Background:Auriculotherapy is a simple,convenient,nonpharmacological option that has shown positive effects for insomnia patients,but its efficacy and safety remain unknown.Objective:The aim of this study is to evalua...Background:Auriculotherapy is a simple,convenient,nonpharmacological option that has shown positive effects for insomnia patients,but its efficacy and safety remain unknown.Objective:The aim of this study is to evaluate the efficacy and safety of auricular therapy for insomnia.Design,setting,participants and intervention:An 18-week(2-week screening,4-week treatment,and 12-week follow-up),recruiter-and assessor-blinded,randomized,sham-controlled trial was performed.Patients were recruited from outpatient settings at three clinical centers in China from 2 February 2023 to 26 January 2024.Participants had insomnia for more than 3 months and met the criteria classified in the Diagnostic and Statistical Manual of Mental Disorders(Fifth Edition).Patients were randomly assigned in a 1:1 ratio to receive auricular therapy(AT)or sham AT(SAT)for 4 consecutive weeks,twice a week,and once every 3 days.Main outcome measures:Primary outcome was the response rate at week 4 after treatment,defined as the percentage of patients with at least a 50%reduction in Pittsburgh Sleep Quality Index(PSQI)from baseline.Secondary outcomes consisted of response rates at other time points,PSQI and 4 related scales,actigraphy-recorded sleep efficiency,sleep arousal frequency,and total sleep time.Results:A total of 234 participants were screened,and 156 patients(122 female[78.2%];mean[standard deviation]age,37.2[13.5]years)were included in the intention-to-treat analysis,with 78 participants randomized to each group.A total of 117 participants(75%)were followed up for 3 months.The AT group had a higher response rate at week 4 than the SAT group(39.73%[29/73]vs 23.29%[17/73],P=0.03),and the response rates for the AT and SAT groups at week 2 were 28.38%(21/74)and 13.33%(10/75),respectively(P=0.02).The differences between the two groups in the PSQI at each time point(P<0.05)and the Insomnia Severity Index at weeks 4 and 12(both P=0.03)were also statistically significant.The other partial outcomes also showed statistically significant differences.Adverse events occurred in 2 cases in the AT group(2.60%).Conclusion:Four-week AT treatment was an effective and safe alternative therapy for insomnia,although the difference with the SAT treatment did not reach the minimal clinically important difference.Adverse events were mild or transient.展开更多
Recent advances in geoscience have underscored the critical role of abiogenic processes in petroleum formation,especially the formation and polymerization of methane.However,whether a direct carbon-H2 reaction can ...Recent advances in geoscience have underscored the critical role of abiogenic processes in petroleum formation,especially the formation and polymerization of methane.However,whether a direct carbon-H2 reaction can produce C2+hydrocarbons(e.g.,ethane and propane)beyond methane remains an open question.Here,we demonstrate the direct synthesis of ethane and propane via reactions between amorphous carbon and H2 under upper mantle conditions(2-10 GPa and 800-1200℃).A systematic investigation reveals that increasing structural disorder in carbon precursors,from graphite to glassy carbon-Ⅱ and carbon black,enhances the production of C2-C3 hydrocarbons.Through integrated X-ray diffraction and reverse Monte Carlo simulations,we establish that the continuous random atomic network structures in amorphous carbon enable one-step synthesis of heavy hydrocarbons with H2.These models establish a direct link between atomic-scale carbon structures and the one-step synthesis of C2+ hydrocarbons under H2-rich,high-pressure,and high-temperature conditions—potentially revealing an efficient mechanism for the abiotic production of C2+ hydrocarbons in the upper mantle.展开更多
Driven by artificial intelligence,the exponential growth in computational and energy demands has spurred exploration of alternative computing paradigms beyond traditional electronic processors.Optical neural networks(...Driven by artificial intelligence,the exponential growth in computational and energy demands has spurred exploration of alternative computing paradigms beyond traditional electronic processors.Optical neural networks(ONNs),a promising neuromorphic platform with light-speed computing,offer inherent advantages for accelerating artificial intelligence,including massive parallelism,ultralow latency,and reduced power consumption.However,most existing ONNs lack physical programmability after deployment due to rigid optical interconnects and limited optical transformations,which restrict their performance across diverse machine learning tasks.We propose a programmable and scalable ONN,an optoelectronic reservoir computing architecture integrated with cascaded linear disordered media.The key novelty lies in its physical kernel optimization via genetic algorithms,which involves programming diffuser orientations within a reduced search space and integrating wavefront shaping for feature preprocessing,thereby enhancing both optical random projection and preprocessing transformations.This platform achieves competitive accuracy in image classification with 93.5%fewer parameters than digital models;it also performs well in graph classification and human action recognition,proving feasibility for non-Euclidean and time-series datasets.Our work addresses the critical programmability bottleneck of ONNs,paving the way for scalable,energyefficient ONNs with programmable physical kernels and opening new paths for high-performance physicsinspired neuromorphic computing in resource-constrained scenarios.展开更多
Objective Previous studies link lower body mass index(BMI)with increased obsessive-compulsive disorder(OCD)risk,yet other body mass indicators may be more etioloically relevant.We dissected the causal association betw...Objective Previous studies link lower body mass index(BMI)with increased obsessive-compulsive disorder(OCD)risk,yet other body mass indicators may be more etioloically relevant.We dissected the causal association between body fat mass(FM)and OCD.Methods Summary statistics from genome-wide association studies of European ancestry were utilized to conduct two-sample Mendelian randomization analysis.Heterogeneity,horizontal pleiotropy,and sensitivity analyses were performed to assess the robustness.Results The inverse variance weighting method demonstrated that a genetically predicted decrease in FM was causally associated with an increased OCD risk[odds ratio(OR)=0.680,95%confidence interval(CI):0.528–0.875,P=0.003].Similar estimates were obtained using the weighted median approach(OR=0.633,95%CI:0.438–0.915,P=0.015).Each standard deviation increases in genetically predicted body fat percentage corresponded to a reduced OCD risk(OR=0.638,95%CI:0.455–0.896,P=0.009).The sensitivity analysis confirmed the robustness of these findings with no outlier instrument variables identified.Conclusion The negative causal association between FM and the risk of OCD suggests that the prevention or treatment of mental disorders should include not only the control of BMI but also fat distribution and body composition.展开更多
Objective Emerging evidence implicates neuroinflammation in the pathogenesis of major depressive disorder(MDD),yet the role of memory B cells remains unclear.In this study,we conducted a bidirectional two-sample Mende...Objective Emerging evidence implicates neuroinflammation in the pathogenesis of major depressive disorder(MDD),yet the role of memory B cells remains unclear.In this study,we conducted a bidirectional two-sample Mendelian randomiza-tion(MR)study and Bayesian colocalization analyses to investigate the causal relationships between memory B-cell traits and MDD risk.Methods MDD summary data were gathered from a meta-analysis of genome-wide association studies(GWASs),whereas memory B-cell genetic variations were sourced from GWASs on immune phenotypes.MR analysis utilized the inverse variance weighted(IVW),MR-Egger,and weighted median methods.Moreover,various sensitivity analyses,including Cochran's Q test,MR Pleiotropy Residual Sum and Outlier(MR-PRESSO),MR-Egger intercept test and Leave-one-out(LOO)analysis,were performed to confirm MR result stability.Bayesian colocalization analyses were also conducted to identify genetic loci shared between memory B cells and MDD.Results Our results indicated that genetically predicted increased CD27 protein expression on memory B cells causally elevated MDD risk(ORs:1.025-1.063,PFDR<0.05).Conversely,MDD did not causally affect memory B-cell traits.Addi-tionally,the colocalization analysis revealed no shared genetic variants,suggesting distinct biological pathways.Conclusions These findings highlight CD27 as a potential novel biomarker and therapeutic target in MDD,warranting fur-ther clinical validation in the future.展开更多
Traditional carbon/carbon composites,such as carbon fiber reinforced carbon matrix composites,are a promising carbon material widely applied in the aerospace field due to their unique mechanical,thermal,and electrical...Traditional carbon/carbon composites,such as carbon fiber reinforced carbon matrix composites,are a promising carbon material widely applied in the aerospace field due to their unique mechanical,thermal,and electrical properties.However,it becomes almost impossible to further enhance their mechanical properties because of the weak van der Waals bonding within or between components.Here,by precisely controlling the graphitization transformation of nanodiamonds,we have successfully synthesized in-situ disordered graphitic carbonanodiamond composites under an industrially achievable pressure of 2 GPa and a moderate temperature of 1200-1400℃.In the composites,nanodiamonds are dispersed in disordered graphitic carbon fragments,featured by a larger spacing compared to that of ideal graphite.Moreover,the nanodiamond and graphitic carbon components are irregularly connected at the interface through random sp2or sp3bonding.Compared with the traditional carbon/carbon composites,the synthesized composites demonstrate exceptional properties:a hardness of up to 17.5 GPa,a compressive strength reaching up to 1.8 GPa,a flexural strength of up to 380 MPa,and an electrical conductivity of around 2290 S/m.This research makes it possible to fabricate high-performance carbon/carbon compos-ites coupled high strength/hardness and electrical conductivity at an industrial achievable pressure.展开更多

