Backgrounds:Somatization and eating-related problems in adolescents living in residential care may be shaped by the interplay of risk and protective factors,including gender,relational trauma,attachment patterns,emoti...Backgrounds:Somatization and eating-related problems in adolescents living in residential care may be shaped by the interplay of risk and protective factors,including gender,relational trauma,attachment patterns,emotional intelligence,and perceived social support.This study examined how gender,relational trauma,attachment dimensions,resilience,and emotional intelligence contribute to the presence of somatic and eating difficulties in this population.Methods:The sample included 46 adolescents(63%female;ages 12–17,Mean=14.85,Standard Deviation(SD)=1.49)residing in child protection institutions in Uruguay.Participants completed self-report measures assessing childhood relational trauma(CaMir),attachment dimensions(anxiety and avoidance),resilience,emotional intelligence(adaptability and stress management),social support(MOS),and psychosocial adjustment(SENA subscales of somatization and eating problems).Using a fuzzy-set Qualitative Comparative Analysis(fsQCA)approach,distinct configurations of risk and protective factors associated with elevated levels of somatization and eating problems were identified.Results:Relational trauma and attachment anxiety showed moderate associations with both somatization and eating problems(r=0.52–0.57,p<0.01),whereas stress management was negatively associated with both outcomes(r=−0.37 to−0.47,p<0.05).FsQCA revealed multiple configurations of risk and protective factors explaining 81–90%of cases,with solution consistencies ranging from 0.83 to 0.87.Results suggest that relational trauma and attachment anxiety are key risk conditions,whereas resilience,emotional regulation,and perceived social support function as protective factors.Conclusions:Findings highlight the importance of considering multifactorial patterns of vulnerability and protection rather than single predictors and underscore the need for tailored interventions that strengthen resilience and emotional skills while addressing the impact of early relational trauma.展开更多

The aim of our stu...Background: Somatic symptoms that are not attributable to organic pathology are common in general practice settings however, data in most parts of Africa including southern Nigeria are still scarce. The aim of our study was to examine such somatic symptoms reported by patients attending a primary care facility at a tertiary hospital in southern Nigeria as well as to motivate future research in this area. Method: The study was conducted at the General Out Patient Clinic (GOPC) of the University of Calabar Teaching Hospital (UCTH). It was a cross sectional study in which data were obtained from the case notes of 115 patients that presented in the clinic with somatic complaints which could not be attributed to organic pathology by their physicians. Results: While 46 of the patients were males, 69 were females. Their mean age was 37.7 (SD = 11.9). Internal heat, crawling sensation, body pains and palpitations were the most prevalent symptoms reported by the patients. When the symptoms were sorted into various groups, the “subjective abnormal bodily sensation” was the most prevalent and far outnumbered the “pseudo neurological symptoms”. Conclusion: A number of patients attending the GOPC of the UCTH seek consultations for medically unexplained somatic symptom. The most prevalent of these symptoms are internal heat and crawling sensations both of which are not stated in the criteria recognized by the International Classification of Diseases—version 10 (ICD-10) for the diagnosis of psychiatric disorders.展开更多

Results: the incidence of somatization disorder in our hospital is not remarkable in gender level, mostly showed chronic delay, the average course of about 7-8 years; somatization disorder usually occurs in junior school culture and below; in clinical symptoms, such as 72.9% and musculoskeletal system 74.9% and 16.9% genitourinary system. Conclusions: somatization disorders mostly occur in poorly educated populations, and clinical symptoms will involve multiple systemic organs in the human body, so targeted research is needed to address the occurrence of the problem.展开更多

Interoception refers to the sensation and perception of internal bodily sensations,and may be related to depressive symptoms.Schemata concerning the body vary across different cultures and may influence interoception ...Interoception refers to the sensation and perception of internal bodily sensations,and may be related to depressive symptoms.Schemata concerning the body vary across different cultures and may influence interoception and symptom presentations of depression.This study explored the relationship between interoception,depressive symptoms,and schema of somatic focus in Chinese people with subsyndromal depression.Thirty-nine individuals with subsyndromal depression(SD)and 40 healthy controls(HCs)were assessed at baseline and after 3 months.Participants completed the self-report questionnaires for assessing interoceptive sensibility,somatic and psychological symptoms of depression,and somatization tendency.They also completed the heartbeat perception behavioral task for estimating interoceptive accuracy.The results showed that both the SD and the HC groups showed similar interoceptive accuracy,although the SD group showed heightened interoceptive sensibility.The discrepancy between interoceptive sensibility and interoceptive accuracy is termed the interoceptive trait prediction error(ITPE).The ITPE was positive in SD participants but was negative in HCs.In the entire sample,interoceptive sensibility and the ITPE were correlated with somatic symptoms rather than with psychological symptoms of depression.Interoceptive sensibility partially mediated the relationship between somatization tendency and somatic symptoms,after controlling for psychological symptoms of depression.These results remained stable after 3 months.The shortcomings of the present study were a lack of clinical interview to ascertain diagnosis and a short follow-up duration.In conclusion,our study suggests that altered interoception occurs in subsyndromal depression.Interoception is related to somatic symptoms of depression.The schema of body was related to depressive symptoms,partially through interoception,in Chinese people with subsyndromal depression.展开更多

