Comparative Study of Auricularia auricula Polysaccharide-Iron Complex,Heme Iron,and Ferrous Glycinate in Iron-Deficient Adults:A Randomized Clinical Trial 认领 引用

Iron deficiency anemia affects approximately 1.62 billion people worldwide,yet traditional iron supplements present bioavailability limitations and gastrointestinal side effects.This randomized,double-blind clinical trial investigated a novel Auricularia auricula polysaccharide-iron complex(AAPIC)compared with heme iron and ferrous glycinate in 180 iron-deficient adults receiving 30 mg elemental iron daily for 12 weeks.AAPIC achieved comparable hemoglobin improvements(from 98.3±8.7 to 126.5±9.2 g/L)to heme iron(from 97.8±9.1 to 128.3±8.6 g/L)and was significantly superior to ferrous glycinate(from 98.6±8.9 to 119.7±10.3 g/L;p<0.001).Iron absorption efficiency showed AAPIC at 23.7±4.2%,heme iron at 25.1±3.8%,and ferrous glycinate at 18.4±5.1%.Toxicological assessments revealed no hepatotoxicity,nephrotoxicity,or mutagenicity.Gastrointestinal adverse events occurred in 8.3%of AAPIC recipients versus 15.0%with ferrous glycinate and 10.0%with heme iron.The polysaccharide component facilitates iron transport through enhanced intestinal uptake mechanisms.AAPIC represents a promising,well-tolerated alternative with clinical efficacy comparable to established iron formulations. 展开更多

Auricularia auricula(AA)and Auricularia polytricha(AP)are popular edible fungi.This study successfully produced hypoglycemic polysaccharides from un-smashed or smashing and sieving(through a 10-mesh sieve)AA and AP(termed as AAP/AAP-10 and APP/APP-10)via scalable processes(water extraction,ethanolic precipitation and deproteinization).This is the first report to compare the effectiveness of AAP and APP in combating streptozotocin-induced oxidative stress and diabetes-related changes in mice(body weight,fasting blood glucose,serum insulin,proinflammatory mediator and cytokines,oxidative stress-related products,antioxidant enzymes).APP and AAP with different molecular weights and monosaccharide molar ratios could be therapeutic options for diabetes with a low dose(100 mg/kg/day)likely working better.At the same dose,APP generally performed more effective than AAP,and AAP-10/APP-10 seemed slightly more beneficial than AAP/APP.One mechanism underlying these antidiabetic functions might involve the NF-κB and associated signalling pathways.AP is cheaper than AA,thereby representing a favorable source of functional polysaccharides. 展开更多

The objective of the study was to examine the physicochemical properties and emulsification stability of three different Auricularia auricula polysaccharides(AAP)obtained through hot water extraction(AAP-W),hot acid extraction(AAP-A),and hot alkaline extraction(AAP-AL),respectively.The findings indicated that AAP-W exhibited superior emulsification stability compared to the other two polysaccharides.AAP-W was employed as a natural emulsifier for emulsion preparation,to examine the influence of varying polysaccharide concentrations and oil-water ratios on emulsion stability.Additionally,an investigation was conducted into the stability of the emulsions with respect to pH and salt ion concentration.The findings revealed that the most favorable polysaccharide concentration for the AAP-W emulsion was determined to be 1%,while the volume fraction of the oil phase was established at 0.5.It was also observed that the emulsion exhibited robust stability even in challenging conditions characterized by strong acidic(pH 3−5)or basic environments(pH 9−11),as well as high concentrations of salt ions(0−500 mM).Furthermore,the construction of an AAP-W emulsion system incorporatingβ-carotene was undertaken to enhance the preservation,bioavailability,and digestive stability ofβ-carotene,thereby expanding the potential applications of Auricularia auricula polysaccharides.This endeavor also presents a novel approach towards the advancement of novel functional food products. 展开更多

