We used seedlings of the two subspecies originated from wild plants from the natural habitats in Sicily and cultivated in the Botanical Garden of Rome. Leaf morphological and physiological traits and growth dynamic were analyzed as well as the response to an imposed water stress experiment. The results underlined a higher relative growth rate in plant height (RGRH, cm·cm-1·d-1) in B. rupestris subsp. rupestris also attested by the highest plant height (H, cm), leaf area (LA, cm2), specific leaf area (SLA, cm·g-1), total leaf area per plant (TLA, cm2) and total plant dry mass (PDM, g plant-1). Moreover, the significantly highest net photosynthetic rates (A, μmol CO2 m-2·s-1) during the study period of this subspecies was related to the high Chlorophyll content (Chl, SPAD units). B. rupestris subsp. rupestris was also the most tolerant subspecies to imposed water stress showing the highest relative water content (RWC, %), A and water use efficiency (WUE, μmol·mmol-1).展开更多
Brassinosteroids(BRs)are extensively distributed in plants and play crucial roles throughout all stages of plant growth.Nevertheless,the molecular mechanism through which BRs influence postharvest senescence in pakcho...Brassinosteroids(BRs)are extensively distributed in plants and play crucial roles throughout all stages of plant growth.Nevertheless,the molecular mechanism through which BRs influence postharvest senescence in pakchoi remains elusive.Previous studies have demonstrated that the application of 1.5μM of the BRs analog 2,4-epibrassinolide(EBR)delayed the leaf senescence in harvested pakchoi.In this study,we constructed the EBR-delayed senescence transcriptome in pakchoi leaves and discovered that EBR modulates the expression of genes involved in the chlorophyll(Chl)metabolism pathway and the BRs pathway in pakchoi.Notably,we identified and characterized an EBR-suppressed,nucleus-localized WRKY transcription factor called BrWRKY8.BrWRKY8 is a highly expressed transcriptional activator in senescent leaves,targeting the promoters of the Chl degradation-associated gene BrSGR2 and the BRs degradation-associated gene BrCHI2,thereby promoting their expression.Overexpression of the BrWRKY8 gene accelerated the senescence process in Arabidopsis leaves,while EBR treatment mitigated the leaf senescence phenotype induced by BrWRKY8 overexpression.Conversely,silencing of BrWRKY8 through the virus-induced gene silencing extended the postharvest storage period of pakchoi.In conclusion,the newly discovered BRs-BrWRKY8 regulatory model in this study provides novel insights into BRs-mediated leaf senescence in pakchoi.展开更多

