The pulse shape discrimination technique plays a pivotal role in neutron field measurements using organic scintillator detectors,and the particle-type labeling accuracy of the pulse waveform dataset has a significant impact on its performance,especially with the growing use of machine learning methods.In this study,a high-accuracy labeling method for pulse waveform datasets based on the time-of-flight(TOF)filtering method,an improved charge comparison method(CCM),and the coincidence measurement method is proposed.The relationship between the experimental parameters and the chance coincidence proportion in the TOF measurement was derived to reduce contamination from chance coincidences at the experimental level.Based on this,an experiment was conducted to obtain raw data using the241AmBe source,and a piled-up identification algorithm based on reference waveform cross-correlation and differential analysis was designed to filter out piled-up pulses.To improve the labeling accuracy,the CCM was optimized,a simple method of selecting the TOF interval for a lower chance coincidence proportion was proposed,and a low-amplitude pulse waveform dataset construction method based on coincidence measurements was developed.To verify these methods,eight pulse waveform datasets were constructed using different combinations of the proposed approaches.Three neural network structures and a corresponding evaluation parameter were designed to test the quality of these datasets.The results showed that the particle identification performance of the CCM was significantly improved after optimization,with the neutron-to-gamma-ray misidentification rate reduced by more than 35%.The proposed accuracy improvement methods reduced ambiguous identification results from these artificial neural networks by more than 50%. 展开更多

Microsatellite markers have become one kind of the most important molecular tools used in various researches. A large number of microsatellite markers are required for the whole genome survey in the fields of molecular ecology,quantitative genetics and genomics. Therefore,it is extremely necessary to select several versatile,low-cost,efficient and time-and labor-saving methods to develop a large panel of microsatellite markers. In this study,we used Zhikong scallop(Chlamys farreri) as the target species to compare the efficiency of the five methods derived from three strategies for microsatellite marker development. The results showed that the strategy of constructing small insert genomic DNA library resulted in poor efficiency,while the microsatellite-enriched strategy highly improved the isolation efficiency. Although the mining public database strategy is time-and cost-saving,it is difficult to obtain a large number of microsatellite markers,mainly due to the limited sequence data of non-model species deposited in public databases. Based on the results in this study,we recommend two methods,microsatellite-enriched library construction method and FIASCO-colony hybridization method,for large-scale microsatellite marker development. Both methods were derived from the microsatellite-enriched strategy. The experimental results obtained from Zhikong scallop also provide the reference for microsatellite marker development in other species with large genomes. 展开更多

Twenty-one typical coupled large samples were chosen from areas within and surrounding nature reserves on the Tibetan Plateau using the large sample comparison method(LSCM).To evaluate the effectiveness of the nature reserves in protecting the ecological environment,the alpine grassland net primary production(NPP) of these coupled samples were compared and the differences between them before and after their establishment as protected areas were analyzed.The results showed that:(1) With respect to the alpine grassland NPP,the ecological and environmental conditions of most nature reserves were more fragile than those of the surrounding areas and also lower than the average values for the Tibetan Plateau.(2) Of the 11 typical nature reserves selected,the positive trend in the NPP for Manzetang was the most significant,whereas there was no obvious trend in Taxkorgan.With the exception of Selincuo,the annual NPP growth rate in the nature reserves covered by alpine meadow and wetland was higher than that in nature reserves consisting of alpine steppe and alpine desert.(3) There were notable findings in 21 typical coupled samples:(a) After the establishment of the nature reserves,the annual rate of increase in the NPP in 76% of samples inside nature reserves and 82% of samples inside national nature reserves was higher than that of the corresponding samples outside nature reserves.(b) The effectiveness of ecological protection of the Mid-Kunlun,Changshagongma,Zoige and Selincuo(Selin Co) nature reserves was significant; the effectiveness of protection was relatively sig-nificant in most parts of the Sanjiangyuan and Qiangtang nature reserves,whereas in south-east Manzetang and north Taxkorgan the protection effectiveness was not obvious.(c) The ecological protection effectiveness was significant in nature reserves consisting of alpine meadow,but was weak in nature reserves covered by alpine steppe.This study also shows that the advantage of large sample comparison method in evaluating regional ecology change.Careful design of the samples used,to ensure comparability between the samples,is crucial to the success of this LSCM. 展开更多

