Anxiety-like behaviors increased in juvenile rats in the streptozotocin-induced diabetes model during the early stage 认领 引用

BACKGROUND Diabetes is an endocrinopathy characterized by the inability to produce insulin or insufficient insulin production because of the destruction of beta cells in the pancreas caused by autoimmune or other epidemiological factors.Diabetes can lead to physical and psychological complications.AIM To investigate early behavioral changes in an experimental diabetes model in juvenile rats.METHODS In this study,a total of 14 juvenile male Wistar rats(7 rats per group)were used.To induce the diabetes model,120 mg/kg streptozotocin(STZ)was administered,while the control group received 1 mL/kg physiological saline.The open field test and the hole-board test were performed for five minutes each.Unpaired comparisons were evaluated using Student’s t test or the Mann-Whitney U test.Correlations were analyzed using the Pearson test.RESULTS Severe hyperglycemia was observed in rats in the diabetes mellitus model induced by STZ,and all diabetic rats displayed glucose levels above 400 mg/dL.In the open-field test,the time spent in the central area,the number of rearings,and the total distance traveled were significantly lower in diabetic rats than in naive rats(P<0.05).In the hole-board test,the duration and frequency of total head dipping were considerably lower in diabetic rats than in healthy rats(P<0.01).The grooming time(P<0.01)and freezing time(P<0.0001)increased dramatically in the diabetes mellitus group.A remarkable correlation was found between behavioral changes and hyperglycemia.CONCLUSION Our data indicate that early-stage diabetes induced by STZ increases anxiety-like behaviors and impairs exploratory behaviors.These behavioral changes may also occur in children with undiagnosed or untreated hyperglycemia and can provide important clues in clinical practice. 展开更多

BACKGROUND Akkermansia muciniphila(A.muciniphila)has been shown to have positive effects on various metabolic diseases and partially prevent the onset of spontaneous type 1 diabetes mellitus(T1DM)in nonobese diabetic mice;however,its therapeutic efficacy in T1DM mice that have already developed T1DM remains unclear.AIM To assess the effects of A.muciniphila intervention on the intestinal barrier,immune parameters[regulatory T(Tregs)cells/T helper 1 cells balance,signal transducer and activator of transcription 1(STAT1)uclear factor kappa-B(NF-κB)signal]and intestinal flora in a streptozotocin(STZ)-induced mouse model of T1DM.METHODS Thirty male C57BL/6 mice were randomized into three groups(n=10 for each):Normal control(NC group),STZ-induced T1DM(STZ group),and A.muciniphilatreated T1DM(A.muciniphila group).T1DM was induced with STZ(50 mg/kg/day intraperitoneally,5 days).Body weight,blood glucose,and water intake were monitored weekly.T1DM was modelled in all STZ-and A.muciniphila-treated mice(random blood glucose>16.7 mmol/L).A.muciniphila group mice received oral A.muciniphila(5×107 colony-forming units/mouse)for 3 days.Following sacrifice,pancreatic histopathology,cytokines in splenic tissue,colonic zonula occludens-1(ZO-1)/zonulin expression,cluster of differentiation(CD)4+/CD8+T-cell ratios,forkhead box P3(FoxP3+)Tregs,STAT1/NF-κB p65 signaling,and gut microbiota composition were analyzed.RESULTS Compared with STZ group alone,A.muciniphila group did not affect metabolic parameters.Histopathologically,STZ group and A.muciniphila group pancreatic islet cells underwent vacuolar degeneration and necrosis,exhibiting reduced counts and significantly decreased insulin positivity(P<0.05 vs NC),with no intergroup differences.Flow cytometry and enzyme-linked immunosorbent assay revealed elevated tumor necrosis factoralpha(TNF-α),transforming growth factor-beta(TGF-β),and zonulin levels and decreased ZO-1 expression in STZ vs NC mice(P<0.05).Compared with STZ alone,A.muciniphila group reduced TNF-α,TGF-β,and zonulin levels while increasing the CD4+/CD8+ratio,FoxP3+Tregs,interleukin-4,and ZO-1(P<0.05).Colonic NF-κB p65 expression was higher in STZ vs NC mice(P<0.05),with no significant A.muciniphila group/NC difference after the intervention of A.muciniphila.The expression of NF-κB p65 in A.muciniphila group was lower than that in STZ group.STAT1 expression was lower in A.muciniphila vs STZ mice(P<0.05).16S sequencing revealed reduced Actinobacteria abundance in A.muciniphila vs STZ mice(P<0.05).CONCLUSION Short-term intervention with A.muciniphila has shown positive effects on immune response parameters,exemplified by Treg-mediated restoration of immune tolerance,which may be associated with intestinal barrier enhancement and the decreased abundance of intestinal Actinobacteria. 展开更多

