Ameloblastin(AMBN, also named "amelin" or "sheathlin") is the second most abundant enamel matrix protein known to have a key role in amelogenesis. Amelogenesis imperfecta(AI [MIM: 104500]) refers to a genetically and phenotypically heterogeneous group of conditions characterized by inherited developmental enamel defects. The hereditary dentin disorders comprise a variety of autosomal-dominant genetic symptoms characterized by abnormal dentin structure affecting either the primary or both the primary and secondary teeth. The vital role of Ambn in amelogenesis has been confirmed experimentally using mouse models. Only two cases have been reported of mutations of AMBN associated with non-syndromic human AI. However, no AMBN missense mutations have been reported to be associated with both human AI and dentin disorders. We recruited one kindred with autosomal-dominant amelogenesis imperfecta(ADAI) and dentinogenesis imperfecta/dysplasia characterized by generalized severe enamel and dentin defects. Whole exome sequencing of the proband identified a novel heterozygous C-T point mutation at nucleotide position 1069 of the AMBN gene, causing a Pro to Ser mutation at the conserved amino acid position 357 of the protein. Exfoliated third molar teeth from the affected family members were found to have enamel and dentin of lower mineral density than control teeth, with thinner and easily fractured enamel, short and thick roots, and pulp obliteration. This study demonstrates, for the first time, that an AMBN missense mutation causes non-syndromic human AI and dentin disorders.展开更多

Nogo proteins, myelin-as- sociated glycoprotein (MAG), oligodendrocyte myelin glycoprotein (OMgp) and B lymphocyte stimulator (BLyS), are 4 inhibitors that commonly interact with the neuronal receptor, Nogo receptor-1 (NgR1), lead- ing to inhibition of axonal growth. Here, we demon- strate that lateral olfactory tract usher substance (LOTUS) binds to NgR1 and blocks the binding of all four ligands to NgR1, resulting in the suppression of axonal growth inhibition induced by these NgR1 li- gands. LOTUS allows neurons to overcome NgRl-me- diated axonal growth inhibition, raising the possibility that LOTUS may be useful in future therapeutic ap- proaches as an endogenous potent inhibitor of NgR1 for promoting neuronal regeneration.展开更多

METHODS: During a 2.5 year period, from September 2003 to March 2006, 12 patients with 13 episodes of SBP were studied. In all cases at the time of admission serum albumin and glucose, urinalysis and urine culture was performed. Analysis [white blood cell (WBC) count with differential, albumin, glucose], gram stain, culture by BACTEC method and antibiogram was done on ascitic fluids. Abdominal paracentesis was repeated after 48 h of antibiotic therapy for bacteriologic assay. The patients were followed for at least three months in a gastroenterology clinic. RESULTS: There were 7 girls (58%) and 5 boys (42%) with a median age of 5.2 years (range, 6 mo to 16 years). All cases had positive ascitic fluid culture. Gram stain was positive in 5 (38.5%) of them. The isolated organisms were S. pneumoniae in 5 (38.5%), E. coli in 2 (15.3%), S. viridans in 2 (15.3%), and K. pneumoniae, H. influenza, Enterococci, and nontypable Streptococcus each in one (7.7%). All of them except Enterococci were sensitive to ciprofloxacin and ceftriaxone. All ascitic fluid cultures were negative after 48 h of antibiotic therapy. CONCLUSION: S. pneumoniae is the most common cause of SBP in the pediatric age group and we recommend a third generation cephalosporine (e.g., Ceftriaxlone or Cefotaxime) for empirical therapy in children with SBP.展开更多

On the basis of results of clinical investigations of affected members of this family disease was diagnosed as Usher syndrome(USH). To identify the locus responsible for the Usher phenotype in this family, genomic DNA from blood sample of each individual was genotyped using microsatellite Short Tandem Repeat(STR) markers for the known Usher syndrome loci. Then direct sequencing was performed to find out disease associated mutations in the candidate gene.· RESULTS: By genetic linkage analysis, the USH phenotype of this family was mapped to PCDH15 locus on chromosome 10q21.1. Three different point mutations in exon 11 of PCDH15 were identified and one of them,c.1304AC was found to be segregating with the disease phenotype in Pakistani family with Usher phenotype. This, c.1304 A C transversion mutation predicts an amino-acid substitution of aspartic acid with an alanine at residue number 435(p.D435A) of its protein product. Moreover, in silico analysis revealed conservation of aspartic acid at position 435 and predicated this change as pathogenic.·CONCLUSION:Theidentificationofc.1304ACpathogenic mutation in PCDH15 gene and its association with Usher syndrome in a consanguineous Pakistani family is thefirst example of a missense mutation of PCDH15 causing USH1 phenotype. In previous reports, it was hypothesized that severe mutations such as truncated protein of PCDH15 led to the Usher I phenotype and that missense variants are mainly responsible for non-syndromic hearing impairment.展开更多