Conventional mechanical method and mechanical method combined with vacuum freeze-drying technology were used to make the ultra-fine powders of edible fungus (Auricularia auricular). The content of basic nutrients, amino acid, micro structure and their properties of raw edible fungus and the edible fungus powders obtained with the two methods were analyzed and compared. The granularity size and micro-structure of the pulverized samples were analyzed by SEM and TEM technology. The average granularity size of the edible fungus powder obtained with mechanical method was 1–5 μm, while that obtained with mechanical method combined with vacuum freeze-drying process was 0.5–1 μm. The ultra-fine powders of edible fungus obtained with the two methods had better water recovery capability and quality, and their preserving time was longer than that of raw edible fungus. All the properties of the ultra-fine powders of edible fungus obtained with the vacuum freeze-drying technology were evidently superior to that of the conventional mechanical method. In order to improve the anticoagulant function of Auricularia auricula, Auricularia auricula polysaccharide (AAP) was converted into its derivatives by the microbial fermentation method and then polysaccharide derivatives with stronger anticoagulant activity were prepared. The optimal conditions for fermenting the polysaccharide from A. auricula were examined and the in vitro anticoagulant activities of transformed and untransformed polysaccharides were compared. Response surface tests and an orthogonal experiment indicated that the best conditions for microbial conversion of AAP3 were an AAP3 concentration of 4.0 mg/ mL, a ratio of substrate (AAP3 ) to donor (p-hydroxybenzoic acid) of 40:1, and a pH of 6.0. Bacillus subtilis Bs-07 was inoculated and then placed on a rotary shaker ( 120 r/min), followed by fermentation for 48 h at 35 ℃. The conversion rate was found to be greater than 40%. The result of in vitro anticoagulant activity showed that the transformed polysaccharide improved activated partial thromboplastin time, prothrombin time, and thrombin time values and greatly enhanced anticoagulant activity compared to the untransformed polysaccharide. 展开更多

We aimed to investigate the synergistic effects of ultrasonic waves and repeated freeze-thaw cycles on cel-wal destruction of Auricularia auric-ula mycelia, and determine the best combination of conditions for cel-wal destruc-tion of Auricularia auricula mycelia. [Method] The effects of destruction time, added water, destruction times, freeze time and number of freeze-thaw cycles during ultra-sonic treatment on polysaccharide yield were investigated by single-factor test in our research. The optimum conditions for cel-wal destruction of Auricularia auricula mycelia by the synergistic effect of ultrasonic waves and repeated freeze-thaw cycles were ascertained by orthogonal test. [Result] The results of single test indicated the best combination of conditions was as fol ows: freeze time, 30 min; destruction time, 20 min; added water, 15 times; destruction times, 2 times; number of freeze-thaw cycles, 3 cycles. The results of orthogonal test indicated the influencing factors ranked as destruction time 〉 destruction times 〉 freeze time. The best combination of conditions was as fol ows: freeze time, 30 min; destruction time, 25 min; destruc-tion times, 2 times. Under above conditions, the polysaccharide yield reached 57.76 mg/g. The functional formula diet AHP, containing polysaccharides from Auricularia auricular, polyphenolic compounds from Hawthorn (Crataegus) and Pueraria radix, has been recently developed as a dietary intervention against dyslipidemia in our previous study. In the present study, its antioxidant activities and protective effects against proliferation of vascular smooth muscle cells (VSMCs) were investigated. AHP possessed the potent radical-scavenging effects against hydroxyl and superoxide radicals, and also the inhibitory effects against peroxidation of low density lipoprotein induced by Cu2+ in vitro. The protective effects of AHP against proliferation of VSMCs were evaluated through the methodology of serum pharmacology. The serum containing AHP significantly inhibited the proliferation of VSMCs induced by oxidized low density lipoprotein in a time- and dose-dependent manner, and also promoted the nitric oxide production of VSMCs. Our study indicated that this functional formula diet would be a potent alternative as a functional diet to prevent atherosclerosis at early stage. 展开更多

