Microbial necromass carbon(MNC)serves a crucial function in the formation and stabilization of soil organic carbon(SOC).Although biochar amendment is recognized as a promising approach for enhancing SOC sequestration,its impact on MNC accumulation across the paddy soil profile remains uncertain.Through a 4-year field experiment,this study examined the effect of biochar amendment on MNC accumulation across three soil layers(0–15,15–30,and 30–45 cm)in a paddy soil profile by combining vertical soil profiling,microbial community dynamics,and biomarker analysis.The results showed that biochar amendment reduced MNC by 10.5%(0–15 cm),7.5%(15–30 cm),and 9.6%(30–45 cm),respectively,compared to the unamended control.In the topsoil(0–15 cm),the reduction in MNC under biochar amendment was attributed to decreases in both fungal and bacterial necromass carbon(C),whereas in the subsoil(15–45 cm),it primarily resulted from the decrease in bacterial necromass C.Biochar amendment reduced MNC content by decreasing microbial biomass and increasing nitrogen(N)acquisition enzyme activities,mainly due to a shift in the microbial community toward K-strategists and intensified microbial N limitation.This study provides novel insights into the microbially-mediated SOC dynamics in response to biochar amendment. 展开更多

Subsea pipelines with egg-shaped profiles may develop cracks during extended operational periods.A cost-effective trenchless rehabilitation method involves installing a thin-walled liner adjacent to the cracked pipeline’s inner surface.This rehabilitation enables the pipeline-liner system to maintain normal liquid/gas transportation functions.The liner can be heated to decrease transportation viscosity and enhance flow rates.However,thermal instability frequently occurs when thin-walled liners are exposed to elevated temperatures.This research presents a computational scheme to analyze the thermal properties of an installed liner with an egg-shaped profile.A modified sine function describes the radial inward deflection,considering the liner’s rigid and tight encasement within the pipeline.Response formulae are derived by combining classic shell principle with the principle of minimum potential energy.These formulae are then solved to determine equilibrium curves and buckling temperature variations.A rigorous verification process validates the computational scheme,demonstrating that the calculated equilibrium curves align well with previous research findings.The study examines the effects of various thickness-to-radius ratios on the egg-shaped liner’s thermostability and compares the thermal buckling resistance between egg-shaped and circular liners.Results indicate superior thermostability in egg-shaped liners compared to circular configurations. 展开更多

Mechanized Huangjiu(Chinese rice wine)is characterized by a stable flavor quality and short fermentation time,yet its flavor quality is inferior to that of traditional Huangjiu.Given the significant market demand for high-quality mechanized Huangjiu and the existing flavor quality gap,this study aimed to isolate and identify highly efficient aroma-producing yeasts from the fermentation mash of traditional Huangjiu for application in mechanized Huangjiu production.From 26 isolated strains,three Saccharomyces cerevisiae strains(OS7302,q1,and NL9)were selected based on their high aroma production capacity in simulated fermentation media.Gas chromatography-mass spectrometry(GC–MS),gas chromatography-olfactometry(GC–O),sensory evaluation,and physicochemical analysis were then combined to systematically verify their abilities to produce aroma compounds.In co-fermentation with mechanized starter cultures,S.cerevisiae OS7302 induced significant increases in the concentrations and aroma intensities of key esters,including ethyl hexanoate,ethyl caprate,and ethyl caprylate,thereby generating an ester aroma profile of mechanized Huangjiu comparable to that of traditional Huangjiu.Furthermore,Huangjiu fermented with S.cerevisiae OS7302 exhibited more intense fruit and ester aromas.In contrast,strains q1 and NL9 promoted the formation of higher alcohols.The use of aroma-producing S.cerevisiae as adjunct cultures for co-fermentation with mechanized starter cultures can improve the flavor quality of mechanized Huangjiu,alleviating the flavor gap between mechanized and traditional Huangjiu. 展开更多