Pseudocapacitive materials employ rapid,non-diffusion-limited faradaic processes to store charge,demonstrating significant potential for fast-charging batteries and supercapacitors.However,the high redox potentials of...Pseudocapacitive materials employ rapid,non-diffusion-limited faradaic processes to store charge,demonstrating significant potential for fast-charging batteries and supercapacitors.However,the high redox potentials of existing pseudocapacitive anodes substantially lower the overall cell voltage and energy density.Here,a cation-disordered rock-salt lithium titanium oxyfluoride(DRX-LixTiOF2,0<x<2)is reported to reversibly accommodate approximately 1.19 mol of Li+(~310 mAh g−1)and delivers high-rate performance(exceeding 64.4 C)via pseudocapacitive Li+storage within a low potential window extending down to 0.1 V vs.Li+/Li.This pseudocapacitive behavior is characterized by several structural and electrochemical features:the absence of phase transformation during Li+intercalation,quasi-rectangular cyclic voltammetry curves,sloping charge/discharge profiles and a surface-controlled current response.We further reveal that the pseudocapacitive characteristics originate from a three-dimensional percolation network that facilitates fast Li+migration with low energy barriers,enabled by a cation/anion-disordered structure arising from the mixed occupancy of Li/Ti cations and O/F anions.Owing to its low operating potential and high-rate capability,DRX-LixTiOF2 allows a lithium-ion capacitor to attain 4.0 V cell voltage and achieve energy and power densities several times higher than those obtained with conventional anodes,such as battery-type Li4Ti5O12 or pseudocapacitive materials like Nb2O5 and TiO2.展开更多