This study explored the therapeutic effects of Auricularia auricula melanin(AAM)on alcoholic liver damage in vitro and in vivo.Human normal liver L02 cells were pre-treated with ethanol and then treated with AAM to explore the therapeutic effect of AAM on ethanol-induced hepatocyte injury.The results show that AAM signifi cantly elevated the cell viability,ameliorated the cell morphology,reduced the ROS and increased the GSH/GSSG of ethanol-pretreated L02 cells.Then,mice were administered with ethanol to induce acute alcoholic liver damage,and administered with AAM to further study the therapeutic effect of AAM on alcoholic liver damage in mice.As a result,AAM reduced the levels of ALT,AST,TG,and MDA,increased the levels of ADH,SOD,and CAT in liver damage mice.The therapeutic effect of AAM may be related to inhibition of CYP2E1 expression and activation of Nrf2 and its downstream antioxidase.The research enriched the bioactivity of AAM and provided some ideas for the development of melanin-related health foods. 展开更多

For evaluating the early quality of the edible fungus, 9 varieties of Auricularia auricular (Au9, CF09, CF05, 29, 916, chang10, chang7, Au.Japanese and 8808) were cultured on the medium consists of potato dextrose agar (PDA) and sawdust to test their mycelium growth rate, endurance to high temperature, resistance to mildew, assimilation of nutriment, and resistance to drought. The result showed that the mycelium of Chang 10, CF09, 29 and Au.Japanese varieties had the eminent characteristics such as short lifespan, stronger assimilation of nutriment, and endurance to high temperature and steady growth. These four varieties are determined as superiority varieties of Auricularia auricular in accordance with the research results. 展开更多

The contents of total flavonoids,polyphenols and triterpenoids in three different alcohol extracts of A. auricula,as well as in the alcohol extracts of polysaccharides and melanin extraction residue of A. auricula were determined. [Results] When90% ethanol was used,the yield of alcohol extract of A. auricula was 10. 7%. In the alcohol extract,the contents of total flavonoids,polyphenols and triterpenoids were 5. 68 mg/100 g,0. 43% and 0. 55%,respectively. When 100% isopropanol was used,the yield of alcohol extract was 10. 1%. In the alcohol extract,the contents of total flavonoids,polyphenols and triterpenoids were 4. 84 mg/100 g,0. 66% and 0. 37%,respectively. When 70% n-butanol was used,the yield of alcohol extract was 5. 4%. In the alcohol extract,the contents of total flavonoids,polyphenols and triterpenoids were 4. 07 mg/100 g,1. 68% and 0. 30%,respectively. [Objectives]This study was conducted to explore the source of substitutive substrate raw materials for cultivation of edible fungi,reduce the cutting and use of woody materials,and realize harmless treatment and resource utilization of byproducts from Chinese medicine production and bamboo processing.[Methods]Traditional Chinese medicine residue and bamboo shavings were used to partially replace hardwood sawdust in the conventional formula,and the growth of Auricularia auricula mycelia in large test tubes under different substrate formulas was studied.[Results]The results showed that the mycelia of A.auricula could grow normally on substrates with byproducts,and the mycelia grew differently with different formulas,and the performance of different strains of the same species was also different.Compared with the conventional formula,the suitable substitution amount of bamboo shavings for A.auricula strains was 10%-30%,and the substitution amount of Chinese medicine residue was 5%-15%.[Conclusions]This study provides reference for the efficient utilization of byproducts and the expansion of raw material sources for production of edible fungi. 展开更多

We examined the rheological and gel properties of Auricularia auricula pulp solution and konjac glucomannan solution at different concentrations(1.2,2.4,3.6,4.8,and 6.0 g/100 mL)as well as A.auricula pulp/konjac glucomannan compound system at different mass ratios(4.8:1.2,3.6:2.4,2.4:3.6,and 1.2:4.8)and discussed the interaction mechanism.The results showed that the apparent viscosity,dynamic viscoelasticity,energy storage modulus(G’),and loss modulus(G”)of the compound system increased with the increase of A.auricula pulp mass ratio.The thixotropic ring decreased with the increase of A.auricula pulp mass ratio,indicating that A.auricula pulp could improve the thixotropy of the compound system.The results of the texture analysis and water retention test showed that konjac glucomannan was the decisive factor for the strength and hardness of A.auricula pulp/konjac glucomannan compound gel,and A.auricula pulp could significantly improve the elasticity,viscosity,and water-holding capacity of the compound system.based on Fourier transform infrared(FTir)spectroscopy and cryo-scanning electron microscopy(cryo-SEM)analyses,we preliminarily concluded that an interaction occurred between A.auricula pulp and konjac glucomannan in the compound system,and a gel network structure was formed. 展开更多