BACKGROUND Laparoscopic appendectomy is the gold standard for treatment of acute appendicitis;however,postoperative pain characterized by a visceral and somatic component with separate neural pathways still interfere ...BACKGROUND Laparoscopic appendectomy is the gold standard for treatment of acute appendicitis;however,postoperative pain characterized by a visceral and somatic component with separate neural pathways still interfere with early recovery.Transversus abdominis plane block(TAPB)relieves somatic pain,and erector spinae plane block(ESPB)may provide additional visceral pain relief by paravertebral and sympathetic chain blockade.Due to the lack of distinction between these two types of pain in previous study comparisons,we performed this retrospective analysis.AIM To compare the analgesic efficacy of ESPB vs TAPB for visceral pain and somatic pain following laparoscopic appendectomy.METHODS A retrospective analysis was conducted on 183 patients who underwent laparoscopic appendectomy with ultrasound-guided nerve blocks between January 2021 and December 2024,including 91 patients in the ESPB group and 92 patients in the TAPB group.Both groups received bilateral blocks with 0.375%ropivacaine at a total volume of 30 mL.Visual Analog Scale(VAS)was used to assess visceral pain and somatic pain scores at rest and during activity at 2,6,12,24,and 48 hours postoperatively.Rescue analgesia within 48 hours postoperatively,postoperative recovery quality indicators,incidence of postoperative nausea and vomiting,and patient satisfaction with analgesia were recorded.RESULTS The ESPB group had significantly lower visceral pain VAS scores at rest and during activity at 2 hours,6 hours,and 12 hours postoperatively compared with the TAPB group(P0.05).There were no significant differences in somatic pain VAS scores between the two groups at any time point(P>0.05).The ESPB group had a lower rescue analgesia rate than the TAPB group(13.2%vs 26.1%,P=0.029),less tramadol consumption(P=0.012),and higher satisfaction scores with analgesia(P=0.013).There were no significant differences between the two groups in time to first ambulation,time to first flatus,postoperative hospital stay,or incidence of nausea and vomiting(P>0.05).No nerve block-related complications occurred in either group.CONCLUSION Compared with TAPB,ESPB provides better analgesia for visceral pain in the early postoperative period following laparoscopic appendectomy,while both techniques demonstrate equivalent analgesic effects for somatic pain.For patients with visceral pain as the predominant component postoperatively,ESPB may be a more appropriate choice.展开更多
Objectives:Recently,how family-related factors influence employees’mental health has garnered increasing attention from researchers and practitioners.Drawing on the cognitive appraisal theory of stress,this study aim...Objectives:Recently,how family-related factors influence employees’mental health has garnered increasing attention from researchers and practitioners.Drawing on the cognitive appraisal theory of stress,this study aims to examine how and when family financial stress affects the employees’mental health and investigate the mediating role of performance stress and the moderating role of workplace competition.Methods:A cross-sectional survey was conducted with 23,520 Chinese employees by using a voluntary and anonymous structured questionnaire,which included family financial stress,performance stress,symptom checklist 90(SCL-90)scale,and workplace competition.The data were analyzed using SPSS 26.0 software and macro PROCESS.Results:The analysis of the mediating effect showed that performance stress mediated the relationship between family financial stress and psychological depression(b=0.064,SE=0.002,p<0.0001)and physical somatization(b=0.042,SE=0.002,p<0.0001),indicating spillover effects of stress from home to workplace.The moderating mediation analysis revealed that the crossover effects were amplified by workplace competition.For psychological depression,index of moderated mediation was:b=0.012,SE=0.001,p<0.001;For physical somatization,index of moderated mediation was:b=0.008,SE=0.001,p<0.001.Conclusion:Performance stress acts as a mediator in the link between family financial stress and mental health.Furthermore,the mediating effects was amplified by workplace competition.These findings suggest that workplace competition may serve to exacerbate the negative spillover effects from home to work through the mechanism of workrelated stress.Organizations should consider implementing supportive measures to mitigate family financial stress,such as providing financial counseling and fostering a collaborative work environment,to reduce the adverse effects of family financial stress on employees’mental health.展开更多
Litchi chinensis Sonn.is an important economic fruit tree in tropical and subtropical regions.Regrettably,the efficiency of plant regeneration via somatic embryogenesis in litchi is typically low due to the poor conve...Litchi chinensis Sonn.is an important economic fruit tree in tropical and subtropical regions.Regrettably,the efficiency of plant regeneration via somatic embryogenesis in litchi is typically low due to the poor conversion of embryos to plants.The purpose of this study was to establish a regeneration system via somatic embryogenesis from immature embryos explants in‘Heiye'cultivar of litchi.Our results demonstrated that MS medium supplemented with 2.0 mg L-12,4-D was optimal for callus induction.For somatic embryo(SE)induction,MS medium containing0.5 g L-1 activated charcoal(AC)was the most effective,while the use of zeatin(ZT)and thidiazuron(TDZ)resulted in abnormal somatic embryos.The rooting and regeneration rate of 2.15%and 17.5%,respectively,were achieved using MS medium supplemented with 0.5 g L-1 AC.Furthermore,transcriptome analysis was performed on embryogenic callus(EC),globular embryo(GE),and heart embryo(HE)to explore the molecular mechanisms of early somatic embryogenesis.2,587 common differentially expressed genes(DEGs)between EC_vs_GE and EC_vs_HE were identified,and the expression patterns of these common DEGs were separated into twelve major clusters.GO annotation and KEGG pathway analysis revealed that these common DEGs were implicated in plant hormone signal transduction,auxin-activated signaling pathway,and other biological processes.Additionally,differentially expressed transcription factors were identified,and the function of LcBBM2 which is specifically highly expressed during early somatic embryogenesis was verified.Overexpression of LcBBM2 in tomato promotes callus and shoot formation.Therefore,this study can provide a theoretical basis and technical support for genetic breeding improvement of litchi.展开更多
Somatic cell nuclear transfer(SCNT)has been successfully employed across various mammalian species,yet cloned animals consistently exhibit low pregnancy rates,primarily due to placental abnormalities such as hyperplas...Somatic cell nuclear transfer(SCNT)has been successfully employed across various mammalian species,yet cloned animals consistently exhibit low pregnancy rates,primarily due to placental abnormalities such as hyperplasia and hypertrophy.This study investigated the involvement of the Hippo signaling pathway in aberrant placentaldevelopmentinSCNT-inducedbovine pregnancies.SCNT-derived cattle exhibited placental hypertrophy,including enlarged abdominal circumference and altered placental cotyledon morphology.RNA sequencing analysis indicated significant dysregulation of Hippo signaling pathway genes in SCNT placentas.Coexpression of YAP1 and CCND1 was observed in cloned blastocysts,placental tissues,and bovine placental mesenchymal stem cells(bPMSCs).Manipulation of YAP1expression demonstrated the capacity to regulate bPMSC proliferation.Experimental assays confirmed the direct binding of YAP1 to CCND1,which subsequently promoted CCND1 expression in bPMSCs.Furthermore,inhibition of CDK6,a downstream target of CCND1,attenuated SCNT bPMSC proliferation.This study identified YAP1 as a key regulatory component within the Hippo signaling pathway that drives placental hyperplasia in cloned cattle through up-regulation of CCND1-CDK6 expression,facilitating cell cycle progression.These findings offer potential avenues for enhancing cloning efficiency,with implications for evolutionary biology and the conservation of valuable germplasm resources.展开更多
Somatic variants in the cancer genome influence gene expression through diverse mechanisms depending on their specific locations.However,a systematic evaluation of the effects of somatic variants located in 3'untr...Somatic variants in the cancer genome influence gene expression through diverse mechanisms depending on their specific locations.However,a systematic evaluation of the effects of somatic variants located in 3'untranslated regions(3'UTRs)on alternative polyadenylation(APA)of m RNA remains lacking.In this study,we analyze 10,199 tumor samples across 32 cancer types and identify 1333 somatic single nucleotide variants(SNVs)associated with abnormal 3'UTR APA.Mechanistically,these 3'UTR SNVs can alter cisregulatory elements,such as the poly(A)signal and UGUA motif,leading to changes in APA.Minigene assays confirm that 3'UTR SNVs in multiple genes,including RPS23 and CHTOP,induce aberrant APA.Among affected genes,62 exhibit differential stability between tandem 3'UTR isoforms,including HSPA4and UCK2,validated by experimental assays.Finally,we establish that SNV-related abnormal APA usage serves as an additional layer of expression regulation for tumor-suppressor gene HMGN2 in breast cancer.Collectively,this study reveals 3'UTR APA as a critical mechanism mediating the functional impact of somatic noncoding variants in human cancers.展开更多