Intrinsically disordered proteins(IDPs)are a class of proteins lacking stable three-dimensional structures and play crucial roles in various kinds of cellular processes.Given that more than half of human proteins are ...Intrinsically disordered proteins(IDPs)are a class of proteins lacking stable three-dimensional structures and play crucial roles in various kinds of cellular processes.Given that more than half of human proteins are at least partially disordered,systematically classifying IDPs helps to better understand the regularities in their functions and mechanisms.However,there is currently no such a systematic classification system for IDPs.In this study,we classified IDPs according to sequence and disorder conservation and explored the distinct properties and functional significance of different types of IDPs.Seven species representing evolutionary nodes of human lineage within the Euarchontoglires were selected,and all human proteins were classified into seven categories based on the homologous gene distribution range and the conservation of sequence and disorder degree.Among them,IDPs with homologous genes in all seven species were classified into four categories based on the conservation of sequence and disorder degree:proteins with both sequence and disorder conservation;proteins with disorder conservation but no sequence conservation;proteins lacking both sequence and disorder conservation;and proteins with sequence conservation but lacking disorder conservation.This study systematically investigates the characteristics of these types of IDPs with respect to physicochemical properties,evolutionary characteristics,functional role,expression pattern.The IDPs with both sequence and disorder conservation(Ho_ConS_ConD)represent the canonical IDP phenotype;those with conserved sequence but non-conserved disorder(Ho_ConS_NoConD)form a seemingly paradoxical yet functionally important class;IDPs lacking conservation in both features(Ho_NoConS_NoConD)exhibit pronounced tissue specificity,particularly in the testis.Overall,we classified IDPs from a novel perspective,constructing a systematic framework to elucidate the functions of different IDPs.展开更多
[Objectives]To investigate the potential causal relationship between psoriasis and common mental disorders,and to provide genetic epidemiological evidence for the early intervention of mental comorbidities.[Methods]Ba...[Objectives]To investigate the potential causal relationship between psoriasis and common mental disorders,and to provide genetic epidemiological evidence for the early intervention of mental comorbidities.[Methods]Based on publicly available large-scale GWAS data,a bidirectional Mendelian randomization(MR)approach was employed to assess the causal associations between psoriasis and major depressive disorder(MDD),bipolar disorder,schizophrenia,and anxiety disorders.The inverse variance weighted(IVW)method served as the primary analytical tool,supplemented by sensitivity analyses using MR-Egger and weighted median methods.Additionally,a subgroup analysis was conducted for psoriatic arthritis(PsA).[Results]Forward MR analysis revealed a significant positive causal association between the genetic predisposition to psoriasis and bipolar disorder as well as MDD,whereas no significant causal relationship was observed with schizophrenia or anxiety disorders.The reverse MR analysis found no causal effect of mental disorders on psoriasis.Subgroup analysis of PsA indicated that its genetic predisposition was significantly associated with the risk of bipolar disorder.The results of various sensitivity analyses and pleiotropy tests supported the robustness of the conclusions.[Conclusions]This study provides genetic evidence supporting a causal link between psoriasis and both MDD and bipolar disorder.In particular,patients with PsA are at a higher risk of developing bipolar disorder,highlighting the need to strengthen early screening and intervention for mental health in clinical management.展开更多
BACKGROUND Anxiety disorders are highly prevalent in patients with bipolar disorder(BD)and are associated with a more severe illness course and poorer outcomes.A significant clinical challenge is the frequent initial ...BACKGROUND Anxiety disorders are highly prevalent in patients with bipolar disorder(BD)and are associated with a more severe illness course and poorer outcomes.A significant clinical challenge is the frequent initial misdiagnosis of BD as anxiety,leading to prolonged diagnostic delays and suboptimal treatment.Growing evidence suggests that early intervention in individuals at high risk for BD can improve prognosis.Established clinical high-risk factors include early onset,family history of BD,and subthreshold manic symptoms.This creates a clinical dilemma whereby the administration of first-line antidepressants(e.g.,sertraline)for anxiety is debated in patients with a bipolar diathesis,given the associated risk of mood destabilization.Conversely,mood stabilizers like lithium are foundational in BD treatment,but their role in treating anxiety in high-risk populations is unproven.Therefore,we conducted this randomized controlled trial to evaluate whether early intervention with a combination of sertraline and lithium is more effective than sertraline monotherapy for anxiety disorder patients with clinical high-risk factors for BD.AIM To investigate whether early intervention has a more positive outcome for anxiety disorders in patients who present with clinical high risk factors for BD.METHODS A total of 66 patients were enrolled in this study from January 2021 and December 2022 in Huzhou Third Municipal Hospital.They were randomly assigned to two groups to receive either an antidepressant(sertraline,n=32)or a combination therapy(sertraline and lithium,n=34).The main variables included alterations in Hamilton Anxiety Rating Scale and Hamilton Depression Rating Scale scores from the initial assessment to the final evaluation.A thorough combined Wald test was conducted to examine the intention-to-treat differences in scale assessment across treatment and time conditions.RESULTS Significant differences in the change of Hamilton Anxiety Rating Scale scores were observed between the two groups at week 1,week 2,and week 4(P<0.05).However,after 8 weeks and 12 weeks of treatment,there were no significant different(P=0.485 and P=0.206).There was no significant difference in the change over time in Hamilton Depression Rating Scale scores between the treatment groups(P=0.2),except at week 12(P=0.034).No significant differences were observed in the adverse effects reported between patients treated with sertraline alone(18%)and those treated with the combination therapy(21%).CONCLUSION This current double-blind,case-controlled study assessed the effectiveness and tolerability of combined therapy vs monotherapy for anxiety disorder in patients with clinical high-risk factors for BD.In light of the constraints associated with this initial study,the results imply that the combination of sertraline and lithium may provide a more favorable prognosis.展开更多
BACKGROUND Disorders of consciousness are critical emergencies requiring timely intervention to improve outcomes.Process reengineering using checklists can enhance efficiency in clinical settings.AIM To evaluate wheth...BACKGROUND Disorders of consciousness are critical emergencies requiring timely intervention to improve outcomes.Process reengineering using checklists can enhance efficiency in clinical settings.AIM To evaluate whether checklist-based process reengineering in the Emergency Department reduces emergency stay duration and hospitalization time,and improves rescue success rate,prognosis,and satisfaction among patients with disorders of consciousness.METHODS From January 2021 to December 2023,our hospital admitted 400 emergency consciousness disorders patients using purposive sampling.The first 200,undergoing rescue without the checklist-based process,formed the control group.The next 200,post-implementation,constituted the observation group.We collected data on emergency stay duration,rescue success rate,hospitalization duration,and patient satisfaction for analysis.Patient prognosis in both groups was compared.RESULTS The observation group showed significantly shorter emergency stay duration,time to recovery,and overall hospitalization duration(intensive care unit,general ward,total)compared to the control group(P<0.05).The observation group had a higher rescue success rate(P<0.05)and better recovery rate(P<0.05)than the control group.Both medical staff and patients’family members reported higher satisfaction with the emergency rescue model after checklist-based process reengineering compared to the conventional emergency rescue model(P<0.05).CONCLUSION Emergency process reengineering based on checklists can effectively reduce the emergency stay duration for patients with consciousness disorders,improve rescue success rates,enhance patient prognosis,and increase overall medical and patient satisfaction.展开更多
Objective:To investigate the clinical efficacy,improvement in social functioning,prevention of relapse,and health economic value of a comprehensive community-based rehabilitation service model in the holistic manageme...Objective:To investigate the clinical efficacy,improvement in social functioning,prevention of relapse,and health economic value of a comprehensive community-based rehabilitation service model in the holistic management of patients with severe mental disorders,providing empirical evidence for the standardized promotion of community rehabilitation.Methods:A parallel-group randomized controlled trial(RCT)was conducted,involving 200 patients with severe mental disorders in a stable phase.They were randomly assigned at a 1:1 ratio to an intervention group(comprehensive community rehabilitation,n=100)and a control group(routine community management,n=100),with continuous intervention for 12 months.The Positive and Negative Syndrome Scale(PANSS)and the Personal and Social Performance Scale(PSP)were used as primary outcome measures to compare differences in symptom improvement,social functioning,relapse rate,medication adherence,hospitalization status,and medical expenses between the two groups.Results:After 12 months of intervention,the PANSS total score in the intervention group(43.0±6.8)was significantly lower than that in the control group(47.9±7.5),while the PSP score(66.0±8.0)was significantly higher than that in the control group(60.8±7.3),with statistically significant differences(p<0.001).The relapse rate in the intervention group(10.0%)was significantly lower than that in the control group(22.0%),while the medication adherence rate(83.0%)was significantly higher than that in the control group(68.0%).The hospitalization rate,number of hospitalization days,and annual total medical expenses were all significantly lower in the intervention group than in the control group(p<0.05).Conclusion:The comprehensive community-based rehabilitation service model can effectively improve psychiatric symptoms and social functioning in patients with severe mental disorders,reduce the risk of relapse and hospitalization,and save medical costs.It represents an efficient and promotable core intervention strategy for the holistic management of severe mental disorders.展开更多
Catalytic doping is one of the economic and efficient strategies to optimize the operating temperature and kinetic behavior of magnesium hydride(MgH2).Herein,efficient regulation of electronic and structural rearra...Catalytic doping is one of the economic and efficient strategies to optimize the operating temperature and kinetic behavior of magnesium hydride(MgH2).Herein,efficient regulation of electronic and structural rearrangements in niobium-rich nickel oxides was achieved through precise compositional design and niobium cation functionalized doping,thereby greatly enhancing its intrinsic catalytic activity in hydrogen storage systems.As the niobium concentration increased,the Ni-Nb catalysts transformed into a mixed state of multi-phase nanoparticles(composed of nickel and niobium-rich nickel oxides)with smaller particle size and uniform distribution,thus exposing more nucleation sites and diffusion channels at the MgH2/Mg interface.In addition,the additional generation of active Ni-Nb-O mixed phase induced numerous highly topical disordered and distorted crystalline,promoting the transfer and reorganization of H atoms.As a result,a stable and continuous multi-phase/component synergistic catalytic microenvironment could be constructed,exerting remarkable enhancement on MgH2’s hydrogen storage performance.After comparative tests,Ni0.7Nb0.3-doped MgH2 presented the optimal low-temperature kinetics with a dehydrogenation activation energy of 78.8 kJ·mol−1.The onset dehydrogenation temperature of MgH2+10 wt%Ni0.7Nb0.3 was reduced to 198℃ and 6.18 wt%H2 could be released at 250℃ within 10 min.In addition,the dehydrogenated MgH2–NiNb composites absorbed 4.87 wt%H2 in 10 min at 125℃ and a capacity retention rate was maintained at 6.18 wt%even after 50 reaction cycles.In a word,our work supplies fresh insights for designing novel defective-state multiphase catalysts for hydrogen storage and other energy related field.展开更多
Background Yueju Pill,a classic traditional Chinese medicine,shows antidepressant effects rapidly.However,biomarkers that can predict its treatment outcomes in major depressive disorder(MDD)are still lacking.Multimoda...Background Yueju Pill,a classic traditional Chinese medicine,shows antidepressant effects rapidly.However,biomarkers that can predict its treatment outcomes in major depressive disorder(MDD)are still lacking.Multimodal magnetic resonance imaging(MRI)offers a promising avenue to identify such biomarkers.Aims This pilot study aimed to explore whether therapeutic responses to Yueju Pill could be predicted by MRI-derived brain networks and to identify drug-specific biomarkers in comparison to escitalopram,a mainstream antidepressant.Methods We collected multimodal MRI data and blood samples from 28 outpatients with MDD from the Fourth People's Hospital of Taizhou,who were randomly divided into two groups to receive either Yueju Pill(23 gime/day)or escitalopram(10 mg,two times a day)for 4 days.Morphological and functional brain networks were constructed and used to predict individual changes in symptoms quantified by the 24-item Hamilton Depression Scale(HAMD-24)scores and serum brain-derived neurotrophic factor(BDNF)levels.Results After the treatment,both groups exhibited significant reductions in the HAMD-24 scores,while only the Yueju Pill group showed significant increases in the BDNF levels.Gyrification Index-based morphological networks predicted change rates of the HAMD-24 scores in both groups,but sulcus depth-based and cortical thickness-based morphological networks predicted change rates of the HAMD-24 scores and BDNF levels,respectively,only in the Yueju Pill group.Subnetwork analyses revealed that the visual network independently predicted the changes in both the HAMD-24 scores(sulcus depth-based networks)and BDNF levels(cortical thickness-based networks)following Yueju Pill treatment.Conclusions Morphological but not functional brain networks can predict symptom improvement and BDNF changes of patients with MDD after Yueju Pill treatment.Sulcus depth-based and cortical thickness-based morphological brain networks,particularly their visual subnetworks,might serve as Yueju Pill-specific biomarkers for predicting the therapeutic responses.These findings have the potential to guide personalised therapy for patients with MDD early in the therapeutic process.展开更多