A survey was conducte...Various bacterial species are known to be agents causing soft rot of potatoes. The results of this study showed that potato soft rot is widely spread in different potato planting areas in Jordan. A survey was conducted through the years 2013-2015 to detect potato soft rot disease in Jordan, two hundred and four rotted potato samples were collected from different potato growing areas through different potato growing seasons. One hundred and thirty one bacterial isolates were isolated, purified on selective media and identified as Pectobacterium carotovorum subspecies carotovorum (Pcc) by different biochemical and physiological tests. Furthermore, 131 Pcc Jordanian (Jo) isolates were identified by PCR analysis of total DNA extracted from isolates that were biochemically identified as Pcc using universal primer Fd1/Rd1. Cloning and sequencing of representative PCR products, amplifying the 16S rDNA region were done. Phylogenetic analysis of the Pcc Jo-isolates revealed other than 90% similarity with different reference Pcc strains available at the GenBank. Different rot causal agents also were detected by PCR amplification and further sequences. The sequencing data revealed similarities to Pseudomonas fluorescence, Enterobacteriaceae genera such as Enterobacter spp., Serratia spp. and Klebsiella spp., in addition to Pectobacterium carotovorum subsp. carotovorum. This study indicated that using molecular techniques such as amplification of 16S rDNA region is a sensitive and specific method for detecting Pcc as potato soft rot causal agent. So far this is the first study where Pcc has been identified by using PCR and sequencing approaches in Jordan.展开更多
Background:The maturation of the immune system is critical during early life,as it involves the differentiation,maturation,and establishment of immune tolerance of immune cells.This process is influenced not only by g...Background:The maturation of the immune system is critical during early life,as it involves the differentiation,maturation,and establishment of immune tolerance of immune cells.This process is influenced not only by genetic factors but also by en-vironmental factors,particularly the symbiotic microbiota.Bifidobacterium animalis subsp.lactis(BB-12),originally found in dairy products,is widely used in infant for-mula and dietary supplements.However,its role and mechanisms in immune develop-ment during early life remain unclear.Methods:Using GF mice as the experimental model,B.animalis subsp.lactis BB-12 was administered via gavage during early life.In the juvenile stage,changes in T-cell subsets in the spleen,thymus,and gut intraepithelial lymphocytes(IEL)were assessed using spectral flow cytometry.Additionally,targeted metabolomics analysis of trypto-phan metabolism and short-chain fatty acid pathways in colonic tissue was conducted to explore how B.animalis subsp.lactis BB-12 influences the immune system through gut microbiota metabolism.Results:BB-12 effectively modulates the gut immune microenvironment,leading to beneficial changes in T-cell subsets in key immune tissues such as the spleen,thymus,and gut IELs.Metabolomics analysis further supports these findings by showing that BB-12 intervention greatly increased the production of tryptophan derivatives and acetic acid in the colon of GF mice.Conclusion:The findings provide theoretical evidence for the role of B.animalis subsp.lactis in immune system development and support its application in dietary supple-ments,suggesting potential as a component for infant immune health and in prevent-ing immune-related diseases.展开更多
Bifidobacterium longum subsp.infantis and Bifidobacterium adolescentis play important roles in the guts of infants and adolescents,respectively.In this study,using a neonatal rat model,we compared the protective effec...Bifidobacterium longum subsp.infantis and Bifidobacterium adolescentis play important roles in the guts of infants and adolescents,respectively.In this study,using a neonatal rat model,we compared the protective effects of these 2 bifidobacterial species against Salmonella infection.The results demonstrated that B.longum subsp.infantis was more effective than B.adolescentis in alleviating the severity of infection in newborn rats exposed to Salmonella enterica serovar Typhimurium strain SL1344.B.longum subsp.infantis attenuated intestinal inflammation and mucosal damage induced by Salmonella infection,as well as protecting intestinal nerves and intestinal barrier function through TLR4/My D88 signalling.B.longum subsp.infantis also displayed the potential to modulate gut metabolites by promoting the biosynthesis of unsaturated fatty acids(arachidonic acid,eicosapentaenoic acid andα-linolenic acid)and purine metabolism(guanine,adenine,inosine and adenosine),thereby regulating metabolic disturbances.Additionally,the benefits of B.longum subsp.infantis were also observed in the liver,spleen and brain,improving nerve reflexes and suppressing hepatosplenomegaly.Overall,these findings provide novel insights into the prevention and treatment of gutrelated diseases in newborns,highlighting the potentially significant role of B.longum subsp.infantis in clinical applications.展开更多

EPS produced from L. bulgaricus contributes to the physical and biological function of dairy products by regulating immune response, and this tendency seems to place EPS with acidic groups. To date, six types of chemical structure have been determined and are basically composed from glucose (Glc), galactose (Gal), and rhamnose (Rha). Eps clusters on chromosome DNA control the EPS synthesis and are transcribed as one mRNA 14 genes with 18kb on L. bulgaricus Lfi5. Furthermore, L. bulgaricus is able to utilize lactose (Lac) as carbohydrate source, repeating units of EPS are synthesized from Glc 6-phosphate, generated by an Embden-Meyerhof (EM) pathway in cellular carbohydrate assimilation. Bifidobacterium longum subsp.infantis is a commensal bacterium that predominates in the infant gut,playing a critical role in both preventing foreign infections and facilitating immune development.This study aimed to ...Bifidobacterium longum subsp.infantis is a commensal bacterium that predominates in the infant gut,playing a critical role in both preventing foreign infections and facilitating immune development.This study aimed to explore the effects of B.longum subsp.infantis supplementation on interferon-beta(IFN-β)secretion and intestinal barrier improvement in growing mice.Female and male mice were orally administered either saline or B.longum subsp.infantis CCFM1269 or I5TI(1×109 CFU/mice per day,n=8)from 1-week-age until 3-,4-,and 5-week-age.RNA sequencing analysis revealed that CCFM1269 exhibited potential antiviral capacity through increasing 2'-5'oligoadenylate synthetase(OAS).Additionally,CCFM1269 supplementation significantly increased colonic IFN-β levels which combined with OAS in 3-week-old female and male mice by activating the TLR4-TRIF-dependent signaling pathway.However,this effect was not observed in 4-and 5-week-old mice.Furthermore,both CCFM1269 were found to modulate the gut microbiota composition and enhance the intestinal barrier function in 3-,4-,and 5-week-old mice.In summary,the results of this study suggested that B.longum subsp.infantis CCFM1269 promoting intestinal barrier and releasing IFN-β in growing mice was in a strain-specific and time-dependent manner.展开更多