Background Cotton is an industrial crop renowned for its multifaceted applications in the textiles,pharmaceuticals,and biofuel industries.Plant regeneration through somatic embryogenesis(SE)plays a crucial role in the...Background Cotton is an industrial crop renowned for its multifaceted applications in the textiles,pharmaceuticals,and biofuel industries.Plant regeneration through somatic embryogenesis(SE)plays a crucial role in the genetic improvement of cotton.There is a strong correlation between SE and zygotic embryogenesis(ZE)in plants.Furthermore,the strategy of ectopic expression of cotton genes into the model plant Arabidopsis has been a widely accepted approach for functional study.Result Based on previous spatial transcriptomics of cotton somatic embryos,two genes,Gh HAT5 and Gh CRK29,were identified.They are highly expressed in cotyledon and epidermal cells of cotton cotyledonary embryos,respectively.In this study,Gh HAT5 and Gh CRK29 were ectopically expressed in Arabidopsis to investigate their functions.The result showed that in Arabidopsis zygotic embryos,the overexpression of Gh HAT5 promoted the development of apical embryonic upper-tier cells and embryonic cotyledon,while the overexpression of Gh CRK29 promoted the development of apical embryonic lower-tier cells and embryonic radicle.Given the similarities between somatic and zygotic embryogenesis,these findings suggest that Gh HAT5 and Gh CRK29 are involved in cotton SE.We also speculate that these genes may promote the expression of the Arabidopsis endogenous gene At SCR,which is crucial for embryonic development.Conclusion These results revealed that Gh HAT5 and Gh CRK29 regulate embryonic development and are essential in advancing our understanding of cotton SE and facilitating targeted genetic manipulation strategies to improve industrial crop traits and agricultural sustainability.展开更多
Litchi has great economic significance as a global fruit crop.However,the advancement of litchi functional genomics has encountered substantial obstacles due to its recalcitrance to stable transformation.Here,we prese...Litchi has great economic significance as a global fruit crop.However,the advancement of litchi functional genomics has encountered substantial obstacles due to its recalcitrance to stable transformation.Here,we present an efficacious Agrobacterium tumefaciens-mediated transformation system in somatic embryos of‘Heiye'litchi.This system was developed through the optimization of key variables encompassing explant selection,A.tumefaciens strain delineation,bacterium concentration,infection duration,and infection methodology.The subsequent validation of the transformation technique in litchi was realized through the ectopic expression of LcMYB1,resulting in the generation of transgenic calli.However,the differentiation of transgenic calli into somatic embryos encountered substantial challenges.To delineate the intricate molecular underpinnings of LcMYB1's inhibitory role in somatic embryo induction,a comprehensive transcriptome analysis was conducted that encompassed embryogenic calli(C),globular embryos(G),and transgenic calli(TC).A total of 1,166 common differentially expressed genes(DEGs)were identified between C-vs.-G and C-vs.-TC.Gene Ontology(GO)annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis revealed that these common DEGs were mostly related to plant hormone signal transduction pathways.Furthermore,RT-qPCR corroborated the pronounced down-regulation of numerous genes that are associated with somatic embryo induction within the transgenic calli.The development of this transformation system provides valuable support for functional genomics research in litchi.展开更多
The CRISPR-Cas-based gene targeting(GT)method has enabled precise modifications of genomic DNA ranging from single base to several kilobase scales through homologous recombination(HR).In plant somatic cells,canonical ...The CRISPR-Cas-based gene targeting(GT)method has enabled precise modifications of genomic DNA ranging from single base to several kilobase scales through homologous recombination(HR).In plant somatic cells,canonical non-homologous end-joining(cNHEJ)is the predominant mechanism for repairing double-stranded breaks(DSBs),thus limiting the HR-mediated GT.In this study,we implemented an approach to shift the repair pathway preference toward HR by using a dominant-negative ku80 mutant protein(KUDN)to disrupt the initiation of cNHEJ.The employment of KUDN conferred a 1.71-to 3.55-fold improvement in GT efficiency at the callus stage.When we screened transformants,there was a more remarkable increase in GT efficiency,ranging from 1.62-to 9.84-fold,at two specific tomato loci,SlHKT1;2 and SlEPSPS1.With practical levels of efficiency,this enhanced KUDN-based GT tool successfully facilitated a 9-bp addition at an additional locus,SlCAB13.These findings provide another promising method for more efficient and precise plant breeding.展开更多