The numerical calculations of localization factors and localization lengths are performed. It can be observed from the results that the band structures can be tuned by exerting the suitable initial stress. For different values of the piezoelectric rod length and the elastic constant, the band structures and the localization phenomena are very different. The waves propagating in directions normal and tangential to the layers are considered. The transfer matrices between two consecutive unit cells are obtained according to the continuity conditions. The expressions of localization factor and localization length in the disordered periodic structures are presented. For the disordered periodic piezoelectric structures, the numerical results of localization factor and localization length are presented and discussed. It can be seen from the results that the frequency passbands and stopbands appear for the ordered periodic structures and the wave localization phenomenon occurs in the disordered periodic ones, and the larger the coefficient of variation is, the greater the degree of wave localization is. The widths of stopbands in the ordered periodic structures are very narrow when the properties of the consecutive piezoelectric materials are similar and the intervals of stopbands become broader when a certain material parameter has large changes. For the wave propagating in the direction normal to the layers the localization length has less dependence on the frequency, but for the wave propagating in the direction tangential to the layers the localization length is strongly dependent on the frequency.展开更多
Background GW117(N-(2-(6-chloro-7-deuteromethoxynaphthalen-1-yl)ethyl)acetamide)is a dual-acting agent(MT1/MT2 agonist,5-HT2Cantagonist)with prior evidence of antidepressant efficacy and favourable safety.Aims To p...Background GW117(N-(2-(6-chloro-7-deuteromethoxynaphthalen-1-yl)ethyl)acetamide)is a dual-acting agent(MT1/MT2 agonist,5-HT2Cantagonist)with prior evidence of antidepressant efficacy and favourable safety.Aims To preliminarily evaluate the efficacy and safety of GW117 in major depressive disorder(MDD)and to explore the optimal dosing.Methods A total of 280 eligible patients aged 18-65years with MDD were randomly assigned(1:1:1:1)to8 weeks of double-blind treatment with fixed doses of GW117 tablets(20,40,60 mg/day)or placebo.The primary endpoint was the change from baseline to Week 8 in the total score of the Hamilton Rating Scale for Depression-17item(HAMD-17).Key secondary endpoints included changes in the Montgomery-?sberg Depression Rating Scale(MADRS)total score over the same period.Results In the full analysis set(n=276),GW117 showed numerically greater reductions versus placebo in the HAMD-17 and MADRS total scores,as well as higher response rates at Week 8.However,these differences did not reach statistical significance,potentially due to a high placebo response and other contributing factors.In a post hoc analysis of an optimal subgroup(baseline HAMD-17>24 or insomnia factor>4),GW117 showed efficacy in improving multidimensional symptoms,including insomnia.The 20 mg dose demonstrated a significant3.66-point greater reduction in MADRS(p=0.026)and a23.16%higher response rate(p=0.013)compared with placebo.GW117 was well-tolerated,with no cases of alanine aminotransferase or aspartate aminotransferase exceeding 3×the upper limit of normal and no concerning safety signals reported.Conclusions This exploratory study found that GW117demonstrated encouraging antidepressant efficacy and a favourable safety profile in patients with MDD.Although differences versus placebo did not reach statistical significance in the overall population,GW11720 mg monotherapy showed significant improvements in multidimensional depressive symptoms,including insomnia,in the optimal response subgroup.No hepatotoxicity was reported,supporting its promising therapeutic potential for further clinical development.展开更多
Background:Scalp electroacupuncture(SA)has been clinically used as a novel treat-ment for ischemic stroke and depression.This study aimed to investigate whether stimulation of SA of the Baihui(GV20)and Yintang(EX-HN3)...Background:Scalp electroacupuncture(SA)has been clinically used as a novel treat-ment for ischemic stroke and depression.This study aimed to investigate whether stimulation of SA of the Baihui(GV20)and Yintang(EX-HN3)acupoints,which regu-late the trigeminal nerve(TN),can alleviate depression and anxiety caused by post-traumatic stress disorder(PTSD)and to identify its therapeutic mechanism.Methods:After stimulation of SA,vasodilation and leakage within the TN area and c-Fos expression in the TN pathway of the brain were analyzed.Also,the anti-inflammatory,antidepressant,and anxiolytic effects of SA were investigated in a mouse model of PTSD induced by single prolonged stress.Results:SA activated the TN pathway,inducing vasodilation along the TN pathway,and increased c-Fos expression in the brain,including the spinal trigeminal nucleus,locus coeruleus(LC),amygdala,and entorhinal cortex.SA significantly alleviated de-pression-and anxiety-related behaviors,decreased pro-inflammatory cytokines,and increased the expression of neurotrophic factors in the hippocampus.Also,SA de-creased the expression of P2X7 receptors in the microglia and norepinephrine trans-porter in the LC and hippocampus.Conclusions:These results demonstrated that SA exhibited anti-inflammatory,anti-depressant,and anxiolytic effects through the P2X7 receptor pathway and the LC-norepinephrine system.展开更多