In this study, the fermentation properties of Streptococcus thermophilus, alone and in combination with Lactobacillus delbrueckii subsp. bulgaricus, were evaluated in skim milk that had been subjected to distinct sterilization conditions. Growth, organic acid generation, and EPS production were determined using pasteurized or autoclaved milk. When S. thermophilus was cultivated in pasteurized skim milk, the growth was strain-dependent. On the other hand, growth of S. thermophilus was accelerated in autoclaved milk. Exocellular polysaccharide (EPS) production by L. bulgaricus was not affected by the combination of S. thermophilus strains. Thus, we observed that yogurt fermented by L. bulgaricus was minimally affected by the combination of S. thermophilus strains;growth of L. bulgaricus was maintained under the constant environment. These results should facilitate the development of fermented milk produced from L. bulgaricus in the dairy industry.展开更多

To emphasize its clinical significance in both digestive and immune system, and to provide health remunerations to the consumer, the cheese microbiota will be investigated. To serve that purpose, the presence of the two potent probiotic Lactic Acid Bacteria (LAB): Lactococcus lactis subsp. lactis and Lactobacillus plantarum will be investigated. For bacterial identification, selection and isolation: culture-dependent techniques that imply the use of laboratory media will be implemented, and culture independent techniques: Polymerase Chain Reaction (PCR) will be applied for further validation. Both strains were immobilized by 4% (w/w) of pectin crude extract from Krung Kha Mao (KKM pectin) leaves, 1.52 mm inner diameters of silicone tube and 5% (v/v) inoculum for immobilization. Efficiency of lactic acid production was compared by immobilized cells ofL. casei subsp, rhamnosus TISTR 108 when KKM pectin, commercial citrus pectin, commercial apple pectin and sodium alginate were used as supporting materials to produce lactic acid 38.50, 38.76, 30.43 and 34.56 g/L, respectively, the productivity of lactic acid has been 0.40, 0.40, 0.36 and 0.36 g/L h, respectively.展开更多

Ohashi et J. Murata and A. ovale Nakai var. ovale are known to have one-leaved phenotype in both males and females;however, we discovered two-leaved individuals of these species. To elucidate the relationship between growth stage and leaf number of A. iyoanum subsp. nakaianum and A. ovale var. ovale, we conducted a morphological analysis of these plants. Our analysis suggested that the two-leaved individuals of A. iyoanum subsp. nakaianum and A. ovale var. ovale appeared only at the female phase. This suggested that one-leaved A. iyoanum subsp. nakaianum and A. ovale var. ovale individuals could not store the resources and hence changed to two-leaved individuals. This transformation could be explained by the fact that these species occur at high altitudes in mountain areas or regions covered in snow of the Japan Sea side, and their flowering time is also late compared with that in other sympatric Arisaema species.展开更多
The aim of this study was to evaluate the possible therapeutic or protective effects of Helichrysum plicatum DC.subsp.plicatum ethanol extract(HPE)against gentamicin-induced nephrotoxicity.Thirty-six Sprague Dawley ...The aim of this study was to evaluate the possible therapeutic or protective effects of Helichrysum plicatum DC.subsp.plicatum ethanol extract(HPE)against gentamicin-induced nephrotoxicity.Thirty-six Sprague Dawley male rats weighing between 200 and 250 g were used as live material.They were formed into six groups containing 6rats each and were allowed to adapt to laboratory conditions for 7 d.Group Ⅰ:control,5%DMSO intraperitoneal(i.p.);Group Ⅱ:HPE 100 mg/(kg·d)i.p.;Group Ⅲ:HPE 200 mg/(kg·d)i.p.;Group Ⅳ:gentamicin as 80 mg/(kg·d)i.p.;Group Ⅴ:gentamicin as 80 mg/(kg·d)i.p.+HPE 100 mg/(kg·d)i.p.;and Group Ⅵ:gentamicin as 80 mg/(kg·d)i.p.+HPE 200 mg/(kg·d)i.p.for 8 d.Following treatment,serum,liver,and kidney tissues were used to assess blood urea nitrogen(BUN),creatinine,enzymatic and non-enzymatic antioxidants,and lipid peroxidation.Gentamicin significantly increased serum BUN,creatinin,and liver and kidney levels of malondialdehyde(MDA).It also decreased the activity of catalase(CAT),glutathione peroxidase(GPx),and superoxide dismutase(SOD).Treatment with the HPE 100 mg/kg reversed gentamicin-induced alterations as evidenced by decreased serum BUN and creatinin,liver and kidney oxidant marker,and tubular necrosis as well as by an increase in antioxidant enzymes.It was found that HPE 200 mg/kg significantly increased liver and kidney tissue MDA levels in nephrotoxicity in rats.As a result,these findings support the proposition that HPE in 100 mg/kg dose demonstrates in the kidney and liver as free radicals and scavenger to prevent the toxic effects of gentamicin in both the biochemical and histopathology parameters.展开更多