We compare three different methods to extract coherent modes from Doppler backscattering (DBS), which are center of gravity (COG) of the complex amplitude spectrum, spectrum of DBS phase derivative (phase derivative method), and phase spectrum, respectively. These three methods are all feasible to extract coherent modes, for example, geodesic acoustic mode oscillation. However, there are stilI differences between dealing with high frequency modes (several hundred kHz) and low frequency modes, (several kHz) hiding in DBS signal. There is a significant amount of power at low frequencies in the phase spectrum, which can be removed by using the phase derivative method and COG. High frequency modes are clearer by using the COO and the phase derivative method than the phase spectrum. The spectrum of DBS amplitude does not show the coherent modes detected by using COG, phase derivative method and phase spectrum. When two Doppler shifted peaks exist, coherent modes and their harmonics appear in the spectrum of DBS amplitude, which are introduced by the DBS phase. 展开更多

Objectives: Dengue virus (DENV) infection is a mosquito-borne disease that stands out as one of the major public health issues and has a wide-ranging geographical distribution throughout the tropics and subtropics. The general alarming increase in the number of cases over the last two decades can be attributed to an extent by the change in national practices to keeping records and reporting dengue to the Ministries of Health, and WHO. Dengue diagnosis is routinely carried out by detection of dengue virus (DENV) antigen NS1 (Non-structural Antigen 1) and/or anti-DENV IgM/IgG antibodies using enzyme-linked immunosorbent assays (ELISAs) and rapid diagnostic tests (RDTs). This study compared the performance of three RDTs and one ELISA used for dengue diagnosis in southeastern, Nigeria. Design: This study adopted a cross-sectional design that included prospective hospital-based surveillance of cases among febrile participants attending two major health facilities within the southeastern region of Nigeria. In this study, 338 HIV-infected participants from two teaching hospitals in Nigeria’s southeast were systematically tested for Dengue with four methods: NS1 RDT, IgG RDT, IgM RDT, and NS1 ELISA. Their specificities and sensitivities were compared, as well as their level of concordance. Their respective performances were also evaluated using the Receivers Operational curve (ROC). Results: Out of the 338 patients, the dengue prevalence from the four dengue diagnostic methods Dengue virus NS1 ELISA, NS1 RDT, IgM and IgG seropositivity were 8.9%, 0.6%, 5.6%, and 8.0%, respectively. The Dengue IgM RDT test indicated 36.8% sensitivity, 92.8% specificity, the IgG anti-dengue specific test indicated 29.6% sensitivity, 92.9% specificity and the dengue NS1 RDT test indicated 3.3% sensitivity, 99.4% specificity when compared with the Dengue NSI Elisa test method as a reference method. Conclusion: The use of NSI ELISA for DENV diagnosis showed good performance and the RDTs showed, to an extent, reliable results compared with ELISA. However, diagnostic laboratories should be aware of performance variations across tests and the possibilities of cross-reactivity that may affect results. 展开更多

AIMTo describe the development and validation of a novel neuronavigation-based method, which allows the quan-tification of the anatomical features that define anapproach, as well as real-time visualization of the surgicapyramid. METHODSThe method was initially developed with commercially-available hardware for coordinate collection (a digitizerand a frameless navigation system) and software forvolume rendering; dedicated neuronavigation software (ApproachViewer, part of GTx-UHN) was then developed. The accuracy of measurements and the possibility of volumetric rendering of surgical approaches simulated in a phantom were compared among three different methods and commercially-available radiological software. In the anatomy laboratory, ApproachViewer was applied to the comparative quantitative analysis of multiple neurosurgical approaches and was used by many surgeons who were untrained for the research method.RESULTSThe accuracy of ApproachViewer is comparable to com-mercially-available radiological software. In the anatomy laboratory, the method appears versatile. The system can be easily used after brief training. ApproachViewer allows for real-time evaluation and comparison of surgical approaches, as well as post-dissection analyses of collected data. The accuracy of the method depends on the navigation registration: with a 1-2 mm registration error, it is adequate for evaluation and comparison of most neurosurgical approaches.CONCLUSIONThis new research method and software allows semi-automated visualization, quantifcation, and comparison of neurosurgical approaches in the anatomy laboratory. 展开更多