Massive black holes surrounded by a gaseous disk have been a prevailing model to explain a wide spectrum of astrophysical phenome-na related to active galactic nuclei(AGNs).However,direct and precise measurements of the disk density profiles remain elusive for current telescopes.In this work,we demonstrate that it is possible to pinpoint the gas density if an inspiralling stellar mass binary black hole is embedded in the AGN disk.Furthermore,if the barycenter of the pair follows an eccentric orbit around an AGN,then space-borne gravitational wave detectors can measure the density of the surrounding disk with multi-year observations by tracking the gravitational wave evolution.The error between the inferred density profile and the injected truth can be constrained to below 2×10-11 g/cm3.Our work opens up an exciting new channel to investigate the very center of galaxies,where disk gas density distributionsρ(r)can be recovered by analyzing time-dependent environmental imprints in gravitational waveforms. 展开更多

Low-level atmospheric turbulence impacts urban air quality and poses substantial risks to aviation safety,underscoring a critical demand for reliable short-term early warning.Radar wind profilers(RWPs)provide continuous vertical observations of wind fields,emerging as a key tool for low-level turbulence monitoring.However,existing turbulence forecasting relies mostly on diagnostic or retrieval approaches,which are often prone to systematic biases and not tailored for short-term operational early warning.In this study,we develop an operational early-warning model for low-level turbulence using a data-driven machine learning approach,leveraging RWP observations.Drawing on long-term observations at Shiyan Station in Shenzhen of Guangdong Province,we integrate RWP measurements with estimates of in situ turbulence dissipation rate(ε),as derived from ultrasonic anemometers installed at 160and 320 m on a meteorological tower.A random forest model is trained to predict the meanεin the subsequent 30minutes,with predictors consisting of preceding half-hourly averaged RWP variables.The results demonstrate that the proposed model effectively captures the nonlinear relationship between RWP observations andε,delivering robust predictive performance.The reliability of model exhibits a clear dependence on atmospheric and seasonal conditions.Under clear-sky scenarios and during autumn and winter,the model achieves enhanced stability,with correlation coefficients exceeding 0.7.In contrast,prediction uncertainty increases under complex conditions characterized by low wind speeds and weak turbulence signals.Further error analysis reveals systematic associations between prediction errors and observed spectral width,wind speed,and true turbulence intensity magnitude.Notably,applying threshold constraints to observed spectral width further improves the reliability of model predictions.This study provides a promising pathway to advancing turbulence monitoring and short-term forecasting in complex urban environments,with direct implications for low-level aviation safety. 展开更多

Accurate retrieval of atmospheric vertical profiles is critical for improving weather prediction and climate monitoring.However,the complexity of atmospheric processes in cloudy regions poses challenges compared to those of clear sky scenarios.This study presents a novel framework that integrates Bayesian optimization and machine learning approaches to retrieve atmospheric vertical profiles—including temperature,humidity,ozone concentration,cloud fraction,ice water content(IWC),and liquid water content(LWC)—from hyperspectral infrared observations.Specifically,a Bayesian method was used to refine ERA5 reanalysis data by minimizing brightness temperature(BT)discrepancies against FY-4B Geostationary Interferometric Infrared Sounder(GIIRS)observations,generating a high-quality profile database(~2.8 million profiles)across diverse weather systems.The optimized profiles improve radiative consistency,reducing BT biases from>40 K to<10 K in cloudy regions.To further overcome the limitations of the Bayesian method,we developed a Transformer-Resnet hybrid model(TERNet),which achieved superior performance with RMSE values of 1.61 K(temperature),5.77%(humidity),and 2.25×10–6/6.09×10–6kg kg–1(IWC/LWC)across the entire vertical levels in all-sky conditions.The TERNet outperforms both ERA5 in cloud parameter retrieval and the GIIRS L2 product in thermodynamic profiling.Independent verification with radiosonde and Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observations(CALIPSO)datasets confirms the framework's reliability across various meteorological regimes.This work demonstrates the capability of combining physics-informed Bayesian methods with data-driven machine learning to fully exploit hyperspectral IR data. 展开更多