本研究利用酶解法提取黑木耳多糖.以黑木耳粗多糖提取率为指标,并通过单因素实验和响应面试验对黑木耳粗多糖的提取工艺条件进行优化;通过DEAE纤维素柱分离出两种多糖组分,分别命名为AAP-1和AAP-2,并对其总糖、蛋白质、糖醛酸及总酚含... 本研究利用酶解法提取黑木耳多糖.以黑木耳粗多糖提取率为指标,并通过单因素实验和响应面试验对黑木耳粗多糖的提取工艺条件进行优化;通过DEAE纤维素柱分离出两种多糖组分,分别命名为AAP-1和AAP-2,并对其总糖、蛋白质、糖醛酸及总酚含量进行测定;通过DPPH自由基、OH自由基、O2-自由基清除率比较AAP-1和AAP-2的抗氧化能力,通过α-淀粉酶和α-葡萄糖苷酶抑制率比较AAP-1和AAP-2的降糖能力.结果表明:黑木耳多糖的最佳提取条件为:酶解pH=5、酶解温度50℃、酶解时间90 min、酶浓度40 mg/g.与粗多糖相比,洗脱后的AAP-1和AAP-2总糖含量明显提高,蛋白质、糖醛酸及总酚等杂质均减少.其中0.8 mg/mL的AAP-1和AAP-2对于DPPH自由基清除率分别为49.05%和62.69%,对于OH自由基清除率分别为34.04%和45.02%、对于O2-自由基清除率分别为38.62%和39.98%.同时,1.0 mg/mL的AAP-1和AAP-2对于α-葡萄糖苷酶抑制率分别为72.24%和87.10%,对于α-淀粉酶抑制率分别为80.51%和89.23%.研究结果表明,AAP-1和AAP-2均具有一定的抗氧化及降糖能力,同时,AAP-2的体外活性较高,为进一步开发木耳多糖提供理论依据. 展开更多

长期高脂饮食会导致机体脂肪堆积、血脂异常、肠道菌群紊乱,增加高血脂等慢性疾病发病率。该文旨在研究菊粉复配黑木耳多糖对高脂饮食小鼠血清指标、肝脏脂肪堆积、肠道菌群的影响。首先评价了不同质量比例(5∶0、4∶1、3∶2、2∶3、1∶... 长期高脂饮食会导致机体脂肪堆积、血脂异常、肠道菌群紊乱,增加高血脂等慢性疾病发病率。该文旨在研究菊粉复配黑木耳多糖对高脂饮食小鼠血清指标、肝脏脂肪堆积、肠道菌群的影响。首先评价了不同质量比例(5∶0、4∶1、3∶2、2∶3、1∶4、0∶5)菊粉复配黑木耳多糖体外降血脂活性,并将小鼠分为正常对照组(饲喂普通饲料),高脂对照组(饲喂高脂饲料),干预组饲喂添加不同比例菊粉复配黑木耳多糖的高脂饲料,干预8周后,研究高脂饮食对小鼠血清、肝脏、肠道菌群的影响。结果表明:在菊粉复配黑木耳多糖(2∶3)条件下,对甘氨胆酸钠、牛磺胆酸钠的结合率最高。不同比例菊粉复配黑木耳多糖对实验小鼠体重、肝体比显著降低,血清总胆固醇、总甘油三酯、低密度脂蛋白胆固醇含量均极显著下降,高密度脂蛋白胆固醇水平显著升高。小鼠肝脏中脂肪滴数量和体积均明显减少,均比单一菊粉、黑木耳多糖干预效果明显。菊粉复配黑木耳多糖干预高脂饮食小鼠,提高了肠道中乳酸杆菌属、杆菌属、副拟杆菌属、拟杆菌属等产生短链脂肪酸菌属相对丰度,降低了脱硫弧菌属、螺杆菌属、扭链瘤胃球菌、Colidextribacter相对丰度,改善了高脂饮食对肠道菌群的影响。该研究表明菊粉复配黑木耳多糖对高脂饮食小鼠具有调控血脂及改善肠道菌群的作用,为预防高血脂及功能性产品开发提供新思路。 展开更多