In Xanthomonas citrisubsp. citri, carA and carB encode the small and large subunits of carbamoyl-phosphate synthase, respectively. The deletion mutation of the coding regions revealed that carA did not affect any of the phenotypes, while carB played multiple roles in pathogenicity. The deletion of carB rendered the loss of pathogenicity in host plants and the ability to induce a hyper- sensitive reaction in the non-hosts. Quantitative reverse transcription-PCR assays indicated that 11 hrp genes coding the type Ill secretion system were suppressed when interacting with citrus plants. The mutation in carB also affected bacterial utilization of several carbon and nitrogen resources in minimal medium MMX and extracellular enzyme activities. These data demonstrated that only the large subunit of carbamoyl-phosphate synthase was essential for canker development by X. citri subsp, citri.展开更多
Water stress and limited aeration imparted by solid-state fermentation (SSF) were reported as crucial factors for the enhancement of endospore production by Bacillus thuringiensis (Bt);and thus, more δ-endotoxin coul...Water stress and limited aeration imparted by solid-state fermentation (SSF) were reported as crucial factors for the enhancement of endospore production by Bacillus thuringiensis (Bt);and thus, more δ-endotoxin could be produced concomitantly with reduced time. Therefore, Bt subsp. kurstaki (Btk) was employed in the present study to evaluate its efficiency for the concomitant production of endospores and δ-endotoxin in LB medium supplemented with various naturally available agricultural products, i.e., flours of soybean, Bengal gram or jack seed at various concentrations (10%, 20%, 30%, 40%, 50%, 60%, 80% or 100%, all w/v). After 12 h fermentation, the supernatant in it was centrifuged off aseptically to obtain solid substrate for subsequent SSF. Of them, soybean (30%) supplemented medium was the best for the enhanced production of endospore and δ-crystals. The maximum yield of endospores during solid-state fermentation was observed 48 h, i.e., compared to submerged fermentation in LB, it was 24 h less gestation period. In control sample, the endospores achieved the maximum length (1.10 ± 0.13 μm) and diameter (0.63 ± 0.07 μm) at 72 h;while in soybean supplemented medium, the maximum length (2.10 ± 0.16 μm) and diameter (1.63 ± 0.16 μm) were at 48 h and 72 h, respectively. Upon staining, acridine orange specifically stained the endospores;malachite green-saffranin stained both δ-crystals and endospores;and coomassie brilliant blue specifically stained δ-endotoxin. Briefly, normal gestation period or harvest time for Btk is 72 h, which could be reduced to 48 h, if SSF is employed as demonstrated in this study.展开更多
Understanding how tree species respond to drought in their natural environment is needed to predict forest adaptation and management practices under global environmental changes.This study was carried out to determine...Understanding how tree species respond to drought in their natural environment is needed to predict forest adaptation and management practices under global environmental changes.This study was carried out to determine and compare physiological and biochemical responses to variations in environmental conditions during summer drought of mixed natural stands of Pinus nigra Arn.subsp.pallasiana(Lamb.)Holmboe and Pinus brutia Ten.Midday xylem water potential(ψmd),water relations,photosynthetic pigments,total soluble sugar and proline contents were investigated during the growing season.ψmd followed a similar seasonal trend in both species but P.nigra subsp.pallasiana had higher ψmd than P.brutia.The ψmd gradually decreased from June,reached its lowest value in August,and then increased again.Gradual decreases in the osmotic potential at turgor loss point(ψnTLP)were observed during the summer.Generally,ψnTLP was lower in P.brutia.Total soluble sugars decreased from April to June for P.brutia,then increased and stayed relatively constant August to October.Similar changes were found at lower values in P.nigra subsp.pallasiana.Prolin accumulation and photosynthetic pigments were higher in P.brutia.The results indicate that physiological and biochemical responses of both species against changing environmental conditions were in different degrees but followed similar trends.P.nigra subsp.pallasiana is more sensitive to summer drought than P.brutia in their natural environment.展开更多

摘要Brassinosteroids(BRs)are extensively distributed in plants and play crucial roles throughout all stages of plant growth.Nevertheless,the molecular mechanism through which BRs influence postharvest senescence in pakchoi remains elusive.Previous studies have demonstrated that the application of 1.5μM of the BRs analog 2,4-epibrassinolide(EBR)delayed the leaf senescence in harvested pakchoi.In this study,we constructed the EBR-delayed senescence transcriptome in pakchoi leaves and discovered that EBR modulates the expression of genes involved in the chlorophyll(Chl)metabolism pathway and the BRs pathway in pakchoi.Notably,we identified and characterized an EBR-suppressed,nucleus-localized WRKY transcription factor called BrWRKY8.BrWRKY8 is a highly expressed transcriptional activator in senescent leaves,targeting the promoters of the Chl degradation-associated gene BrSGR2 and the BRs degradation-associated gene BrCHI2,thereby promoting their expression.Overexpression of the BrWRKY8 gene accelerated the senescence process in Arabidopsis leaves,while EBR treatment mitigated the leaf senescence phenotype induced by BrWRKY8 overexpression.Conversely,silencing of BrWRKY8 through the virus-induced gene silencing extended the postharvest storage period of pakchoi.In conclusion,the newly discovered BRs-BrWRKY8 regulatory model in this study provides novel insights into BRs-mediated leaf senescence in pakchoi.