Gas chromatography equipped with an electron capture detector (GC-ECD) has been widely used for measuring atmospheric N2O,but nonlinear response and the influence of atmospheric CO2 have been recognized as defects for quantification.An original GCECD method using N 2 as carrier gas was improved by introducing a small flow rate of CO2 makeup gas into the ECD,which could well remedy the above defects.The N2O signal of the improved method was 4-fold higher than that of the original method and the relative standard deviation was reduced from 〉 1% to 0.31%.N2O concentrations with different CO2 concentrations (172.2×10-6-1722×10-6mol/mol) measured by the improved GC-ECD method were in line with the actual N2O concentrations.However,the N2O concentrations detected by the original method were largely biased with a variation range of-4.5%~7%.The N2O fluxes between an agricultural field and the atmosphere measured by the original method were greatly overestimated in comparison with those measured by the improved method.Good linear correlation (R2=0.9996) between the response of the improved ECD and N2O concentrations (93×10-9-1966×10-9mol/mol) indicated that atmospheric N2O could be accurately quantified via a single standard gas.Atmospheric N2O concentrations comparatively measured by the improved method and a high precision GC-ECD method were in good agreement. 展开更多

该研究系统对比了傅里叶变换显微红外光谱法(μ-FTIR)与热裂解-气相色谱-质谱联用法(Py-GCMS)的核心性能、数据差异及适用场景。以实际空气降尘样品为研究对象,经统一前处理后,通过粒径梯度、基质干扰、老化效应及实际样品验证实验,多... 该研究系统对比了傅里叶变换显微红外光谱法(μ-FTIR)与热裂解-气相色谱-质谱联用法(Py-GCMS)的核心性能、数据差异及适用场景。以实际空气降尘样品为研究对象,经统一前处理后,通过粒径梯度、基质干扰、老化效应及实际样品验证实验,多维度剖析两种方法的检测优劣。结果表明:μ-FTIR可实现微塑料无损检测,精准获取颗粒形貌与聚合物类型,适用于>50μm大粒径微塑料定性分析,但对<20μm超微塑料的检出率仅18.6%,抗基质干扰能力弱;Py-GC-MS不受颗粒粒径、形貌限制,全粒径的检出率超90%,定量精度与稳定性优异,可有效适配复杂老化样品,但无法获取形貌信息、难以区分结构相似聚合物。实际样品检测中,两种方法识别的微塑料优势组分(PE、PP、PET)高度一致,但颗粒丰度与质量浓度仅呈中等相关(R²=0.724),受粒径分布与聚合物密度差异的影响无法直接换算。综上,两种方法优势互补,建立二者标准化联用体系,可实现空气降尘微塑料“形貌-组分-质量”全维度表征,为微塑料检测标准化、污染溯源及风险评价提供技术支撑。 展开更多

[目的]分析水沙变化趋势和原因,探究水沙变化之间的关系曲线,为科学认识水沙变化机制,促进黄河流域高质量发展提供支撑。[方法]基于1956—2023年月尺度径流量、输沙量等数据资料,采用M-K检验、Budyko方程和累积量斜率变化率比较法分析了年输沙量、年径流量变化趋势,诊断突变年份,构建水沙关系曲线,量化水沙变化原因。[结果](1)1956—2023年洮河流域年径流量、年输沙量均呈显著下降趋势。其中,红旗水文站年径流量和年输沙量下降速率最大,分别为-0.248亿m3/a和-46.68万t/a。岷县水文站年径流量和年输沙量下降速率最小,分别为-0.191亿m3/a和-3.38万t/a。(2)1956—2023年洮河流域各水文站年径流量和年输沙量均发生了突变。其中,红旗水文站年径流量和年输沙量的突变时间分别为1987年、2003年。(3)1956—2023年洮河流域各水文站的水沙关系曲线均呈幂函数。(4)气候变化和人类活动对洮河流域径流减少的贡献率分别为42.54%和57.47%,对洮河流域输沙量减少的贡献率分别为8.53%和91.47%。[结论]洮河流域径流量、输沙量均呈显著减少趋势,人类活动对其的影响超过了50%,人类活动是洮河流域减水、减沙的主要影响因子。 展开更多