AIM: To clarify the mechanism underlying the antidiabetic activities of cortex cinnamomi extract (CCE). METHODS: To induce in vivo diabetes, mice were injected with streptozotocin (STZ) via a tail vein (100 mg STZ/kg body weight). To determine the effects of CCE, mice were administered CCE twice daily for 7 d by oral gavage starting 1 wk before the STZ injection. Blood glucose and plasma insulin concentration were measured as an index of diabetes. Also, to induce cytotoxicity of RINm5F cells, we treated with cytokines (IL-1β (2.0 ng/mL) and IFN-γ (100 U/mL)). Cell viability and nitric oxide production were measured colorimetrically. Inducible nitric oxide synthase (iNOS) mRNA and protein expression were determined by RT-PCR and Western blotting, respectively. The activation of NF-κβ was assayed by using gel mobility shift assays of nuclear extracts. RESULTS: Treatment of mice with STZ resulted in hyperglycemia and hypoinsulinemia, which was further evidenced by immunohistochemical staining of islets. However, the diabetogenic effects of STZ were completely prevented when mice were pretreated with CCE. The inhibitory effect of CCE on STZ-induced hyperglycemia was mediated through the suppression of iNOS expression. In rat insulinoma RINm5F cells,CCE completely protected against interleukin-1β and interferon-y-mediated cytotoxicity. Moreover, RINm5F cells incubated with CCE showed significant reductions in interleukin-1β and interferon-y-induced nitric oxide production and in iNOS mRNA and protein expression, and these findings correlated well with in vivo observations. CONCLUSION: The molecular mechanism by which CCE inhibits iNOS gene expression appears to involve the inhibition of NF-κβ activation, These results reveal the possible therapeutic value of CCE for the prevention of diabetes mellitus progression, 展开更多

METHODS:The antioxidant effect of COSs on pancreatic islet cells was detected under optical microscopy and with colorimetric assay and gel electrophoresis.The activities of glutathione peroxidase and superoxide dismutase,total antioxidant capacity,and content of malondialdehyde in serum and tissue slices of pancreas were examined after 60 d to determine the effect of COSs in streptozotocin-induced diabetes in rats. RESULTS:COSs can prohibit the apoptosis of pancreatic islet cells.All concentrations of COSs can improve the capability of total antioxidant capacity and activity of superoxide dismutase and decrease the content of malondialdehyde drastically.Morphological investigation in the pancreas showed that COSs have resulted in the reduction of islets,loss of pancreatic cells,and nuclear pyknosis of pancreatic cells. CONCLUSION:COSs possess various biological activities and can be used in the treatment of diabetes mellitus. 展开更多