Economic activities including agriculture, mining, and infra-structure improvement to meet basic human needs continuously degrade the natural and forest resources. The rate of deforestation in Ghana is alarming due to over-reliance on forest resources by forest-dependent communities. Perceived causes of deforestation differ from individuals and groups perspec-tive. This depends on factors including environmental knowledge, education level, market demand and socio-economic activities. Simple random sampling and key informant interview with the aid of semi-structured questionnaire was used to elicit information from the indigenes of Sissala West District to determine their perceptions of causes of deforestation. The study indicated that poverty, high illiteracy, population growth and lack of alternative source of livelihood were the indirect causes which trigger livelihood economic activities such as farming, charcoal burning, wood logging and hunting leading to degradation of the ecosystem. It was also realized that majority of the indigenes are uneducated and this contributes to their unawareness of rate of de-forestation. Recommendations suggested to address challenges were enforce-ment of bye-laws and stringent government environmental policies to deter people from degrading the forest. Education, agroforestry, afforestation, and provision of alternative livelihood were also good interventions suggested.展开更多

Actinomyces israelii is usually responsible for the infections and causes chronic suppurative and granulomatous infections. The most common disease form is cervicofascial infection. Liver is involved in 5% of all actinomycosis infections. Liver involvement secondary to any primary infection site in the abdomen frequently occurs. In 15% of abdominal infections the liver is also involved. However, in very rare cases, a primary focus cannot be found, and these cases are called primary or isolated hepatic actinomycosis (IHA). IHA which is frequently seen as a solitary abscess can be confused with a malignancy both clinically and radiologically.展开更多

摘要Staphylococcus aureus(S.aureus)is the third most common pathogen causing 10.6%of bacterial foodborne illnesses in China in 2021[1].Heat-stable Staphylococcal Enterotoxins(SEs)produced by S.aureus are the main contributors to staphylococcal food poisoning(SFP),causing vomiting, diarrhea, abdominal pain, headache, muscle cramps, and other acute gastroenteritis symptoms.More than 25 SEs and staphylococcal enterotoxin-like toxins(SE/s)have been described and which together comprise a superfamily of pyrogenic toxin superantigens(SAgs)[2].
摘要Against the backdrop of the accelerating global clean energy transition and the rapid increase in renewable energy source(RES)penetration, temporal imbalances between supply and demand in power systems are becoming increasingly prominent.Consequently, the phenomenon of negative electricity prices has evolved from sporadic occurrences into an increasingly prevalent market signal. This paper focuses on case studies of negative electricity prices in major international electricity markets(the Netherlands,Germany,Australia)and selected provincial markets in China(Shandong,Zhejiang). It systematically analyzes their underlying causal mechanisms, assesses their multidimensional impacts, and comparatively examines the mitigation strategies adopted across these diverse markets. The study reveals that the core driver of negative prices is the concentrated high generation feed-in from RES during low-load periods. This is concurrently influenced by multiple factors, including insufficient system flexibility,inter-zonal transmission congestion, and constraints imposed by specific market rules such as subsidies and bidding strategies.Major international electricity markets are widely exploring mitigation measures such as optimizing market clearing mechanisms, incentivizing energy storage and demand-side response, and adjusting subsidy policies. In contrast, the occurrence of negative prices in China’s electricity markets, while sharing common drivers, also exhibits complex correlations with domestic factors, notably medium-and long-term contracts and market power dynamics.Consequently, formulating and implementing differentiated and systematic mitigation strategies, drawing upon international experience while being tailored to China’s specific national context,is crucial for the long-term sustainable development of its electricity markets.

摘要This paper proposes a Bayesian Network-based framework for risk assessment and probability estimation of vessel-platform allision accidents, using a novel technique that derives probabilities from incidental data.A dataset of 557 allision incidents collected from multiple open source agencies is analysed to identify causation patterns.Basic causes could only be determined for 375 incidents, with supply vessels involved in 61%of cases.Statistical analysis revealed that vessel type and the month of occurrences are significantly associated, and most incidents arose during cargo transfer operations.Fixed installation accounted for the majority of allisions with moving vessels, and human error emerged as the leading contributor(30%).Building on these insights,a Bayesian Network model is developed incorporating 42 identified causes, three causal factors and four consequence levels.Using a recent probabilistic approach, probabilities of basic causes are derived from annual allision occurrence rates. The BN model is then applied to predict annual allision probabilities and to conduct sensitivity analyses. Results show that weather-related causes and misalignment errors exert the strongest influences on accident probabilities. The methodology is transparent and holistic in providing better discernment of the causation probability of allision accidents.
摘要Echinococcus is a zoonotic parasite and 1 of 17 neglected tropical diseases with a worldwide distribution. The World Health Organization(WHO)targeted for control or elimination by 2050[1,2].Two main species of Echinococcus infect humans:Echinococcus granulosus, causing cystic echinococcosis(CE),and Echinococcus multilocularis(EM),causing alveolar echinococcosis(AE)[3].AE is much rarer but far more severe than CE and ranks as one of the most dangerous helminthic zoonoses in the world[4,5].Humans are rare aberrant intermediate hosts and typically become infected through the ingestion of EM eggs shed in the feces of definitive hosts[6].After an incubation period of many years, humans may develop AE[7].While slow-growing,AE is a devastating clinical condition characterized by silent progression and infiltrative proliferation of the parasite, mimicking a malignancy[7].Without appropriate treatment,AE has a death rate of more than 90%within 10 years of diagnosis[5].For this reason,AE must be considered in the differential diagnosis of patients presenting with a hepatic mass or malignancy.
摘要The clinical picture of actinomycosis was first described in 1878.Actinomycosis agents are found in the natural flora of the oral cavity, upper gastrointestinal system and female genital systems. IHA which is frequently seen as a solitary abscess can be confused with a malignancy both clinically and radiologically.