探明甘肃省康县代表性食用菌种类--香菇及黑木耳在不同栽培方式下的品质差异,为甘肃省康县食用菌主流栽培方式的选择及代料基质的优化提供参考。以甘肃省康县代料和段木2种主要栽培方式的香菇和黑木耳为试验材料,测定并对比其子实体大... 探明甘肃省康县代表性食用菌种类--香菇及黑木耳在不同栽培方式下的品质差异,为甘肃省康县食用菌主流栽培方式的选择及代料基质的优化提供参考。以甘肃省康县代料和段木2种主要栽培方式的香菇和黑木耳为试验材料,测定并对比其子实体大小、质构特性等感官指标,以及脂肪、蛋白质、粗多糖等营养物质含量和矿质元素含量差异。结果表明,甘肃省康县代料栽培香菇品质优于段木栽培香菇,表现为菇体的大小形状规则均匀美观,主要营养成分蛋白质、膳食纤维、粗多糖含量和矿质元素钙、钾、磷含量均高于段木栽培香菇;其中,粗多糖含量为22.6 g/kg,较段木栽培香菇显著增加85.25%;钙含量20.1 mg/kg、磷含量1.68 g/kg,分别较段木栽培香菇显著高91.43%、52.73%。代料栽培黑木耳总体品质优于段木栽培黑木耳,表现为朵形大小规则均匀美观,质构特性表现出的商品性和适口性较好,主要营养成分脂肪、蛋白质、粗多糖、黑色素、总氨基酸含量和矿质元素钙、磷含量均不同程度高于段木栽培黑木耳;其中,脂肪、蛋白质、氨基酸含量分别为2.0、14.9、10.1 g/kg,分别较段木栽培黑木耳显著高100%、77.38%、741.67%。综上所述,认为代料栽培更适合甘肃康县香菇、黑木耳食用菌产业发展。 展开更多

Lentinus edodes and Auricularia auricula are the first and second largest edible fungi in China with high nutritional and medicinal values.Research about phytochemicals of L.edodes and A.auricula mainly focuses on polysaccharides,other phytochemicals are not fully studied.Phytochemicals of L.edodes and A.auricula were investigated by ultra-high performance liquid chromatography tandem hybrid quadrupole-orbitrap mass spectrometry(UPLC-Q-Exactive-Orbitrap MS).Twenty-three compounds were identified in L.edodes,mainly including amino acids and analogs,flavonoids,nucleosides and analogs,phenols and steroids.Twenty-nine compounds were identified in A.auricula,mainly including amino acids and analogs,nucleosides and analogs,flavonoids and phenols.There are 9 common ingredients of L.edodes and A.auricula,including 4 amino acids and analogs,2 flavonoids,2 nucleosides and analogs and 1 phenol. 展开更多