Objective:To investigate the antihyperglycemic and antihyperlipidemie properties of hydroalcoholic extract of fruits of Sapindus mukorossi Caerlen and its beneficial effect on haeniatological parameters with histopathological analysis in streptpzotocin induced diabetic rats.Methods:Sapindus mukorossi fruits extract(250 and 500 mg/kg body weight) and standard drug glybenclamide(0.5 mg/kg body weight) were administered to diabetic rats.Effect of extract on hyperglycemia,hyperlipidemia and hematological parameters was studied in diabetic rats.Histopathological changes in diabetic rat pancreas were also observed after extract and glybenclamide treatment.Results:Daily oral administration of Sapindus mukorossi fruits extract(250 and 500 mg/kg body weight) and glybenclamide for 20 days showed beneficial effects on blood glucose level(P【0.01) and lipid level.The extract has a favorable effect on the histopathological changes of the pancreas in streptozotocin induced diabetes.Conclusions: These findings reveal that the hydroalcoholic extract of Sapindus mukorossi fruits extract possesses antihyperglycemic and antihyperlipidemie properties.In addition.the extract can prevent various complications of diabetes and improve some haeniatological parameters. 展开更多

Objective: The present study aims at determining the stability of a popular type 2 diabetes rat model induced by a high-fat diet combined with a low-dose streptozotocin injection. Methods: Wistar rats were fed with a high-fat diet for 8 weeks followed by a one-time injection of 25 or 35 mg/kg streptozotocin to induce type 2 diabetes. Then the diabetic rats were fed with regular diet/high-fat diet for 4 weeks. Changes in biochemical parameters were monitored during the 4 weeks. Results: All the rats developed more severe dyslipidemia and hepatic dysfunction after streptozotocin injection. The features of 35 mg/kg streptozotocin rats more resembled type 1 diabetes with decreased body weight and blood insulin. Rats with 25 mg/kg streptozotocin followed by normal diet feeding showed normalized blood glucose level and pancreatic structure, indicating that normal diet might help recovery from certain symptoms of type 2 diabetes. In comparison, diabetic rats fed with high-fat diet presented decreased but relatively stable blood glucose level, and this was significantly higher than that of the control group(P〈0.05). Conclusions: This model easily recovers with normal diet feeding. A high-fat diet is suggested as the background diet in future pharmacological studies using this model. 展开更多

Objective To investigate the pretreatment effects of Rhodiola rosea(R.rosea)extract on cognitive dysfunction,oxidative stress in hippocampus and hippocampal neuron injury in a rat model of Alzheimer's disease(AD).Methods Male Sprague-Dawley rats were pretreated with R.rosea extract at doses of 1.5,3.0,and 6.0 g/kg for 3 weeks,followed by bilateral intracerebroventricular injection with streptozotocin(1.5 mg/kg)on days 1 and 3.Behavioral alterations were monitored after 2 weeks from the lesion using Morris water maze task.Three weeks after the lesion,the rats were sacrificed for measuring the malondialdehyde(MDA),glutathione reductase(GR)and reduced glutathione(GSH)levels in hippocampus and histopathology of hippocampal neurons.Results The MDA level was significantly increased while the GR and GSH levels were significantly decreased with striking impairments in spatial learning and memory and severe damage to hippocampal neurons in the model rat induced by intracerebroventricular injection of streptozotocin.These abnormalities were significantly improved by pretreatment with R.rosea extract(3.0 g/kg).Conclusion R.rosea extract can protect rats against cognitive deficits,neuronal injury and oxidative stress induced by intracerebroventricular injection of streptozotocin,and may be used as a potential agent in treatment of neurodegenerative diseases such as AD. 展开更多

METHODS: Mice were injected with streptozotocin viaa tail vein to induce diabetes. Rat insulinoma RINm5F cells and isolated rat islets were treated with interleukin1β and interferon-γ to induce cytotoxicity. RESULTS: Treatment of mice with streptozotocin resulted in hyperglycemia and hypoinsulinemia, which was conf irmed by immunohistochemical staining of the islets. The diabetogenic effects of streptozotocin were completely abolished when mice were pretreated with NJE. Inhibition of streptozotocin-induced hyperglycemia by NJE was mediated by suppression of nuclear factor (NF)-κB activation. In addition, NJE protected against cytokine-mediated cytotoxicity. Incubation of RINm5F cells and islets with NJE resulted in a signif icant reduction in cytokine-induced NF-κB activation and downstream events, inducible nitric oxide synthase expression and nitric oxide production. The protective effect of NJE was further demonstrated by the normal insulin secretion of cytokine-treated islets in response to glucose. CONCLUSION: NJE provided resistance to pancreatic β-cell damage from cytokine or streptozotocin treatment. The β-cell protective effect of NJE is mediated by suppressing NF-κB activation. 展开更多