Conifers pose challenges for breeding programs due to their extended vegetative phases.Despite the critical role of APETALA2(AP2)in regulating phase transitions,the AP2/ERF superfamily remains largely unexplored in gy...Conifers pose challenges for breeding programs due to their extended vegetative phases.Despite the critical role of APETALA2(AP2)in regulating phase transitions,the AP2/ERF superfamily remains largely unexplored in gymnosperms.Here,the first genome-wide analysis of the AP2/ERF superfamily in a conifer,Larix kaempferi(Japanese larch)is presented,and 374 members were identified.Among all eight paralogs,four euAP2 lineage genes,TARGET OF EATs(TOEs),exhibit age-decreased expression patterns.Functional characterization of LkTOE1-2 demonstrates its involvement in somatic embryogenesis and seed germination.Importantly,the RUBY reporter system confirmed an enhanced promoter activity in somatic embryo maturation.Over-expression of LkTOE1-2 in Arabidopsis accelerates seed germination by 23.77%,bolting by 6.93%,and flowering by 5.92%.This study provides not only an expanded genomic resource for gymnosperms but also a candidate gene(LkTOE1-2)to shorten the juvenile phase of larch via molecular breeding.展开更多
摘要Backgrounds:Somatization and eating-related problems in adolescents living in residential care may be shaped by the interplay of risk and protective factors,including gender,relational trauma,attachment patterns,emotional intelligence,and perceived social support.This study examined how gender,relational trauma,attachment dimensions,resilience,and emotional intelligence contribute to the presence of somatic and eating difficulties in this population.Methods:The sample included 46 adolescents(63%female;ages 12–17,Mean=14.85,Standard Deviation(SD)=1.49)residing in child protection institutions in Uruguay.Participants completed self-report measures assessing childhood relational trauma(CaMir),attachment dimensions(anxiety and avoidance),resilience,emotional intelligence(adaptability and stress management),social support(MOS),and psychosocial adjustment(SENA subscales of somatization and eating problems).Using a fuzzy-set Qualitative Comparative Analysis(fsQCA)approach,distinct configurations of risk and protective factors associated with elevated levels of somatization and eating problems were identified.Results:Relational trauma and attachment anxiety showed moderate associations with both somatization and eating problems(r=0.52–0.57,p<0.01),whereas stress management was negatively associated with both outcomes(r=−0.37 to−0.47,p<0.05).FsQCA revealed multiple configurations of risk and protective factors explaining 81–90%of cases,with solution consistencies ranging from 0.83 to 0.87.Results suggest that relational trauma and attachment anxiety are key risk conditions,whereas resilience,emotional regulation,and perceived social support function as protective factors.Conclusions:Findings highlight the importance of considering multifactorial patterns of vulnerability and protection rather than single predictors and underscore the need for tailored interventions that strengthen resilience and emotional skills while addressing the impact of early relational trauma.
摘要Background: Somatic symptoms that are not attributable to organic pathology are common in general practice settings however, data in most parts of Africa including southern Nigeria are still scarce. The aim of our study was to examine such somatic symptoms reported by patients attending a primary care facility at a tertiary hospital in southern Nigeria as well as to motivate future research in this area. Method: The study was conducted at the General Out Patient Clinic (GOPC) of the University of Calabar Teaching Hospital (UCTH). It was a cross sectional study in which data were obtained from the case notes of 115 patients that presented in the clinic with somatic complaints which could not be attributed to organic pathology by their physicians. Results: While 46 of the patients were males, 69 were females. Their mean age was 37.7 (SD = 11.9). Internal heat, crawling sensation, body pains and palpitations were the most prevalent symptoms reported by the patients. When the symptoms were sorted into various groups, the “subjective abnormal bodily sensation” was the most prevalent and far outnumbered the “pseudo neurological symptoms”. Conclusion: A number of patients attending the GOPC of the UCTH seek consultations for medically unexplained somatic symptom. The most prevalent of these symptoms are internal heat and crawling sensations both of which are not stated in the criteria recognized by the International Classification of Diseases—version 10 (ICD-10) for the diagnosis of psychiatric disorders.
摘要Objective: this paper mainly analyzes the clinical characteristics of somatization disorder, and to enhance the understanding of medical staff. Methods: use the medical staff to investigate 90 patients with somatization disorder and analyze them based on the survey results. Results: the incidence of somatization disorder in our hospital is not remarkable in gender level, mostly showed chronic delay, the average course of about 7-8 years; somatization disorder usually occurs in junior school culture and below; in clinical symptoms, such as 72.9% and musculoskeletal system 74.9% and 16.9% genitourinary system. Conclusions: somatization disorders mostly occur in poorly educated populations, and clinical symptoms will involve multiple systemic organs in the human body, so targeted research is needed to address the occurrence of the problem.