摘要Rodent-borne leptospirosis is by far the most common bacterial zoonosis and it is an important emerging global public health concern in Southeast Asia.Bacterial pathogens associated with rodents,especially those that live in close association with humans have been underreported.To fill this knowledge gap,the present study was undertaken to explore other neglected disease agents that can naturally infect synanthropic rodents.

摘要Microbial food is an important direction of sustainable food development in the future.Microorganisms such as lactic acid bacteria(LAB)are important sources of natural radioprotectors.Moderate environmental stress can induce stress response and improve biological activity of LAB.In the previous study,Lactococcus lactis subsp.lactis IL1403(L.lactis IL1403)cell-free extract induced by ionizing radiation(IR)of 500 Gy(IR-CFE)presented the stronger radioprotective effect than untreated cell-free extract in mice.To explore the radioprotective active substances of IR-CFE,the key protein was screened by proteomics and its radioprotective effect in vivo was further evaluated.The Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment showed that two-component systems(TCS)were enhanced to adapt the IR induction.Meanwhile,the protein LlrG belonged to TCS was screened by the protein-protein interaction.Furthermore,the recombinant protein LlrG(rLlrG)could markedly alleviate the 60Coγ-induced damage to the hematopoietic system,oxidative stress and inflammation in mice,thereby exerting its radioprotective function.These results suggested LlrG protein not only played an important role in the adaptation of L.lactis IL1403 to IR environment,but also exerted a good radioprotective effect,which could be applied in the development of protein-based radioprotectors.
摘要Various bacterial species are known to be agents causing soft rot of potatoes. The results of this study showed that potato soft rot is widely spread in different potato planting areas in Jordan. A survey was conducted through the years 2013-2015 to detect potato soft rot disease in Jordan, two hundred and four rotted potato samples were collected from different potato growing areas through different potato growing seasons. One hundred and thirty one bacterial isolates were isolated, purified on selective media and identified as Pectobacterium carotovorum subspecies carotovorum (Pcc) by different biochemical and physiological tests. Furthermore, 131 Pcc Jordanian (Jo) isolates were identified by PCR analysis of total DNA extracted from isolates that were biochemically identified as Pcc using universal primer Fd1/Rd1. Cloning and sequencing of representative PCR products, amplifying the 16S rDNA region were done. Phylogenetic analysis of the Pcc Jo-isolates revealed other than 90% similarity with different reference Pcc strains available at the GenBank. Different rot causal agents also were detected by PCR amplification and further sequences. The sequencing data revealed similarities to Pseudomonas fluorescence, Enterobacteriaceae genera such as Enterobacter spp., Serratia spp. and Klebsiella spp., in addition to Pectobacterium carotovorum subsp. carotovorum. This study indicated that using molecular techniques such as amplification of 16S rDNA region is a sensitive and specific method for detecting Pcc as potato soft rot causal agent. So far this is the first study where Pcc has been identified by using PCR and sequencing approaches in Jordan.

摘要Background:The maturation of the immune system is critical during early life,as it involves the differentiation,maturation,and establishment of immune tolerance of immune cells.This process is influenced not only by genetic factors but also by en-vironmental factors,particularly the symbiotic microbiota.Bifidobacterium animalis subsp.lactis(BB-12),originally found in dairy products,is widely used in infant for-mula and dietary supplements.However,its role and mechanisms in immune develop-ment during early life remain unclear.Methods:Using GF mice as the experimental model,B.animalis subsp.lactis BB-12 was administered via gavage during early life.In the juvenile stage,changes in T-cell subsets in the spleen,thymus,and gut intraepithelial lymphocytes(IEL)were assessed using spectral flow cytometry.Additionally,targeted metabolomics analysis of trypto-phan metabolism and short-chain fatty acid pathways in colonic tissue was conducted to explore how B.animalis subsp.lactis BB-12 influences the immune system through gut microbiota metabolism.Results:BB-12 effectively modulates the gut immune microenvironment,leading to beneficial changes in T-cell subsets in key immune tissues such as the spleen,thymus,and gut IELs.Metabolomics analysis further supports these findings by showing that BB-12 intervention greatly increased the production of tryptophan derivatives and acetic acid in the colon of GF mice.Conclusion:The findings provide theoretical evidence for the role of B.animalis subsp.lactis in immune system development and support its application in dietary supple-ments,suggesting potential as a component for infant immune health and in prevent-ing immune-related diseases.

