Efficient H2O2 photosynthesis through linker engineering of benzotrithiophene-based covalent organic frameworks 认领 引用

The photogenerated carrier separation efficiency and material wettability are of critical importance for aqueous-phase photocatalytic reactions,achieving both simultaneously poses a significant challenge owing to the inherent interdependencies and trade-offs involved.In this work,a series of isoreticular benzotrithiophene-based covalent organic frameworks(COFs)were successfully synthesized by incorporating diverse hydrophobic and hydrophilic functional groups(-OH,-F,-H)onto their skeletons,thereby modulating their characteristic charge separation and transport as well as their wettability,and systematically studied their photocatalytic H2O2 production performance in O2-saturated water under visible-light irradiation.Remarkably,the synthesized hydrophilic BTT-BD-OH-COF demonstrates the highest H2O2 production rate of 6105μmol g-1 h-1 in the absence of any sacrificial agent in pure water,attributed to its extended light absorption range,improved hydrophilicity,and enhanced photo-induced charge separation and transport efficiency.Combined experimental results and the density functional theory calculations elucidate the reaction mechanism,revealing the overall H2O2 photosynthesis via both oxygen reduction reaction and water oxidation reaction dual pathways.This study demonstrates that functional-group-mediated linker engineering is a powerful approach for significantly enhancing the efficiency of COF-based photocatalysts. 展开更多

Base editing enables efficient nucleotide conversions without inducing DNA double-strand breaks(DSBs)or requiring exogenous donor DNA templates.However,its broader editing window often causes bystander editing,increasing the risk of unintended mutations.In this study,we find that linker length significantly influences the editing window,and base editors with a 7-amino-acid linker reduce bystander editing by an average of 54.4%across 13 endogenous genomic sites in both rice and human cell lines.We further develop an optimized strategy by modulating the linker length between various deaminases and Cas9 nickases,which effectively reduces bystander editing across multiple applications,including functional studies,precise crop breeding,and correction of pathogenic variants.Our work reveals that shortening the linker enhances the specificity of base editing,addressing a key safety concern for its agricultural and therapeutic applications. 展开更多

Dentin hypersensitivity(DH)originates from collagen exposure and dentinal tubules(DTs)patency caused by mechanical abrasion or demineralization.For an effective long-term therapeutic desensitizing agent,the ability to achieve deep tubular occlusion,stable remineralization,and satisfactory biocompatibility simultaneously is essential.In this study,we designed a novel α-hairpin peptide,TKH,engineered by incorporating a flexible GKG linker into an α-helical template,and provide an analysis of its liquid-liquid phase separation(LLPS)and self-assembly properties.TKH effectively stabilized amorphous calcium phosphate and facilitated intrafibrillar mineralization within collagen fibrils.In vitro and in vivo remineralization studies combined with molecular dynamics(MD)simulations demonstrated that TKH promoted the peptide self-assembly through LLPS,hydroxyapatite(HA)adsorption,and amorphous calcium phosphate(ACP)stabilization,ultimately achieving intrafibrillar and extrafibrillar mineralization to seal DTs,alongside excellent biosafety.These findings highlight the potential of TKH as a promising peptide-based biomaterial for dentin remineralization and the treatment of dentin hypersensitivity. 展开更多

Nucleosomes play a vital role in chromatin organization and gene regulation,acting as key hubs that inter-act with various chromatin-associated factors through diverse binding mechanisms.Recent research has highlighted the prevalence of mutations in linker histones across different types of cancer,emphasizing their critical involvement in cancer progression.These cancer-associated mutations in linker histones have been shown to disrupt nucleosome stacking and the formation of higher-order chromatin structures,which in turn significantly affect epigenetic regulatory processes.In this review,we provide a comprehensive analysis of how cancer-associated linker histone mutations alter their physicochemical properties,influencing their binding to nucleosomes,and overall chromatin architecture.Additionally,we explore the significant impact of mutations near post-translational modification sites,which further modulate chromatin dynamics and regulatory functions,offering insights into their role in oncogenesis and potential therapeutic targets. 展开更多