摘要Somatic hybridization offers a promising approach to enhance genetic diversity in cauliflower,a crop characterized by a narrow genetic base.In this study,we investigated epigenetic remodeling and associated phenotypic changes in the Brassica nigra-derived somatic hybrid introgression line‘Y15-15-116'and its recipient parent‘Korso'.‘Y15-15-116'exhibits early maturity,pale-green curd,and loose texture,whereas‘Korso'displays late maturity with white,compact curd.Combined analysis of wholegenome bisulfite sequencing and transcriptome revealed that somatic hybridization induced significant alterations in CHG and CHH methylation levels,particularly in transposable elements(TEs).

摘要The etiopathogenesis of gastrointestinal diseases is varied in nature.Various etiogenic factors described are infective,inflammatory,viral,bacterial,parasitic,dietary and lifestyle change.Rare causative agents are immunological,and others associated as idiopathic,are undiagnosed by all possible means.Some of the rare diseases are congenital in nature,passing from the parent to the child.Many of the undiagnosed diseases are now being diagnosed as genetic and the genes have been implicated as a causative agent.There is a search for newer treatments for such diseases,which is called genomic medicine.Genomic medicine is an emerging medical discipline that involves the use of genomic information about an individual.This is used both for diagnostic as well as therapeutic decisions to improve the current health domain and pave the way for policymakers for its clinical use.In the developing era of precision medicine,genomics,epigenomics,environmental exposure,and other data would be used to more accurately guide individual diagnosis and treatment.Genomic medicine is already making an impact in the fields of oncology,pharmacology,rare,infectious and many undiagnosed diseases.It is beginning to fuel new approaches in certain medical specialties.Oncology is at the leading edge of incorporating genomics,as diagnostics for genetic and genomic markers are increasingly included in cancer screening,and to guide tailored treatment strategies.Genetics and genetic medicine have been reported to play a role in gastroenterology in several ways,including genetic testing(hereditary pancreatitis and hereditary gastrointestinal cancer syndromes).Genetic testing can also help subtype diseases,such as classifying pancreatitis as idiopathic or hereditary.Gene therapy is a promising approach for treating gastrointestinal diseases that are not effectively treated by conventional pharmaceuticals and surgeries.Gene therapy strategies include gene addition,gene editing,messenger RNA therapy,and gene silencing.Understanding genetic determinants,advances in genetics,have led to a better understanding of the genetic factors that contribute to human disease.Family-member risk stratification and genetic diagnosis can help identify family members who are at risk,which can lead to preventive treatments,lifestyle recommendations,and routine follow ups.Selecting target genes helps identify the gene targets associated with each gastrointestinal disease.Common gastrointestinal diseases associated with genetic abnormalities include-inflammatory bowel disease,gastroesophageal reflux disease,non-alcoholic fatty liver disease,and irritable bowel syndrome.With advancing tools and technology,research in the search of newer and individualized treatment,genes and genetic medicines are expected to play a significant role in human health and gastroenterology.

摘要Tumor is generally believed to originate from a single cell,which develops into a genetically heterogeneous population after continuous cell divisions under the principle of“survival of the fittest”.Tumor evolution is a dynamic process,and reconstruction of the disappeared history is challenging but crucial for both understanding the mechanism and personalized therapy.Genetic alterations can be used as molecular clues to tumor evolution,with somatic mutations as a key tracer.Although bulk-level sequencing of a large number of tumor specimens has provided useful information on the mutational landscape,it is not easy to utilize such data for evolutionary history study.This is because bulk-level mutation prevalence may be confounded by various factors,such as tumor purity,copy number alterations,and more severely,the overlapping of prevalence ranges for mutations from similarly sized subclones.Single-cell mutational profiling overcomes such challenges,enabling precise identification of co-mutation groups and tumor subclones.This further enables successful reconstruction of tumor evolution,even for a solid tumor sample collected at a single time point.Technical and analytical advancements in single-cell profiling,such as mutation calling from single-cell RNA-Seq or spatial transcriptomic data,will provide further insights into tumor evolution and cancer treatment.