摘要Background:Auriculotherapy is a simple,convenient,nonpharmacological option that has shown positive effects for insomnia patients,but its efficacy and safety remain unknown.Objective:The aim of this study is to evaluate the efficacy and safety of auricular therapy for insomnia.Design,setting,participants and intervention:An 18-week(2-week screening,4-week treatment,and 12-week follow-up),recruiter-and assessor-blinded,randomized,sham-controlled trial was performed.Patients were recruited from outpatient settings at three clinical centers in China from 2 February 2023 to 26 January 2024.Participants had insomnia for more than 3 months and met the criteria classified in the Diagnostic and Statistical Manual of Mental Disorders(Fifth Edition).Patients were randomly assigned in a 1:1 ratio to receive auricular therapy(AT)or sham AT(SAT)for 4 consecutive weeks,twice a week,and once every 3 days.Main outcome measures:Primary outcome was the response rate at week 4 after treatment,defined as the percentage of patients with at least a 50%reduction in Pittsburgh Sleep Quality Index(PSQI)from baseline.Secondary outcomes consisted of response rates at other time points,PSQI and 4 related scales,actigraphy-recorded sleep efficiency,sleep arousal frequency,and total sleep time.Results:A total of 234 participants were screened,and 156 patients(122 female[78.2%];mean[standard deviation]age,37.2[13.5]years)were included in the intention-to-treat analysis,with 78 participants randomized to each group.A total of 117 participants(75%)were followed up for 3 months.The AT group had a higher response rate at week 4 than the SAT group(39.73%[29/73]vs 23.29%[17/73],P=0.03),and the response rates for the AT and SAT groups at week 2 were 28.38%(21/74)and 13.33%(10/75),respectively(P=0.02).The differences between the two groups in the PSQI at each time point(P<0.05)and the Insomnia Severity Index at weeks 4 and 12(both P=0.03)were also statistically significant.The other partial outcomes also showed statistically significant differences.Adverse events occurred in 2 cases in the AT group(2.60%).Conclusion:Four-week AT treatment was an effective and safe alternative therapy for insomnia,although the difference with the SAT treatment did not reach the minimal clinically important difference.Adverse events were mild or transient.

摘要Recent advances in geoscience have underscored the critical role of abiogenic processes in petroleum formation,especially the formation and polymerization of methane.However,whether a direct carbon-H2 reaction can produce C2+hydrocarbons(e.g.,ethane and propane)beyond methane remains an open question.Here,we demonstrate the direct synthesis of ethane and propane via reactions between amorphous carbon and H2 under upper mantle conditions(2-10 GPa and 800-1200℃).A systematic investigation reveals that increasing structural disorder in carbon precursors,from graphite to glassy carbon-Ⅱ and carbon black,enhances the production of C2-C3 hydrocarbons.Through integrated X-ray diffraction and reverse Monte Carlo simulations,we establish that the continuous random atomic network structures in amorphous carbon enable one-step synthesis of heavy hydrocarbons with H2.These models establish a direct link between atomic-scale carbon structures and the one-step synthesis of C2+ hydrocarbons under H2-rich,high-pressure,and high-temperature conditions—potentially revealing an efficient mechanism for the abiotic production of C2+ hydrocarbons in the upper mantle.

摘要Driven by artificial intelligence,the exponential growth in computational and energy demands has spurred exploration of alternative computing paradigms beyond traditional electronic processors.Optical neural networks(ONNs),a promising neuromorphic platform with light-speed computing,offer inherent advantages for accelerating artificial intelligence,including massive parallelism,ultralow latency,and reduced power consumption.However,most existing ONNs lack physical programmability after deployment due to rigid optical interconnects and limited optical transformations,which restrict their performance across diverse machine learning tasks.We propose a programmable and scalable ONN,an optoelectronic reservoir computing architecture integrated with cascaded linear disordered media.The key novelty lies in its physical kernel optimization via genetic algorithms,which involves programming diffuser orientations within a reduced search space and integrating wavefront shaping for feature preprocessing,thereby enhancing both optical random projection and preprocessing transformations.This platform achieves competitive accuracy in image classification with 93.5%fewer parameters than digital models;it also performs well in graph classification and human action recognition,proving feasibility for non-Euclidean and time-series datasets.Our work addresses the critical programmability bottleneck of ONNs,paving the way for scalable,energyefficient ONNs with programmable physical kernels and opening new paths for high-performance physicsinspired neuromorphic computing in resource-constrained scenarios.

摘要Objective Previous studies link lower body mass index(BMI)with increased obsessive-compulsive disorder(OCD)risk,yet other body mass indicators may be more etioloically relevant.We dissected the causal association between body fat mass(FM)and OCD.Methods Summary statistics from genome-wide association studies of European ancestry were utilized to conduct two-sample Mendelian randomization analysis.Heterogeneity,horizontal pleiotropy,and sensitivity analyses were performed to assess the robustness.Results The inverse variance weighting method demonstrated that a genetically predicted decrease in FM was causally associated with an increased OCD risk[odds ratio(OR)=0.680,95%confidence interval(CI):0.528–0.875,P=0.003].Similar estimates were obtained using the weighted median approach(OR=0.633,95%CI:0.438–0.915,P=0.015).Each standard deviation increases in genetically predicted body fat percentage corresponded to a reduced OCD risk(OR=0.638,95%CI:0.455–0.896,P=0.009).The sensitivity analysis confirmed the robustness of these findings with no outlier instrument variables identified.Conclusion The negative causal association between FM and the risk of OCD suggests that the prevention or treatment of mental disorders should include not only the control of BMI but also fat distribution and body composition.
摘要Objective Emerging evidence implicates neuroinflammation in the pathogenesis of major depressive disorder(MDD),yet the role of memory B cells remains unclear.In this study,we conducted a bidirectional two-sample Mendelian randomiza-tion(MR)study and Bayesian colocalization analyses to investigate the causal relationships between memory B-cell traits and MDD risk.Methods MDD summary data were gathered from a meta-analysis of genome-wide association studies(GWASs),whereas memory B-cell genetic variations were sourced from GWASs on immune phenotypes.MR analysis utilized the inverse variance weighted(IVW),MR-Egger,and weighted median methods.Moreover,various sensitivity analyses,including Cochran's Q test,MR Pleiotropy Residual Sum and Outlier(MR-PRESSO),MR-Egger intercept test and Leave-one-out(LOO)analysis,were performed to confirm MR result stability.Bayesian colocalization analyses were also conducted to identify genetic loci shared between memory B cells and MDD.Results Our results indicated that genetically predicted increased CD27 protein expression on memory B cells causally elevated MDD risk(ORs:1.025-1.063,PFDR<0.05).Conversely,MDD did not causally affect memory B-cell traits.Addi-tionally,the colocalization analysis revealed no shared genetic variants,suggesting distinct biological pathways.Conclusions These findings highlight CD27 as a potential novel biomarker and therapeutic target in MDD,warranting fur-ther clinical validation in the future.