Stratospheric aerosol extinction profiles are crucial for studying climate change and various atmospheric physical and chemical processes.Limb observations are among the most effective methods for obtaining global stratospheric aerosol extinction profiles.In this paper,based on the color-ratio and optimal estimation methods,aerosol extinction profiles between 10 and 35 km were successfully extracted from limb-scattering signals measured by the Backward Limb Spectrometer(BLS)onboard the Tiangong-2 space laboratory.The aerosol extinction profiles derived from BLS were compared with those from the OMPS-LP v2.0 aerosol product,and the results showed good consistency.Using the average profile of each pair of BLS and OMPS-LP cross-observations as a reference,the mean relative errors of BLS are generally within±15%between 15 and 30 km,with a correlation coefficient of 0.955.However,larger errors may occur below 15 km.Sensitivity analysis indicates that the larger errors at lower altitudes are attributed to inaccuracies in the tangent height and the stratospheric aerosol particle size distribution model,both of which affect the accuracy of the final retrievals. 展开更多

Small extracellular vesicles(sEVs)membrane protein profile(sEVpp)is a novel biomarker for cancer,and it can reveal the in-depth phenotype information.The point-of-care testing(POCT)of sEVpp holds great significance for mass screening of cancer,so the cost-effective and simple detection methods of sEVpp are urgently demanded.Herein,we constructed a paper-based multichannel sEVpp POCT device(sEVpp-PAD)enabled by functional DNA probes and metal-organic framework(MOF).The core components are aptamer/MOF-modified paper chips.The modified aptamers can immunocapture the sEV expressing corresponding proteins,while the modified MOF can provide abundant sites for aptamer-modification,reduce the nonspecific protein absorption,and act as reference for ratiometric detection.Simply powered by two syringes,the sEVpp-PAD can efficiently capture sEVs expressing corresponding protein from cell culture media and sera.Furthermore,a detection probe(DP)consisted of CD63 aptamer and G-quadruplex was developed for the colorimetric detection of captured sEVs.Utilizing this device,the sEVpp in various hepatocellular carcinoma cell culture medium and,more importantly,in human sera can be accurately determined,only with$2 device,$0.2 detection reagents and 1.8 h procedure.This simple strategy for sEVpp detection can innovatively promote the POCT and subtyping of cancer based on sEV-related liquid biopsy. 展开更多

BACKGROUND Perianal abscesses(PAs)are associated with significant complications,such as recurrent infections,pain,anal fistulas,rectovaginal fistulas,rectourethral fistulas,and rectovesical fistulas.However,established primary and secondary prevention strategies for PAs are lacking.AIM To explore the relationships between obesity and lipid metabolites,including perianal abscess onset.METHODS We conducted two independent studies under a unified research question.Casecontrol analysis was conducted at a single hospital between May 2023 and November 2023.Inpatients diagnosed with a perianal abscess and matched healthy controls were included.Body dimensions and serum metabolites were measured.Genome-wide association study data regarding genetic variants of PAs,obesity,and serum metabolites were obtained for the Mendelian randomization(MR)analysis.The study outcomes were perianal abscess onset and the number and location of PAs.RESULTS In the case-control study,higher body mass index(BMI),waist-to-hip ratio(WHR),waist-to-height ratio(WHtR),blood glucose levels,uric acid(UA)levels,total cholesterol levels,triglyceride levels,and low-density lipoprotein(LDL)levels were associated with increased risk of PAs.Higher high-density lipoprotein levels were associated with reduced risk of PAs.The BMI,WHR,WHtR,UA level,triglyceride level,and LDL level were associated with the number and severity of PAs.In MR analysis,the BMI,WHR,body fat percentage,whole body fat mass,limb fat percentage,limb fat mass,and various lipid profiles were significantly associated with the risk of PAs.CONCLUSION A hospital-based case-control study and an independent MR analysis consistently support obesity and lipid metabolism profiles are associated with an increased risk of perianal abscess.These findings provide a basis for developing primary and secondary prevention strategies for perianal abscess. 展开更多