摘要Guangdong Province experienced heavy to extremely heavy rainstorms during Super Typhoon Saola(2023).Areas such as Yunfu, which are typically drier, experienced anomalously extreme rainfall during this event. We analyzed the convective structure and microphysical characteristics of the typhoon using dual-polarization radar, disdrometer, and ERA5 reanalysis data. The results show that following Saola’s second landfall, convection redeveloped on the northern side, which was characterized by rich mid-level ice particles and low rain mixing ratios. This resulted in a phase difference in the precipitation particles along Saola’s path.Stations that recorded heavy rain were categorized into four types based on raindrop spectral, spatial, and temporal variables by employing the K-means clustering algorithm.Specifically, the particles observed at Doumen(DM)were classified into a different category from those observed at Yangchun(YC)and Luoding(LD).The microphysical characteristics of raindrop particles evolve under the influence of vertical motion. At DM,the convective structure exhibited a single center of vertical motion with fewer high-altitude ice particles, resulting in a uniform oceanic raindrop type.By contrast,YC was sequentially influenced by two vertically separated centers of vertical motion in the upper and lower atmospheres. These centers alternately dominated the ice-phase and warm-rain processes, resulting in a complex raindrop evolution that transitioned from an intermediate state to a continental type before rapidly reverting to an oceanic type.At LD,the structure showed more pronounced baroclinicity with a higher concentration of water vapor in lower levels, with raindrops tending towards an intermediate continental type. The evolution of vertical motion was driven by changes in environmental conditions. The addition of cold air complicated raindrop evolution in western Guangdong Province compared with that in the Pearl River Estuary. The upper-level vertical-motion center, induced by the uplifting effect of the frontal zone associated with cold air, promoted the development of ice-phase processes and an increase in raindrop size.Subsequently, the lower-level upward-motion center became dominant, which enhanced precipitation concentration.A unique circulation pattern created by the interaction of twin typhoons with each other and with cold air intensified the low-level easterly winds and moisture influx, contributing to extreme rainfall in a typically drier region.Orographic convergence and uplift also played a role in the microphysical evolution of raindrops.

摘要Brachinite is a group of primitive achondrites that enables investigating the evolution of asteroids not fully diff erentiated in the early stage of the solar system.Kumtag 061 is a new meteorite sample collected on October 27,2019,in Kumtag Desert,Xinjiang Province,China. The oxygen isotope composition(δ18O=5.086‰,δ17O=2.396‰,Δ’17O=-0.298‰)and petrologic and mineralogic analysis suggest Kumtag 061 is a heavy-impacted brachinite(S4-S5).The geochemical composition suggests Kumtag 061 represents a partial melting residue of the brachinite parent body.Based on the noble gas composition, the cosmic ray exposure age of Kumtag 061 is 60.9±9.0 Ma.Combined with the gas retention ages, they indicate a(series of)thermal events on the parent body of brachinites before Kumtag 061 was ejected into space.

摘要Human reproduction requires the fertilization of a mature oocyte with a sperm to form a high-quality embryo.Oogenesis, folliculogenesis, and the activities of the endocrine system play essential roles in female fertility, and disturbances in these processes can result in female infertility, which has become an urgent public health issue worldwide.Genetic studies have identified multiple variants in key genes underlying these processes in infertile females, and these patients are mainly diagnosed with disorders of sex development, premature ovarian insufficiency, congenital hypogonadotropic hypogonadism, central precocious puberty, resistant ovary syndrome, oocyte maturation arrest, fertilization failure, and early embryonic defects.Notably, the known genes account for about 13.2%cases of oocyte and embryo defects(479/3627)and 18.7%cases of premature ovarian insufficiency(193/1030).Here,we review the critical events in female reproduction and highlight the single gene variants with Mendelian inheritance patterns that are responsible for female infertility.

摘要Primary liver cancer is the fourth most common malignancy and the second leading cause of cancer-related death in China[1].Most patients are diagnosed at an advanced stage, which renders them ineligible for curative surgical resection. Thus, improving the resectability rate of hepatocellular carcinoma(HCC)remains a critical challenge in liver cancer research.Selective internal radiotherapy(SIRT)is currently utilized to downstage unresectable HCC,with objective response rates ranging from 85% to 93%[2].