Auricularia auricula polysaccharides(AHP)have potential immunomodulatory activities.However,the relationship between polysaccharides,intestinal microbiota,and immune activity remains unclear.Therefore,to explore this relationship,we performed immunosuppressive donor,fecal microbiota transplantation(FMT),and pseudo-sterile mouse validation experiments.Our results showed that AHP enhanced immunomodulatory activity,as evidenced by the alleviation of cyclophosphamide(Cy)-induced body weight loss,spleen and thymus atrophy,and splenic and colonic lesions.Additionally,AHP effectively regulated antioxidant enzyme activities(SOD,T-AOC,CAT,GSH-Px),lipid peroxidation product content(MDA),levels of immune factors(TNF-α,IFN-γ,IL-2,IL-4,IL-6),immunoglobulins(sIgA),and the expression of tight junction proteins(Muc2,ZO-1,Occuldin,and Claudin-1)in the colon of immunosuppressed mice.FMT results demonstrated that FAHP displayed superior immunomodulatory ability and could further affect the abundance of Dehydroepiandrosterone,Prostaglandin E2,Cholesterol and Pregnenolone by increasing the abundance of unclassified_Muribaculaceae,Desulfovibrio,Alloprevotella,and unclassified_Oscillospiraceae and decreasing that of Staphylococcus,Bacteroides,and Corynebacterium.Pseudo-sterilization experiments showed that AHP was unable to increase antioxidant enzyme activity,reduce lipid peroxidation content,or promote the release of immune factors.In brief,AHP did not alleviate Cy-induced immunosuppression after microbiota elimination.In summary,AHP exerted a protective effect against Cy-induced immunosuppression and intestinal damage by modulating intestinal microbiota and its metabolites.These findings provide new insights into the mechanisms by which AHP,as a functional food or drug,promotes immune response regulation and maintains intestinal health. 展开更多

In this paper,using to the comprehensive scoring method,pressure shear assisted extraction was used to prepare polysaccharide from Auricularia auricula(PS-AAP)by orthogonal test to optimize the extraction.The highest comprehensive score(90.70)was obtained under 1:70 g/mL solid-liquid ratio,110°C reaction temperature,60 min reaction time and 1.0 MPa reaction pressure.Compared to AAP by water bath(WB-AAP),PS-AAP led to a higher extraction efficiency and decreased the relative molecular weight.In addition,infrared spectrum(FT-IR)analysis showed that pressure shear assisted did not change the polysaccharide structure.Scanning electron microscope(SEM)showed that PS-AAP was smaller and more broken by pressure shear assisted.Furthermore,the ratio of glucose to mannose in PS-AAP was relatively higher than WB-AAP,which also proved that PS-AAP had certain antioxidant and hypoglycemic activities.Compared with WB-AAP,the inhibitionα-glucosidase andα-amylase activity of PS-AAP increased by 11.37%and 24.43%respectively.These results revealed that PS-AAP has the poten-tial value of being a antioxidant and hypoglycemic agent. 展开更多

针对目前我国木耳菌包生产机械自动化程度低、生产效率低、劳动强度大、产出菌包品质参差不齐等问题,结合普遍应用的木耳袋料栽培工艺,研制了一种全自动木耳菌包装袋窝口插棒一体机,实现木耳菌包制作所必要的套袋、灌料、倒浮料、窝口... 针对目前我国木耳菌包生产机械自动化程度低、生产效率低、劳动强度大、产出菌包品质参差不齐等问题,结合普遍应用的木耳袋料栽培工艺,研制了一种全自动木耳菌包装袋窝口插棒一体机,实现木耳菌包制作所必要的套袋、灌料、倒浮料、窝口和插棒等工序的全程自动化作业。介绍了其整机结构与工作原理;选取生产率、密度为试验指标,以绞龙转速、绞龙螺距、绞龙直径为试验因素,进行了中心复合表面组合试验,利用Design-Expert8.0.6软件对试验数据进行分析处理,得到了可信的回归模型和响应曲面,分析了各试验因素及其交互作用对试验指标的影响规律,得到了最优参数组合。验证试验测得,在绞龙转速为1314 r/min、绞龙螺距为65 mm、绞龙直径为86 mm参数条件下,生产率为1215袋/h、密度为649 kg/m3,表明全自动木耳菌包装袋窝口插棒一体机能够有效提高生产效率、保证菌包品质,极大降低了劳动强度与用工成本,为全自动木耳菌包生产机械的进一步研究提供了依据。 展开更多