Background:Exposure to environmental vulnerability poses significant threats to adolescent suicidal ideation,while individual resilience can mitigate these adverse effects with notable gender commonalities and differences.However,research examining how these factors co-configure at the individual level remains limited,particularly from a gender-specific perspective.Thus,the present study aims to adopt a person-centered analytic approach to identify gender-specific configurations of environmental vulnerability and individual resilience associated with suicidal ideation among Chinese adolescents.Methods:Data were collected from 2616 Chinese primary and secondary school students(aged 10–17;1223 girls).Participants completed validated scales measuring environmental vulnerability,individual resilience,and suicidal ideation.Latent profile analysis(LPA)was conducted separately by gender.Results:Gender differences were prominent:males exhibited higher resilience and lower suicidal ideation,while females reported higher environmental vulnerability and elevated levels of suicidal ideation.LPA identified three distinct profiles for males:Low Vulnerable–High Protective–Low Risk(LHL),Medium Vulnerable–Low Protective–Low Risk(MLL),and High Vulnerable–Low Protective–High Risk(HLH).Four profiles emerged for females:LHL,MLL,Medium Vulnerable–Low Protective–Medium Risk(MLM),and HLH.Crucially,within the HLH profile,males exhibited particularly deficient humor(η2=0.19)and confidence(η2=0.16),while females formed a distinct subgroup characterized by severe academic and family stressors(η2=0.30–0.36).Conclusion:The study underscores developing gender-specific mental health interventions using a nuanced,person-centered approach that considers both environmental risk and individual resilience factors,which allows for targeted suicide prevention strategies addressing the unique needs of male and female adolescents. 展开更多

In the realm of composite materials,the machining accuracy of Carbon Fiber Reinforced Polymer(CFRP)circular cell honeycombs plays a critical role in determining the performance of sandwich components.However,due to the discontinuous nature and relatively weak stiffness characteristics of this material,achieving precise control over the complicate surface profile accuracy becomes a tough task,often accompanied by the problem of low processing efficiency.This study proposes a novel topological hierarchy-based toolpath strategy specifically designed for the grinding process,combined with compensating CFRP circular cell honeycomb surfaces.The proposed methodology involves sequentially processing each circular cell,wherein those cells are treated as discrete units arranged within a two-dimensional plane.To enhance the surface profile accuracy of the workpiece while maintaining high processing efficiency,a compensatory machining approach has been employed.Based on this approach,algorithms for the extraction of the discontinuous compensatory machining regions have been subsequently developed.Consequently,an optimized toolpath and a serialized machining process has been established,enabling the efficient and high accuracy grinding of the CFRP circular cell honeycombs.Experimental verifications demonstrate that the proposed method achieves a significant improvement in machining efficiency and successfully confines the surface profile error within a tolerance of 20μm.Additionally,the peak-to-valley on crosssection lines after compensatory machining decreases from 0.20 to 0.05 mm.Overall,this innovative method presents a practical solution for the fabrication of large-scale honeycomb sandwich components,thereby making significant contributions to the field of advanced composite material machining. 展开更多

Using fully kinetic particle-in-cell simulations,we investigate the stability and performance of autoresonant plasma beat-wave excitation in plasmas with tailored density profiles.We show that a prescribed spatial variation of the background density sustains continuous phase locking between the driving laser beat and the excited plasma mode,thereby enabling precise control of the shape and group velocity of the plasma wavepacket and providing an alternative to frequency chirping of the drive lasers.The density-gradient scale is found to govern the nonlinear autoresonant growth,and the attainable saturation amplitude can exceed the classical Rosenbluth-Liu prediction and,for appropriate laser intensities,approach the nonrelativistic wave-breaking limit.We show that a four-laser configuration in a steep parabolic density profile can generate a specially confined two-phase quasi-periodic plasma lattice.The generation of such structures may lead to novel applications in plasma photonics. 展开更多