[目的]构建木聚糖酶(XynB)与甘露聚糖酶(ManA)的融合酶基因,使其能在哺乳动物表达系统中表达并分泌兼有木聚糖酶和甘露聚糖酶活性的双功能融合酶。[方法]利用基因融合技术(SOE—PCR),把11条Linker融合到木聚糖酶(XynB)和甘露聚糖酶(ManA)基因间,构建真核表达载体,经转染猪肾细胞(pKl5)收集细胞培养液,用DNS法测定其酶活并进行酶学分析。[结果]经表达分析12条不同Linker构建的融合酶与亲本酶酶活,发现用Linkera3、pS3和具有“自我剪切”能力T2A构建的融合酶在木聚糖酶和甘露聚糖酶活性都显著高于两亲本酶。融合酶XynB-a3-ManA的木聚糖酶和甘露聚糖酶酶活比亲本酶XynB和ManA分别提高了54.06%和104.40%;该融合酶在酸性环境3.0—7.0起作用,对pH3.08.0具有一定的耐受力。[结论]首次获得可在哺乳动物系统表达分泌的增强型双功能XynB—ManA融合酶。 展开更多

How ATP binding initiates the docking process of kinesin's neck linker is a key question in understanding kinesin mechanisms. By exploiting a molecular dynamics method, we investigate the initial conformation of kinesin's neck linker in its docking process. We find that, in the initial conformation, the neck linker has interactions with /30 and forms a 'cover-neck bundle' structure with/30. From this initial structure, the formation of extra turns and the docking of the cover- neck bundle structure can be achieved. The motor head provides a forward force on the initial cover-neck bundle structure through ATP-induced rotation. This force, together with the hydrophobic interaction of ILE327 with the hydrophobic pocket on the motor head, drives the formation of the extra turn and initiates the neck linker docking process. Based on these findings, a pathway from ATP binding-induced motor head rotation to neck linker docking is proposed. 展开更多

Two new different metal-organic frameworks(MOFs)[Ba(L1)(H2O)2]n·nH2O(MOF 1)and[Ba(L2)(-n2 O)2]n·0.5 nDMF·0.5 nH2O(MOF 2)were yielded by the assembly of oxygen-friendly Ba(Ⅱ)ions and two similar linkers,namely 2-(imidazol-1-yl)terephthalic acid(H2L1)and 2-(1 H-1,2,4-triazol-1-yl)terephthalic acid(H2L2).Single-crystal X-ray diffractions(XRD)indicate that MOF1 is a new three-dimensional(3 D)stacking dense network formed by the one-dimensional(1 D)rod-shaped chains and L1 linkers,whereas MOF 2 presents a 3 D nanotube porous framework with cylindrical tunnels based on the 1 D loop chains as the secondary building units(SBUs)by replacing the imidazole group in H2L1 with the triazole group in H2L2.As a result,MOF 2 has a higher density of active sites and Lewis acid sites in the porous surface of nanotube than MOF 1.Thereby,the CO2capture and separation capacity of MOF 2 is great higher than that of CH4at298 K. 展开更多

In this work,semirigid linkers of the alkyl-thiophene-alkyl structure are developed to construct double-cable polymers.Three alkyl units,propyl(C3H6),hexyl(C6H12),and dodecyl(C12H24),are applied as semirigid linkers,yielding three double-cable polymers:PBC6-T,PBC12-T,and PBC24-T,respectively.PBC12-T which uses C6H12-thiophene-C6H12 linkers is found to exhibit the best device efficiency of 5.56%,while PBC6-T and PBC24-T with shorter or longer linkers yield device efficiencies of only 2.65%and 1.09%in single-component organic solar cells(SCOSCs).Further studies reveal that PBC12-T exhibits higher crystallinity and improved charge transport,resulting in better efficiencies.Our work provides an approach to construct double-cable conjugated polymers with long alkyl linkers,and it shows the importance of the linker length for the photovoltaic performance of SCOSCs. 展开更多