目的系统性评价罗氏Cobas e801、普门eCL9000、西门子Atellica IM 1600及新产业MAGLUMI X84种化学发光免疫分析平台测定血清铁蛋白(SF)结果的一致性及可比性。方法收集296例临床剩余血清样本,结果覆盖整个检测范围。所有样本在4种分析... 目的系统性评价罗氏Cobas e801、普门eCL9000、西门子Atellica IM 1600及新产业MAGLUMI X84种化学发光免疫分析平台测定血清铁蛋白(SF)结果的一致性及可比性。方法收集296例临床剩余血清样本,结果覆盖整个检测范围。所有样本在4种分析平台上平行检测SF。采用Wilcoxon符号秩检验比较中位数差异;使用Passing-Bablok回归分析系统偏差,并用Bland-Altman分析评估一致性界限;根据各平台参考范围进行结果分类,计算符合率与Kappa值以评价分类一致性。结果4种分析平台检测结果的中位数差异具有统计学意义(均P<0.05),其中西门子Atellica IM1600测定值较其余3种平台系统性偏低。Passing-Bablok回归显示,罗氏Cobas e801、普门eCL9000与新产业MAGLUMI X8三者间斜率接近1(0.940~1.020),截距接近0,而西门子Atellica IM1600与其他分析平台存在显著比例偏差(斜率:0.638~0.658)。Bland-Altman分析表明,西门子Atellica IM1600与其他平台的均值差最大(-47.9~-50.9ng/mL)。一致性分析显示,罗氏Cobas e801、普门eCL9000、新产业MAGLUMI X8两两间Kappa值均大于0.80,一致性良好;而涉及西门子Atellica IM1600的比对,Kappa值仅为0.371~0.479,一致性一般。结论罗氏Cobas e801、普门eCL9000与新产业MAGLUMI X83个平台检测SF结果具有良好的一致性。西门子Atellica IM1600平台结果存在系统性负偏差,与其他平台一致性较差,临床结果不具有可比性。实验室更换分析平台时必须进行方法学比对并建立平台特异性参考区间。 展开更多

目的 比较研究超高效液相色谱-串联质谱法测定茶叶中黄曲霉毒素的前处理方法。方法 采用免疫亲和柱法、QuEChERS法与多功能净化柱法3种不同前处理方法对茶叶进行前处理,结合超高效液相色谱-串联质谱法对茶叶中黄曲霉毒素B1(aflatoxin... 目的 比较研究超高效液相色谱-串联质谱法测定茶叶中黄曲霉毒素的前处理方法。方法 采用免疫亲和柱法、QuEChERS法与多功能净化柱法3种不同前处理方法对茶叶进行前处理,结合超高效液相色谱-串联质谱法对茶叶中黄曲霉毒素B1(aflatoxin B1, AFB1)、黄曲霉毒素B2(aflatoxin B2, AFB2)、黄曲霉毒素G1(aflatoxin G1, AFG1)、黄曲霉毒素G2(aflatoxin G2, AFG2)进行检测,比较3种前处理方法对检测结果的影响。通过考察净化效果、基质效应、稳定性、重复性、检出限、回收率,对3种前处理方法进行了对比。结果 免疫亲和柱法处理样品,杂质较少,基质效应影响最小。3种不同前处理方法在1~60μg/L范围内4种黄曲霉毒素均具有良好线性关系,相关系数均不小于0.999。加标回收率在78.6%~107.1%之间,相对标准偏差1.3%~3.1%,方法检出限为0.04~0.22μg/kg。检出限对比结果,免疫亲和柱法与多功能净化柱法差别不大,但均优于QuEChERS法。3种前处理方法对茶叶质控样品检测结果的差异性在可接受范围内, AFB1、AFB2、AFG1、AFG2检测结果 3种方法均符合要求。结论 3种前处理方法均适用于茶叶中AFB1、AFB2、AFG1、AFG2的测定。免疫亲和柱法基质效应影响小,灵敏度高,适合检测低浓度样品。在操作便捷性及对仪器维护成本方面,QuEChERS法优于多功能净化柱法。QuEChERS法简单高效,适用于大批量样品的检测。 展开更多