The health benefits of teas and tea preparations in assisting with weight loss have been well documented in numerous studies.Nevertheless,there is a paucity of population-based research on the effects of consumption of tea and tea preparations on human dietary intake,gut microbial and metabolic profiles,and thus on weight loss.This study therefore focused on changes in dietary intake,gut microbiota and metabolism in healthy adults who consumed black tea for 3 consecutive weeks.Our findings showed that the consumption of black tea was associated with a notable increase in satiety prior to meals,which was observed to be more pronounced in female participants than in male participants.The consumption of black tea has been shown to increase the species richness of gut microorganisms,as evidenced by a higher number of observed operational taxonomic units(OTUs)compared the test group with the control group(203±35 vs.229±18,respectively).Moreover,it improves the microbiological structure of the gut,enhances the abundance of beneficial bacteria such as Faecalibacterium and Parabacteroides,and reduces the abundance of Prevotella.Metabolomic analysis revealed 13 significantly different metabolites and 9 enriched metabolic pathways in individuals who consumed black tea.Correlation analyses of body mass index(BMI),satiety,gut flora and metabolic profiles indicate that the modulatory effects of black tea on gut flora and metabolic profiles are more effective in high BMI participant.This study may provide new insights into the relationship between black tea interventions and host satiety,gut microbiota and metabolic profiles.Furthermore,it may offer new directions and options for clinical strategies and personalized nutrition for tea and tea preparations to assist in weight loss and lipid reduction. 展开更多

BACKGROUND Diabetic gastrointestinal autonomic neuropathy(DGAN)is a common yet underrecognized manifestation of diabetic neuropathy.However,the clinical correlations and associated alterations of serum bile acid(BA)metabolism with DGAN remain unclear.AIM To identify potential metabolite biomarkers capable of identifying DGAN among patients with type 2 diabetes mellitus(T2DM).METHODS This cross-sectional study included 26 patients with clinically defined DGAN and 69 patients with uncomplicated T2DM.Fifteen individual BAs were quantified in fasting serum using liquid chromatography-tandem mass spectrometry.Gastrointestinal symptoms were scored using the Gastrointestinal Symptom Rating Scale.Spearman’s correlation,multivariable logistic regression,receiver operating characteristic,and decision curve analyses were used to explore the associations and build a predictive nomogram.RESULTS Orthogonal partial least squares discriminant analysis of serum BAs revealed clear separation between the T2DM and DGAN groups,identifying taurolithocholic acid(TLCA)as the key discriminator(variable importance in projection>1,fold change<0.5).Univariate logistic regression identified age,body mass index,hemoglobin,fasting/2-h C-peptide,albumin,and TLCA levels as protective factors and the urinary albumin-to-creatinine ratio as a risk factor.After multivariate adjustment age,fasting C-peptide levels,and TLCA levels remained independently associated with DGAN.Receiver operating characteristic analysis yielded areas under the curve of 0.651,0.760,and 0.678 for age,fasting C-peptide,and TLCA,respectively,and the three variables collectively had an area under the curve of 0.970(95%confidence interval:0.937-1.000).Decision curve analysis confirmed the clinical net benefit across threshold probabilities of 9%-68%.The derived nomogram displayed excellent calibration and net clinical benefits for the model.CONCLUSION These findings highlighted the association between altered BA profiles and DGAN in patients with T2DM.Combining BA profiling with conventional clinical data could facilitate the early identification of DGAN and offer new insights into early screening and BA-targeted interventions.While these findings offer valuable insights,they should nevertheless be viewed as hypothesis-generating and require further validation in larger,multicenter cohorts. 展开更多

