and safety in treating liver cancer.However,its clinical application is hindered by the risk of residual disease and metastasis resulting from incomplete ablation,while H-FIRE’s inherent capacity to induce immunogenic cell death(ICD)provides a potential direction for optimizing its efficacy.AIM To leverage H-FIRE’s ICD capacity,and investigate the efficacy and safety of H-FIRE combined with immunotherapy in the treatment of liver cancer.METHODS A murine subcutaneous hepatocellular carcinoma model was established using hepatoma 22 cells.Mice were randomized into eight groups:Control,BMS-1,R848,BMS-1+R848,H-FIRE,H-FIRE+BMS-1,H-FIRE+R848,and H-FIRE+BMS-1+R848.Treatments comprised:Intraperitoneal BMS-1 and intratumoral R848 administered every 2 days(4 total doses);H-FIRE ablation performed once when tumors reached 7-8 mm in long diameter.Efficacy was assessed by monitoring survival,primary tumor volume,primary tumor cell proliferation and damageassociated molecular patterns expression,immune microenvironment alterations in primary tumors and spleens,distant secondary tumor apoptosis,and treatment safety.RESULTS H-FIRE robustly induced ICD in primary liver tumors.Notably,the triple-combination therapy(H-FIRE+BMS-1+R848)exerted the most potent antitumor efficacy:It significantly reduced primary tumor burden relative to all other monotherapy or dual-combination groups,with a mean tumor volume of 305.3±35.0 mm3 in the triplecombination group.This therapeutic benefit was further confirmed by markedly extended overall survival,with median survival times of 17,24.5,25,28.5,28.5,33.5,37.5,and 53 days,respectively.Mechanistically,the triplecombination strategy was accompanied by enhanced tumor cell apoptosis and suppressed cell proliferation,underscoring its multi-faceted antitumor effects.In primary tumors and spleens,this combination markedly increased proportions of CD3+CD4+T cells,CD3+CD8+T cells,CD11c+CD80+CD86+dendritic cells,F4/80+CD11b+CD86+M1 macrophages,and the M1/M2 macrophage ratio.Conversely,it reduced proportions of CD4+CD25+FOXP3+regulatory T cells and F4/80+CD11b+CD206+M2 macrophages,with no significant toxicity observed.Furthermore,the triple therapy effectively promoted apoptosis and inhibited growth in distant secondary tumors.CONCLUSION The triple therapy synergistically reverses local and systemic immune tolerance,eliciting robust anti-tumor immunity.This strategy not only effectively inhibits primary and secondary tumor progression but also exhibits favorable safety,underscoring its potential as a novel combinatorial cancer treatment.

摘要To address challenges in architectural extensibility and cross-module collaboration of CAE software,this study proposes OPFEM(open-source Python-based finite element modeling)—an open-source framework featuring a unified four-layer architecture.The geometric modeling framework achieves plug-in support for geometric kernels through an interface abstraction layer and adapter patterns,decoupling kernel-specific implementations while enabling state machine-driven interaction design and parametric sketching.The pre-processing modules establish multi-level associations among materials,sections,and geometric entities using Composite and Factory patterns,while implementing Observer pattern to ensure geometric-mesh consistency and employing finite-state machines to optimize boundary workflows.The computational modules implement a modular finite element library that decouples topology from element attributes,along with a surface boundary element technique for load conversion and task-scheduling management,validated through a cantilever beam large-deformation case.The post-processing module facilitates standardized data storage and dynamic field visualization through architecture-level standardized interface definitions and hierarchical component design.Collectively,OPFEM achieves full-process integration from parametric modeling to nonlinear solving and visualization,enhancing configuration efficiency and providing an extensible,pattern-driven solution for complex CAE challenges.