Carbon super-heterostructures with high nitrogen contents from the covalent hybrid precursors of covalent triazine frameworks(CTFs)and zeolitic imidazolic frameworks(ZIFs)are scarcely explored because of CTF's ordered structure and toxic superacid that dissolves or destabilizes the metal nodes.To solve this problem,herein,we report a straightforward two-step pathway for the covalent hybridization of disordered CTF(d–CTF)–ZIF composites via preincorporation of an imidazole(IM)linker into ordered CTFs,followed by the imidazole-site-specific covalent growth of ZIFs.Direct carbonization of these synthesized d–CTF−IM−ZIF hybrids results in unique hollow carbon super-heterostructures with ultrahigh nitrogen content(>18.6%),high specific surface area(1663m2g−1),and beneficial trace metal(Co/Zn NPs)contents for promoting the redox pseudocapacitance.As proof of concept,the obtained carbon super-heterostructure(Co–Zn–NCSNH–800)is used as a positive electrode in an asymmetric supercapacitor,demonstrating a remarkable energy density of 61Wh kg−1and extraordinary cyclic stability of 97%retention after 30,000 cycles at the cell level.Our presynthetic modifications of CTF and their covalent hybridization with ZIF crystals pave the way toward new design strategies for synthesizing functional porous carbon materials for promising energy applications. 展开更多

Objective To design and develop a novel, sensitive and versatile method for in vivo foot printing and studies of DNA damage, such as DNA adducts and strand breaks. Methods Starting with mammalian genomic DNA, single-stranded products were made by repeated primer extension, these products were ligated to a double-stranded linker having a randomized 3 overhang, and used for PCR. DNA breaks in p53 gene produced by restriction endonuclease AfaI were detected by using this new method followed by Southern hybridization with DIG-labeled probe. Results This randomized terminal linker-dependent PCR (RDPCR) method could generate band signals many-fold stronger than conventional ligation-mediated PCR (LMPCR), and it was more rapid, convenient and accurate than the terminal transferase-dependent PCR (TDPCR). Conclusion DNA strand breakage can be detected sensitively in the gene level by RDPCR. Any lesion that blocks primer extension should be detectable. 展开更多

Electro-reduction of carbon dioxide(ERCO2) is considered an effective method to alleviate the greenhouse effect and produce value-added chemicals.Achieving the dominant selectivity of Zn-based catalysts for formate remains a challenge.In this article,the ZnIn-E12 catalyst is successfully prepared by solvent assisted ligand exchange(SALE) method to convert organic ligands,achieving a Faradaic efficiency of 72.28% for formate at-1.26 V vs.RHE(VRHE),which is 1.42 times higher than the original catalyst.Evidence shows that the successful conversion of organic ligands can transform the catalyst from the original large size polyhedron to cross-linked network of particles with a diameter of about 30 nm.The increased specific surface area can expose more active sites and facilitate the electrocatalytic conversion of CO2 to formate.This work is expected to provide inspiration for the regulation of formate selectivity and catalyst size in Zn-based catalysts. 展开更多

目的:探讨重组毒素IL6D24LinkerPE40融合蛋白及其接头设计的合理性。方法:应用核酸和蛋白质序列分析软件系统GOLDKEY软件,对IL6D24LinkerPE40重组毒素融合蛋白的柔性、抗原性、亲水性、表位等分子生物学特性进行计算... 目的:探讨重组毒素IL6D24LinkerPE40融合蛋白及其接头设计的合理性。方法:应用核酸和蛋白质序列分析软件系统GOLDKEY软件,对IL6D24LinkerPE40重组毒素融合蛋白的柔性、抗原性、亲水性、表位等分子生物学特性进行计算机模拟预测,并做Westernblot分析进行验证。结果:IL6D24LinkerPE40分别保留了IL6D24和PE40的表位特征,没有新的抗原表位出现,而且柔性接头处具有极低的抗原性。Westernblot分析结果表明融合蛋白能分别与IL6和PEA的单克隆抗体发生特异结合,这说明融合蛋白保留了IL6和PE40的空间结构特征。 展开更多