摘要Bifidobacterium longum subsp.infantis and Bifidobacterium adolescentis play important roles in the guts of infants and adolescents,respectively.In this study,using a neonatal rat model,we compared the protective effects of these 2 bifidobacterial species against Salmonella infection.The results demonstrated that B.longum subsp.infantis was more effective than B.adolescentis in alleviating the severity of infection in newborn rats exposed to Salmonella enterica serovar Typhimurium strain SL1344.B.longum subsp.infantis attenuated intestinal inflammation and mucosal damage induced by Salmonella infection,as well as protecting intestinal nerves and intestinal barrier function through TLR4/My D88 signalling.B.longum subsp.infantis also displayed the potential to modulate gut metabolites by promoting the biosynthesis of unsaturated fatty acids(arachidonic acid,eicosapentaenoic acid andα-linolenic acid)and purine metabolism(guanine,adenine,inosine and adenosine),thereby regulating metabolic disturbances.Additionally,the benefits of B.longum subsp.infantis were also observed in the liver,spleen and brain,improving nerve reflexes and suppressing hepatosplenomegaly.Overall,these findings provide novel insights into the prevention and treatment of gutrelated diseases in newborns,highlighting the potentially significant role of B.longum subsp.infantis in clinical applications.
摘要A new species of Gentiana (Gentianaceae), Gentiana ciliolata is described and illustrated;it has a spectacular characteristics of calyx lobe ciliolate, distributed in Yunnan and Sichuan Provinces on the two sides of Jinsha River, and then two subspecies have been further re-classified as Gentiana ciliolata subsp. ciliolata and Gentiana ciliolata subsp. longiloba respectively. The subsp. ciliolata idistributes on the right side of Jinsha River in Yiliang County, Yunnan Province and the subsp. longiloba distributes itself on the southwest and west part of Sichuan Province, such as Leibo County, Wenchuan County and Mabian Yi Autonomous Prefacture, in the grassland under forests and moist rock slopes at an altitude of 1300 - 3200 m.
摘要Lactobacillus delbrueckii subsp. bulgaricus, which has been widely used as a fermented milk starter, is a type of probiotic, and certain strains are able to produce exopolysaccharide (EPS). EPS produced from L. bulgaricus contributes to the physical and biological function of dairy products by regulating immune response, and this tendency seems to place EPS with acidic groups. To date, six types of chemical structure have been determined and are basically composed from glucose (Glc), galactose (Gal), and rhamnose (Rha). Eps clusters on chromosome DNA control the EPS synthesis and are transcribed as one mRNA 14 genes with 18kb on L. bulgaricus Lfi5. Furthermore, L. bulgaricus is able to utilize lactose (Lac) as carbohydrate source, repeating units of EPS are synthesized from Glc 6-phosphate, generated by an Embden-Meyerhof (EM) pathway in cellular carbohydrate assimilation. This review discusses EPS produced from L. bulgaricus.