摘要Interoception refers to the sensation and perception of internal bodily sensations,and may be related to depressive symptoms.Schemata concerning the body vary across different cultures and may influence interoception and symptom presentations of depression.This study explored the relationship between interoception,depressive symptoms,and schema of somatic focus in Chinese people with subsyndromal depression.Thirty-nine individuals with subsyndromal depression(SD)and 40 healthy controls(HCs)were assessed at baseline and after 3 months.Participants completed the self-report questionnaires for assessing interoceptive sensibility,somatic and psychological symptoms of depression,and somatization tendency.They also completed the heartbeat perception behavioral task for estimating interoceptive accuracy.The results showed that both the SD and the HC groups showed similar interoceptive accuracy,although the SD group showed heightened interoceptive sensibility.The discrepancy between interoceptive sensibility and interoceptive accuracy is termed the interoceptive trait prediction error(ITPE).The ITPE was positive in SD participants but was negative in HCs.In the entire sample,interoceptive sensibility and the ITPE were correlated with somatic symptoms rather than with psychological symptoms of depression.Interoceptive sensibility partially mediated the relationship between somatization tendency and somatic symptoms,after controlling for psychological symptoms of depression.These results remained stable after 3 months.The shortcomings of the present study were a lack of clinical interview to ascertain diagnosis and a short follow-up duration.In conclusion,our study suggests that altered interoception occurs in subsyndromal depression.Interoception is related to somatic symptoms of depression.The schema of body was related to depressive symptoms,partially through interoception,in Chinese people with subsyndromal depression.

摘要Genetic transformation and somatic embryogenesis(SE)are essential biotechnological tools for cotton improvement.Recent advances in transcriptomics,proteomics,epigenetics,and RNA interference(RNAi)have enabled us to better understand the SE regulatory pathways.This review summarizes the findings of several key research studies,focusing on the roles of transcription factors(e.g.,GhAGL15s,GhL1L1,GhWOX),hormonal signaling(auxin and cytokinin),DNA methylation,and small RNAs in SE initiation and embryogenic callus formation.In addition,we discuss recent breakthroughs in Agrobacterium-mediated and RNAi-based transformation.Challenges such as genotype recalcitrance and somaclonal variation,along with new approaches to improve SE efficiency,are also addressed.
摘要BACKGROUND Laparoscopic appendectomy is the gold standard for treatment of acute appendicitis;however,postoperative pain characterized by a visceral and somatic component with separate neural pathways still interfere with early recovery.Transversus abdominis plane block(TAPB)relieves somatic pain,and erector spinae plane block(ESPB)may provide additional visceral pain relief by paravertebral and sympathetic chain blockade.Due to the lack of distinction between these two types of pain in previous study comparisons,we performed this retrospective analysis.AIM To compare the analgesic efficacy of ESPB vs TAPB for visceral pain and somatic pain following laparoscopic appendectomy.METHODS A retrospective analysis was conducted on 183 patients who underwent laparoscopic appendectomy with ultrasound-guided nerve blocks between January 2021 and December 2024,including 91 patients in the ESPB group and 92 patients in the TAPB group.Both groups received bilateral blocks with 0.375%ropivacaine at a total volume of 30 mL.Visual Analog Scale(VAS)was used to assess visceral pain and somatic pain scores at rest and during activity at 2,6,12,24,and 48 hours postoperatively.Rescue analgesia within 48 hours postoperatively,postoperative recovery quality indicators,incidence of postoperative nausea and vomiting,and patient satisfaction with analgesia were recorded.RESULTS The ESPB group had significantly lower visceral pain VAS scores at rest and during activity at 2 hours,6 hours,and 12 hours postoperatively compared with the TAPB group(P0.05).There were no significant differences in somatic pain VAS scores between the two groups at any time point(P>0.05).The ESPB group had a lower rescue analgesia rate than the TAPB group(13.2%vs 26.1%,P=0.029),less tramadol consumption(P=0.012),and higher satisfaction scores with analgesia(P=0.013).There were no significant differences between the two groups in time to first ambulation,time to first flatus,postoperative hospital stay,or incidence of nausea and vomiting(P>0.05).No nerve block-related complications occurred in either group.CONCLUSION Compared with TAPB,ESPB provides better analgesia for visceral pain in the early postoperative period following laparoscopic appendectomy,while both techniques demonstrate equivalent analgesic effects for somatic pain.For patients with visceral pain as the predominant component postoperatively,ESPB may be a more appropriate choice.