The dynamic regulatory mechanisms by which Lactobacillus casei 139 fermentation modulates the nutritional components and flavor characteristics of Aronia melanocarpa(Michx)Elliott juice were investigated through a systematic evaluation of temporal changes in nutritional and flavor profiles during fermentation.The results indicated that the viable cell density peaked at 6.01 log CFU/mL within 24 h.During fermentation,the pH of A.melanocarpa juice initially increased slightly within 48 h before gradually decreasing,while titratable acidity consistently decreased,reflecting the metabolic activity of Lactobacillus casei 139.Concurrently,total sugar content exhibited a decline followed by a leveling off to a steady state.It was found that the contents of total phenolic and flavonoid contents decreased initially,while the color difference a*first decreased,then increased.Combined electronic nose and sensory analyses revealed that fermentation enhanced aroma complexity,and mitigated astringency by promoting aromatic compound accumulation.The research confirms that fermentation with strain L.casei 139 can effectively improve both the sensory quality and nutritional value of fermented A.melanocarpa beverages. 展开更多

We read with interest the recent study by Zeber-Lubecka et al,published in the World Journal of Hepatology,investigating transcriptome profiles of peripheral blood mononuclear cells(PBMCs)in male adolescents with non-alcoholic fatty liver disease(NAFLD).To maintain consistency with the original publication,we retain the term“NAFLD”throughout this commentary,while recognizing the recent shift toward the MASLD nomenclature.The authors stimulated PBMCs ex vivo with autologous fecal extracts to explore immune-microbiota interactions.Their results revealed distinct transcriptomic and cytokine patterns,including elevated pro-inflammatory mediators indicative of early immune dysregulation.These findings underscore the impact of host-specific gut microbiota on systemic immunity and suggest that PBMC-based immune signatures could serve as minimally invasive biomarkers.This work provides valuable insights into gutimmune crosstalk in pediatric NAFLD and offers a foundation for future research on early diagnosis and therapeutic strategies. 展开更多

BACKGROUND Tangwang formula(TWF),a traditional Chinese medicine,has been shown to delay the progression of diabetic retinopathy(DR)over the past 20 years.However,the potential therapeutic mechanisms and effective components of TWF remain unclear.AIM To investigate the effective components of TWF and elucidate the mechanism underlying TWF treatment for DR.METHODS The chemical ingredients of TWF were detected using high-pressure liquid chromatography.Network pharmacology and molecular docking were applied to identify the targets and pathways associated with TWF and DR.A DR mouse model was established by streptozotocin to evaluate the therapeutic effect of TWF in vivo.High glucose treatment was used to induce injury in mouse retinal endothelial cells(mRECs)to verify the mechanism of TWF in vitro.The 16S ribosomal RNA sequencing and untargeted metabolomics were performed to detect intestinal bacteria and fecal metabolites in DR mice.RESULTS Eight important active constituents were identified by high-pressure liquid chromatography.Network pharmacology results showed that Bcl2 and Casp3 were key targets between TWF and DR.Isorhamnetin-3-O-neohespeidoside,naringenin,ononin,and calycosin-7-O-β-D-glucoside from TWF had strong affinities with Bcl2 and Casp3.TWF significantly reduced random blood glucose,inhibited the levels of proinflammatory factors in DR mice and mRECs,and alleviated retinal damage by downregulating the expressions of vascular endothelial growth factor and receptor advanced glycation end products,and upregulating the expressions of zonula occludens-1 and RBP-3.TWF inhibited the apoptosis in mouse retinal tissues and mRECs.Analysis of intestinal bacteria and metabolites revealed that TWF increased the richness and evenness of intestinal microbiota and improved fecal metabolic profiles in DR mice.Importantly,seven genus bacteria were closely correlated with amino acids,fatty acids,glycerophosphocholine,and bile acid,which could participate in ameliorating retinal injury.CONCLUSION These findings suggest that TWF may ameliorate retinal damage in DR mice via anti-inflammatory and antiapoptotic activities,which may be associated with modulation of the intestinal microbiota and metabolic profiles. 展开更多