摘要Bifidobacterium longum subsp.infantis is a commensal bacterium that predominates in the infant gut,playing a critical role in both preventing foreign infections and facilitating immune development.This study aimed to explore the effects of B.longum subsp.infantis supplementation on interferon-beta(IFN-β)secretion and intestinal barrier improvement in growing mice.Female and male mice were orally administered either saline or B.longum subsp.infantis CCFM1269 or I5TI(1×109 CFU/mice per day,n=8)from 1-week-age until 3-,4-,and 5-week-age.RNA sequencing analysis revealed that CCFM1269 exhibited potential antiviral capacity through increasing 2'-5'oligoadenylate synthetase(OAS).Additionally,CCFM1269 supplementation significantly increased colonic IFN-β levels which combined with OAS in 3-week-old female and male mice by activating the TLR4-TRIF-dependent signaling pathway.However,this effect was not observed in 4-and 5-week-old mice.Furthermore,both CCFM1269 were found to modulate the gut microbiota composition and enhance the intestinal barrier function in 3-,4-,and 5-week-old mice.In summary,the results of this study suggested that B.longum subsp.infantis CCFM1269 promoting intestinal barrier and releasing IFN-β in growing mice was in a strain-specific and time-dependent manner.
摘要The sterilization conditions between experiment and dairy industrial level are different and concern about influence on product’s qualities. In this study, the fermentation properties of Streptococcus thermophilus, alone and in combination with Lactobacillus delbrueckii subsp. bulgaricus, were evaluated in skim milk that had been subjected to distinct sterilization conditions. Growth, organic acid generation, and EPS production were determined using pasteurized or autoclaved milk. When S. thermophilus was cultivated in pasteurized skim milk, the growth was strain-dependent. On the other hand, growth of S. thermophilus was accelerated in autoclaved milk. Exocellular polysaccharide (EPS) production by L. bulgaricus was not affected by the combination of S. thermophilus strains. Thus, we observed that yogurt fermented by L. bulgaricus was minimally affected by the combination of S. thermophilus strains;growth of L. bulgaricus was maintained under the constant environment. These results should facilitate the development of fermented milk produced from L. bulgaricus in the dairy industry.
摘要Raw goat milk cheeses are known for their natural microflora linked to many biodiversity factors such as the use of raw milk. That microflora serves probiotic attribution conferring a beneficial health impact on the consumer. Darfiyeh is an artisanal raw goat milk cheese manufactured traditionally in Northern Lebanese Mountains. To emphasize its clinical significance in both digestive and immune system, and to provide health remunerations to the consumer, the cheese microbiota will be investigated. To serve that purpose, the presence of the two potent probiotic Lactic Acid Bacteria (LAB): Lactococcus lactis subsp. lactis and Lactobacillus plantarum will be investigated. For bacterial identification, selection and isolation: culture-dependent techniques that imply the use of laboratory media will be implemented, and culture independent techniques: Polymerase Chain Reaction (PCR) will be applied for further validation. Both bacteria were further verified as Lactococcus lactis subsp. lactis and Lactobacillus plantarum by implementing specie-specific primers for the qualitative PCR.
摘要The research was first to perform the utilization of novel and cheap pectin crude extract from Krung Kha Mao leaves (Cissampelos pareira L.) to immobilize cells of Lactobacillus casei subsp, rhamnosus TISTR 108 and Lactobacillus delbrueckii subsp, bulgaricus TISTR 1339 for lactic acid production, the optimum condition of immobilized cells was produced significantly at 5% of probability, that the highest lactic acid has been 38.50 and 33.66 g/L in steady state of whey medium for 96 and 108 h, respectively. Both strains were immobilized by 4% (w/w) of pectin crude extract from Krung Kha Mao (KKM pectin) leaves, 1.52 mm inner diameters of silicone tube and 5% (v/v) inoculum for immobilization. Efficiency of lactic acid production was compared by immobilized cells ofL. casei subsp, rhamnosus TISTR 108 when KKM pectin, commercial citrus pectin, commercial apple pectin and sodium alginate were used as supporting materials to produce lactic acid 38.50, 38.76, 30.43 and 34.56 g/L, respectively, the productivity of lactic acid has been 0.40, 0.40, 0.36 and 0.36 g/L h, respectively.
摘要Arisaema iyoanum Makino subsp. nakaianum (Ohba) H. Ohashi et J. Murata and A. ovale Nakai var. ovale are known to have one-leaved phenotype in both males and females;however, we discovered two-leaved individuals of these species. To elucidate the relationship between growth stage and leaf number of A. iyoanum subsp. nakaianum and A. ovale var. ovale, we conducted a morphological analysis of these plants. Our analysis suggested that the two-leaved individuals of A. iyoanum subsp. nakaianum and A. ovale var. ovale appeared only at the female phase. This suggested that one-leaved A. iyoanum subsp. nakaianum and A. ovale var. ovale individuals could not store the resources and hence changed to two-leaved individuals. This transformation could be explained by the fact that these species occur at high altitudes in mountain areas or regions covered in snow of the Japan Sea side, and their flowering time is also late compared with that in other sympatric Arisaema species.

