Here we explore an unusual distylous flower associated with self- and intramorph compatibility in Mussaenda macrophylla Wall. The species did not exhibit a precise reciprocal herkogamy, and the populations studied had a slight bias in favor of the short-styled morph (S-morph). In addition, pollen tube growth in intermorph pollination was faster than either in intramorph or self-pollination, indicating that cryptic heteromorphic self-incompatibility occurred in M. macrophylla. Specifically, pollen tube growth rates after legitimate and illegitimate pollination were much more highly differentiated in long-styled morph (L-morph) than in S-morph, which might be attributed to the increased de- esterified pectin in the tip of L-morph pollen tube wall in comparison with S-morph. Transmission electron microscopic observation revealed illegitimate self-pollinated pollen tubes of two morphs were seriously degraded with the large autolysosome-like vacuoles in cytoplasm of L-morph pollen tube but accumulating small vacuoles in that of S-morph pollen tube, suggesting that incompatible pollen tubes might undergo autophagic programmed cell death except self-incompatible S-morph pollen tubes. Our results indicate that M. macrophylla is morphologically distylous with a cryptic heteromorphic self-incompatibility breeding system, which functions differently in S-morph with self-pollination compared with other illegitimate pollination. 展开更多

Reconstructions of phylogenetic relationships in the flowering plant family Rubiaceae have up until now relied heavily on single-or multi-gene data,primarily from the plastid compartment.With the availability of cost-and time-efficient techniques for generating complete genome sequences,the opportunity arises to resolve some of the relationships that,up until now,have proven problematic.Here,we contribute new data from complete 58 plastid genome sequences,representing 55 of the currently 65 recognized tribes of the Rubiaceae.Also contributed are new data from the nuclear rDNA cistrons for corresponding taxa.Phylogenetic analyses are conducted on two plastid data sets,one including data from the protein coding genes only,and a second where protein coding data are combined with non-coding regions,and on a nuclear rDNA data set.Our results clearly show that simply adopting a“more characters”approach does not resolve the relationships in the Rubiaceae.More importantly,we identify conflicting phylogenetic signals in the data.Analyses of the same plastid data,treated as nucleotides or as codon-degenerated data,resolve and support conflicting topologies in the subfamily Cinchonoideae.As these analyses use the same data,we interpret the conflict to result from erroneous assumptions in the models used to reconstruct our phylogenies.Conflicting signals are also identified in the analyses of the plastid versus the nuclear rDNA data sets.These analyses use data from different genomic compartments,with different inheritance patterns,and we interpret the conflicts as representing“real”conflicts,reflecting biological processes of the past. 展开更多

The potential application of DNA barcodes of plastid (matK, trnH-psbA, petD, and rbcL) and nuclear (internal transcribed spacer (ITS) of rDNA) DNA regions was investigated for 25 Hedyotis taxa. The ITS showed the best species discrimination by resolving 23 of the species as exclusive lineages with no shared alleles between any of the 24 distinct species (H. assimilis and/-/, mellii are not supported as distinct species based on our molecular and morphological data). Conversely, rbcL performed the worst and only resolved 10 of the species as exclusive lineages, and 10 species with shared alleles. Using ITS has the advantage of high PCR amplification success and it provides good intra- and interspecific variation distribution patterns. The most powerful plastid markers were petD and trnH-psbA, but we could amplify and sequence trnH-psbA for only 83% of the accessions sampled. Combination of ITS and petD performed extremely well, with all 24 of the distinct species resolved as exclusive lineages and no shared alleles between any of the distinct species. We therefore recommend ITS, or a combination of ITS andpetD, as the standard DNA barcode in Hedyotis, but acknowledge that there are no shared alleles between distinct species for matK and rbcL combined. 展开更多

Cinchona officinalis (Rubiaceae) is an endemic species of the Loja Valley in southern Ecuador with medicinal uses. Because of over-exploitation in the nine- teenth century and more recent disturbances to its ecosystem, C. officinalis populations are threatened. Currently, natural regeneration of the populations is low, despite its high plant regeneration and seed formation capacity. In the present study, an efficient protocol for germination, shoot proliferation and plantlets regeneration was developed for this species. Phenolic content and germination rate of C. officinalis seeds were compared with a control species, C. pubescens. Nodal segments from seedlings of C. officinalis were cultured on Gamborg medium supplemented with different combinations of plant growth regulators. Because the phenol content is high in C. officinalis, the phenolic should be removed with hydrogen peroxide or water washes to stimulate germination. Shoots and callus developed from nodal segments within 45 days using most of the tested combinations of plant growth regulators. The best rates of shoot proliferation, callus formation and adventitious buds were obtained in medium supplemented with 5.0 mg L^-1 6-benzyl-aminopurine and 3.0 mg L^-1 indole- 3-butyfic acid. 展开更多