摘要Traditional carbon/carbon composites,such as carbon fiber reinforced carbon matrix composites,are a promising carbon material widely applied in the aerospace field due to their unique mechanical,thermal,and electrical properties.However,it becomes almost impossible to further enhance their mechanical properties because of the weak van der Waals bonding within or between components.Here,by precisely controlling the graphitization transformation of nanodiamonds,we have successfully synthesized in-situ disordered graphitic carbonanodiamond composites under an industrially achievable pressure of 2 GPa and a moderate temperature of 1200-1400℃.In the composites,nanodiamonds are dispersed in disordered graphitic carbon fragments,featured by a larger spacing compared to that of ideal graphite.Moreover,the nanodiamond and graphitic carbon components are irregularly connected at the interface through random sp2or sp3bonding.Compared with the traditional carbon/carbon composites,the synthesized composites demonstrate exceptional properties:a hardness of up to 17.5 GPa,a compressive strength reaching up to 1.8 GPa,a flexural strength of up to 380 MPa,and an electrical conductivity of around 2290 S/m.This research makes it possible to fabricate high-performance carbon/carbon compos-ites coupled high strength/hardness and electrical conductivity at an industrial achievable pressure.

摘要The ability to simulate physical systems is essential for an understanding of their physical properties and advancing related technologies.However,simulating certain complex systems at scale,including many-body spin models,is often a significant challenge for conventional computing approaches.Quantum simulators,employing controlled quantum systems that mimic the dynamics of a target system,can offer a more efficient approach.This work demonstrates the use of photonic circuits for quantum simulation of disordered spin systems,where quantum properties such as superposition and interference can be leveraged for a more efficient simulation.We fabricate and characterize a low-loss silicon-on-insulator chip with a buried aluminum mirror and a reconfigurable interferometer,which is employed in conjunction with two input photons to map the dynamics of a spin Hamiltonian with four-body interactions.Our results demonstrate several characteristics associated with the target system,such as spin-degenerate ground states and phase transitions,confirming the device's functionality as a quantum simulator of the spin system.

摘要Pseudocapacitive materials employ rapid,non-diffusion-limited faradaic processes to store charge,demonstrating significant potential for fast-charging batteries and supercapacitors.However,the high redox potentials of existing pseudocapacitive anodes substantially lower the overall cell voltage and energy density.Here,a cation-disordered rock-salt lithium titanium oxyfluoride(DRX-LixTiOF2,0<x<2)is reported to reversibly accommodate approximately 1.19 mol of Li+(~310 mAh g−1)and delivers high-rate performance(exceeding 64.4 C)via pseudocapacitive Li+storage within a low potential window extending down to 0.1 V vs.Li+/Li.This pseudocapacitive behavior is characterized by several structural and electrochemical features:the absence of phase transformation during Li+intercalation,quasi-rectangular cyclic voltammetry curves,sloping charge/discharge profiles and a surface-controlled current response.We further reveal that the pseudocapacitive characteristics originate from a three-dimensional percolation network that facilitates fast Li+migration with low energy barriers,enabled by a cation/anion-disordered structure arising from the mixed occupancy of Li/Ti cations and O/F anions.Owing to its low operating potential and high-rate capability,DRX-LixTiOF2 allows a lithium-ion capacitor to attain 4.0 V cell voltage and achieve energy and power densities several times higher than those obtained with conventional anodes,such as battery-type Li4Ti5O12 or pseudocapacitive materials like Nb2O5 and TiO2.

摘要To the editor:Methamphetamine use disorder(MUD) is associated with deficits in cognitive control,decision-making and reward processing,which can contribute to the maintenance of addictive behaviours.1 Substantial evidence from neuroimaging,neuropsychological and computational modelling studies also indicates dysregulation of the neural circuits underlying reward processing.2,3 Reward learning,crucial in reward processing,may be impaired in MUD,affecting individuals' ability to learn action values and guide goal-directed behaviours,potentially contributing to substance use disorders.4 Recently,there has been growing interest in computational neuroscience approaches to understanding the mechanisms of reward learning deficits in addiction.5 Reinforcement learning models have been used to parse the specific components of reward learning that may be impaired in addiction,such as value representation,prediction error signalling and decisionmaking.
摘要Intrinsically disordered proteins(IDPs)are a class of proteins lacking stable three-dimensional structures and play crucial roles in various kinds of cellular processes.Given that more than half of human proteins are at least partially disordered,systematically classifying IDPs helps to better understand the regularities in their functions and mechanisms.However,there is currently no such a systematic classification system for IDPs.In this study,we classified IDPs according to sequence and disorder conservation and explored the distinct properties and functional significance of different types of IDPs.Seven species representing evolutionary nodes of human lineage within the Euarchontoglires were selected,and all human proteins were classified into seven categories based on the homologous gene distribution range and the conservation of sequence and disorder degree.Among them,IDPs with homologous genes in all seven species were classified into four categories based on the conservation of sequence and disorder degree:proteins with both sequence and disorder conservation;proteins with disorder conservation but no sequence conservation;proteins lacking both sequence and disorder conservation;and proteins with sequence conservation but lacking disorder conservation.This study systematically investigates the characteristics of these types of IDPs with respect to physicochemical properties,evolutionary characteristics,functional role,expression pattern.The IDPs with both sequence and disorder conservation(Ho_ConS_ConD)represent the canonical IDP phenotype;those with conserved sequence but non-conserved disorder(Ho_ConS_NoConD)form a seemingly paradoxical yet functionally important class;IDPs lacking conservation in both features(Ho_NoConS_NoConD)exhibit pronounced tissue specificity,particularly in the testis.Overall,we classified IDPs from a novel perspective,constructing a systematic framework to elucidate the functions of different IDPs.
摘要[Objectives]To investigate the potential causal relationship between psoriasis and common mental disorders,and to provide genetic epidemiological evidence for the early intervention of mental comorbidities.[Methods]Based on publicly available large-scale GWAS data,a bidirectional Mendelian randomization(MR)approach was employed to assess the causal associations between psoriasis and major depressive disorder(MDD),bipolar disorder,schizophrenia,and anxiety disorders.The inverse variance weighted(IVW)method served as the primary analytical tool,supplemented by sensitivity analyses using MR-Egger and weighted median methods.Additionally,a subgroup analysis was conducted for psoriatic arthritis(PsA).[Results]Forward MR analysis revealed a significant positive causal association between the genetic predisposition to psoriasis and bipolar disorder as well as MDD,whereas no significant causal relationship was observed with schizophrenia or anxiety disorders.The reverse MR analysis found no causal effect of mental disorders on psoriasis.Subgroup analysis of PsA indicated that its genetic predisposition was significantly associated with the risk of bipolar disorder.The results of various sensitivity analyses and pleiotropy tests supported the robustness of the conclusions.[Conclusions]This study provides genetic evidence supporting a causal link between psoriasis and both MDD and bipolar disorder.In particular,patients with PsA are at a higher risk of developing bipolar disorder,highlighting the need to strengthen early screening and intervention for mental health in clinical management.