从PUBMED中查找人心脏cTnI和cTnC的cDNA序列,在两者之间加上15个中性氨基酸残基(G4S)3的linker编码序列,并分别构建原核表达质粒pET32a-cTnI-linker-TnC和pET28a-cTnI-linker-TnC,转化入大肠杆菌表达菌株BL21(DE3)中,用异丙基硫代-β-D... 从PUBMED中查找人心脏cTnI和cTnC的cDNA序列,在两者之间加上15个中性氨基酸残基(G4S)3的linker编码序列,并分别构建原核表达质粒pET32a-cTnI-linker-TnC和pET28a-cTnI-linker-TnC,转化入大肠杆菌表达菌株BL21(DE3)中,用异丙基硫代-β-D-半乳糖苷(IPTG)诱导其表达。通过SDS-PAGE电泳和间接ELISA对目的蛋白进行初步鉴定,并优化表达条件以实现目的蛋白的可溶性高表达。用标准抗cTnI单克隆抗体和标准抗cTnC单克隆抗体对目的蛋白进行Western blot鉴定和双抗体夹心ELISA鉴定,同时用八位心肌梗塞患者血清做间接ELISA鉴定。结果表明,cTnI-linker-TnC融合蛋白在表达载体pET32a中实现了可溶性高表达,最佳表达条件为28℃、0.4mmol/L IPTG、诱导表达5h;且融合蛋白具有较好的免疫原性,保存了cTnI和cTnC单体的天然结构,有望成为天然cTnI-TnC复合物的替代物,为下一步制备高特异性的抗cTnI-TnC复合物的单克隆抗体奠定了基础。 展开更多

In this work,we report a novel octa-nuclear uranyl(U8)motif[(UO2)8O4(μ3-OH)2(μ2-OH)2]4+embedded in a uranyl-oxalate coordination polymer(compound 1)based on a U-shaped linker with extra-long xylylene chains for stabilizing the resulting high-nuclear motif through additional cross-linking connectivity.A comparison with dimeric and monomeric uranyl compounds obtained at different pH value from the same hydrothermal system reveals that,solution pH plays a vital role in formation of this octa-nuclear uranyl motif by promoting hydrolysis of uranyl source.Since high similarity of eight uranium centers in this nearly planar U8 motif here,overlapping and broadening of signals in fluorescence,infra-red(IR)and Raman spectra can be found. 展开更多

Self-immolative linkers have been widely used to construct prodrugs to improve their efficacy and safety.In this study,we report the use of phenoxysilyl linker as a self-immolative unit to prepare antibody-drug conjugates(ADCs).Phenoxysily based ADC Ate-PPS-CA4 was prepared and its release was systematically investigated by mass spectrometry.Biological evaluation showed that Ate-PPS-CA4 displayed the ability to target delivery and self-immolative release the active payload CA4 on PD-L1 positive cells MDA-MB-231.As the same with its payload CA4,it could arrest the cell cycle to the G2/M phase and induced changes in cell morphology at the dose of its IC50.The development of this linker with novel drug release mechanisms will expand the methodology to construct ADCs,especially for non-internalizing ADCs by extracellular cleavage. 展开更多

Linkers were assembled on a glass surface based on the hydrolysis and condensation of 3-glycidoxy ̄propyltrimethoxysilane (GPS). After the assembly of GPS, four approaches were tried to open the ending epoxide group of GPS or to further elongate the linkers. The effect of these approaches on DNA in situ synthesis and hybridization was investigated. For the spacing of the synthesis initiation sites, the wettability of the support and the length of the linking group that attaches the initiation site to the surface have direct influences on the yield of coupling reactions and the subsequent hybridization events. X-ray photoelectron spectroscopy (XPS) and mean contact angles of deionized water of the above slides were measured to assess the linker's characteristics in each procedure. It was proved that the glass slides were successfully modified and became excellent supports for the oligonucleotides synthesis. In addition, it proved best for the in situ oligonucleotides synthesis that a glass slide was in turn treated with ethylenediamine, glutaradehyde, ethanolamine and sodium borohydride solution at ambient temperature after silanized with GPS. 展开更多