摘要The aim of this study was to evaluate the possible therapeutic or protective effects of Helichrysum plicatum DC.subsp.plicatum ethanol extract(HPE)against gentamicin-induced nephrotoxicity.Thirty-six Sprague Dawley male rats weighing between 200 and 250 g were used as live material.They were formed into six groups containing 6rats each and were allowed to adapt to laboratory conditions for 7 d.Group Ⅰ:control,5%DMSO intraperitoneal(i.p.);Group Ⅱ:HPE 100 mg/(kg·d)i.p.;Group Ⅲ:HPE 200 mg/(kg·d)i.p.;Group Ⅳ:gentamicin as 80 mg/(kg·d)i.p.;Group Ⅴ:gentamicin as 80 mg/(kg·d)i.p.+HPE 100 mg/(kg·d)i.p.;and Group Ⅵ:gentamicin as 80 mg/(kg·d)i.p.+HPE 200 mg/(kg·d)i.p.for 8 d.Following treatment,serum,liver,and kidney tissues were used to assess blood urea nitrogen(BUN),creatinine,enzymatic and non-enzymatic antioxidants,and lipid peroxidation.Gentamicin significantly increased serum BUN,creatinin,and liver and kidney levels of malondialdehyde(MDA).It also decreased the activity of catalase(CAT),glutathione peroxidase(GPx),and superoxide dismutase(SOD).Treatment with the HPE 100 mg/kg reversed gentamicin-induced alterations as evidenced by decreased serum BUN and creatinin,liver and kidney oxidant marker,and tubular necrosis as well as by an increase in antioxidant enzymes.It was found that HPE 200 mg/kg significantly increased liver and kidney tissue MDA levels in nephrotoxicity in rats.As a result,these findings support the proposition that HPE in 100 mg/kg dose demonstrates in the kidney and liver as free radicals and scavenger to prevent the toxic effects of gentamicin in both the biochemical and histopathology parameters.

摘要Carbamoyl-phosphate synthase plays a vital role in the carbon and nitrogen metabolism cycles. In Xanthomonas citrisubsp. citri, carA and carB encode the small and large subunits of carbamoyl-phosphate synthase, respectively. The deletion mutation of the coding regions revealed that carA did not affect any of the phenotypes, while carB played multiple roles in pathogenicity. The deletion of carB rendered the loss of pathogenicity in host plants and the ability to induce a hyper- sensitive reaction in the non-hosts. Quantitative reverse transcription-PCR assays indicated that 11 hrp genes coding the type Ill secretion system were suppressed when interacting with citrus plants. The mutation in carB also affected bacterial utilization of several carbon and nitrogen resources in minimal medium MMX and extracellular enzyme activities. These data demonstrated that only the large subunit of carbamoyl-phosphate synthase was essential for canker development by X. citri subsp, citri.
摘要Water stress and limited aeration imparted by solid-state fermentation (SSF) were reported as crucial factors for the enhancement of endospore production by Bacillus thuringiensis (Bt);and thus, more δ-endotoxin could be produced concomitantly with reduced time. Therefore, Bt subsp. kurstaki (Btk) was employed in the present study to evaluate its efficiency for the concomitant production of endospores and δ-endotoxin in LB medium supplemented with various naturally available agricultural products, i.e., flours of soybean, Bengal gram or jack seed at various concentrations (10%, 20%, 30%, 40%, 50%, 60%, 80% or 100%, all w/v). After 12 h fermentation, the supernatant in it was centrifuged off aseptically to obtain solid substrate for subsequent SSF. Of them, soybean (30%) supplemented medium was the best for the enhanced production of endospore and δ-crystals. The maximum yield of endospores during solid-state fermentation was observed 48 h, i.e., compared to submerged fermentation in LB, it was 24 h less gestation period. In control sample, the endospores achieved the maximum length (1.10 ± 0.13 μm) and diameter (0.63 ± 0.07 μm) at 72 h;while in soybean supplemented medium, the maximum length (2.10 ± 0.16 μm) and diameter (1.63 ± 0.16 μm) were at 48 h and 72 h, respectively. Upon staining, acridine orange specifically stained the endospores;malachite green-saffranin stained both δ-crystals and endospores;and coomassie brilliant blue specifically stained δ-endotoxin. Briefly, normal gestation period or harvest time for Btk is 72 h, which could be reduced to 48 h, if SSF is employed as demonstrated in this study.

摘要Understanding how tree species respond to drought in their natural environment is needed to predict forest adaptation and management practices under global environmental changes.This study was carried out to determine and compare physiological and biochemical responses to variations in environmental conditions during summer drought of mixed natural stands of Pinus nigra Arn.subsp.pallasiana(Lamb.)Holmboe and Pinus brutia Ten.Midday xylem water potential(ψmd),water relations,photosynthetic pigments,total soluble sugar and proline contents were investigated during the growing season.ψmd followed a similar seasonal trend in both species but P.nigra subsp.pallasiana had higher ψmd than P.brutia.The ψmd gradually decreased from June,reached its lowest value in August,and then increased again.Gradual decreases in the osmotic potential at turgor loss point(ψnTLP)were observed during the summer.Generally,ψnTLP was lower in P.brutia.Total soluble sugars decreased from April to June for P.brutia,then increased and stayed relatively constant August to October.Similar changes were found at lower values in P.nigra subsp.pallasiana.Prolin accumulation and photosynthetic pigments were higher in P.brutia.The results indicate that physiological and biochemical responses of both species against changing environmental conditions were in different degrees but followed similar trends.P.nigra subsp.pallasiana is more sensitive to summer drought than P.brutia in their natural environment.