摘要BACKGROUND Anxiety disorders are highly prevalent in patients with bipolar disorder(BD)and are associated with a more severe illness course and poorer outcomes.A significant clinical challenge is the frequent initial misdiagnosis of BD as anxiety,leading to prolonged diagnostic delays and suboptimal treatment.Growing evidence suggests that early intervention in individuals at high risk for BD can improve prognosis.Established clinical high-risk factors include early onset,family history of BD,and subthreshold manic symptoms.This creates a clinical dilemma whereby the administration of first-line antidepressants(e.g.,sertraline)for anxiety is debated in patients with a bipolar diathesis,given the associated risk of mood destabilization.Conversely,mood stabilizers like lithium are foundational in BD treatment,but their role in treating anxiety in high-risk populations is unproven.Therefore,we conducted this randomized controlled trial to evaluate whether early intervention with a combination of sertraline and lithium is more effective than sertraline monotherapy for anxiety disorder patients with clinical high-risk factors for BD.AIM To investigate whether early intervention has a more positive outcome for anxiety disorders in patients who present with clinical high risk factors for BD.METHODS A total of 66 patients were enrolled in this study from January 2021 and December 2022 in Huzhou Third Municipal Hospital.They were randomly assigned to two groups to receive either an antidepressant(sertraline,n=32)or a combination therapy(sertraline and lithium,n=34).The main variables included alterations in Hamilton Anxiety Rating Scale and Hamilton Depression Rating Scale scores from the initial assessment to the final evaluation.A thorough combined Wald test was conducted to examine the intention-to-treat differences in scale assessment across treatment and time conditions.RESULTS Significant differences in the change of Hamilton Anxiety Rating Scale scores were observed between the two groups at week 1,week 2,and week 4(P<0.05).However,after 8 weeks and 12 weeks of treatment,there were no significant different(P=0.485 and P=0.206).There was no significant difference in the change over time in Hamilton Depression Rating Scale scores between the treatment groups(P=0.2),except at week 12(P=0.034).No significant differences were observed in the adverse effects reported between patients treated with sertraline alone(18%)and those treated with the combination therapy(21%).CONCLUSION This current double-blind,case-controlled study assessed the effectiveness and tolerability of combined therapy vs monotherapy for anxiety disorder in patients with clinical high-risk factors for BD.In light of the constraints associated with this initial study,the results imply that the combination of sertraline and lithium may provide a more favorable prognosis.

摘要BACKGROUND Disorders of consciousness are critical emergencies requiring timely intervention to improve outcomes.Process reengineering using checklists can enhance efficiency in clinical settings.AIM To evaluate whether checklist-based process reengineering in the Emergency Department reduces emergency stay duration and hospitalization time,and improves rescue success rate,prognosis,and satisfaction among patients with disorders of consciousness.METHODS From January 2021 to December 2023,our hospital admitted 400 emergency consciousness disorders patients using purposive sampling.The first 200,undergoing rescue without the checklist-based process,formed the control group.The next 200,post-implementation,constituted the observation group.We collected data on emergency stay duration,rescue success rate,hospitalization duration,and patient satisfaction for analysis.Patient prognosis in both groups was compared.RESULTS The observation group showed significantly shorter emergency stay duration,time to recovery,and overall hospitalization duration(intensive care unit,general ward,total)compared to the control group(P<0.05).The observation group had a higher rescue success rate(P<0.05)and better recovery rate(P<0.05)than the control group.Both medical staff and patients’family members reported higher satisfaction with the emergency rescue model after checklist-based process reengineering compared to the conventional emergency rescue model(P<0.05).CONCLUSION Emergency process reengineering based on checklists can effectively reduce the emergency stay duration for patients with consciousness disorders,improve rescue success rates,enhance patient prognosis,and increase overall medical and patient satisfaction.
摘要Objective:To investigate the clinical efficacy,improvement in social functioning,prevention of relapse,and health economic value of a comprehensive community-based rehabilitation service model in the holistic management of patients with severe mental disorders,providing empirical evidence for the standardized promotion of community rehabilitation.Methods:A parallel-group randomized controlled trial(RCT)was conducted,involving 200 patients with severe mental disorders in a stable phase.They were randomly assigned at a 1:1 ratio to an intervention group(comprehensive community rehabilitation,n=100)and a control group(routine community management,n=100),with continuous intervention for 12 months.The Positive and Negative Syndrome Scale(PANSS)and the Personal and Social Performance Scale(PSP)were used as primary outcome measures to compare differences in symptom improvement,social functioning,relapse rate,medication adherence,hospitalization status,and medical expenses between the two groups.Results:After 12 months of intervention,the PANSS total score in the intervention group(43.0±6.8)was significantly lower than that in the control group(47.9±7.5),while the PSP score(66.0±8.0)was significantly higher than that in the control group(60.8±7.3),with statistically significant differences(p<0.001).The relapse rate in the intervention group(10.0%)was significantly lower than that in the control group(22.0%),while the medication adherence rate(83.0%)was significantly higher than that in the control group(68.0%).The hospitalization rate,number of hospitalization days,and annual total medical expenses were all significantly lower in the intervention group than in the control group(p<0.05).Conclusion:The comprehensive community-based rehabilitation service model can effectively improve psychiatric symptoms and social functioning in patients with severe mental disorders,reduce the risk of relapse and hospitalization,and save medical costs.It represents an efficient and promotable core intervention strategy for the holistic management of severe mental disorders.

摘要Catalytic doping is one of the economic and efficient strategies to optimize the operating temperature and kinetic behavior of magnesium hydride(MgH2).Herein,efficient regulation of electronic and structural rearrangements in niobium-rich nickel oxides was achieved through precise compositional design and niobium cation functionalized doping,thereby greatly enhancing its intrinsic catalytic activity in hydrogen storage systems.As the niobium concentration increased,the Ni-Nb catalysts transformed into a mixed state of multi-phase nanoparticles(composed of nickel and niobium-rich nickel oxides)with smaller particle size and uniform distribution,thus exposing more nucleation sites and diffusion channels at the MgH2/Mg interface.In addition,the additional generation of active Ni-Nb-O mixed phase induced numerous highly topical disordered and distorted crystalline,promoting the transfer and reorganization of H atoms.As a result,a stable and continuous multi-phase/component synergistic catalytic microenvironment could be constructed,exerting remarkable enhancement on MgH2’s hydrogen storage performance.After comparative tests,Ni0.7Nb0.3-doped MgH2 presented the optimal low-temperature kinetics with a dehydrogenation activation energy of 78.8 kJ·mol−1.The onset dehydrogenation temperature of MgH2+10 wt%Ni0.7Nb0.3 was reduced to 198℃ and 6.18 wt%H2 could be released at 250℃ within 10 min.In addition,the dehydrogenated MgH2–NiNb composites absorbed 4.87 wt%H2 in 10 min at 125℃ and a capacity retention rate was maintained at 6.18 wt%even after 50 reaction cycles.In a word,our work supplies fresh insights for designing novel defective-state multiphase catalysts for hydrogen storage and other energy related field.