目的评价北京九强金斯尔(Gcell)血清淀粉样蛋白A(serum amyloid A,SAA)乳胶免疫比浊法检测试剂盒的分析性能,并与德国SIEMENS SAA检测试剂进行方法学比对,探讨其在常规临床实验室中的适用性。方法依据CNAS-GL037:2019《临床化学定量检... 目的评价北京九强金斯尔(Gcell)血清淀粉样蛋白A(serum amyloid A,SAA)乳胶免疫比浊法检测试剂盒的分析性能,并与德国SIEMENS SAA检测试剂进行方法学比对,探讨其在常规临床实验室中的适用性。方法依据CNAS-GL037:2019《临床化学定量检验程序性能验证指南》及WS/T 402-2012《临床实验室检验项目参考区间的制定》,对Gcell SAA试剂盒的正确度、精密度、线性范围、可报告范围及参考区间进行验证。与SIEMENS SAA检测系统进行方法学比对,采用Passing-Bablok回归与Pearson相关系数评价两方法的线性相关性,同时采用Bland-Altman法评价一致性。结果Gcell SAA试剂盒对厂家工作标准品的相对偏倚分别为0.66%和-0.33%;两水平质控品批内CV为0.82%和1.15%,批间CV为1.49%和1.47%,均符合厂家声明要求。验证线性范围为5.2~180.5mg/L,最大稀释倍数为32倍,可报告上限为5776.00mg/L。30例健康体检人群检测结果均位于厂家声明参考区间(0~10mg/L)内。与SIEMENS SAA检测结果具有良好的线性相关性(r=0.964);Bland-Altman分析显示,平均偏倚为-8.10mg/L,95%一致性界限为-31.28~15.07mg/L。结论Gcell SAA乳胶免疫比浊法试剂盒的各项分析性能指标符合相关标准要求,与SIEMENS检测系统结果具有良好一致性,适用于常规临床实验室SAA检测。 展开更多

目的验证5种小而密低密度脂蛋白胆固醇(sdLDL‑C)试剂的分析性能及检测结果一致性,为临床实验室方法学选择和标准化提供参考。方法在Beckman AU 5800全自动生化分析仪上对国内已获国家药品监督管理局医疗器械注册证的5种sdLDL‑C试剂(标为... 目的验证5种小而密低密度脂蛋白胆固醇(sdLDL‑C)试剂的分析性能及检测结果一致性,为临床实验室方法学选择和标准化提供参考。方法在Beckman AU 5800全自动生化分析仪上对国内已获国家药品监督管理局医疗器械注册证的5种sdLDL‑C试剂(标为A、B、C、D、E)进行验证。参考国家卫生行业标准及美国临床和实验室标准协会文件的要求分别对5种sdLDL‑C试剂进行精密度、线性、参考区间验证及方法学比对。精密度采用高低浓度混合血清及第三方质控物。线性范围经梯度混合血清样本验证。从北京协和医院体检人群临床检测剩余样本中选取70例表观健康人,对参考区间进行验证。选取2022年11月至2023年1月临床检测剩余血清246例,用于5种试剂比对。结果5种sdLDL‑C试剂的重复性为1.1%~5.5%,实验室内不精密度为3.2%~7.0%。除试剂C外,其余4种试剂在说明书声称的分析测量范围内均呈线性(r≥0.990),且检测值与理论值的偏差均在15%以内。A~E各试剂结果落在各自提供参考区间的比例依次为100%(70/70)、94.3%(66/70)、92.9%(65/70)、98.6%(69/70)、80.0%(56/70)。5种试剂与其测定均值的相关系数r分别在0.933~0.981之间,平均绝对百分偏倚在8.3%~32.3%之间,与均值比较的平均偏倚在‑0.12~0.21 mmol/L之间。结论应用于自动生化分析仪的5种sdLDL‑C酶法试剂,具有较高的精密度,但一致性仍需提高。 展开更多