AIM:To investigate the mechanism behindβ-cell regeneration in neonatal rat pancreas treated with strep-tozotocin(STZ).METHODS:Neonatal Sprague Dawley rats were intra-peritoneally injected with 70 mg/kg STZ.Body weight,pancreas weight and blood glucose were recorded every two days after the treatment.To identify the expression and location of transcription factors in the rat pancreas,double immunofluorescent staining was performed using antibodies to specific cell markers and transcription factors.RESULTS:Expression of Neurogenin 3(Ngn3),a marker for endocrine precursor cells,was observed by immunofluorescence in a fewβ-cells and many a-cells.The expression reached a peak 12 d after treatment.Pax4,a transcription factor that lies downstream of Ngn3 and plays an important role inβ-cell differentiation,was also expressed in theα-cells of STZ-treated rats.We did not observe significant changes in Nkx6.1,which is essential forβ-cell maturation in the treated rats.CONCLUSION:α-cells dedifferentiated into endocrine precursor cells and acquired the ability to dedifferentiate in the neonatal rat pancreas after STZ treatment. 展开更多

Objective:To justify the use of African mistletoe(AM) Viscum album {V.album) in folkoric medicine to treat diabetes.Methods:In one experiment,the fasting blood glucose(FBG) leveb of diabetic rats were monitored for 4 h.Diabetic rats were treated with AM at doses of 50 mg/kg(AM1) and 100 mg/kg(AM2),glibenclamide(GB)(positive control) and saline solution(SS).In another experiment,diabetic rats were treated with AM2,GB and SS daily for 3 weeks.Results:AM1 and AM2 elicited significant(P【0.05) hypoglycaemic effects within 4 h of extract administration. AM1 and AM2 decreased the FBG by 41%and 49%,respectively,at 2 h.AM2 was found to lower FBC by 51%,relative to baseline,which was comparable to GB at 3 h.In the second experiment, AM2 and GB significantly(P【0.05) decreased the FBG by 34%and 51%,respectively.This was followed by marked decrease in levels of HbAlC in AM2- and GB- treated diabetic rats.AM2 significantly(P【0.05) decreased the STZ-induced increase in levels of serum triglyceride,urea, lactate dehydrogenase,α-amylase and low-density lipoprotein-cholesterol.Furthermore, diabetic rats treated with AM2 had significanlly(P【0.05) elevated high-density lipoprotein-cholesterol. In contrast,STZ administration produced insignificant(P【0.05) effect on the levels of serum creatinine and total bilirubin.Conclusions:Extract of African mistletoe has anti-diabetic and anti-hyperlipidemic effects in STZ-diabetic rats.AM may find clinical application in the amelioration of diabetes-induced lipid disorders. 展开更多

Painful diabetic neuropathy(PDN)is a diabetes mellitus complication.Unfortunately,the mechanisms underlying PDN are still poorly understood.Adenosine triphosphate(ATP)-gated P2X7 receptor(P2X7R)plays a pivotal role in non-diabetic neuropathic pain,but little is known about its effects on streptozotocin(STZ)-induced peripheral neuropathy.Here,we explored whether spinal cord P2X7R was correlated with the generation of mechanical allodynia(MA)in STZ-induced type 1 diabetic neuropathy in mice.MA was assessed by measuring paw withdrawal thresholds and western blotting.Immunohistochemistry was applied to analyze the protein expression levels and localization of P2X7R.STZ-induced mice expressed increased P2X7R in the dorsal horn of the lumbar spinal cord during MA.Mice injected intrathecally with a selective antagonist of P2X7R and P2X7R knockout(KO)mice both presented attenuated progression of MA.Double-immunofluorescent labeling demonstrated that P2X7R-positive cells were mostly co-expressed with Iba1(a microglia marker).Our results suggest that P2X7R plays an important role in the development of MA and could be used as a cellular target for treating PDN. 展开更多