摘要Background Yueju Pill,a classic traditional Chinese medicine,shows antidepressant effects rapidly.However,biomarkers that can predict its treatment outcomes in major depressive disorder(MDD)are still lacking.Multimodal magnetic resonance imaging(MRI)offers a promising avenue to identify such biomarkers.Aims This pilot study aimed to explore whether therapeutic responses to Yueju Pill could be predicted by MRI-derived brain networks and to identify drug-specific biomarkers in comparison to escitalopram,a mainstream antidepressant.Methods We collected multimodal MRI data and blood samples from 28 outpatients with MDD from the Fourth People's Hospital of Taizhou,who were randomly divided into two groups to receive either Yueju Pill(23 gime/day)or escitalopram(10 mg,two times a day)for 4 days.Morphological and functional brain networks were constructed and used to predict individual changes in symptoms quantified by the 24-item Hamilton Depression Scale(HAMD-24)scores and serum brain-derived neurotrophic factor(BDNF)levels.Results After the treatment,both groups exhibited significant reductions in the HAMD-24 scores,while only the Yueju Pill group showed significant increases in the BDNF levels.Gyrification Index-based morphological networks predicted change rates of the HAMD-24 scores in both groups,but sulcus depth-based and cortical thickness-based morphological networks predicted change rates of the HAMD-24 scores and BDNF levels,respectively,only in the Yueju Pill group.Subnetwork analyses revealed that the visual network independently predicted the changes in both the HAMD-24 scores(sulcus depth-based networks)and BDNF levels(cortical thickness-based networks)following Yueju Pill treatment.Conclusions Morphological but not functional brain networks can predict symptom improvement and BDNF changes of patients with MDD after Yueju Pill treatment.Sulcus depth-based and cortical thickness-based morphological brain networks,particularly their visual subnetworks,might serve as Yueju Pill-specific biomarkers for predicting the therapeutic responses.These findings have the potential to guide personalised therapy for patients with MDD early in the therapeutic process.

摘要The elastic wave localization in disordered periodic piezoelectric rods with initial stress is studied using the transfer matrix and Lyapunov exponent method. The electric field is approximated as quasi-static. The effects of the initial stress on the band gap characteristics are investigated. The numerical calculations of localization factors and localization lengths are performed. It can be observed from the results that the band structures can be tuned by exerting the suitable initial stress. For different values of the piezoelectric rod length and the elastic constant, the band structures and the localization phenomena are very different. Larger disorder degree can lead to more obvious localization phenomenon.

摘要Considering the mechnoelectrical coupling, the localization of SH-waves in disordered periodic layered piezoelectric structures is studied. The waves propagating in directions normal and tangential to the layers are considered. The transfer matrices between two consecutive unit cells are obtained according to the continuity conditions. The expressions of localization factor and localization length in the disordered periodic structures are presented. For the disordered periodic piezoelectric structures, the numerical results of localization factor and localization length are presented and discussed. It can be seen from the results that the frequency passbands and stopbands appear for the ordered periodic structures and the wave localization phenomenon occurs in the disordered periodic ones, and the larger the coefficient of variation is, the greater the degree of wave localization is. The widths of stopbands in the ordered periodic structures are very narrow when the properties of the consecutive piezoelectric materials are similar and the intervals of stopbands become broader when a certain material parameter has large changes. For the wave propagating in the direction normal to the layers the localization length has less dependence on the frequency, but for the wave propagating in the direction tangential to the layers the localization length is strongly dependent on the frequency.
摘要Background GW117(N-(2-(6-chloro-7-deuteromethoxynaphthalen-1-yl)ethyl)acetamide)is a dual-acting agent(MT1/MT2 agonist,5-HT2Cantagonist)with prior evidence of antidepressant efficacy and favourable safety.Aims To preliminarily evaluate the efficacy and safety of GW117 in major depressive disorder(MDD)and to explore the optimal dosing.Methods A total of 280 eligible patients aged 18-65years with MDD were randomly assigned(1:1:1:1)to8 weeks of double-blind treatment with fixed doses of GW117 tablets(20,40,60 mg/day)or placebo.The primary endpoint was the change from baseline to Week 8 in the total score of the Hamilton Rating Scale for Depression-17item(HAMD-17).Key secondary endpoints included changes in the Montgomery-?sberg Depression Rating Scale(MADRS)total score over the same period.Results In the full analysis set(n=276),GW117 showed numerically greater reductions versus placebo in the HAMD-17 and MADRS total scores,as well as higher response rates at Week 8.However,these differences did not reach statistical significance,potentially due to a high placebo response and other contributing factors.In a post hoc analysis of an optimal subgroup(baseline HAMD-17>24 or insomnia factor>4),GW117 showed efficacy in improving multidimensional symptoms,including insomnia.The 20 mg dose demonstrated a significant3.66-point greater reduction in MADRS(p=0.026)and a23.16%higher response rate(p=0.013)compared with placebo.GW117 was well-tolerated,with no cases of alanine aminotransferase or aspartate aminotransferase exceeding 3×the upper limit of normal and no concerning safety signals reported.Conclusions This exploratory study found that GW117demonstrated encouraging antidepressant efficacy and a favourable safety profile in patients with MDD.Although differences versus placebo did not reach statistical significance in the overall population,GW11720 mg monotherapy showed significant improvements in multidimensional depressive symptoms,including insomnia,in the optimal response subgroup.No hepatotoxicity was reported,supporting its promising therapeutic potential for further clinical development.

摘要Background:Scalp electroacupuncture(SA)has been clinically used as a novel treat-ment for ischemic stroke and depression.This study aimed to investigate whether stimulation of SA of the Baihui(GV20)and Yintang(EX-HN3)acupoints,which regu-late the trigeminal nerve(TN),can alleviate depression and anxiety caused by post-traumatic stress disorder(PTSD)and to identify its therapeutic mechanism.Methods:After stimulation of SA,vasodilation and leakage within the TN area and c-Fos expression in the TN pathway of the brain were analyzed.Also,the anti-inflammatory,antidepressant,and anxiolytic effects of SA were investigated in a mouse model of PTSD induced by single prolonged stress.Results:SA activated the TN pathway,inducing vasodilation along the TN pathway,and increased c-Fos expression in the brain,including the spinal trigeminal nucleus,locus coeruleus(LC),amygdala,and entorhinal cortex.SA significantly alleviated de-pression-and anxiety-related behaviors,decreased pro-inflammatory cytokines,and increased the expression of neurotrophic factors in the hippocampus.Also,SA de-creased the expression of P2X7 receptors in the microglia and norepinephrine trans-porter in the LC and hippocampus.Conclusions:These results demonstrated that SA exhibited anti-inflammatory,anti-depressant,and anxiolytic effects through the P2X7 receptor pathway and the LC-norepinephrine system.