摘要Objectives:Recently,how family-related factors influence employees’mental health has garnered increasing attention from researchers and practitioners.Drawing on the cognitive appraisal theory of stress,this study aims to examine how and when family financial stress affects the employees’mental health and investigate the mediating role of performance stress and the moderating role of workplace competition.Methods:A cross-sectional survey was conducted with 23,520 Chinese employees by using a voluntary and anonymous structured questionnaire,which included family financial stress,performance stress,symptom checklist 90(SCL-90)scale,and workplace competition.The data were analyzed using SPSS 26.0 software and macro PROCESS.Results:The analysis of the mediating effect showed that performance stress mediated the relationship between family financial stress and psychological depression(b=0.064,SE=0.002,p<0.0001)and physical somatization(b=0.042,SE=0.002,p<0.0001),indicating spillover effects of stress from home to workplace.The moderating mediation analysis revealed that the crossover effects were amplified by workplace competition.For psychological depression,index of moderated mediation was:b=0.012,SE=0.001,p<0.001;For physical somatization,index of moderated mediation was:b=0.008,SE=0.001,p<0.001.Conclusion:Performance stress acts as a mediator in the link between family financial stress and mental health.Furthermore,the mediating effects was amplified by workplace competition.These findings suggest that workplace competition may serve to exacerbate the negative spillover effects from home to work through the mechanism of workrelated stress.Organizations should consider implementing supportive measures to mitigate family financial stress,such as providing financial counseling and fostering a collaborative work environment,to reduce the adverse effects of family financial stress on employees’mental health.

摘要Litchi chinensis Sonn.is an important economic fruit tree in tropical and subtropical regions.Regrettably,the efficiency of plant regeneration via somatic embryogenesis in litchi is typically low due to the poor conversion of embryos to plants.The purpose of this study was to establish a regeneration system via somatic embryogenesis from immature embryos explants in‘Heiye'cultivar of litchi.Our results demonstrated that MS medium supplemented with 2.0 mg L-12,4-D was optimal for callus induction.For somatic embryo(SE)induction,MS medium containing0.5 g L-1 activated charcoal(AC)was the most effective,while the use of zeatin(ZT)and thidiazuron(TDZ)resulted in abnormal somatic embryos.The rooting and regeneration rate of 2.15%and 17.5%,respectively,were achieved using MS medium supplemented with 0.5 g L-1 AC.Furthermore,transcriptome analysis was performed on embryogenic callus(EC),globular embryo(GE),and heart embryo(HE)to explore the molecular mechanisms of early somatic embryogenesis.2,587 common differentially expressed genes(DEGs)between EC_vs_GE and EC_vs_HE were identified,and the expression patterns of these common DEGs were separated into twelve major clusters.GO annotation and KEGG pathway analysis revealed that these common DEGs were implicated in plant hormone signal transduction,auxin-activated signaling pathway,and other biological processes.Additionally,differentially expressed transcription factors were identified,and the function of LcBBM2 which is specifically highly expressed during early somatic embryogenesis was verified.Overexpression of LcBBM2 in tomato promotes callus and shoot formation.Therefore,this study can provide a theoretical basis and technical support for genetic breeding improvement of litchi.

摘要Somatic cell nuclear transfer(SCNT)has been successfully employed across various mammalian species,yet cloned animals consistently exhibit low pregnancy rates,primarily due to placental abnormalities such as hyperplasia and hypertrophy.This study investigated the involvement of the Hippo signaling pathway in aberrant placentaldevelopmentinSCNT-inducedbovine pregnancies.SCNT-derived cattle exhibited placental hypertrophy,including enlarged abdominal circumference and altered placental cotyledon morphology.RNA sequencing analysis indicated significant dysregulation of Hippo signaling pathway genes in SCNT placentas.Coexpression of YAP1 and CCND1 was observed in cloned blastocysts,placental tissues,and bovine placental mesenchymal stem cells(bPMSCs).Manipulation of YAP1expression demonstrated the capacity to regulate bPMSC proliferation.Experimental assays confirmed the direct binding of YAP1 to CCND1,which subsequently promoted CCND1 expression in bPMSCs.Furthermore,inhibition of CDK6,a downstream target of CCND1,attenuated SCNT bPMSC proliferation.This study identified YAP1 as a key regulatory component within the Hippo signaling pathway that drives placental hyperplasia in cloned cattle through up-regulation of CCND1-CDK6 expression,facilitating cell cycle progression.These findings offer potential avenues for enhancing cloning efficiency,with implications for evolutionary biology and the conservation of valuable germplasm resources.

摘要Somatic variants in the cancer genome influence gene expression through diverse mechanisms depending on their specific locations.However,a systematic evaluation of the effects of somatic variants located in 3'untranslated regions(3'UTRs)on alternative polyadenylation(APA)of m RNA remains lacking.In this study,we analyze 10,199 tumor samples across 32 cancer types and identify 1333 somatic single nucleotide variants(SNVs)associated with abnormal 3'UTR APA.Mechanistically,these 3'UTR SNVs can alter cisregulatory elements,such as the poly(A)signal and UGUA motif,leading to changes in APA.Minigene assays confirm that 3'UTR SNVs in multiple genes,including RPS23 and CHTOP,induce aberrant APA.Among affected genes,62 exhibit differential stability between tandem 3'UTR isoforms,including HSPA4and UCK2,validated by experimental assays.Finally,we establish that SNV-related abnormal APA usage serves as an additional layer of expression regulation for tumor-suppressor gene HMGN2 in breast cancer.Collectively,this study reveals 3'UTR APA as a critical mechanism mediating the functional impact of somatic noncoding variants in human cancers.