目的对免疫透射比浊法和免疫散射比浊IgG4检测结果的一致性进行评价.方法方法学评价,免疫透射比浊法为评估方法,免疫散射比浊法为参比方法.收集首都医科大学附属北京同仁医院检验科2024年3月至2024年9月期间临床常规检测剩余血清40份,... 目的对免疫透射比浊法和免疫散射比浊IgG4检测结果的一致性进行评价.方法方法学评价,免疫透射比浊法为评估方法,免疫散射比浊法为参比方法.收集首都医科大学附属北京同仁医院检验科2024年3月至2024年9月期间临床常规检测剩余血清40份,分别由评估方法和参比方法检测.采用SPSS 25.0及MedCalc 20.2进行统计学分析,配对秩和检验对两种检测方法的结果进行差异性分析,Passing-Bablok回归和一致性相关系数(CCC)评估两种检测结果的一致性和相关性,Bland-Altman图分析结果的相对平均偏差.结果配对秩和检验Z=2.19,P=0.029,两种检测方法结果在总体分布上差异存在统计学意义.将免疫散射比浊法检测结果定义为自变量,免疫透射比浊法检测结果定义为因变量,两种检测结果的Passing-Bablok回归方程为Y=0.242+0.874X,斜率为0.242(95%CI:0.115~0.323),截距为0.874(95%CI:0.829~0.955),截距和斜率的95%CI(不含0和1)表明两种检测结果存在系统偏差和比例偏差.CCC为0.981,结果一致性较好.Bland-Altman分析对两种检测方法结果进行一致性评估,结果显示其平均相对偏差为-2.6%,以1/3允许总误差(1/3 TEa=8.3%)为判断标准,偏差在可接受范围内.结论免疫透射比浊法和免疫散射比浊法检测结果存在系统偏差和比例偏差,偏差在可接受范围内. 展开更多

在钢铁行业高端化、智能化、绿色化转型加速推进的背景下,铁水预处理作为提升钢材品质的关键工艺环节,其重要性日益凸显。概述了国内外铁水预处理脱硫工艺的发展历程,重点分析了复合喷吹脱硫与机械搅拌脱硫2种主流技术的原理、应用现状... 在钢铁行业高端化、智能化、绿色化转型加速推进的背景下,铁水预处理作为提升钢材品质的关键工艺环节,其重要性日益凸显。概述了国内外铁水预处理脱硫工艺的发展历程,重点分析了复合喷吹脱硫与机械搅拌脱硫2种主流技术的原理、应用现状和优缺点,梳理了高效脱硫技术的发展趋势。热力学和动力学分析表明,喷吹脱硫和机械搅拌脱硫各自优缺点明显,且具有较强的互补性。融合喷吹法与机械搅拌法各自优势的集成技术成为新型高效脱硫技术发展的趋势。攀钢西昌钢钒通过多次技术迭代研究,成功研发了旋转喷吹高效脱硫新技术,充分融合了复合喷吹技术脱硫剂脱硫能力强、铁水深层浸入喷吹脱硫剂、脱硫剂粒度小和用量少的优势与KR(Kambara reactor)技术机械搅拌能力强、流场控制好的优势。通过机械搅拌实现喷入粉气泡的充分细化与弥散化,在高硫铁水脱硫处理中表现出脱硫效率高、温降低、铁损低、成本低的明显优势,有望成为新一代高硫铁水预处理脱硫新技术。 展开更多