OBJECTIVE: To evaluate the effect of Nelumbo Nucifera leaf water extract (NNLE) on insulinoma (RIN) cells induced by interleukin-1β (1L-1β) and interferon-g (IFN-γ), and injured pancreatic β-cells induced by Streptozotocin (STZ) in rats. METHODS: The anti-oxidative effects of NNLE were assessed using 1,1 -diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide (NO) scavenging assays. The inhibitory effect of NNLE on a-glucosidase and DPP (dipeptidyl peptidase)-IV was measured in vitro. Pancreatic 1β-cell protective and insulin secretory effects were assessed, using 1L-1β and IFN-γ-inducedrat RIN cells. STZ-induced diabetic rats were treated with 50, 100, and 400 mg/kg NNLE for 4 weeks. The effects of NNLE on blood glucose (BG), body weight (BW), and lipid profiles were measured. RESULTS: NNLE inhibited DPPH, NO, aglucosidase, and DPP-Ⅳ which were directly linked to the function of β-cells. Furthermore, NNLE protected RIN cells from toxicity induced by 11-11β and IFN-γ, decreased NO production, and increased insulin secretion. NNLE caused a significant reduction in blood glucose, triglyceride (TG), total cholesterol (TC), blood urea nitrogen (BUN), and creatinine in STZ-induced diabetic rats. Furthermore, it significantly decreased BW loss in STZ-induced diabetic rats. CONCLUSION: Our results suggest that NNLE reduced the toxicity in insulinoma cells and increased insulin secretion in pancreatic β-cells in STZ-induced diabetic rats. 展开更多

Objective: To evaluate the antidiabetic and antioxidant ef ects of aqueous ethanolic extract of Caesalpinia ferrea(C. ferrea) leaf in normal and streptozotocin(STZ) induced diabetic rats.Methods: Male Sprague-Dawley rats divided into 6 groups of 6 rats each were assigned into diabetic and non-diabetic groups. Diabetes was induced in rats by single intraperitoneal administration of STZ(65 mg/kg body weight). C. ferrea extract at the doses of 250 and 500 mg/kg body weight was orally administered to both diabetic and non-diabetic animals for a period of 30 days. After completion of experimental duration serum, liver and pancreas were used for evaluating biochemical and histopathological changes.Results: Oral administration of C. ferrea leaf extract significantly reduced elevated serum glucose, α-amylase, liver function levels and signii cantly increased serum insulin, total protein and body weight as well as improved lipid proi le due to diabetes. Furthermore, the treatment resulted in a marked increase in glutathione peroxidase, superoxide dismutase, catalase and reduced glutathione, and diminished levels of lipid peroxidation in liver and pancreas of diabetic rats. Histopathological studies demonstrated the reduction in the pancreas and liver damage and coni rmed the biochemical i ndings.Conclusions: From the present study, it can be concluded that the C. ferrea leaf extract ef ectively improved hyperglycaemia while inhibiting the progression of oxidative stress in STZ-induced diabetic rats. Hence, it can be used in the management of diabetes mellitus. 展开更多

The polysaccharides from pumpkin fruit (PP) were obtained and purified by hot-water extraction, anion-exchange chromatography, and gel column chromatography. The physicochemical properties of PP were determined by gel filtration chromatography, gas chromatography, fourier transform infrared (FTIR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. Results indicated that the molecular weight of PP was about 23 kDa and PP was composed of D-Arabinose, D-Marmose, D-Glucose, and D-Galactose with a molar ratio of 1 : 7.79 : 70.32 : 7.05. FTIR and NMR spectra indicated that PP was the polysaccharide containing pyranose ring. Additionally, PP protected islets cells from streptozotocin (STZ) injury in vitro via increasing the levels of super-oxide dismutase (SOD) and malondialdehyde (MDA) and reducing the production of NO. The experiment of reverse transcriptase-polymerase chain reaction further proved that PP inhibited apoptosis via modulating the expression of Bax/Bcl-2 in STZ-damaged islet cells. In conclusion, PP could be explored as a novel agent for the treatment of diabetes mellitus. 展开更多