摘要Bananas(Musa spp.)are among the top-produced food crops,serving as a primary source of food for millions of people.Cultivated bananas originated primarily from the wild diploid species Musa acuminata(A genome)and Musa balbisiana(B genome)through intraand interspecific hybridization and selections via somatic variation.Following the publication of complete A-and B-genome sequences,prospects for complementary studies on S-and T-genome traits,key gene identification for yield,ripening,quality,and stress resistance,and advances in molecular breeding have significantly expanded.In this review,latest research progress on banana A,B,S,and T genomes is briefly summarized,highlighting key advances in banana cytoplasmic inheritance,flower and fruit development,sterility,and parthenocarpy,postharvest ripening and quality regulation,and biotic and abiotic stress resistance associated with desirable economic traits.We provide updates on transgenic,gene editing,and molecular breeding.We also explore future directions for banana breeding and genetic improvement.

摘要Background Cotton is an industrial crop renowned for its multifaceted applications in the textiles,pharmaceuticals,and biofuel industries.Plant regeneration through somatic embryogenesis(SE)plays a crucial role in the genetic improvement of cotton.There is a strong correlation between SE and zygotic embryogenesis(ZE)in plants.Furthermore,the strategy of ectopic expression of cotton genes into the model plant Arabidopsis has been a widely accepted approach for functional study.Result Based on previous spatial transcriptomics of cotton somatic embryos,two genes,Gh HAT5 and Gh CRK29,were identified.They are highly expressed in cotyledon and epidermal cells of cotton cotyledonary embryos,respectively.In this study,Gh HAT5 and Gh CRK29 were ectopically expressed in Arabidopsis to investigate their functions.The result showed that in Arabidopsis zygotic embryos,the overexpression of Gh HAT5 promoted the development of apical embryonic upper-tier cells and embryonic cotyledon,while the overexpression of Gh CRK29 promoted the development of apical embryonic lower-tier cells and embryonic radicle.Given the similarities between somatic and zygotic embryogenesis,these findings suggest that Gh HAT5 and Gh CRK29 are involved in cotton SE.We also speculate that these genes may promote the expression of the Arabidopsis endogenous gene At SCR,which is crucial for embryonic development.Conclusion These results revealed that Gh HAT5 and Gh CRK29 regulate embryonic development and are essential in advancing our understanding of cotton SE and facilitating targeted genetic manipulation strategies to improve industrial crop traits and agricultural sustainability.

摘要Litchi has great economic significance as a global fruit crop.However,the advancement of litchi functional genomics has encountered substantial obstacles due to its recalcitrance to stable transformation.Here,we present an efficacious Agrobacterium tumefaciens-mediated transformation system in somatic embryos of‘Heiye'litchi.This system was developed through the optimization of key variables encompassing explant selection,A.tumefaciens strain delineation,bacterium concentration,infection duration,and infection methodology.The subsequent validation of the transformation technique in litchi was realized through the ectopic expression of LcMYB1,resulting in the generation of transgenic calli.However,the differentiation of transgenic calli into somatic embryos encountered substantial challenges.To delineate the intricate molecular underpinnings of LcMYB1's inhibitory role in somatic embryo induction,a comprehensive transcriptome analysis was conducted that encompassed embryogenic calli(C),globular embryos(G),and transgenic calli(TC).A total of 1,166 common differentially expressed genes(DEGs)were identified between C-vs.-G and C-vs.-TC.Gene Ontology(GO)annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis revealed that these common DEGs were mostly related to plant hormone signal transduction pathways.Furthermore,RT-qPCR corroborated the pronounced down-regulation of numerous genes that are associated with somatic embryo induction within the transgenic calli.The development of this transformation system provides valuable support for functional genomics research in litchi.

摘要The CRISPR-Cas-based gene targeting(GT)method has enabled precise modifications of genomic DNA ranging from single base to several kilobase scales through homologous recombination(HR).In plant somatic cells,canonical non-homologous end-joining(cNHEJ)is the predominant mechanism for repairing double-stranded breaks(DSBs),thus limiting the HR-mediated GT.In this study,we implemented an approach to shift the repair pathway preference toward HR by using a dominant-negative ku80 mutant protein(KUDN)to disrupt the initiation of cNHEJ.The employment of KUDN conferred a 1.71-to 3.55-fold improvement in GT efficiency at the callus stage.When we screened transformants,there was a more remarkable increase in GT efficiency,ranging from 1.62-to 9.84-fold,at two specific tomato loci,SlHKT1;2 and SlEPSPS1.With practical levels of efficiency,this enhanced KUDN-based GT tool successfully facilitated a 9-bp addition at an additional locus,SlCAB13.These findings provide another promising method for more efficient and precise plant breeding.

摘要Conifers pose challenges for breeding programs due to their extended vegetative phases.Despite the critical role of APETALA2(AP2)in regulating phase transitions,the AP2/ERF superfamily remains largely unexplored in gymnosperms.Here,the first genome-wide analysis of the AP2/ERF superfamily in a conifer,Larix kaempferi(Japanese larch)is presented,and 374 members were identified.Among all eight paralogs,four euAP2 lineage genes,TARGET OF EATs(TOEs),exhibit age-decreased expression patterns.Functional characterization of LkTOE1-2 demonstrates its involvement in somatic embryogenesis and seed germination.Importantly,the RUBY reporter system confirmed an enhanced promoter activity in somatic embryo maturation.Over-expression of LkTOE1-2 in Arabidopsis accelerates seed germination by 23.77%,bolting by 6.93%,and flowering by 5.92%.This study provides not only an expanded genomic resource for gymnosperms but also a candidate gene(LkTOE1-2)to shorten the juvenile phase of larch via molecular breeding.