Wang (Rubioideae, Rubiaceae), are described from eastern Guangdong, China. It is characterized by its subshrub habit, pentamerous and heterostylous flowers, 2-1ocular ovary with many ovules, and apically dehiscent capsules with numerous angulated seeds. Phylogenetic analysis of four chloroplast DNA regions (rbcL, rps16, ndhF, and atpB-rbcL) revealed that the new genus is nested in the Spermacoceae alliance and is sister to Dunnia. Morphological comparison between these two genera indicated that they had few synapomorphies; it was therefore inappropriate to classify the new genus in the existing tribe Dunnieae, and a new tribe, Foonchewieae R. Antherostele Bremek. is an understudied genus of Rubiaceae endemic to the Philippines formerly comprising four species of shrubs or small trees distributed in Luzon and Visayas. In this first molecular study of the genus, the ITS (nrDNA) was utilized to determine the monophyly of Antherostele, reconstruct its phylogeny, and resolve its placement in Rubiaceae. Furthermore, an update on the assessment of its conservation status of the included species in the genus was done. Antherostele is monophyletic and is nested within the tribe Urophylleae of subfamily Rubioideae which confirms its segregation from Urophyllum and its placement within Urophylleae as proposed by earlier authors based on morphology. However, the relationships of Urophylleae members remain unresolved. Antherostele is united by reflexed corolla, hairs on upper side of the corolla lobes and syngenesious stamens. The montane A. luzoniensis with small leaves is found to be sister to the rest of Antherostele species. Antherostele is hypothesized to originate from montane regions and diversified by forming larger leaves following dispersal to the shaded forest understory. From the five species of Antherostele currently recognized in the study, two are endangered (A. banahaensis, A. grandistipula) and three are critically endangered including a new species (A. callophylla, A. luzoniensis, A. sp. nov.) following the criteria of IUCN. 展开更多

METHOD: Compounds were isolated by various chromatographic methods. Their structures were elucidated on the basis of spectroscopic techniques(IR, UV, MS, and NMR), and they were evaluated for their cytotoxic activities by the MTT method. RESULTS: Eight triterpenes were isolated and identified as 3α, 6β, 19α-trihydroxy-urs-12-en-28-oic acid(1), 3β, 6β, 19α- trihydroxy-urs-12-en-28-oic acid(2), 3-oxo-6β-19α-dihydroxy-urs-12-en-28-oic acid(3), 3β, 6β, 19α-trihydroxy-urs-12-en-24, 28-dioic acid 24-methyl ester(4), 3β, 6β, 19α, 24-tetrahydroxy-urs-12-en-28-oic acid(5), rotundic acid(6), 23-nor-24-exomethylene- 3β, 6β, 19α-trihydroxy-urs-12-en-28-oic acid(7), and pololic acid(8), respectively. All of the isolates were tested against two human tumor cell lines, MCF-7(breast) and HT-29(colon). CONCLUSION: Compound 1 was a new triterpene. Compounds 5-7 exhibited potent inhibitory effects on the growth of MCF-7 and HT-29 cells, and the others showed no cytotoxicity. 展开更多

Tribe Condamineeae appears to be well supported in recent phylogenetic studies.However,the species of Bathysa were divided into two clades,leading to restoration of Schizocalyx.We studied the reproductive biology of one species from each clade,which occur sympatrically in the montane Brazilian Atlantic forest.Flowering overlap was short(from December to March in B.australis and from February to June in S.cuspidatus).The flowers of both species are protogynous and homostylous and last for about 3 days.The unit of pollination in B.australis is the inflorescence.Its flowers have a greenish hypocrateriform corolla(tube about 5 mm long)and were mainly pollinated by bees and wasps in search of nectar.Schizocalyx cuspidatus has white flowers with an infundibuliform corolla(tube about 8 mm long),and its main pollinators were stingless bees in search of pollen.The pollination systems of the two species did not correspond to their pollination syndromes.Morphological differences between Bathysa and Schizocalyx were reflected in their pollination systems,with greater phenotypic specialization in S.cuspidatus,the flowers of which offer pollen as the main resource,an unusual feature within Rubiaceae.Schizocalyx cuspidatus showed higher reproductive capacity by having more inflorescences per plant,more ovules per flower,and twice the proportion of flowering individuals.However,the reproductive efficiency(fruit set,seed/ovule ratio)did not differ between the species,despite the higher frequency of visits by pollinators to S.cuspidatus.Self-compatibility in B.australis and self-incompatibility in S.cuspidatus seem to explain these results. 展开更多

Brazil is the first producer and second consumer in the world. Flower evocation triggered by environmental signals is essential for adaptability and productivity, and despite that it is neglected and barely considered as a part of the reproductive cycle. Aiming to review molecular mechanisms producing phenological responses observed in the fields, orthologs to A. thaliana CO, FLC/FLM, FLT, SOC and VRN genes were identified in silico for C. arabica and its ancestors C. canephora and C. eugenioides. Protein structures and conserved domains, regulatory elements in promoters, and the related literature in both genera were accessed and compared. Hypotheses regarding Coffea spp. orthologs responsiveness to light and temperature signals at the tropics are proposed. Preliminary analysis of phenological data taken from early, intermediary and late C. arabica plants are included to illustrate the diversity regarding flower bud emission, which quite certainly is defined during flower evocation. 展开更多

They were characterized as adinaic acid 3 beta-O-[alpha-L-rhamnopyranosyl(1-->2)-beta-D-glucopyranosyl(1-->2)-beta-D-glucurono-pyranoside-6-O-methyl ester]-28-O-beta-D-glucopyranoside, adinaic acid 3 beta-O-[alpha-L-rhamnopyranosyl(1-->2)-beta-D-glucopyranosyl(1-->2)-beta-D-glucuronopyranoside-6-O-butyl ester]-28-O-beta-D-glucopyranoside, adinaic acid 3 beta-O-[beta-D-glucopyranosyl(1-->2)-beta-D-glucopyrnaosyl]-(28-->1)-beta-D-glucopyranosyl(1-->6)-beta-D-glucopyranosyl ester, 27-hydroxyursolic acid 3 beta-O-[alpha-L-rhamnopyranosyl (1-->2)-beta-D-glucopyranosyl(1-->2)-beta-D-glucuronopyranoside-6-O-methyl ester]-28-O-beta-D-glycopyranoside.