Objective:To investigate the antidiabetic and anti-lipemic effects of Cassia siamea methanolic leaves extract.Methods:The antidiabetic study was performed by measuring blood glucose level with elegance glucometer at weekly intervals i.e.0,7,14 and 21 in normal and streptozotocin induced diabetic rats.Total cholesterol,triglyceride and HDL-cholesterol were determined in normal and streptozotocin induces diabetic rats by autoanalyser.Glibenclamide was used as a reference drug at a dose of 10 mg/kg.Results:After the oral administration of extracts at doses of 250 mg/kg and 500 mg/kg for three weeks,blood glucose levels and body weights were significantly improved(P【0.01).Daily oral treatment with the extract also resulted in significantly reduction of serum cholesterol and triglycerides.HDL-cholesterol level was found to be improved to(P【0.01).Conclusions:The Cassia siamea leaf extract is useful in controlling blood glucose level as well as improving lipid metabolism and body weight in rats with induced diabetes. 展开更多

Objective To analyze the effects of Dan Huang Ming Mu Recipe(DHMMR)(a pharmaceutical preparation from herbs and having the function of replenishing vital essence,removing heat,promoting blood circulation and excreting pathogenic water) on diabetic retinopathy(DR) after retinal laser photocoagulation through regulating the expression of vascular endothelial growth factor(VEGF) and pigment epithelium-derived factor(PEDF).Methods Forty male Brown Norway(BN) rats were randomly divided into blank group(group A,10 BN rats) and model group(30 BN rats).DR models were induced by 40 mg/kg streptozotocin(STZ) and the body weight and blood glucose of rats were monitored.After 12-week injection,fluorescein fundus angiography(FFA) and histopathological examination were detected to confirm the successful establishment of DR models.Subsequently,the right eyes of model group rats were conducted retinal laser photocoagulation with the left eyes having no retinal laser photocoagulation and rats of the model group were randomly divided into group B(the model control group),group C(the positive control group),and group D(the DHMMR group),with 10 rats in each group.Rats of the group A and the group B were given vehicle,and the group C were given calcium dobesilate suspension by gavage,and the group D were given DHMMR by gavage.After 4-week gavage,FFA was carried out to observe the fundus microvascular change.Then,the rats were sacrificed to do histopathological examinations and the serum levels of P-selectin,cAMP,cGMP,and the relative expression of the protein of VEGF and PEDF in retina were detected.Results After 12-week STZ injection,the blood glucose of the model group were pronouncedly higher than the blank group(P < 0.01).Fundus micro-hemangioma changes were observed in the rats of the model group through FFA,and the rats of the blank group did not see any changes.The pictures of HE staining showed that the retinal structure of the model group was more disordered than the blank group.After 4-week gavage,treatments with DHMMR showed dramatic reduction of serum levels of Pselectin,cAMP and cGMP compared with the group B(P < 0.01).FFA and histopathological examinations of DHMMR-treated rats revealed significantly suppression of retinal edema,fundus microvascular destruction and retinal destruction distinguishing from the group B.And the relative expression of VEGF protein of the group D and the group A was markedly lower than that of the group B(P < 0.01).The relative expression of PEDF protein of the group D and the group A was dramatically higher than that of the group B to the contrary(P < 0.01).Conclusions DHMMR demonstrates pronounced suppressive effects on the progression of DR after retinal laser photocoagulation,through down-regulating VEGF and upregulating PEDF. 展开更多
